Nel corso di questa Tesi sono state studiate le reazioni di cluster carbonil anionici di Platino con fosfine. In particolare, sono state investigate nel dettaglio le reazioni dei cluster [Pt3n(CO)6n]2– (n = 3,4) e [Pt19(CO)22]n– (n = 3,4) con PPh3. Sono state poi preliminarmente studiate anche le reazioni di [Pt24(CO)30]2– e [Pt38(CO)44]2– con PPh3. Questo ha portato alla sintesi e alla completa caratterizzazione mediante diffrazione di raggi X su cristallo singolo delle specie [NBu4]2[Pt9(CO)16(PPh3)2] e [NBu4]x[Pt22(CO)22(PPh3)6]2•yCH3CN, contenenti gli anioni [Pt9(CO)16(PPh3)2]2– e [Pt22(CO)22(PPh3)6]2–. È stato inoltre preparato un nuovo composto tentativamente formulato come [Pt19(CO)20(PPh3)2]4–, sulla base dei dati IR, 31P NMR e ESI-MS. Questi rappresentano i primi esempi di cluster carbonilici anionici di Platino contenenti fosfine. Nel caso delle reazioni di [Pt24(CO)30]2– e [Pt38(CO)44]2– con PPh3 i prodotti sono stati caratterizzati al momento solo mediante spettroscopia IR, e quindi è molto difficile ipotizzare una loro struttura. I composti [Pt9(CO)16(PPh3)2]2– e [Pt19(CO)20(PPh3)2]4– sono stati investigati mediante spettroscopia 31P NMR in soluzione a temperatura variabile. Il primo mostra un unico segnale 31P NMR, in accordo con la struttura allo stato solido, mentre [Pt19(CO)20(PPh3)2]4– è risultato essere flussionale. È stato poi studiato nel dettaglio il comportamento fotochimico dei cluster [Pt3n(CO)6n]2– (n = 3-6) in funzione della concentrazione, confermando la loro natura di “Double emitting quantum dots”. Infine è stato preparato e caratterizzato strutturalmente il sale [DAMS]2[Pt9(CO)18]•dmf, contenente il catione [DAMS]+.

In den letzten Jahrzehnten wurde eine deutliche, anhaltende Veränderung des globalen Klimas beobachtet, die in Zukunft zu einer Erhöhung der durchschnittlichen Oberflächentemperatur, erhöhten Niederschlagsmengen und anderen gravierenden Umweltveränderungen führen wird (IPCC 2001). Der Klimawandel wird in Flüssen sowohl mehr Extremereignisse verursachen als auch das Abflussregime bisher schmelzwasserdominierter Flüsse zu grundwassergespeisten hin ändern; dies gilt insbesondere für den Rhein (MIDDELKOOP et al. 2001). Um die möglichen Auswirkungen dieser Veränderungen auf die genetische Populationsstruktur von Makrozoobenthosorganismen vorhersagen zu können, wurden in den grundwassergespeisten Flüssen Main und Mosel sowie im Rhein Entnahmestellen oberhalb und unterhalb von Staustufen beprobt, die durch kontrastierende Strömungsverhältnisse als Modell für die zu erwartenden Änderungen dienten. Als Untersuchungsobjekt wurden Dreissena polymorpha PALLAS 1771 sowie Dikerogammarus villosus SOWINSKI 1894 herangezogen. Sie zeichnen sich durch hohe Abundanzen aus, sind aber unterschiedlich u.a. hinsichtlich ihrer Besiedlungsstrategie und –historie. Bei beiden Spezies sind die phylogeographischen Hintergründe bekannt; daher wurde auch versucht, die Einwanderungsrouten in der Populationsstruktur nachzuweisen (phylogeographisches Szenario). Dies konkurrierte mit der möglichen Anpassung der Spezies an das Abflussregime des jeweiligen Flusses (Adaptations-Szenario). Die Populationen wurden molekulargenetisch mit Hilfe der AFLP-Methode („Amplified-Fragment Length Polymorphism“) untersucht. Die Ergebnisse zeigen, dass D. polymorpha deutlich durch die Abflussregimes der Flüsse (Schmelz- oder Grundwasserdominanz) beeinflusst wird. Die Allelfrequenzen in Populationen des Rheins sind von denen der beiden grundwassergespeisten Flüsse Main und Mosel deutlich unterscheidbar (Adaptations-Szenario). Jedoch ist kein Unterschied der genetischen Diversitäten zu beobachten; das ist auf die lange Adaptation an ihre jeweiligen Habitate durch die lange Besiedlungsdauer zurückzuführen. Dies ist auch der Grund, warum die Einwanderungsrouten anhand der Populationsstruktur nicht mehr nachzuweisen waren. Die kontrastierenden Strömungsverhältnisse um die Staustufen hatten ebenfalls keine konsistenten Auswirkungen auf die genetische Diversität der Populationen. Diese Ergebnisse zeigen eine hohe phänotypische Plastizität der Spezies und dadurch eine große Anpassungsfähigkeit an wechselnde Umweltbedingungen, die unter anderem für den großen Erfolg dieser Spezies verantwortlich ist. D. villosus wanderte erst vor Kurzem in das Untersuchungsgebiet ein; die Einwanderungsroute war anhand der genetischen Diversität nachvollziehbar (phylogeographisches Szenario); durch die kurze Besiedlungsdauer war eine Adaptation an die divergenten Abflussregime der Flüsse nicht zu erwarten und wurde auch nicht gefunden. Dagegen war ein deutlicher negativer Einfluss von starker Strömung auf die genetische Diversität nachweisbar. Die Ergebnisse weisen darauf hin, dass die zukünftigen Auswirkungen des Klimawandels auf die Strömungsgeschwindigkeit negative Konsequenzen auf die genetische Diversität von D. villosus haben werden, während D. polymorpha hier keine Auswirkungen erkennen lässt. Die Auswirkungen des veränderten Abflussregimes im Rhein sind für D. villosus mit den vorliegenden Daten aufgrund der kurzen Besiedlungsdauer nicht vorhersagbar; D. polymorpha wird durch die Veränderung des Rheins zu einem grundwassergespeisten Fluss zwar einen Wandel in der genetischen Struktur erfahren, aber auch hier keine Einbußen in der genetischen Diversität erleiden.

Sheet pile walls are one of the oldest earth retention systems utilized in civil engineering projects. They are used for various purposes; such as excavation support system, cofferdams, cut-off walls under dams, slope stabilization, waterfront structures, and flood walls. Sheet pile walls are one of the most common types of quay walls used in port construction. The worldwide increases in utilization of large ships for transportation have created an urgent need of deepening the seabed within port areas and consequently the rehabilitation of its wharfs. Several methods can be used to increase the load-carrying capacity of sheet-piling walls. The use of additional anchored tie rods grouted into the backfill soil and arranged along the exposed wall height is one of the most practical and appropriate solutions adopted for stabilization and rehabilitation of the existing quay wall. The Ravenna Port Authority initiated a project to deepen the harbor bottom at selected wharves. An extensive parametric study through the finite element program, PLAXIS 2D, version 2012 was carried out to investigate the enhancement of using submerged grouted anchors technique on the load response of sheet-piling quay wall. The influence of grout-ties area, length of grouted body, anchor inclination and anchor location were considered and evaluated due to the effect of different system parameters. Also a comparative study was conducted by Plaxis 2D and 3D program to investigate the behavior of these sheet pile quay walls in terms of horizontal displacements induced along the sheet pile wall and ground surface settlements as well as the anchor force and calculated factor of safety. Finally, a comprehensive study was carried out by using different constitutive models to simulate the mechanical behavior of the soil to investigate the effect of these two models (Mohr-Coulomb and Hardening Soil) on the behavior of these sheet pile quay walls.

Bovine Neonatal Pancytopenia (BNP) is a novel haemorrhagic disease in sucking calves, characterised by bleeding, haematological changes and high mortality. Dams that gave birth to BNP affected calves were immunized with PregSure® BVD, a highly adjuvanted vaccine against Bovine Viral Diarrhoea (BVD). We can show that bioprocess impurities in the vaccine, originating from the cell line used for vaccine production induces alloantibodies in vaccinated cattle. Via flow cytometry and immunoprecipitation we can demonstrate that PregSure® BVD immunization leads to BNP alloantibody production. BNP alloantibodies target highly polymorphic bovine MHC-I molecules (BoLA I). We sequenced eight BoLA I variants expressed by the production cell line and identified three alleles which are responsible for the majority of PregSure® BVD induced BoLA I reactivity. The BoLA I alleles of BNP unaffected calves are not recognized by the BNP associated alloantibodies of their respective dams. We also examined whether BNP alloantibodies cross-react with human cells, thus being a potential hazard for human colostrum consumers and could show that BNP alloantibodies are cross-reactive to human MHC-I and can even be found in commercial colostrum powder manufactured from cows immunized with PregSure® BVD. Overall we can demonstrate that BNP is a vaccine induced alloimmune disease.

Sexual selection by female mating preference for male nuptial coloration has been suggested as a driving force in the rapid speciation of Lake Victoria cichlid fish. This process could have been facilitated or accelerated by genetic associations between female preference loci and male coloration loci. Preferences, as well as coloration, are heritable traits and are probably determined by more than one gene. However, little is known about potential genetic associations between these traits. In turbid water, we found a population that is variable in male nuptial coloration from blue to yellow to red. Males at the extreme ends of the phenotype distribution resemble a reproductively isolated species pair in clear water that has diverged into one species with blue-grey mates and one species with bright red males. Females of the turbid water population vary in mating preference coinciding with the male phenotype distribution. For the current study, these females were mated to blue males. We measured the coloration of the sires and male offspring. Parents-offspring regression showed that the sires did not affect male offspring coloration, which confirms earlier findings that the blue species breeds true. In contrast, male offspring coloration was determined by the identity of the dams, which suggests that there is heritable variation in male color genes between females. However, we found that mating preferences of the dams were not correlated with male offspring coloration. Thus, there is no evidence for strong genetic linkage between mating preference and the preferred trait in this population [Current Zoology 56 (1): 57-64 2010].

Many foals develop diarrhoea within the first two weeks of life which has been suggested to coincide with postpartum oestrus in their dams. To analyse the pathogenesis of this diarrhoea we have determined faecal bacteria in foals and their dams (n=30 each), and serum IGF-1 and gamma-globulins for 6 weeks after birth. In addition, effects of beta-carotene supplementation to mares (group 1: 1000 mg/day, n=15, group 2: control, n=15) on diarrhoea in foals were studied. Diarrhoea occurred in 92 and 79% of foals in groups 1 and 2, respectively, but was not correlated with oestrus in mares. Beta-carotene supplementation was without effect on foal diarrhoea. In mares, bacterial flora remained stable. The percentage of foals with cultures positive for E. coli was low at birth but increased within one day, the percentage positive for Enterococcus sp. was low for 10 days and for Streptococcus sp. and Staphylococcus sp. was low for 2-4 weeks. By 4 weeks of age, bacterial flora in foals resembled an adult pattern. Concentration of serum IGF-1 was low at birth (group 1: 149 +/- 11, group 2: 166 +/- 17ng/ml), increased after day 1 (day 7 group 1: 384 +/- 30, group 2: 372 +/- 36) but at no time differed between groups. Serum gamma-globulin concentration in foals was low before colostrum intake and highest on day 1 (p<0.001 over time). In conclusion, neonatal diarrhoea in foals does not coincide with postpartum oestrus in their dams but with changes in intestinal bacteria and is not influenced by beta-carotene supplementation given to mares.

This study investigated the effects of different environmental treatments and personality types on aggression at mixing of newly weaned domestic piglets. From birth to weaning, 16 litters were housed with their dams in either barren (B) or larger, substrate-enriched (E) environments. At 15 days old, piglets were classified as 'high' (HR) or low resistant' (LR) in a manual restraint test (backtest), which is thought to identify proactive (HR) and reactive (LR) stress coping strategies that may reflect different personality types. At 30 days old, 128 piglets were weaned, relocated and mixed into 32 pens comprising two HR and two LR unfamiliar pigs, balanced for sex and weaning weight. Eight B and eight E groups changed environmental condition whereas the others remained in the same type of environment. Number and duration of fights. fight outcomes and unilateral fighting were scored for 5 h post-mixing and skin lesions were counted before and 5 h, 1 day and 2 days after mixing. On the day following weaning, fighting and also exploratory and oral manipulative behaviours were measured for 6 h. Generalized Linear Mixed Model analyses suggested interactions between pre-weaning environment, post-weaning environment and personality type. Overall, pre-weaning E pigs had longer fights at weaning and mixing (P=0.01) and fought for longer on the next day (P=0.02) than pre-weaning B pigs, and inflicted more skin lesions (P=0.02). Post-weaning enrichment did not affect fighting at mixing but reduced the time spent fighting the next day (P=0.03). Personality had subtle and environment-dependent effects on fighting, and influenced the "structure" rather than the amount of aggressive behaviour. HR pigs, for instance, bullied (i.e. chased surrendering pigs) more often (P=0.009) and their fighting behaviour was less affected by their relative body weight than that of LR pigs. Post-weaning E pigs showed relatively higher levels of exploratory behaviour (P=0.02) and less oral manipulative behaviour (P=0.04) than post-weaning B pigs. In particular, switching from a good quality environment (E) to a worse quality one (B) at weaning decreased exploratory behaviour on the next day, especially for LR pigs, who also tended to fight with and orally manipulate their pen mates more in that condition, and seemed to be more affected by a deterioration of the environment. Overall, pre-weaning enrichment increased aggression after weaning whereas post-weaning enrichment reduced it, and personality type related to some aspects of fighting behaviour. While having the highest vitamin C (VitC) concentrations in the body, specific functions of VitC in the brain have only recently been acknowledged. We have shown that postnatal VitC deficiency in guinea pigs causes impairment of hippocampal memory function and leads to 30% less neurons. This study investigates how prenatal VitC deficiency affects postnatal hippocampal development and if any such effect can be reversed by postnatal VitC repletion. Eighty pregnant Dunkin Hartley guinea pig dams were randomized into weight stratified groups receiving High (900 mg) or Low (100 mg) VitC per kg diet. Newborn pups (n = 157) were randomized into a total of four postnatal feeding regimens: High/High (Control); High/Low (Depleted), Low/Low (Deficient); and Low/High (Repleted). Proliferation and migration of newborn cells in the dentate gyrus was assessed by BrdU labeling and hippocampal volumes were determined by stereology. Prenatal VitC deficiency resulted in a significant reduction in postnatal hippocampal volume (P<0.001) which was not reversed by postnatal repletion. There was no difference in postnatal cellular proliferation and survival rates in the hippocampus between dietary groups, however, migration of newborn cells into the granular layer of the hippocampus dentate gyrus was significantly reduced in prenatally deficient animals (P<0.01). We conclude that a prenatal VitC deficiency in guinea pigs leads to persistent impairment of postnatal hippocampal development which is not alleviated by postnatal repletion. Our findings place attention on a yet unrecognized consequence of marginal VitC deficiency during pregnancy.

The major route of transmission of Neospora caninum in cattle is transplacentally from an infected cow to its progeny. Therefore, a vaccine should be able to prevent both the horizontal transmission from contaminated food or water and the vertical transmission. We have previously shown that a chimeric vaccine composed of predicted immunogenic epitopes of NcMIC3, NcMIC1 and NcROP2 (recNcMIC3-1-R) significantly reduced the cerebral infection in BALB/c mice. In this study, mice were first vaccinated, then mated and pregnant mice were challenged with 2×10(6)N. caninum tachyzoites at day 7-9 of pregnancy. Partial protection was only observed in the mice vaccinated with a tachyzoite crude protein extract but no protection against vertical transmission or cerebral infection in the dams was observed in the group vaccinated with recNcMIC3-1-R. Serological and cytokine analysis showed an overall lower cytokine level in sera associated with a dominant IL-4 expression and high IgG1 titers. Thus, the Th2-type immune response observed in the pregnant mice was not protective against experimental neosporosis, in contrary to the mixed Th1-/Th2-type immune response observed in the non-pregnant mouse model. These results demonstrate that the immunomodulation that occurs during pregnancy was not favorable for the protection against N. caninum infection conferred by vaccination with recNcMIC3-1-R.

Synthetic glucocorticoids (GC) are used as a clinical therapeutic to stimulate lung development in fetuses that present the risk of preterm delivery. Previous studies have shown that a prenatal exposure to Dexamethasone (DEX) causes a disturbance in normal GC mediation of neuritic outgrowth, cell signaling, and serotonergic systems. Our hypothesis is that a prenatal exposure to DEX during the third trimester of pregnancy alters 5HT1A receptor function. Pregnant dams were injected daily with 150μg/ml/kg of DEX from gestation day 14 through 19. Control dams were treated with and equal volume of saline. Swim stress followed by elevated plus maze testing was conducted on male rats an hour and a half prior to being sacrificed to induce postnatal acute stress. The non-stressed group was also tested and allowed to return to baseline before sacrifice. Hippocampi were analyzed using a radioligand-receptor binding assay and GTPγS35 incorporation (3H-MPPF antagonist and 8-OH-DPAT agonist, respectively).
