Background Lifestyle factors such as diet and physical activity have been shown to modify the association between fat mass and obesity–associated (FTO) gene variants and metabolic traits in several populations; however, there are no gene-lifestyle interaction studies, to date, among Asian Indians living in India. In this study, we examined whether dietary factors and physical activity modified the association between two FTO single nucleotide polymorphisms (rs8050136 and rs11076023) (SNPs) and obesity traits and type 2 diabetes (T2D). Methods The study included 734 unrelated T2D and 884 normal glucose-tolerant (NGT) participants randomly selected from the urban component of the Chennai Urban Rural Epidemiology Study (CURES). Dietary intakes were assessed using a validated interviewer administered semi-quantitative food frequency questionnaire (FFQ). Physical activity was based upon the self-report. Interaction analyses were performed by including the interaction terms in the linear/logistic regression model. Results There was a significant interaction between SNP rs8050136 and carbohydrate intake (% energy) (Pinteraction = 0.04), where the ‘A’ allele carriers had 2.46 times increased risk of obesity than those with ‘CC’ genotype (P = 3.0 × 10−5) among individuals in the highest tertile of carbohydrate intake (% energy, 71 %). A significant interaction was also observed between SNP rs11076023 and dietary fibre intake (Pinteraction = 0.0008), where individuals with AA genotype who are in the 3rd tertile of dietary fibre intake had 1.62 cm lower waist circumference than those with ‘T’ allele carriers (P = 0.02). Furthermore, among those who were physically inactive, the ‘A’ allele carriers of the SNP rs8050136 had 1.89 times increased risk of obesity than those with ‘CC’ genotype (P = 4.0 × 10−5). Conclusions This is the first study to provide evidence for a gene-diet and gene-physical activity interaction on obesity and T2D in an Asian Indian population. Our findings suggest that the association between FTO SNPs and obesity might be influenced by carbohydrate and dietary fibre intake and physical inactivity. Further understanding of how FTO gene influences obesity and T2D through dietary and exercise interventions is warranted to advance the development of behavioral intervention and personalised lifestyle strategies, which could reduce the risk of metabolic diseases in this Asian Indian population.

O objetivo deste estudo foi analisar o papel do polimorfismo de I/D do gene da Enzima Conversora de Angiotensina (ECA) e o polimorfismo K121Q da PC-1 nas modificações das taxas de filtração glomerular (TFG), excreção urinária de albumina (EUA) e pressão arterial em uma coorte de pacientes diabéticos tipo 1 normoalbuminúricos (EUA<20μg/min) em um estudo com seguimento de 10,2 ± 2,0anos (6,5 a 13,3 anos). A EUA (imunoturbidimetria), TFG (técnica da injeção única de 51Cr-EDTA), HbA1c (cromatografia de troca iônica) e pressão arterial foram medidas no início do estudo e a intervalos de 1,7 ± 0,6 anos. O polimorfismo I/D e K121Q foram determinados através da PCR e restrição enzimática. Onze pacientes apresentaram o genótipo II, 13 o ID e 6 apresentaram o genótipo DD. Pacientes com o alelo D (ID/DD) desenvolveram mais freqüentemente hipertensão arterial e retinopatia diabética. Os 3 pacientes do estudo que desenvolveram nefropatia diabética apresentaram o alelo D. Nos pacientes ID/DD (n=19) ocorreu maior redução da TFG quando comparados com os pacientes II (n=11) (-0,39 ± 0,29 vs – 0,12 ± 0,37 ml/min/mês; P=0,035). A presença do alelo D, em análise de regressão múltipla linear (R2=0,15; F=4,92; P=0,035) foi o único fator associado à redução da TFG (-0,29 ± 0,34 ml/min/mês; P<0,05). Já o aumento da EUA (log EUA = 0,0275 ± 0,042 μg/min/mês; P=0,002) foi associado somente aos níveis iniciais de EUA (R2=0,17; F=5,72; P=0,024). Um aumento significativo (P<0,05) no desenvolvimento de hipertensão arterial e de novos casos de retinopatia diabética foi observado somente nos pacientes com os genótipos ID/DD. Vinte e dois pacientes apresentaram genótipo KK, 7 KQ e 1 apresentou genótipo QQ. Pacientes com os genótipos KQ/QQ apresentaram um aumento significativo (P=0,045) de novos casos de retinopatia diabética. Em conclusão a presença do alelo D nesta amostra de pacientes DM tipo 1 normoalbuminúricos e normotensos está associada com aumento na proporção de complicações microvasculares e hipertensão arterial.

O trabalho tem por objetivo trazer elementos da corrente filosófica do pragmatismo para a discussão metodológica em economia, particularmente no que diz respeito a teoria da verdade e suas implicações para o debate epistemológico. Após apresentar as contribuições dos pioneiros do pragmatismo William James, John Dewey e Charles S. Peirce e discutir aspectos da obra de W. Quine e Richard Rorty, procuramos identificar influências dessa corrente filosófica em importantes economistas tais como J. M. Keynes, M. Friedman e T. Veblen. Por fim concluímos com algumas reflexões possivelmente úteis para a prática da ciência econômica.

The main purpose of this exploratory work is to investigate the attributes that motivate customers to choose services within beauty and aesthetic centers. Everyday, we are taking by surprise due to the discovery of new procedures that will make many dreams come true regarding youth¿s prolong. Customers are much more demanding now, so the beauty industry must emphasize the importance of beauty in our day lives. The old age fears, the need of men and women in getting close to the perfect beauty and looking great explaining the reason many customer looking for beauty and aesthetic centers to ¿postpone¿ the inevitable old age.This work focus on the increase of beauty and aesthetic industry market position, the quality of service provide by these centers, emphasize the concepts of service quoted by different authors, for instance, Lovelock (2006), Las Casas (2002), Kotler (1998), the characteristics of services, the behaviour of customers, the conceptualized models of service quality from Albrecht and Zemke (1998), moments of truth according to Carlzon (1994).This work presents a summary of beauty along the years; shows the methodology used and the interviews results showing the main quality service attributes in beauty and aesthetic centers; the difficulty faced and suggestion for future research.

The objective of this paper is to bring elements from the philosophical movement of hermeneutics and pragmatism to the discussion on methodology in economics, with a specific concern on the theory of truth. Our aim is to present the concept of the hermeneutic space, developed by the philosopher Richard Rorty, as a rational justification for pluralism in economics. We consider the hermeneutic space an interesting concept which should allow us to overcome the void left by the incapacity of epistemological theories to explain the evolution of sciences. It defends the idea that our culture, values and ways of interpreting things are what build the sciences, not any closed epistemological method. In this sense, pluralism is nothing more than letting the hermeneutic space work, without epistemological barriers, and understanding that this is desirable for the future development of economics as a science. This approach differs from all other methodological justifications for pluralism because it does not rely on any epistemological method, but assumes that the hermeneutic space can entirely fulfill the gap created by them

This article discusses some issues in communicating experience, based on a life history interview with 83-year-old Brazilian jurist Evandro Lins e Silva conducted by the Getúlio Vargas Foundation’s oral history program (Centro de Pesquisa e Documentação de História Contemporânea do Brasil, or CPDOC) between August 1994 and January 1995.1The text focuses especially on two images used by the interviewee, which consolidate both the experiences that have been communicated to him and the experience that he himself endeavors to communicate regarding his activities as an attorney and the status of truth within the field of law.

Este trabalho analisa a utilização do estágio probatório no Brasil para avaliar se houve mudanças na sua utilização a partir das reformas empreendidas na década de 1990. A literatura indica a inoperância desta avaliação segundo as mesmas dificuldades apresentadas em outros instrumentos da política de recursos humanos do Estado, tais como a seleção por meio do concurso público e a implementação de sistemas de avaliação de desempenho. A pesquisa delineou o histórico da utilização do estágio probatório no Brasil, identificou os aspectos que o constituem e, por meio de três estudos de caso, avaliou a utilização do estágio probatório em três carreiras do governo do Estado de São Paulo nas quais se buscou entender se houve mudanças na sua utilização após as alterações estabelecidas pela emenda constitucional nº 19, de 1998. O estudo indicou que o estágio probatório se fez presente e foi reforçado pelas normas gerais relacionadas aos servidores públicos, mas nunca houve um movimento para torná-lo uma avaliação efetiva, o que o manteve a margem das discussões da seleção por mérito, que enfatizava a importância do concurso público; e desvalorizado pela extensão do direito à estabilidade a todos os funcionários independentemente do ingresso por concurso ou de um processo de avaliação efetivo. Além disso, este incorporou as dificuldades da implementação de avaliações de desempenho no setor público brasileiro. No entanto, os três casos avaliados demonstraram incorporar mudanças incrementais na utilização do estágio probatório após 1998, com características distintas de acordo com as carreiras correspondentes e a política pública desenvolvida pelas Secretarias, não sendo possível afirmar sua inoperância.

A Salmonella sp. é uma das principais causas de doenças transmitidas por alimentos, principalmente de origem animal. Os suínos podem ser portadores assintomáticos de Salmonella sp. e, nessa condição, serem levados ao abate. Desta forma, a entrada de Salmonella sp. na cadeia de produção é uma preocupação para a indústria. Sabendo-se da importância deste microrganismo, pelo seu impacto na indústria e na saúde pública, este trabalho teve como objetivo determinar a prevalêncía de Salmonella sp. em suínos levados ao abate em um frigorífico sob inspeção federal no Rio Grande do Sul, e correlacionar com a contaminação de cortes de pernil. Para tanto, 64 amostras de conteúdo intestinal e 121 amostras de cortes de pernil coletadas em 4 visitas, foram submetidas ao isolamento e identificação de Salmonella sp. As amostras de Salmonella sp. foram avaliadas quanto a resistência a 14 antimicrobianos, através da técnica de difusão em ágar. Salmonella sp. foi isolada de 46,87% das amostras de conteúdo intestinal e de 49,59% dos cortes de pernil. Foi encontrado grande número de sorovares, sendo os mais prevalentes Panama e Bredeney. Das amostras isoladas, 60,8% apresentaram perfil de multiresistência, sendo os maiores índices de resistência a sulfonamida, ácido nalidíxico e tetraciclina. Utilizando o perfil de resistência a antimicrobianos de amostras de um mesmo sorovar forma comparadas quanto a sua similaridade e agrupadas em dendrogramas, observou-se grande diversidade nas linhagens isoladas. Os resultados demonstraram que a entrada de animais no frigorífico excretando Salmonella sp., pode contribuir para a contaminação do produto final. A diversidade de sorovares e linhagens pode representar que são várias as fontes de contaminação, tanto dos animais quanto do produto final.

Os caracteres produtivos são normalmente influenciados por muitos fatores, sendo difícil determinar todos os locos envolvidos em um fenótipo específico. Por isso, a seleção animal tem se baseado principalmente em uma estimativa direta ou indireta do fenótipo. A leptina é um importante regulador do metabolismo energético, da adiposidade e da reprodução. E por desempenhar diferentes funções, pode ser considerado um bom gene candidato para o estudo de associações entre marcadores moleculares e a eficiência reprodutiva ou ganho de peso. Em várias espécies, têm sido descritos diversos polimorfismos no gene da leptina, influenciando o ganho de peso, a reprodução, e outras características produtivas. Em bovinos, o STR IDVGA51 e o SNP LEPSau3A1, foram descritos por afetarem a performance reprodutiva, os alelos IDVGA51*181 e LEPSau3A1*2 estando associados a um aumento no intervalo entre partos de 79 e 81 dias, respectivamente, e os STRs BMS1074 e BM1500 afetam o ganho de peso, em vacas, no pós-parto: os alelos BMS1074*151 e BM1500*135 reduzindo e aumentando, respectivamente, o ganho de peso diário. Para confirmar o efeito ou não destes alelos na expressão do gene da leptina, este trabalho comparou os níveis de mRNA de leptina em animais portadores e não portadores dos alelos IDVGA51*181, LEPSau3A1*2, BMS1074*151 e BM1500*135, com os objetivos de: 1. Verificar as distribuições genotípicas e alélicas dos marcadores IDVGA51, BMS1074, BM1500 e LEPSau3A1, em uma amostra de 137 bovinos da raça Brangus-Ibagé, provenientes da Empresa Brasileira de Pesquisa Agropecuária (EMBRAPA-Pecuária Sul, Bagé-RS). 2. Identificar, no rebanho, animais portadores e não portadores dos alelos IDVGA51*181, LEPSau3A1*2, BMS1074*151 ou BM1500*135, previamente descritos como associados à eficiência reprodutiva e ganho de peso. 3. Avaliar os animais, criados em campo nativo, com pastagem natural, quanto à sua condição corporal. 4. Realizar procedimento cirúrgico, para obtenção de amostras de tecido adiposo subcutâneo e omental. 5. Dosar os níveis de mRNA de leptina nos adipócitos destes dois tecidos, através do método quantitativo em tempo real (Real-Time RT-PCR), comparando a expressão do gene LEP entre indivíduos portadores e não portadores dos alelos de predisposição a ganho de peso e desempenho reprodutivo.

In vitro and in animal models, APE1, OGG1, and PARP-1 have been proposed as being involved with inflammatory response. In this work, we have investigated if the SNPs APE1 Asn148Glu, OGG1 Ser326Cys, and PARP-1 Val762Ala are associated to meningitis and also developed a system to enable the functional analysis of polymorphic proteins. Patients with bacterial meningitis (BM), aseptic meningitis (AM) and controls (non-infected) genotypes were investigated by PIRA-PCR or PCR-RFLP. DNA damages were detected in genomic DNA by Fpg treatment. IgG and IgA were measured from plasma and the cytokines and chemokines were measured from cerebrospinal fluid samples using Bio-Plex assays. The levels of NF-κB and c-Jun were measured in CSF by dot blot assays. A significant (P<0.05) increase in the frequency of APE1 148Glu allele in BM and AM patients was observed. A significant increase in the genotypes Asn/Asn in control group and Asn/Glu in BM group was also found. For the SNP OGG1 Ser326Cys, the genotype Cys/Cys was more frequent (P<0.05) in BM group. The frequency of PARP-1 Val/Val genotype was higher in control group (P<0.05). The occurrence of combined SNPs increased significantly in BM patients, indicating that these SNPs may be associated to the disease. Increasing in sensitive sites to Fpg was observed in carriers of APE1 148Glu allele or OGG1 326Cys allele, suggesting that SNPs affect DNA repair activity. Alterations in IgG production were observed in the presence of SNPs APE1Asn148Glu, OGG1Ser326Cys or PARP-1Val762Ala. Reductions in the levels ofIL-6, IL-1Ra, MCP-1/CCL2and IL-8/CXCL8 were observed in the presence of APE1148Glu allele in BM patients, however no differences were observed in the levels of NF-κB and c-Jun considering genotypes and analyzed groups. Using APE1 as model, a system to enable the analysis of cellular effects and functional characterization of polymorphic proteins was developed using strategies of cloning APE1 cDNA in pIRES2-EGFP vector, cellular transfection of the construction obtained, siRNA for endogenous APE1 and cellular cultures genotyping. In conclusion, we obtained evidences of an effect of SNPs in DNA repair genes on the regulation of immune response. This is a pioneering work in the field that shows association of BER variant enzymes with an infectious disease in human patients, suggesting that the SNPs analyzed may affect immune response and damage by oxidative stress level during brain infection. Considering these data, new approaches of functional characterization must be developed to better analysis and interactions of polymorphic proteins in response to this context

In a hospital environment, these bacteria can be spread by insects such as ants, which are characterized by high adaptability to the urban environment. Staphylococcus is a leading cause of hospital infection. In Europe, Latin America, USA and Canada, the group of coagulase negative staphylococci (CoNS) is the second leading cause of these infections, according to SENTRY (antimicrobial surveillance program- EUA). In this study, we investigated the potential of ants (Hymenoptera: Formicidae) as vehicle mechanics of Staphylococcus bacteria in a public hospital, in Natal-RN. The ants were collected, day and night, from June 2007 to may 2008, in the following sectors: hospitals, laundry, kitchen, blood bank. The ants were identified according to the identification key of Bolton, 1997. For the analysis of staphylococci, the ants were incubated in broth Tryptic Soy Broth (TSB) for 24 hours at 35 º C and then incubated on Mannitol Salt Agar. The typical colonies of staphylococci incubated for 24 hours at 35 ° C in Tryptic Soy Agar for the characterization tests (Gram stain, catalase, susceptibility to bacitracin and free coagulase). The identification of CoNS was performed through biochemical tests: susceptibility to novobiocin, growth under anaerobic conditions, presence of urease, the ornithine decarboxylation and acid production from the sugars mannose, maltose, trehalose, mannitol and xylose. The antimicrobial susceptibility examined by disk-diffusion technique. The technique of Polymerase Chain Reaction was used to confirm the presence of mecA gene and the ability to produce biofilm was verified by testing in vitro using polystyrene inert surface, in samples of resistant staphylococci. Among 440 ants, 85 (19.1%) were carrying coagulase-negative staphylococci (CoNS) of the species Staphylococcus saprophyticus (17), Staphylococcus epidermidis (15), Staphylococcus xylosus (13), Staphylococcus hominis hominis (10), Staphylococcus lugdunensis (10), Staphylococcus warneri (6), Staphylococcus cohnii urealyticum (5), Staphylococcus haemolyticus (3), Staphylococcus simulans (3), Staphylococcus cohnii cohnii (2), and Staphylococcus capitis (1). No Staphylococcus aureus was found. Among the isolates, 30.58% showed resistance to erythromycin. Two samples of CoNS (2.35%), obtained from the ant Tapinoma melanocephalum collected in the post-surgical female ward, S. Hominis hominis and S. lugdunensis harbored the mecA gene and were resistant to multiple antibiotics, and the specie S. hominis hominis even showed to be a biofilm producer. This study proves that ants act as carriers of multidrug-resistant coagulase-negative Staphylococci and biofilm producers and points to the risk of the spreading of pathogenic microorganisms by this insect in the hospital environment

Physiological changes induced by the aging process is dynamic and progressive, reducing the adaptability and independence of older people and may be influenced by genetic and environmental factors. Thus the aim of this thesis was to investigate the association between polymorphism of the ACE gene ID and the phenotypes of muscular strength and blood pressure of 62 elderly Brazilian (67.35 ± 5.66 years) during a 16-week program of supervised training. The elderly women were stratified by age, with the group 1 (G1, n = 34) <70 years and group 2 (G2 n = 28) ≥ 70 years, and in three groups by ACE, ACE-II (n = 8) ACE- DD (n = 35) and ACE-ID (n = 19). The level of muscle strength was evaluated by the method of maximum repetitions and measures of blood pressure (BP) were measured before and after training (PAPré1 and PAPós1) and before and after each training session (PAPre2 and PAPós2), in place of training. DNA samples were isolated from peripheral blood leukocytes polymorphism and insertion / deletion (ID) of the ACE gene (rs1800795) was genotyped by polymerase chain reaction (PCR) plus PCR-confirmatory. The genotype distribution of the polymorphism ID attended the prerogatives of Hardy-Weitíherg. There was variation in power levels before and after training and the age between groups (t-test) and the ACE polymorphism (ANOVA) (p <0.05). Depending on the results it was concluded that resistance training helps to reduce SBP and increased muscle strength of upper and lower limbs when considering the age and ACE polymorphism. In this study the Elderly carriers of the D allele were more reactive to changes in BP resistance training. This study was multidisciplinary project involving researchers in the areas Medical, Physical Education, Pharmacy, Nutrition, Gerontology and Statistics. This fulfilled the requirements of the multidisciplinary Graduate Program in Health Sciences

Propomo-nos a mostrar que, ao contrário do que supõe o senso comum, é possível separar o problema da cientificidade do problema da verdade, ou seja, podem-se estabelecer critérios de acordo com os quais um conhecimento é legítimo, embora não se possa nunca saber se ele ou qualquer outro é verdadeiro; pode-se, ainda, aplicar esses mesmos critérios para identificar e desqualificar pseudoconhecimentos. Para tratar dessa temática familiar à Filosofia da Ciência, tomamos como exemplo um pensador que lhe é precursor, o filósofo empirista do século XVIII, David Hume.

Beta thalassemia arises as a consequence of the reduction (β+, β++, βsilent) or absence (β0) of beta globin chain synthesis and results from a number of mechanisms that lead to genetic defects. The inheritance of beta thalassemia is characterized by the existence of heterozygous individuals, compound heterozygotes, homozygotes and those with coinheritance of beta thalassemia allele and other thalassemias and/or hemoglobin variants. The aim of this study was to perform molecular and laboratory characterization of beta thalassemia in heterozygous and homozygous individuals and in those with coinheritance of S beta thalassemia. A total of 48 individuals were included (35 heterozygotes, 4 homozygotes and 9 S beta thalessemia carriers) referred to the Integrated Laboratory of Clinical Analyses of the Federal University of Rio Grande do Norte (UFRN) and the Hematology Ambulatory Facility of the Dalton Barbosa Cunha Hemocenter (Hemonorte Natal, Brazil). Peripheral blood samples form each patient underwent the following laboratory examinations: erythrogram, hemoglobin electrophoresis at alkaline pH, measurements of Hb A2, Fetal Hb and serum ferritin. DNA was extracted using the illustra blood genomicPrep Mini Spin Kit and molecular characterization was performed by the PCR/RFLP technique, which involves digestion with specific restriction enzymes for IVS-1 nt 1 (G®A), IVS-1 nt 6 (T®C) and codon 39 (CAG®TAG) mutations. Of the 35 heterozygotes, 37.1% showed IVS-1 nt 6 mutation, 42.9% IVS-1 nt 1 and 20% were carriers of other mutations not identified by the technique used. The four homozygous patients presented with the IVS-1 nt 6 mutation, while 66.7% of the individuals with S beta thalassemia had the IVS-1 nt 1 mutation. Codon 39 was not detected in any of the patients investigated. Of the thallasemic alleles found, 40.4% were IVS- 1 nt 1, 40.4% IVS-1 nt 6 and 19.2% were not identified. Laboratory data showed that the heterozygotes exhibited microcytosis and hypochromia, evidenced by MCV ranging from 57 to 75fL and MCH from 15.9 to 23.6 pg. Hemoglobin A2 varied between 3.7 and 7.2%. The homogygotes also showed reduced MCV and MCH and elevated HbA2.. Comparison of laboratory data between heterozygous individuals with IVS-1 nt 1 and IVS-1 nt 6 mutations showed that heterozygotes for the IVS1-1 mutation had significantly lower mean MCV and MCH (p = 0.023 and 0.007, respectively) and significantly higher hemoglobin A2 (p < 0.001) when compared to heterozygotes for the IVS-1 nt 6 mutation. PCR/RFLP was useful in identifying the presence or absence of IVS-1 nt 6, IVS-1 nt 1 and codon 39 mutations in most of the patients investigated here. This is the first study conducted in the state of Rio Grande do Norte, Brazil aimed at identifying beta thalassemia mutations and represents an important contribution to the knowledge regarding the molecular profile of beta thalassemia in our country
