Objective: Our research program has focused on the development of promising, soft alkylating N-phenyl-N’-(2-chloroethyl)urea (CEU) compounds which acylate the glutamic acid-198 of β-tubulin, near the binding site of colchicum alkaloids. CEUs inhibit the motility of cancerous cells in vitro and, interestingly, exhibit antiangiogenic and anticancer activity in vivo. Mitotic arrest induced by microtubule-interfering agents such as CEUs remains the major mechanism of their anticancer activity, leading to apoptosis. However, we recently demonstrated that microtubule disruption by CEUs and other common antimicrotubule agents greatly alters the integrity and organization of microtubule-associated structures, the focal adhesion contact, thereby initiating anoikis, an apoptosis-like cell death mechanism caused by the loss of cell contact with the extracellular matrix. Methods: To ascertain the activated signaling pathway profile of CEUs, flow cytometry, Western blot, immunohistochemistry and transfection experiments were performed. Wound-healing and chick embryo assays were carried out to evaluate the antiangiogenic potency of CEUs. Results: CEU-induced apoptosis involved early cell cycle arrest in G2/M and increased level of CDK1/cycline B proteins. These signaling events were followed by the specific activation of the intrinsic apoptosis pathway, involving loss of mitochondrial membrane potential (Δψm) and ROS production, cytochrome c release from mitochondria, caspase activation, AIF nuclear translocation, PARP cleavage and nuclear fragmentation. CEUs maintained their efficacy on cells plated on pro-survival extracellular matrices or exhibiting overexpression of P-glycoprotein or the anti-apoptotic protein Bcl-2. Conclusion: Our results suggest that CEUs represent a promising new class of antimicrotubule, antiangiogenic and pro-anoikis agents.

La division asymétrique est essentielle pour générer la diversité au cours du développement et permet aussi de réguler la balance entre renouvellement et différenciation des cellules souches chez l’adulte. Dans ces deux cas de figure, elle dépend respectivement d’une polarité intrinsèque ou d’une polarité extrinsèque. C. elegans est un excellent modèle pour étudier les mécanismes cellulaires et moléculaires de la division asymétrique in vivo. Chez l’embryon, le maintien d’un axe de polarité antéro-postérieur dépend des protéines PAR conservées et localisées de façon asymétrique en deux groupes mutuellement exclusifs; le groupe antérieur avec PAR-3, PAR-6, PKC-3 et le groupe postérieur avec PAR-2 et PAR-1. L’absence d’une protéine PAR entraine une perte de polarité et une létalité embryonnaire. Lors d’un crible par ARN interférence mené par Jean-Claude Labbé pour identifier les suppresseurs de la létalité associée à la perte de PAR-2, deux cyclines de type B, CYB-2.1 et CYB-2.2 ont été trouvées. J’ai déterminé que CYB-2.1 et CYB-2.2 interviennent dans la polarité sans perturber le cycle cellulaire et agissent vraisemblablement avec leur kinase associée, CDK-1, pour stabiliser les niveaux protéiques de PAR-6. Ces travaux permettent de mieux définir les liens étroits entre polarité et cycle cellulaire. La lignée germinale de C. elegans est un excellent modèle pour étudier les divisions des cellules souches germinales in vivo. Par contre, l’absence d’orientation préférentielle de ces divisions laisse envisager que la complexité morphologique de la niche pourrait engendrer une diversité d’axe possible. J’ai étudié la régulation morphologique de cette niche, une unique cellule somatique appelée distal tip cell (DTC), qui arborise de longues extensions au stade adulte. Mes résultats préliminaires favorisent un modèle dans lequel les cellules souches et progéniteurs germinaux (CSPG) supportent la formation de ces extensions. Enfin, j’ai obtenu des conditions favorables à l’étude de la division asymétrique extrinsèque dans ce modèle, en simplifiant l’architecture de la niche dans des conditions qui préservent les divisions cellulaires des cellules souches. Mes travaux ont permis de mieux comprendre les liens unissant les différents processus biologiques impliqués dans la division asymétrique, d’une part par l’étude du rôle qu’y jouent des régulateurs clés du cycle cellulaire au cours du développement et d’autre part par la caractérisation d’une communication bidirectionnelle entre la niche et les cellules souches chez l’adulte.

Les avancées techniques et méthodologiques de la neuroscience ont permis de caractériser le sommeil comme un état actif et dynamique où des événements neuronaux cohésifs organisent les fonctions cérébrales. Les fuseaux de sommeil et les ondes lentes sont les marqueurs électroencéphalographiques de ces événements, et la mesure de leurs paramètres reflète et nuance les interactions neuronales à l’oeuvre pendant le sommeil lent. Considérant leur implication dans les fonctions hypniques et cognitives, les événements du sommeil lent sont particulièrement pertinents à l’étude du vieillissement, où l’intégrité de ces fonctions est mise au défi. Le vieillissement normal s’accompagne non seulement de réductions importantes des paramètres composant les événements du sommeil lent, mais aussi de modifications précises de l’intégrité anatomique et fonctionnelle du cerveau. Récemment, les études ont souligné la régulation locale des événements du sommeil lent, dont l’évolution avec l’âge demeure toutefois peu explorée. Le présent ouvrage se propose de documenter les liens unissant la neurophysiologie du sommeil, le vieillissement normal et l’activité régionale du cerveau par l’évaluation topographique et hémodynamique des événements du sommeil lent au cours du vieillissement. Dans une première étude, la densité, la durée, l’amplitude et la fréquence des fuseaux de sommeil ont été évaluées chez trois groupes d’âge au moyen de l’analyse topographique et paramétrique de l’électroencéphalogramme. Dans une seconde étude, les variations hémodynamiques associées à l’occurrence et modulées par l’amplitude des ondes lentes ont été évaluées chez deux groupes d’âge au moyen de l’électroencéphalographie combinée à l’imagerie par résonance magnétique fonctionnelle. Globalement, les résultats obtenus ont indiqué : 1) une dichotomie des aires corticales antérieures et postérieures quant aux effets d’âge sur les paramètres des fuseaux de sommeil; 2) des variations de la réponse hémodynamique associées aux ondes lentes dans une diversité de régions corticales et sous-corticales chez les personnes âgées. Ces résultats suggèrent la réorganisation fonctionnelle de l’activité neuronale en sommeil lent à travers l’âge adulte, soulignent l’utilité et la sensibilité des événements du sommeil lent comme marqueurs de vieillissement cérébral, et encouragent la recherche sur l’évolution des mécanismes de plasticité synaptique, de récupération cellulaire et de consolidation du sommeil avec l’âge.

Un dérèglement du cycle cellulaire peut causer le cancer. Lors de la cytocinèse un anneau contractile d’actine et de myosine se forme, se contracte, et donne un anneau du midbody qui mène à l’abscision. Le processus de cytocinèse est sous le contrôle de protéines telles que la GTPase Rho qui active la cytocinèse et les cyclines-Cdks qui l'inhibent. La Drosophile possède 3 cyclines mitotiques CycA/ CycB/ CycB3 qui sont successivement dégradées en fin de mitose et permettent l'initiation de la cytocinèse. La dernière étape d’abscission est un phénomène qui reste encore peu connu. Les protéines Vps4 et CHMP4C liées à ANCHR vont, sous la dépendance de la kinase Aurora B, promouvoir l’abscision mais, suite à quelques études récentes, il semble y avoir une implication de la cycline B. Ici, le but était de tester l’implication de cette cycline dans les processus de cytocinèse et d’abscision, elle a été menée par microscopie à haute résolution en temps réel avec des cellules S2 de l’organisme Drosophila melanogaster par le suivi de protéines recombinantes fluorescentes. L’étude a été divisée en deux axes : gain et perte de fonction par l’intermédiaire respectivement de la protéine Cycline B recombinante stable, non dégradable (CycBstable-GFP) et l’inhibition par l’utilisation d’ARN double brin (ARNdb) sur l’endogène. La CycBstable-GFP a perturbé la cytocinèse en induisant plusieurs anneaux contractiles et midbodies. En revanche la réduction de l’expression de CycB n'a pas eu d’effet observable, et elle ne semble pas avoir d’action sur l’abscission malgré le recrutement de CycB-GFP au midbody tardif. En revanche la protéine Cdk1 semble avoir un rôle dans l'abscision puisque sa réduction d’expression a induit un délai. Elle a donc une implication potentielle sur la cytocinèse.

In this thesis, the applications of the recurrence quantification analysis in metal cutting operation in a lathe, with specific objective to detect tool wear and chatter, are presented.This study is based on the discovery that process dynamics in a lathe is low dimensional chaotic. It implies that the machine dynamics is controllable using principles of chaos theory. This understanding is to revolutionize the feature extraction methodologies used in condition monitoring systems as conventional linear methods or models are incapable of capturing the critical and strange behaviors associated with the metal cutting process.As sensor based approaches provide an automated and cost effective way to monitor and control, an efficient feature extraction methodology based on nonlinear time series analysis is much more demanding. The task here is more complex when the information has to be deduced solely from sensor signals since traditional methods do not address the issue of how to treat noise present in real-world processes and its non-stationarity. In an effort to get over these two issues to the maximum possible, this thesis adopts the recurrence quantification analysis methodology in the study since this feature extraction technique is found to be robust against noise and stationarity in the signals.The work consists of two different sets of experiments in a lathe; set-I and set-2. The experiment, set-I, study the influence of tool wear on the RQA variables whereas the set-2 is carried out to identify the sensitive RQA variables to machine tool chatter followed by its validation in actual cutting. To obtain the bounds of the spectrum of the significant RQA variable values, in set-i, a fresh tool and a worn tool are used for cutting. The first part of the set-2 experiments uses a stepped shaft in order to create chatter at a known location. And the second part uses a conical section having a uniform taper along the axis for creating chatter to onset at some distance from the smaller end by gradually increasing the depth of cut while keeping the spindle speed and feed rate constant.The study concludes by revealing the dependence of certain RQA variables; percent determinism, percent recurrence and entropy, to tool wear and chatter unambiguously. The performances of the results establish this methodology to be viable for detection of tool wear and chatter in metal cutting operation in a lathe. The key reason is that the dynamics of the system under study have been nonlinear and the recurrence quantification analysis can characterize them adequately.This work establishes that principles and practice of machining can be considerably benefited and advanced from using nonlinear dynamics and chaos theory.

The effect of lasers of three wavelengths in the visible region - 476, 488 and 514 nm on mitotic and meiotic cell divisions, growth, yield and activity of specific enzymes were studied in two taxonomically diverse plant species — A/lium cepa L. and Vicia faba. The effect of laser exposures was compared with the effect of two physical mutagens (Gamma and Ultraviolet radiations) and two chemical mutagens (Ethyl Methane Sulphonate and Hydroxyl amine). The study indicated that lasers could be mutagenic causing aberration in the mitotic and meiotic cell divisions while also producing changes in the growth and yield of the plants. Lasers of higher wavelengths 488 and 514 nm caused aberrations in the early stages of mitotic cell division whereas lasers of lower wavelengths (476 nm) caused more aberrations in the later stages of mitotic cell division. Laser exposure of 488 nm wavelength at power density 400 mW induced higher mitotic and meiotic aberrations and also induced higher pollen sterility than lasers of 476 and 514 nm. The frequency of mitotic aberrations induced by lasers was lesser than that caused by y-irradiation but comparable to that induced by EMS and HA. Lasers cause mutations in higher frequencies than UV. Lasers had a stimulatory effect on growth and yield in both plant species. This stimulatory effect of lasers on germination could not however be correlated to the activity of amylase and protease, the key enzymes in seed gennination. Enzymes such as peroxidase and catalase, involved in scavenging of free oxygen radicals often produced by irradiation, did not show increased activity in laser irradiated samples. Further studies are required for elucidating the exact mechanisms by which lasers cause mutations

The collection of X chromosome insertions (PX) lethal lines, which was isolated from a screen for essential genes on the X chromosome, was characterized by means of cloning the insertion sites, mapping the sites within genomic DNA and determination of the associated reporter gene expresssion patterns. The established STS flanking the P element insertion sites were submitted to EMBL nucleotide databases and their in situ data together with the enhancer trap expression patterns have been deposited in the FlyView database. The characterized lines are now available to be used by the scientific community for a detailed analysis of the newly established lethal gene functions. One of the isolated genes on the X chromosome was the Drosophila gene Wnt5 (DWnt5). From two independent screens, one lethal and three homozygous viable alleles were recovered, allowing the identification of two distinct functions for DWnt5 in the fly. Observations on the developing nervous system of mutant embryos suggest that DWnt5 activity affects axon projection pattern. Elevated levels of DWNT5 activity in the midline cells of the central nervous system causes improper establishment and maintenance of the axonal pathways. Our analysis of the expression and mutant phenotype indicates that DWnt5 function in a process needed for proper organization of the nervous system. A second and novel function of DWnt5 is the control of the body size by regulation of the cell number rather than affecting the size of cells. Moreover, experimentally increased DWnt5 levels in a post-mitotic region of the eye imaginal disc causes abnormal cell cycle progression, resulting in additional ommatidia in the adult eye when compared to wild type. The increased cell number and the effects on the cell cycle after exposure to high DWNT5 levels is the result of a failure to downregulate cyclin B and therefore the unsuccessful establishment of a G1 arrest.

Heterochromatin Protein 1 (HP1) is an evolutionarily conserved protein required for formation of a higher-order chromatin structures and epigenetic gene silencing. The objective of the present work was to functionally characterise HP1-like proteins in Dictyostelium discoideum, and to investigate their function in heterochromatin formation and transcriptional gene silencing. The Dictyostelium genome encodes three HP1-like proteins (hcpA, hcpB, hcpC), from which only two, hcpA and hcpB, but not hcpC were found to be expressed during vegetative growth and under developmental conditions. Therefore, hcpC, albeit no obvious pseudogene, was excluded from this study. Both HcpA and HcpB show the characteristic conserved domain structure of HP1 proteins, consisting of an N-terminal chromo domain and a C-terminal chromo shadow domain, which are separated by a hinge. Both proteins show all biochemical activities characteristic for HP1 proteins, such as homo- and heterodimerisation in vitro and in vivo, and DNA binding activtity. HcpA furthermore seems to bind to K9-methylated histone H3 in vitro. The proteins thus appear to be structurally and functionally conserved in Dictyostelium. The proteins display largely identical subnuclear distribution in several minor foci and concentration in one major cluster at the nuclear periphery. The localisation of this cluster adjacent to the nucleus-associated centrosome and its mitotic behaviour strongly suggest that it represents centromeric heterochromatin. Furthermore, it is characterised by histone H3 lysine-9 dimethylation (H3K9me2), which is another hallmark of Dictyostelium heterochromatin. Therefore, one important aspect of the work was to characterise the so-far largely unknown structural organisation of centromeric heterochromatin. The Dictyostelium homologue of inner centromere protein INCENP (DdINCENP), co-localized with both HcpA and H3K9me2 during metaphase, providing further evidence that H3K9me2 and HcpA/B localisation represent centromeric heterochromatin. Chromatin immunoprecipitation (ChIP) showed that two types of high-copy number retrotransposons (DIRS-1 and skipper), which form large irregular arrays at the chromosome ends, which are thought to contain the Dictyostelium centromeres, are characterised by H3K9me2. Neither overexpression of full-length HcpA or HcpB, nor deletion of single Hcp isoforms resulted in changes in retrotransposon transcript levels. However, overexpression of a C-terminally truncated HcpA protein, assumed to display a dominant negative effect, lead to an increase in skipper retrotransposon transcript levels. Furthermore, overexpression of this protein lead to severe growth defects in axenic suspension culture and reduced cell viability. In order to elucidate the proteins functions in centromeric heterochromatin formation, gene knock-outs for both hcpA and hcpB were generated. Both genes could be successfully targeted and disrupted by homologous recombination. Surprisingly, the degree of functional redundancy of the two isoforms was, although not unexpected, very high. Both single knock-out mutants did not show any obvious phenotypes under standard laboratory conditions and only deletion of hcpA resulted in subtle growth phenotypes when grown at low temperature. All attempts to generate a double null mutant failed. However, both endogenous genes could be disrupted in cells in which a rescue construct that ectopically expressed one of the isoforms either with N-terminal 6xHis- or GFP-tag had been introduced. The data imply that the presence of at least one Hcp isoform is essential in Dictyostelium. The lethality of the hcpA/hcpB double mutant thus greatly hampered functional analysis of the two genes. However, the experiment provided genetic evidence that the GFP-HcpA fusion protein, because of its ability to compensate the loss of the endogenous HcpA protein, was a functional protein. The proteins displayed quantitative differences in dimerisation behaviour, which are conferred by the slightly different hinge and chromo shadow domains at the C-termini. Dimerisation preferences in increasing order were HcpA-HcpA << HcpA-HcpB << HcpB-HcpB. Overexpression of GFP-HcpA or a chimeric protein containing the HcpA C-terminus (GFP-HcpBNAC), but not overexpression of GFP-HcpB or GFP-HcpANBC, lead to increased frequencies of anaphase bridges in late mitotic cells, which are thought to be caused by telomere-telomere fusions. Chromatin targeting of the two proteins is achieved by at least two distinct mechanisms. The N-terminal chromo domain and hinge of the proteins are required for targeting to centromeric heterochromatin, while the C-terminal portion encoding the CSD is required for targeting to several other chromatin regions at the nuclear periphery that are characterised by H3K9me2. Targeting to centromeric heterochromatin likely involves direct binding to DNA. The Dictyostelium genome encodes for all subunits of the origin recognition complex (ORC), which is a possible upstream component of HP1 targeting to chromatin. Overexpression of GFP-tagged OrcB, the Dictyostelium Orc2 homologue, showed a distinct nuclear localisation that partially overlapped with the HcpA distribution. Furthermore, GFP-OrcB localized to the centrosome during the entire cell cycle, indicating an involvement in centrosome function. DnmA is the sole DNA methyltransferase in Dictyostelium required for all DNA(cytosine-)methylation. To test for its in vivo activity, two different cell lines were established that ectopically expressed DnmA-myc or DnmA-GFP. It was assumed that overexpression of these proteins might cause an increase in the 5-methyl-cytosine(5-mC)-levels in the genomic DNA due to genomic hypermethylation. Although DnmA-GFP showed preferential localisation in the nucleus, no changes in the 5-mC-levels in the genomic DNA could be detected by capillary electrophoresis.

Bauchspeicheldrüsenkrebs ist die vierthäufigste Krebstodesursache in Deutschland. Durch die tiefe Lage des Organs im Körperinneren und das späte Auftreten von Symptomen erfolgt die Diagnose meist zu einem sehr späten Zeitpunkt, zu dem eine Resektion des Tumors in 80% der Fälle nicht mehr möglich ist. Die Hälfte der Patienten verstirbt bereits im ersten Jahr nach Diagnosestellung. Nach heutiger Erkenntnis entwickeln sich Adenokarzinome der Bauchspeicheldrüse über schrittweise histologische Veränderungen, die sogenannten PanIN Läsionen (pancreatic intraepithelial neoplasia). Bis heute fehlen jedoch klinisch einsetzbare Parameter für die Früherkennung des Karzinoms und seiner Vorstufen. Bisher ist nicht vollständig geklärt, welche molekularen Veränderungen hierbei eine wesentliche Rolle spielen. Das Ziel der vorliegenden Arbeit ist, die molekular- und zytogenetische Mutationsinzidenz und -Sequenz im Verlauf der neoplastischen Progression in der PanIN-Sequenz aufzuklären. Unter Anwendung der Fluoreszenz-in-situ-Hybridisierung (FISH) wird weiterführend die Frage beantwortet, ob sich der Nachweis von zytogenetischen Veränderungen in Zellen, die endoskopisch aus dem Pankreassekret gewonnen wurden, als neuartiger Ansatz für eine Frühdiagnostik nutzen lassen. Die molekulargenetischen Analysen zeigen, dass die PanIN-Läsionen aus Geweben mit chronischer Pankreatitis denen aus Geweben mit Karzinomen gleichzusetzen sind. Veränderungen in der Anzahl einzelner Chromosomen, sowie Basenmutationen finden sich bereits in den frühesten Gangläsionen. Die diffuse Verteilung von Genmutationen lässt einen mutagenen Feldeffekt vermuten, in welchem endogene (z.B. Pankreasenzyme, oxidativer Stress) und/oder exogene (z.B. Nikotin, Alkohol) Noxen auf verschiedene Pankreasgänge während einer langen Zeit einwirken. Auf der Basis der erhaltenen Daten kann angenommen werden, dass die prä-neoplastischen Läsionen in Geweben mit chronischer Pankreatitis eine Progression durchlaufen, in der sich sporadische Defekte wie Basenmutationen und Mitosefehler (Aneuplodien) akkumulieren. Die biologische Relevanz für die Tumorentstehung sollte jedoch immer im klinischen Kontext betrachtet werden. In Kombination mit weiteren Parametern (z.B. Alter, Dauer der Pankreatitis) könnte dies eine Möglichkeit bieten, das Risiko für die Entstehung eines Karzinoms individuell zu bestimmen und somit Patienten frühzeitig genug vor der Manifestation eines Tumors einer (Teil-)Resektion des Organs zuzuführen.

Después de los ataques del 11 de septiembre de 2001, Pakistán se ha convertido en uno de los principales aliados de Estados Unidos para combatir las fuerzas terroristas de Al-Qaeda y los talibán en el centro y sur de Asia. La alianza bilateral no ha manifestado resultados determinantes para aliviar los problemas de seguridad en Pakistán, por el contrario, la yihad islámica se ha fortalecido en su población y los ataques terroristas que atentan contra la población civil y el aparato estatal se vuelven cada vez mas frecuentes, en razón de lo anterior, los grupos insurgentes en Pakistán han fortalecido su capacidad operativa y expandido su escenario de influencia.

El trasplante de órganos y/o tejidos es considerado como una opción terapéutica viable para el tratamiento tanto de enfermedades crónicas o en estadios terminales, como de afectaciones no vitales, pero que generen una disminución en la calidad de vida percibida por el paciente. Este procedimiento, de carácter multidimensional, está compuesto por 3 actores principales: el donante, el órgano/tejido, y el receptor. Si bien un porcentaje significativo de investigaciones y planes de intervención han girado en torno a la dimensión biológica del trasplante, y a la promoción de la donación; el interés por la experiencia psicosocial y la calidad de vida de los receptores en este proceso ha aumentado durante la última década. En relación con esto, la presente monografía se plantea como objetivo general la exploración de la experiencia y los significados construidos por los pacientes trasplantados, a través de una revisión sistemática de la literatura sobre esta temática. Para ello, se plantearon unos objetivos específicos derivados del general, se seleccionaron términos o palabras claves por cada uno de estos, y se realizó una búsqueda en 5 bases de datos para revistas indexadas: Ebsco Host (Academic Search; y Psychology and Behavioral Sciences Collection); Proquest; Pubmed; y Science Direct. A partir de los resultados, se establece que si bien la vivencia de los receptores ha comenzado a ser investigada, aún es necesaria una mayor exploración sobre la experiencia de estos pacientes; exploración que carecería de objetivo si no se hiciera a través de las narrativas o testimonios de los mismos receptores

Introdução: Os sarcomas associados aos locais de inoculação têm vindo a ganhar destaque na clinica veterinária devido à sua frequência e dificuldade de tratamento. A obtenção de fatores de prognóstico é uma ferramenta essencial para determinar a terapêutica mais apropriada e auxiliar na determinação da sobrevida nos diferentes casos. Objetivo: Estudar uma população de 26 gatos diagnosticados com sarcomas associados aos locais de inoculação, que foram submetidos a tratamento cirúrgico. Material e Métodos: É um estudo retrospetivo de casos que ocorreram entre o ano 2005 e 2011, onde treze casos procedem do Hospital Veterinário do Porto, e treze casos correspondem a clínicas localizadas na zona de Lisboa, Amadora e Margem Sul. Foram avaliados fatores como idade, sexo, raça, localização, índice mitótico, presença de tecido necrosado, margens cirúrgicas, estadiamento pré-cirúrgico com tomografia computadorizada, tempo de recidiva local e sobrevida global. Resultados: Tendo em conta a nossa baixa amostra nenhum dos resultados foi totalmente conclusivo, foi obtida uma sobrevida global dos animais operados de 1120 dias. Discussão/Conclusão: Apesar dos resultados não serem conclusivos, o estudo reforça a importância da avaliação pré-cirúrgica para poder implementar uma terapêutica que garanta a maior sobrevida possível. É necessário um estudo mais controlado dos diferentes fatores e com uma amostra mais significativa para a obtenção de resultados mais conclusivos.

BACKGROUND: Mammalian cardiac myocytes withdraw from the cell cycle during post-natal development, resulting in a non-proliferating, fully differentiated adult phenotype that is unable to repair damage to the myocardium, such as occurs following a myocardial infarction. We and others previously have shown that forced expression of certain cell cycle molecules in adult cardiac myocytes can promote cell cycle progression and division in these cells. The mitotic serine/threonine kinase, Polo-like kinase-1 (Plk1), is known to phosphorylate and activate a number of mitotic targets, including Cdc2/Cyclin B1, and to promote cell division. PRINCIPAL FINDINGS: The mammalian Plk family are all differentially regulated during the development of rat cardiac myocytes, with Plk1 showing the most dramatic decrease in both mRNA, protein and activity in the adult. We determined the potential of Plk1 to induce cell cycle progression and division in cultured rat cardiac myocytes. A persistent and progressive loss of Plk1 expression was observed during myocyte development that correlated with the withdrawal of adult rat cardiac myocytes from the cell cycle. Interestingly, when Plk1 was over-expressed in cardiac myocytes by adenovirus infection, it was not able to promote cell cycle progression, as determined by cell number and percent binucleation. CONCLUSIONS: We conclude that, in contrast to Cdc2/Cyclin B1 over-expression, the forced expression of Plk1 in adult cardiac myocytes is not sufficient to induce cell division and myocardial repair.

AtTRB1, 2 and 3 are members of the SMH (single Myb histone) protein family, which comprises double-stranded DNA-binding proteins that are specific to higher plants. They are structurally conserved, containing a Myb domain at the N-terminus, a central H1/H5-like domain and a C-terminally located coiled-coil domain. AtTRB1, 2 and 3 interact through their Myb domain specifically with telomeric double-stranded DNA in vitro, while the central H1/H5-like domain interacts non-specifically with DNA sequences and mediates protein–protein interactions. Here we show that AtTRB1, 2 and 3 preferentially localize to the nucleus and nucleolus during interphase. Both the central H1/H5-like domain and the Myb domain from AtTRB1 can direct a GFP fusion protein to the nucleus and nucleolus. AtTRB1–GFP localization is cell cycle-regulated, as the level of nuclear-associated GFP diminishes during mitotic entry and GFP progressively re-associates with chromatin during anaphase/telophase. Using fluorescence recovery after photobleaching and fluorescence loss in photobleaching, we determined the dynamics of AtTRB1 interactions in vivo. The results reveal that AtTRB1 interaction with chromatin is regulated at two levels at least, one of which is coupled with cell-cycle progression, with the other involving rapid exchange.

Six nutrient formulations were studied for their efficacy in inducing mitosis in white lupin seedling cotyledon protoplasts of which the formulations of Schafer-Menuhr & Sturmer (AS) and Kao (K8p) were found to be superior over the other four when supplemented with 6-benzylaminopurine and alpha-naphthaleneacetic acid (alpha-NAA). An unltrafiltration treatment of K8p increased mitotic frequency by 130% when compared with the untreated control. Medium enrichment with 0.2% bovine serum albumin (BSA) brought about a dramatic 1341% rise in protoplast division in comparison with BSA-free medium but only when the enrichment was carried out in Kao and Michayluk (KM8p) background containing 2, 4-dichlorophenoxyacetic acid, alpha-NAA and zeatin. A higher number of protocolonies (each proliferating from single protoplast following multiple divisions) were seen in 0.4% BSA. With this breakthrough in white lupin protoplast research, it is now possible to reproducibly obtain protocolonies that was hitherto not possible.
