Dans cet article, nous présenterons le modèle biopsychosocial du trouble de la personnalité borderline et le mettrons en lien avec les critères développés dans les manuels diagnostiques (DSM-IV-TR, CIM-70). Seront ensuite explicité les principaux cadres de prise en charge de la thérapie comportementale-dialectique (TCD), tels que conçus par Marsha M. Linehan. Le modèle des dimensions de l'ouverture émotionnelle permettra d'enrichir la conceptualisation des émotions de la TCD. Nous insisterons particulièrement sur le groupe thérapeutique de Gestion des Émotions, inspiré des principes constitutifs de la TCD. Les pruniers résultats d'une étude pilote et la présentation d'une étude randomisée contrôlée seront discutés, en tenant compte des exigences cliniques et des considérations méthodologiques de l'évaluation des psychothérapies.

In 2008 we published the first set of guidelines for standardizing research in autophagy. Since then, research on this topic has continued to accelerate, and many new scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Accordingly, it is important to update these guidelines for monitoring autophagy in different organisms. Various reviews have described the range of assays that have been used for this purpose. Nevertheless, there continues to be confusion regarding acceptable methods to measure autophagy, especially in multicellular eukaryotes. A key point that needs to be emphasized is that there is a difference between measurements that monitor the numbers or volume of autophagic elements (e.g., autophagosomes or autolysosomes) at any stage of the autophagic process vs. those that measure flux through the autophagy pathway (i.e., the complete process); thus, a block in macroautophagy that results in autophagosome accumulation needs to be differentiated from stimuli that result in increased autophagic activity, defined as increased autophagy induction coupled with increased delivery to, and degradation within, lysosomes (in most higher eukaryotes and some protists such as Dictyostelium) or the vacuole (in plants and fungi). In other words, it is especially important that investigators new to the field understand that the appearance of more autophagosomes does not necessarily equate with more autophagy. In fact, in many cases, autophagosomes accumulate because of a block in trafficking to lysosomes without a concomitant change in autophagosome biogenesis, whereas an increase in autolysosomes may reflect a reduction in degradative activity. Here, we present a set of guidelines for the selection and interpretation of methods for use by investigators who aim to examine macroautophagy and related processes, as well as for reviewers who need to provide realistic and reasonable critiques of papers that are focused on these processes. These guidelines are not meant to be a formulaic set of rules, because the appropriate assays depend in part on the question being asked and the system being used. In addition, we emphasize that no individual assay is guaranteed to be the most appropriate one in every situation, and we strongly recommend the use of multiple assays to monitor autophagy. In these guidelines, we consider these various methods of assessing autophagy and what information can, or cannot, be obtained from them. Finally, by discussing the merits and limits of particular autophagy assays, we hope to encourage technical innovation in the field.

Hatching is an important niche shift, and embryos in a wide range of taxa can either accelerate or delay this life-history switch in order to avoid stage-specific risks. Such behavior can occur in response to stress itself and to chemical cues that allow anticipation of stress. We studied the genetic organization of this phenotypic plasticity and tested whether there are differences among populations and across environments in order to learn more about the evolutionary potential of stress-induced hatching. As a study species, we chose the brown trout (Salmo trutta; Salmonidae). Gametes were collected from five natural populations (within one river network) and used for full-factorial in vitro fertilizations. The resulting embryos were either directly infected with Pseudomonas fluorescens or were exposed to waterborne cues from P. fluorescens-infected conspecifics. We found that direct inoculation with P. fluorescens increased embryonic mortality and induced hatching in all host populations. Exposure to waterborne cues revealed population-specific responses. We found significant additive genetic variation for hatching time, and genetic variation in trait plasticity. In conclusion, hatching is induced in response to infection and can be affected by waterborne cues of infection, but populations and families differ in their reaction to the latter.

The aim of this research was to structure a conceptual model of hope and hopelessness based on dictionary definitions, and to verify this model on the basis of the experiences of the severely depressive and non-depressive elderly. This research has produced a substantive theory of hope and hopelessness which is based on the experiences of the depressive and non-depressive elderly, and on the concept analysis of hope and hopelessness based on English dictionary definitions. The patients who participated in the research were 65 years old and older men and women (n=22) who had been admitted to a psychiatric hospital because of major depression, and another group: the non-depressive elderly (n=21), who were recruited from the pensioners’ clubs. The data were collected in interviews using the Clinical Assessment Tool, developed by Farran, Salloway and Clark (1990) and Farran, Wilken and Popovich (1992), and it produced 553 pages of written text, which were analysed using the ATLAS/ti programme. ATLAS/ti is a tool for analysing qualitative data and is based on Grounded Theory. The medical and nursing records of the depressive elderly completed source triangulation. The concept analysis of hope and hopelessness was made on the basis of the definitions of English dictionaries (n=103), using semantic analysis and the ATLAS/ti programme. The most important hope-promoting factors were human relations, health and managing in everyday living. Autonomy, self-determination and feeling of security were highly appreciated among the elderly. Hopelessness, on the other hand, was most often associated with the same factors: human relations, health and everyday living. Especially, losses of significant others were experienced as strongly hope-diminishing. Old age had brought freedom from duties concerning others, but now, when you finally had an opportunity to enjoy yourself, you could not accomplish anything; you were clasped in the arms of total inability, depression had come. The most obvious difference in the life course of the depressive and nondepressive elderly was the abundance of traumatic experiences in the childhood and youth of the depressive elderly. The continuous circulation of fearful thoughts was almost touchable, and suicidality was described in connection with these thoughts. You were afraid to be awake and also to go to sleep. Managing day by day was the goal. The research produced the Basic Social Process (BSP) of hope: achieving - maintaining - losing, which expresses a continuous balancing between Being without and Being with. The importance of the object of hope was combined with the amount of hope and disappointment. The process of approaching defined the realisation of hope and the process of withdrawal that of losing. Joy and security versus grief and insecurity defined the Being with and Being without. Two core categories were found. The first one “If only I could�? reflects lack of energy, lack of knowledge, lack of courage and lack of ability. The other one “There is always a loophole�? reflects deliberate tracing of possibilities and the belief in finding solutions, and managing.

The aim of this study was to determine whether multiwalled carbon nanotubes (MWNCT) are taken up by and are toxic to human intestinal enterocytes using the Caco-2 cell model. Caco-2 cells were exposed to 50 ?g/ml MWCNT (oxidized or pristine) for 24 h, and experiments were repeated in the presence of 2.5 mg/L natural organic matter. Cells displayed many of the properties that characterize enterocytes, such as apical microvilli, basolateral basement membrane, and glycogen. The cell monolayers also displayed tight junctions and electrical resistance. Exposure to pristine and oxidized MWCNT, with or without natural organic matter, did not markedly affect viability, which was assessed by measuring activity of released lactate dehydrogenase (LDH) and staining with propidium iodide. Ultrastructural analysis revealed some damage to microvilli colocalized with the MWCNT; however, neither type of MWCNT was taken up by Caco-2 cells. In contrast, pristine and oxidized MWCNT were taken up by the macrophage RAW 264.7 line. Our study suggests that intestinal enterocytes cells do not take up MWCNT. [Authors]

Iowa’s land was mapped long before it was declared a state. Since Lewis and Clark published their journey across the North American west in 1814, many different uses for maps have been found. Today there are maps of Iowa’s roads, waterways, landscape features, geology, and land use. One of the more recent mapping efforts has involved using a technology called LiDAR. This technology creates a topographic map of Iowa’s elevation that is accurate to within eight inches, ten times higher resolution than in previous elevation maps.

Miniature diffusion size classifiers (miniDiSC) are novel handheld devices to measure ultrafine particles (UFP). UFP have been linked to the development of cardiovascular and pulmonary diseases; thus, detection and quantification of these particles are important for evaluating their potential health hazards. As part of the UFP exposure assessments of highwaymaintenance workers in western Switzerland, we compared a miniDiSC with a portable condensation particle counter (P-TRAK). In addition, we performed stationary measurements with a miniDiSC and a scanning mobility particle sizer (SMPS) at a site immediately adjacent to a highway. Measurements with miniDiSC and P-TRAK correlated well (correlation of r = 0.84) but average particle numbers of the miniDiSC were 30%âeuro"60% higher. This difference was significantly increased for mean particle diameters below 40 nm. The correlation between theminiDiSC and the SMPSduring stationary measurements was very high (r = 0.98) although particle numbers from the miniDiSC were 30% lower. Differences between the three devices were attributed to the different cutoff diameters for detection. Correction for this size dependent effect led to very similar results across all counters.We did not observe any significant influence of other particle characteristics. Our results suggest that the miniDiSC provides accurate particle number concentrations and geometric mean diameters at traffic-influenced sites, making it a useful tool for personal exposure assessment in such settings.

This publication of our 2008 Annual Report. This past year was beset with challenges and disappointments, but we were still able to forge ahead and register some significant accomplishments along the way. Our vision to organize our collective genius in a systematic way is beginning to take shape, and these efforts are reflected in this report. From various developments, like the making of a documentary and film of the Alexander Clark, Sr. story, to the partnership between the division and principal leadership in the faith community around the OCBI’s “Ten Point Plan”, one can envision the proverbial Black “phoenix” rising, renewed from the ashes of banal ideologies and practices. We will seize this opportunity, in conjunction with all of Iowa, to work together around the entire state, to produce a plan that reflects our collective interests, passions, and needs as we help create the Governor and Lt Governor’s vision of “One Iowa with One Unlimited Future.”

Animals and plants are associated with symbiotic microbes whose roles range from mutualism to commensalism to parasitism. These roles may not only be taxon-specific but also dependent on environmental conditions and host factors. To experimentally test these possibilities, we drew a random sample of adult whitefish from a natural population, bred them in vitro in a full-factorial design in order to separate additive genetic from maternal environmental effects on offspring, and tested the performance of the resulting embryos under different environmental conditions. Enhancing the growth of symbiotic microbes with supplemental nutrients released cryptic additive genetic variance for viability in the fish host. These effects vanished with the concurrent addition of the water mould Saprolegnia ferax. Our findings demonstrate that the heritability of host fitness is environment-specific and critically depends on the interaction between symbiotic microbes.

O objetivo deste trabalho foi avaliar os efeitos de micorriza arbuscular, do estado nutricional de P da planta e de concentrações crescentes de P em solução nutritiva na toxidez de Zn para Trema micrantha (L.) Blum. Em um primeiro experimento, mudas de trema foram formadas em substrato que continha doses crescentes de P [0, 100, 200 e 400 mg dm-3 na forma de Ca(H2PO4)2] e um tratamento de inoculação com Glomus etunicatum (Ge). Após crescimento por 60 dias, as mudas foram transferidas para vasos com solução nutritiva de Clark, que continha 2, 75, 150 e 225 µmol L-1 de Zn, e mantidas por mais 40 dias, quando foram colhidas e avaliadas. Os efeitos do P na amenização da fitotoxidez de Zn foram avaliados em outro experimento, aplicando-se, simultaneamente e de forma combinada em solução, doses de P (0,07; 0,5; 1 e 2 mmol L-1 fornecido por diferentes fontes) e de Zn (2, 75, 150 e 225 µmol L-1 na forma de ZnSO4.7H2O), nas quais foram cultivadas mudas de trema por 40 dias. Houve acentuada inibição no crescimento e na colonização micorrízica da trema em doses elevadas de Zn em solução (150 e 225 µmol L-1). Constatou-se que a melhoria da nutrição fosfática reduziu a translocação do Zn das raízes para a parte aérea, mas isto, assim como a colonização micorrízica, não resultou em favorecimento do crescimento da planta em condições de excesso deste metal em solução. No segundo experimento, verificou-se que a elevação na concentração de P em solução nutritiva promoveu melhoria no estado nutricional de P, conferindo proteção à planta do excesso de Zn. Como a especiação química da solução indicou que a aplicação de P não interferiu, de modo significativo, nas formas de Zn em solução, os resultados indicam que a ação amenizante do P ocorre na planta, possivelmente reduzindo a translocação do Zn das raízes para a parte aérea.
