In this study, we report the effect of fatty acids on the Thy-1 antigen mRNA decay. Low serum and synthetic medium culture conditions were used to demonstrate that fatty acids, which are important metabolites involved as second messengers in signal transduction, also influence the steady-state mRNA level. Detailed analysis demonstrated that polyunsaturated lipids attached to bovine serum albumin, such as linoleic, linolenic, and arachidonic acids, modulate gene expression specifically in the S1A T lymphoma cell line by inducing a 3-5-fold increase in the steady-state Thy-1 mRNA level, concomitant with a twofold increase in cell surface expression. A similar modulation was observed in the immature CD4-CD8- T cell precursors but not in mature thymocytes. Nuclear run-on and transfection experiments indicated that the observed Thy-1 mRNA level is post-transcriptionally regulated and that the presence of the coding region is sufficient for this adaptive response. A mechanism without a requirement for protein kinase C activation, but involving Ca2+ entry, could account for this difference in Thy-1 mRNA stability.

Purpose: To evaluate whether parametric imaging with contrast material-enhanced ultrasonography (US) is superior to visual assessment for the differential diagnosis of focal liver lesions (FLLs). Materials and Methods: This study had institutional review board approval, and verbal patient informed consent was obtained. Between August 2005 and October 2008, 146 FLLs in 145 patients (63 women, 82 men; mean age, 62.5 years; age range, 22-89 years) were imaged with real-time low-mechanical-index contrast-enhanced US after a bolus injection of 2.4 mL of a second-generation contrast agent. Clips showing contrast agent uptake kinetics (including arterial, portal, and late phases) were recorded and subsequently analyzed off-line with dedicated image processing software. Analysis of the dynamic vascular patterns (DVPs) of lesions with respect to adjacent parenchyma allowed mapping DVP signatures on a single parametric image. Cine loops of contrast-enhanced US and results from parametric imaging of DVP were assessed separately by three independent off-site readers who classified each lesion as benign, malignant, or indeterminate. Sensitivity, specificity, accuracy, and positive and negative predictive values were calculated for both techniques. Interobserver agreement (κ statistics) was determined. Results: Sensitivities for visual interpretation of cine loops for the three readers were 85.0%, 77.9%, and 87.6%, which improved significantly to 96.5%, 97.3%, and 96.5% for parametric imaging, respectively (P < .05, McNemar test), while retaining high specificity (90.9% for all three readers). Accuracy scores of parametric imaging were higher than those of conventional contrast-enhanced US for all three readers (P < .001, McNemar test). Interobserver agreement increased with DVP parametric imaging compared with conventional contrast-enhanced US (change of κ from 0.54 to 0.99). Conclusion: Parametric imaging of DVP improves diagnostic performance of contrast-enhanced US in the differentiation between malignant and benign FLLs; it also provides excellent interobserver agreement.

Cet article se propose de prendre pour objet deux assemblées pentecôtistes de l'Est de la France - l'Église Évangélique de Pentecôte de Besançon (Franche-Comté) et la Porte Ouverte Chrétienne de Mulhouse (Alsace) - qui ont fait l'objet d'investigations ethnographiques dans le cadre de notre thèse de doctorat. Après avoir situé ces deux assemblées dans la mouvance du pentecôtisme français, nous nous sommes concentré sur la figure du pasteur pentecôtiste. L'exemplarité de l'histoire de vie des pasteurs, le rôle centrale que jouent ces derniers dans la « mise en présence » du Saint Esprit durant le culte et l'inspiration divine dont sont supposés être imprégnés leurs enseignements bibliques légitiment ce personnage dans sa fonction d'intermédiaire incontournable dans la relation entre le fidèle et son Dieu, alors même que le pentecôtisme s'inscrit historiquement dans la mouvance évangélique qui valorise la relation personnelle du chrétien né de nouveau avec son créateur. This paper considers two Pentecostal assemblies in Eastern France - the Église Évangélique de Pentecôte of Besançon (Franche-Comté) and the Porte Ouverte Chrétienne of Mulhouse (Alsace) - I have investigated for my PhD research, following an ethnographical method. After located these two assemblies in the sphere of influence of the French Pentecostalism, we focused on the Pentecostal pastor figure. The exemplary nature of the story of life of the pastors, the central role that pastors play in encounters with divine during the cult and the divine inspiration of their biblical educations legitimize this figure like inescapable intermediary between the believer and his God. Even if Pentecostalism is historically part of the evangelical sphere of influence which values personal relationships of the born again Christian with his creator.

Supernatants from cell cultures (also called conditioned media, CMs) are commonly analyzed to study the pool of secreted proteins (secretome). To reduce the exogenous protein background, serum-free media are often used to obtain CMs. Serum deprivation, however, can severely affect cell viability and phenotype, including protein secretion. We present a strategy to analyze the proteins secreted by cells in fetal bovine serum-containing CMs, which combines the advantage of metabolic labeling and protein concentration linearization techniques. Incubation of CMs with a hexapeptide ligand library was used to reduce the dynamic range of the samples and led to the identification of 3 times more proteins than in untreated CM samples. Labeling with a deuterated amino acid was used to distinguish between cellular proteins and homologous bovine proteins contained in the medium. Application of the strategy to two breast cancer cell lines led to the identification of proteins secreted in different amounts and which could correlate with their varying degree of aggressiveness. Selected reaction monitoring (SRM)-based quantitation of three proteins of interest in the crude samples yielded data in good agreement with the results from concentration-equalized samples.
