Smoking cue-provoked craving is an intricate behavior associated with strong changes in neural networks. Craving is one of the main reasons subjects continue to smoke; therefore interventions that can modify activity in neural networks associated with craving can be useful tools in future research investigating novel treatments for smoking cessation. The goal of this study was to use a neuromodulatory technique associated with a powerful effect on spontaneous neuronal firing - transcranial direct current stimulation (tDCS) - to modify cue-provoked smoking craving. Based on preliminary data showing that craving can be modified after a single tDCS session, here we investigated the effects of repeated tDCS sessions on craving behavior. Twenty-seven subjects were randomized to receive sham or active tDCS (anodal tDCS of the left DLPFC). Our results show a significant cumulative effect of tDCS on modifying smoking cue-provoked craving. In fact, in the group of active stimulation, smoking cues had an opposite effect on craving after stimulation - it decreased craving - as compared to sham stimulation in which there was a small decrease or increase on craving. In addition, during these 5 days of stimulation there was a small but significant decrease in the number of cigarettes smoked in the active as compared to sham tDCS group. Our findings extend the results of our previous study as they confirm the notion that tDCS has a specific effect on craving behavior and that the effects of several sessions can increase the magnitude of its effect. These results open avenues for the exploration of this method as a therapeutic alternative for smoking cessation and also as a mean to change stimulus-induced behavior. (C) 2009 Elsevier Ireland Ltd. (c) 2012 John Wiley & Sons A/S Background and Objective: Smokers are more predisposed than nonsmokers to infection with Porphyromonas gingivalis, one of the most important pathogens involved in the onset and development of periodontitis. It has also been observed that tobacco, and tobacco derivatives such as nicotine and cotinine, can induce modifications to P. gingivalis virulence. However, the effect of the major compounds derived from cigarettes on expression of protein by P.gingivalis is poorly understood. Therefore, this study aimed to evaluate and compare the effects of nicotine and cotinine on the P.gingivalis proteomic profile. Material and Methods: Total proteins of P gingivalis exposed to nicotine and cotinine were extracted and separated by two-dimensional electrophoresis. Proteins differentially expressed were successfully identified through liquid chromatography-mass spectrometry and primary sequence databases using MASCOT search engine, and gene ontology was carried out using DAVID tools. Results: Of the approximately 410 protein spots that were reproducibly detected on each gel, 23 were differentially expressed in at least one of the treatments. A particular increase was seen in proteins involved in metabolism, virulence and acquisition of peptides, protein synthesis and folding, transcription and oxidative stress. Few proteins showed significant decreases in expression; those that did are involved in cell envelope biosynthesis and proteolysis and also in metabolism. Conclusion: Our results characterized the changes in the proteome of P.gingivalis following exposure to nicotine and cotinine, suggesting that these substances may modulate, with minor changes, protein expression. The present study is, in part, a step toward understanding the potential smokepathogen interaction that may occur in smokers with periodontitis.

Background: The purpose of this study was to estimate the genetic influences on the initiation of cigarette smoking, the persistence, quantity and age-at-onset of regular cigarette use in Brazilian families. Methods: The data set consisted of 1,694 individuals enrolled in the Baependi Heart Study. The heritability and the heterogeneity in genetic and environmental variance components by gender were estimated from variance components approaches, using the SOLAR (Sequential Oligogenic Linkage Analysis Routines) computer package. The mixed-effects Cox model was used for the genetic analysis of the age-at onset of regular cigarette use. Results: The heritability estimates were high (> 50%) for smoking initiation and were intermediate, ranging from 23.4 to 31.9%, for smoking persistence and quantity. Significant evidence for heterogeneity in variance components by gender was observed for smoking initiation and age-at-onset of regular cigarette use. Genetic factors play an important role in the interindividual variation of these phenotypes in females, while in males there is a predominant environmental component, which could be explained by greater social influences in the initiation of tobacco use. Conclusions: Significant heritabilities were observed in smoking phenotypes for both males and females from the Brazilian population. These data add to the literature and are concordant with the notion of significant biological determination in smoking behavior. Samples from the Baependi Heart Study may be valuable for the mapping of genetic loci that modulate this complex biological trait.

Background Tax policy is considered the most effective strategy to reduce tobacco consumption and prevalence. Tax avoidance and tax evasion therefore undermine the effectiveness of tax policies and result in less revenue for governments, cheaper prices for smokers and increased tobacco use. Tobacco smuggling and illicit tobacco trade have probably always existed, since tobacco's introduction as a valuable product from the New World, but the nature of the trade has changed. Methods This article clarifies definitions, reviews the key issues related to illicit trade, describes the different ways taxes are circumvented and looks at the size of the problem, its changing nature and its causes. The difficulties of data collection and research are discussed. Finally, we look at the policy options to combat illicit trade and the negotiations for a WHO Framework Convention on Tobacco Control (FCTC) protocol on illicit tobacco trade. Results Twenty years ago the main type of illicit trade was large-scale cigarette smuggling of well known cigarette brands. A change occurred as some major international tobacco companies in Europe and the Americas reviewed their export practices due to tax regulations, investigations and lawsuits by the authorities. Other types of illicit trade emerged such as illegal manufacturing, including counterfeiting and the emergence of new cigarette brands, produced in a rather open manner at well known locations, which are only or mainly intended for the illegal market of another country. Conclusions The global scope and multifaceted nature of the illicit tobacco trade requires a coordinated international response, so a strong protocol to the FCTC is essential. The illicit tobacco trade is a global problem which needs a global solution.

PURPOSE: Investigate the morphological effects of chronic exposure to tobacco smoke inhalation and alcohol consumption on the lungs and on the growth of rats. METHODS: Sixty male Wistar rats were divided into four groups: control, tobacco, alcohol, tobacco + alcohol, for a period of study 260 days. Morphological analysis was conducted by optical and electron microscopy. Rat growth was investigated by measuring the snout-anus length, body mass index and body weight. RESULTS: The three groups exposed to the drugs presented lower growth and lower weight than the control group. The percentages of alveolitis, bronchiolitis and the mean alveolar diameter were greater, particularly in the groups exposed to tobacco smoke, but were not significantly different from the control group. Electron microscopy revealed more intense apoptotic and degenerative lesions in the smoking group, while degenerative lesions in the lamellar bodies were more intense with the association of both drugs. CONCLUSIONS: This experimental model showed morphological alterations observed by electron microscopy, principally due to tobacco smoke exposure. Alcohol and tobacco hindered the growth of rats, such that tobacco showed a greater effect on body length and alcohol on body weight.

Die Gesundheitseffekte von Aerosolpartikeln werden stark von ihren chemischen und physikalischen Eigenschaften und somit den jeweiligen Bildungsprozessen und Quellencharakteristika beeinflusst. Während die Hauptquellen der anthropogenen Partikelemissionen gut untersucht sind, stellen die spezifischen Emissionsmuster zahlreicher kleiner Aerosolquellen, welche lokal und temporär zu einer signifikanten Verschlechterung der Luftqualität beitragen können, ein Forschungsdesiderat dar.rnIn der vorliegenden Arbeit werden in kombinierten Labor- und Feldmessungen durch ein integratives Analysekonzept mittels online (HR-ToF-AMS ) und filterbasierter offline (ATR-FTIR-Spektroskopie ) Messverfahren die weitgehend unbekannten physikalischen und chemischen Eigenschaften der Emissionen besonderer anthropogener Aerosolquellen untersucht. Neben einem Fußballstadion als komplexe Mischung verschiedener Aerosolquellen wie Frittieren und Grillen, Zigarettenrauchen und Pyrotechnik werden die Emissionen durch Feuerwerkskörper, landwirtschaftliche Intensivtierhaltung (Legehennen), Tief- und Straßenbauarbeiten sowie abwasserbürtige Aerosolpartikel in die Studie mit eingebunden. Die primären Partikelemissionen der untersuchten Quellen sind vorrangig durch kleine Partikelgrößen (dp < 1 µm) und somit eine hohe Lungengängigkeit gekennzeichnet. Dagegen zeigen die Aerosolpartikel im Stall der landwirtschaftlichen Intensivtierhaltung sowie die Emissionen durch die Tiefbauarbeiten einen hohen Masseanteil von Partikeln dp > 1 µm. Der Fokus der Untersuchung liegt auf der chemischen Charakterisierung der organischen Partikelbestandteile, welche für viele Quellen die NR-PM1-Emissionen dominieren. Dabei zeigen sich wichtige quellenspezifische Unterschiede in der Zusammensetzung der organischen Aerosolfraktion. Die beim Abbrand von pyrotechnischen Gegenständen freigesetzten sowie die abwasserbürtigen Aerosolpartikel enthalten dagegen hohe relative Gehalte anorganischer Substanzen. Auch können in einigen spezifischen Emissionen Metallverbindungen in den AMS-Massenspektren nachgewiesen werden. Über die Charakterisierung der Emissionsmuster und -dynamiken hinaus werden für einige verschiedenfarbige Rauchpatronen sowie die Emissionen im Stall der Intensivtierhaltung Emissionsfaktoren bestimmt, die zur quantitativen Bilanzierung herangezogen werden können. In einem weiteren Schritt werden anhand der empirischen Daten die analytischen Limitierungen der Aerosolmassenspektrometrie wie die Interferenz organischer Fragmentionen durch (Hydrogen-)Carbonate und mögliche Auswertestrategien zur Überwindung dieser Grenzen vorgestellt und diskutiert.rnEine umfangreiche Methodenentwicklung zur Verbesserung der analytischen Aussagekraft von organischen AMS-Massenspektren zeigt, dass für bestimmte Partikeltypen einzelne Fragmentionen in den AMS-Massenspektren signifikant mit ausgewählten funktionellen Molekülgruppen der FTIR-Absorptionsspektren korrelieren. Bedingt durch ihre fehlende Spezifität ist eine allgemeingültige Interpretation von AMS-Fragmentionen als Marker für verschiedene funktionelle Gruppen nicht zulässig und häufig nur durch die Ergebnisse der komplementären FTIR-Spektroskopie möglich. Des Weiteren wurde die Verdampfung und Ionisation ausgewählter Metallverbindungen im AMS analysiert. Die Arbeit verdeutlicht, dass eine qualitative und quantitative Auswertung dieser Substanzen nicht ohne Weiteres möglich ist. Die Gründe hierfür liegen in einer fehlenden Reproduzierbarkeit des Verdampfungs- und Ionisationsprozesses aufgrund von Matrixeffekten sowie der in Abhängigkeit vorangegangener Analysen (Verdampferhistorie) in der Ionisationskammer und auf dem Verdampfer statt-findenden chemischen Reaktionen.rnDie Erkenntnisse der Arbeit erlauben eine Priorisierung der untersuchten anthropogenen Quellen nach bestimmten Messparametern und stellen für deren Partikelemissionen den Ausgangpunkt einer Risikobewertung von atmosphärischen Folgeprozessen sowie potentiell negativen Auswirkungen auf die menschliche Gesundheit dar. rn

Smoke spikes occurring during transient engine operation have detrimental health effects and increase fuel consumption by requiring more frequent regeneration of the diesel particulate filter. This paper proposes a decision tree approach to real-time detection of smoke spikes for control and on-board diagnostics purposes. A contemporary, electronically controlled heavy-duty diesel engine was used to investigate the deficiencies of smoke control based on the fuel-to-oxygen-ratio limit. With the aid of transient and steady state data analysis and empirical as well as dimensional modeling, it was shown that the fuel-to-oxygen ratio was not estimated correctly during the turbocharger lag period. This inaccuracy was attributed to the large manifold pressure ratios and low exhaust gas recirculation flows recorded during the turbocharger lag period, which meant that engine control module correlations for the exhaust gas recirculation flow and the volumetric efficiency had to be extrapolated. The engine control module correlations were based on steady state data and it was shown that, unless the turbocharger efficiency is artificially reduced, the large manifold pressure ratios observed during the turbocharger lag period cannot be achieved at steady state. Additionally, the cylinder-to-cylinder variation during this period were shown to be sufficiently significant to make the average fuel-to-oxygen ratio a poor predictor of the transient smoke emissions. The steady state data also showed higher smoke emissions with higher exhaust gas recirculation fractions at constant fuel-to-oxygen-ratio levels. This suggests that, even if the fuel-to-oxygen ratios were to be estimated accurately for each cylinder, they would still be ineffective as smoke limiters. A decision tree trained on snap throttle data and pruned with engineering knowledge was able to use the inaccurate engine control module estimates of the fuel-to-oxygen ratio together with information on the engine control module estimate of the exhaust gas recirculation fraction, the engine speed, and the manifold pressure ratio to predict 94% of all spikes occurring over the Federal Test Procedure cycle. The advantages of this non-parametric approach over other commonly used parametric empirical methods such as regression were described. An application of accurate smoke spike detection in which the injection pressure is increased at points with a high opacity to reduce the cumulative particulate matter emissions substantially with a minimum increase in the cumulative nitrogrn oxide emissions was illustrated with dimensional and empirical modeling.

The conclusions were based principally on the field relationships and lithology because no fossils were found.

In this article, we analyze political parties' campaign communication during the 2009 European Parliamentary election in 11 countries (Austria, Bulgaria, Czech Republic, Germany, Hungary, The Netherlands, Poland, Portugal, Spain, Sweden, and the UK). We study which types of issues Euroskeptic fringe and Euroskeptic mainstream parties put on their campaign agendas and the kind and extent of EU opposition they voice. Further, we seek to understand whether Euroskeptic and non-Euroskeptic parties co-orient themselves toward each other within their national party systems with regard to their campaigns. To understand the role of Euroskeptic parties in the 2009 European Parliamentary elections, we draw on a systematic content analysis of parties' posters and televised campaign spots. Our results show that it is Euroskeptic parties at the edges of the political spectrum who discuss polity questions of EU integration and who most openly criticize the union. Principled opposition against the project of EU integration, however, can only be observed in the UK. Finally, we find indicators for co-orientation effects regarding the tone of EU mobilization: In national political environments where Euroskeptic parties strongly criticize the EU, pro-European parties at the same time publicly advance pro-EU positions.
