Tropical cyclogenesis is generally considered to occur in regions devoid of baroclinic structures; however, an appreciable number of tropical cyclones (TCs) form in baroclinic environments each year. A global climatology of these baroclinically influenced TC developments is presented in this study. An objective classification strategy is developed that focuses on the characteristics of the environmental state rather than on properties of the vortex, thus allowing for a pointwise “development pathway” classification of reanalysis data. The resulting climatology shows that variability within basins arises primarily as a result of local surface thermal contrasts and the positions of time-mean features on the subtropical tropopause. The pathway analyses are sampled to generate a global climatology of 1948–2010 TC developments classified by baroclinic influence: nonbaroclinic (70%), low-level baroclinic (9%), trough induced (5%), weak tropical transition (11%), and strong tropical transition (5%). All basins other than the North Atlantic are dominated by nonbaroclinic events; however, there is extensive interbasin variability in secondary development pathways. Within each basin, subregions and time periods are identified in which the relative importance of the development pathways also differs. The efficiency of tropical cyclogenesis is found to be highly dependent on development pathway. The peak efficiency defined in the classification subspace straddles the nonbaroclinic/trough-induced boundary, suggesting that the optimal environment for TC development includes a baroclinic contribution from an upper-level disturbance. By assessing the global distribution of baroclinically influenced TC formations, this study identifies regions and pathways whose further study could yield improvements in our understanding of this important subset of TC developments.

In this article we argue that it is pressing to study the “hybrid media system” at the intersection of online and offl ine communication and its potential for agenda building. The topic is relevant because it is argued that the internet off ers new opportunities of public infl uence for challengers without access to political decision making. Except for single case studies, little is known about the conditions under which these actors succeed. Informed by the research on agenda building we tackle with the mechanisms of online-offline media agenda building and the conditions under which challengers succeed to produce issue spill-over into conventional mass media. We develop a theoretical framework for investigating the linkage between online communication and traditional mass media and discuss how our model translates into empirical research. We conclude that the nature of online networks is critical for spill-over, but also the issue itself and the structure of the political system.

Die traverse lancierte im Jahr 2010 eine Reihe von vier historiographischen Sonderheften. Nach der Wirtschaftsgeschichte (1/2010), der Sozialgeschichte (1/2011) und der Kulturgeschichte (1/2012) wird die Reihe mit einem Heft zur Politikgeschichte (1/2013) abgeschlossen. In 14 Artikeln werden Themen aus dem Mittelalter, der Neuzeit, der Sattelzeit und der Zeitgeschichte aufgenommen. Die Beiträge bieten Forschungsüberblicke zur Geschichte der Parteien und der politischen Bewegungen, der Militärgeschichte, der Rechtsgeschichte, der staatsbürgerlichen Rechte und der Staatsbürgerschaft, den auswärtigen Beziehungen, der Neutralität, der schweizerischen Mitarbeit in internationalen Organisationen, des Service Publics, des Parlamentes und der Verfassung, der Religionsgeschichte und des Säkularismus, der Umwelt und zur Konstruktion von Geschichte und nationaler Indentität. Zusätzlich werden die beiden Arbeitsinstrumente Diplomatische Dokumente der Schweiz und das Jahrbuch Schweizerische Politik (L'Année politique suisse) vorgestellt und am Beispiel des Bergier-Berichts wird die politische Instrumentalisierung der Geschichte diskutiert.

SSE1 and SSE2 encode the essential yeast members of the Hsp70-related Hsp110 molecular chaperone family. Both mammalian Hsp110 and the Sse proteins functionally interact with cognate cytosolic Hsp70s as nucleotide exchange factors. We demonstrate here that Sse1 forms high-affinity (Kd approximately 10-8 M) heterodimeric complexes with both yeast Ssa and mammalian Hsp70 chaperones and that binding of ATP to Sse1 is required for binding to Hsp70s. Sse1.Hsp70 heterodimerization confers resistance to exogenously added protease, indicative of conformational changes in Sse1 resulting in a more compact structure. The nucleotide binding domains of both Sse1/2 and the Hsp70s dictate interaction specificity and are sufficient for mediating heterodimerization with no discernible contribution from the peptide binding domains. In support of a strongly conserved functional interaction between Hsp110 and Hsp70, Sse1 is shown to associate with and promote nucleotide exchange on human Hsp70. Nucleotide exchange activity by Sse1 is physiologically significant, as deletion of both SSE1 and the Ssa ATPase stimulatory protein YDJ1 is synthetically lethal. The Hsp110 family must therefore be considered an essential component of Hsp70 chaperone biology in the eukaryotic cell.

BACKGROUND: Quantitative myocardial PET perfusion imaging requires partial volume corrections. METHODS: Patients underwent ECG-gated, rest-dipyridamole, myocardial perfusion PET using Rb-82 decay corrected in Bq/cc for diastolic, systolic, and combined whole cycle ungated images. Diastolic partial volume correction relative to systole was determined from the systolic/diastolic activity ratio, systolic partial volume correction from phantom dimensions comparable to systolic LV wall thicknesses and whole heart cycle partial volume correction for ungated images from fractional systolic-diastolic duration for systolic and diastolic partial volume corrections. RESULTS: For 264 PET perfusion images from 159 patients (105 rest-stress image pairs, 54 individual rest or stress images), average resting diastolic partial volume correction relative to systole was 1.14 ± 0.04, independent of heart rate and within ±1.8% of stress images (1.16 ± 0.04). Diastolic partial volume corrections combined with those for phantom dimensions comparable to systolic LV wall thickness gave an average whole heart cycle partial volume correction for ungated images of 1.23 for Rb-82 compared to 1.14 if positron range were negligible as for F-18. CONCLUSION: Quantitative myocardial PET perfusion imaging requires partial volume correction, herein demonstrated clinically from systolic/diastolic absolute activity ratios combined with phantom data accounting for Rb-82 positron range.

Divergent relatives of the Hsp70 protein chaperone such as the Hsp110 and Grp170 families have been recognized for some time, yet their biochemical roles remained elusive. Recent work has revealed that these "atypical" Hsp70s exist in stable complexes with classic Hsp70s where they exert a powerful nucleotide-exchange activity that synergizes with Hsp40/DnaJ-type cochaperones to dramatically accelerate Hsp70 nucleotide cycling. This represents a novel evolutionary transition from an independent protein-folding chaperone to what appears to be a dedicated cochaperone. Contributions of the atypical Hsp70s to established cellular roles for Hsp70 now must be deciphered.

The macronuclear genome of the ciliate Oxytricha trifallax displays an extreme and unique eukaryotic genome architecture with extensive genomic variation. During sexual genome development, the expressed, somatic macronuclear genome is whittled down to the genic portion of a small fraction (∼5%) of its precursor "silent" germline micronuclear genome by a process of "unscrambling" and fragmentation. The tiny macronuclear "nanochromosomes" typically encode single, protein-coding genes (a small portion, 10%, encode 2-8 genes), have minimal noncoding regions, and are differentially amplified to an average of ∼2,000 copies. We report the high-quality genome assembly of ∼16,000 complete nanochromosomes (∼50 Mb haploid genome size) that vary from 469 bp to 66 kb long (mean ∼3.2 kb) and encode ∼18,500 genes. Alternative DNA fragmentation processes ∼10% of the nanochromosomes into multiple isoforms that usually encode complete genes. Nucleotide diversity in the macronucleus is very high (SNP heterozygosity is ∼4.0%), suggesting that Oxytricha trifallax may have one of the largest known effective population sizes of eukaryotes. Comparison to other ciliates with nonscrambled genomes and long macronuclear chromosomes (on the order of 100 kb) suggests several candidate proteins that could be involved in genome rearrangement, including domesticated MULE and IS1595-like DDE transposases. The assembly of the highly fragmented Oxytricha macronuclear genome is the first completed genome with such an unusual architecture. This genome sequence provides tantalizing glimpses into novel molecular biology and evolution. For example, Oxytricha maintains tens of millions of telomeres per cell and has also evolved an intriguing expansion of telomere end-binding proteins. In conjunction with the micronuclear genome in progress, the O. trifallax macronuclear genome will provide an invaluable resource for investigating programmed genome rearrangements, complementing studies of rearrangements arising during evolution and disease.

The Ssel/Hsp110 molecular chaperones are a poorly understood subgroup of the Hsp70 chaperone family. Hsp70 can refold denatured polypeptides via a carboxyl-terminal peptide binding domain (PBD), which is regulated by nucleotide cycling in an amino-terminal ATPase domain. However, unlike Hsp70, both Sse1 and mammalian Hsp110 bind unfolded peptide substrates but cannot refold them. To test the in vivo requirement for interdomain communication, SSE1 alleles carrying amino acid substitutions in the ATPase domain were assayed for their ability to complement sse1Δ phenotypes. Surprisingly, all mutants predicted to abolish ATP hydrolysis complemented the temperature sensitivity of sse1Δ, whereas mutations in predicted ATP binding residues were non-functional. Remarkably, the two domains of Ssel when expressed in trans functionally complement the sse1Δ growth phenotype and interact by coimmunoprecipitation analysis, indicative of a novel type of interdomain communication. ^ Relatively little is known regarding the interactions and cellular functions of Ssel. Through co-immunoprecipitation analysis, we found that Ssel forms heterodimeric complexes with the abundant cytosolic Hsp70s Ssa and Ssb in vivo. Furthermore, these complexes can be efficiently reconstituted in vitro using purified proteins. The ATPase domains of Ssel and the Hsp70s were found to be critical for interaction as inactivating point mutations severely reduced interaction efficiency. Ssel stimulated Ssal ATPase activity synergistically with the co-chaperone Ydj1 via a novel nucleotide exchange activity. Furthermore, FES1, another Ssa nucleotide exchange factor, can functionally substitute for SSE1/2 when overexpressed, suggesting that Hsp70 nucleotide exchange is the fundamental role of the Sse proteins in yeast, and by extension, the Hsp110 homologs in mammals. ^ Cells lacking SSE1 were found to accumulate prepro-α-factor, but not the cotranslationally imported protein Kar2, similar to mutants in the Ssa chaperones. This indicates that the interaction between Ssel and Ssa is functionally significant in vivo. In addition, sse10 cells are compromised for cell wall strength, likely a result of decreased Hsp90 chaperone activity with the cell integrity MAP kinase SIC. Taken together, this work established that the Hsp110 family must be considered an essential component of Hsp70 chaperone biology in the eukaryotic cell.^

Primary Objectives - Describe and quantify the present strength and variability of the circulation and oceanic processes of the Nordic Seas regions using primarily observations of the long term spread of a tracer purposefully released into the Greenland Sea Gyre in 1996. - Improve our understanding of ocean processes critical to the thermaholine circulation in the Nordic Seas regions so as to be able to predict how this region may respond to climate change. - Assess the role of mixing and ageing of water masses on the carbon transport and the role of the thermohaline circulation in carbon storage using water transports and mixing coefficients derived from the tracer distribution. Specific Objectives Perform annual hydrographic, chemical and SF6 tracer surveys into the Nordic regions in order to: - Measure lateral and diapycnal mixing rates in the Greenland Sea Gyre and in the surrounding regions. - Document the depth and rates of convective mixing in the Greenland Sea using the SF6 and the water masses characteristics. - Measure the transit time and transport of water from the Greenland Sea to surrounding seas and outflows. Document processes of water mass transformation and entrainment occurring to water emanating from the central Greenland Sea. - Measure diapycnal mixing rates in the bottom and margins of the Greenland Sea basin using the SF6 signal observed there. Quantify the potential role of bottom boundary-layer mixing in the ventilation of the Greenland Sea Deep Water in absence of deep convection. Monitor the variability of the entrainment of water from the Greenland Sea using time series auto-sampler moorings at strategic positions i.e., sill of the Denmark Strait, Labrador Sea, Jan Mayen fracture zone and Fram Strait. Relate the observed variability of the tracer signal in the outflows to convection events in the Greenland Sea and local wind stress events. Obtain a better description of deepwater overflow and entrainment processes in the Denmark Strait and Faeroe Bank Channel overflows and use these to improve modelling of deepwater overflows. Monitor the tracer invasion into the North Atlantic using opportunistic SF6 measurements from other cruises: we anticipate that a number of oceanographic cruises will take place in the north-east Atlantic and the Labrador Sea. It should be possible to get samples from some cruises for SF6 measurements. Use process models to describe the spread of the tracer to achieve better parameterisation for three-dimensional models. One reason that these are so resistant to prediction is that our best ocean models are as yet some distance from being good enough, to predict climate and climate change.

El turismo es una actividad económica que crece en la Argentina y en la provincia de Misiones. Sin embargo, se desconoce el grado de crecimiento del empleo en general en el sector y especialmente de los profesionales (Licenciados, Técnicos y Guías) matriculados en el Colegio profesional. Asimismo se destaca la movilidad y la temporalidad de este empleo que es condicionado por la estacionalidad y otras variables económicas. En esta investigación se estudiará mediante encuestas la percepción de los alumnos avanzados de las carreras universitarias de turismo de la Provincia acerca de su inserción laboral, se analizará la situación de empleo de los profesionales, tanto desempleados, en relación de dependencia, free lance y los emprendedores matriculados, a fin de medir su inserción, movilidad, evaluación de su formación académica por competencias y proponer mecanismos de formación continua. Por último se busca medir mediante el Índice de Demanda Laboral, la demanda de empleo turístico en la provincia en el periodo 2011-2012 y se realizarán entrevistas a funcionarios, empresarios y referentes institucionales acerca de la situación actual y evolución del mercado laboral del turismo en Misiones.

The Arctic Ocean is connected with the North Atlantic Ocean by the Fram Strait between Greenland and Svalbard. The strait is located in the northern part of the Greenland Sea. In the eastern part of the strait, warm saline water flows northward as the West Spitsbergen Current; while in the western part, cold less-saline water flows southward as the East Greenland Current. The northwestern part of the Greenland Sea is normally covered with sea ice even in summer. Furthermore, this region is regarded as a major area where the Arctic sea ice is discharged into mid latitude oceans. Thus, this area plays an important role in heat and salt exchange processes in the Arctic marine system. The reveal exchange processes of water masses and ocean-atmosphere interaction in high-latitude oceans, a number of international research programs have been focused on the Greenland Sea and its surrounding waters. As one of the international Arctic research programs, oceanographic studies have been executed in cooperation with the Norsk Polarinstitutt and other institutes under the leadership of the National Institute of Polar Research since 1991. Japanese scientists have been carrying out field observations in and around Svalbard. The observations include not only physical measurements but also biological surveys. This report presents physical oceanographic data obtained in the Greenland Sea in 1992 and 1993, and data around Svalbard from 1991 to 1993.
