Objective: To evaluate the influence of Everolimus (RAD001) on chemically induced urothelial lesions in mice and its influence on in vitro human bladder cancer cell lines. Methods: ICR male mice were given N-butyl-N-(4-hydroxybutyl) nitrosamine in drinking water for a period of 12 weeks. Subsequently, RAD001 was administered via oral gavage, for 6 weeks. At the end of the experiment, all the animals were sacrificed and tumor development was determined by means of histopathologic evaluation; mammalian target of rapamycin (mTOR) expressivity was evaluated by immunohistochemistry. Three human bladder cancer cell lines (T24, HT1376, and 5637) were treated using a range of RAD001 concentrations. MTT assay, terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL), and flow cytometry were used to assess cell proliferation, apoptosis index, and cell cycle analysis, respectively. Immunoblotting analysis of 3 cell line extracts using mTOR and Akt antibodies was performed in order to study the expression of Akt and mTOR proteins and their phosphorylated forms. Results: The incidence of urothelial lesions in animals treated with RAD001 was similar to those animals not treated. RAD001 did not block T24 and HT1376 cell proliferation or induce apoptosis. A reduction in cell proliferation rate and therefore G0/G1 phase arrest, as well as a statistically significant induction of apoptosis (P 0.001), was only observed in the 5637 cell line. Conclusion: RAD001 seems not to have a significant effect on chemically induced murine bladder tumors. The effect of RAD001 on tumor proliferation and apoptosis was achieved only in superficial derived bladder cancer cell line, no effect was observed in invasive cell lines.

Purpose/Objective: The purpose of this work was to determine biologically equivalent alternative regimens for the treatment of prostate cancer using External Beam Radiotherapy (EBRT) and Low Dose-Rate Brachytherapy (LDRBT) with 125I implants and to evaluate the sensitivity of these regimens to different sets of radiobiological parameters of the Linear-Quadratic (LQ) model.

Recentemente, várias aplicações da radioterapia estereotáxica têm sido identificadas, especialmente nos tumores do pulmão e fígado. A radioterapia estereotáxica tornou possível aplicar uma única irradiação com elevada dose sem causar morbilidades tardias, mesmo em órgãos sensíveis como o sistema nervoso central. Como tal, é possível extrapolar que a mesma fracção única com dose elevada possa ser realizada em tumores de pulmão desde que o PTV seja suficientemente pequeno e que o tratamento seja realizado a 3D.

Sendo a radioterapia (RT) uma modalidade terapêutica cada vez mais complexa e precisa em oncologia, torna-se pertinente refletir sobre alguns conceitos e procedimentos acerca da qualidade e do controlo da qualidade a ela associados, uma vez que se encontram profundamente aplicados nos centros de radioterapia já existentes. Estes conceitos e procedimentos estão bem sustentados em muitas experiências clinicas e em estudos acessíveis numa vasta série de artigos científicos publicados em revistas e outras fontes credíveis e científicas. A metodologia utilizada para a elaboração deste trabalho passa pela pesquisa e revisão de material bibliográfico existente não só em livros de referência como também em artigos de revistas da área da física médica, radioterapia e oncologia. A estrutura do trabalho passa pela descrição do “estado-da-arte” atual, sendo que, e nesta perspetiva, se tenha recorrido ao percurso evolutivo dos conceitos e procedimentos tidos ao longo do desenvolvimento das tecnologias e técnicas em radioterapia. O que se observa neste trabalho é que “Qualidade em Radioterapia” envolve uma complexa estrutura não só de recursos físicos como também humanos, onde uma pequena falha de comunicação no elo de ligação entre os diversos membros da equipa compromete afinal aquilo que é o seu objetivo major: tratar com qualidade e dignidade o doente oncológico.

Ionic Liquids (ILs) are ionic compounds that possess melting temperature below 100ºC and they have been a topic of great interest since the mid-1990s due to their unique properties. The range of IL uses has been broadened, due to a significant increase in the variety of physical, chemical and biological ILs properties. They are now used as Active Pharmaceutical Ingredients (APIs) and recent interests are focused on their application as innovative solutions in new medical treatment and delivery options.1 In this work, our principal objective was the synthesis and investigation of physicochemical and medical properties of ionic liquids (ILs) and organic salts from ampicillin. This approach is of huge interest in pharmaceutical industry as cation and anion composition of ILs and organic salts can greatly alter their desired properties, namely the melting temperature and even synergistic effects can be obtained.2,3 For the synthesis of these compounds we used a recently developed method proposed by Ohno et al.4 for the preparation of quaternary ammonium and phosphonium hydroxides, that were neutralized by ampicillin. After purification we obtained pure ILs and salts in good yields. These ILs shows good antimicrobial and antifungal activities. As it is well known that some ionic liquids containing phosphonium and ammonium cation also shows anti-cancer activity1,5 we also decided to study these compounds against some cancer cell lines.

A radioterapia (RT) desempenha um papel fundamental na terapêutica curativa ou paliativa do cancro, sendo aplicada no tratamento de 50 a 60% dos doentes diagnosticados. O seu objectivo é administrar uma dose prescrita de radiação ionizante a um volume alvo com um mínimo de dano para os tecidos sãos circundantes, preservando a sua função. Os desvios relativamente à prescrição poderão comprometer o objectivo do tratamento, levando a possíveis sobredosagens nos tecidos saudáveis e subdosagens no volume de tratamento. A maioria dos tratamentos ocorre sem incidentes e contribui para a qualidade de vida ou cura do doente. No entanto, a RT é reconhecida como um procedimento de alto risco devido à tecnologia e ambiente complexos, bem como ao elevado número de etapas e grupos profissionais envolvidos que trabalham em equipa para prescrever, planear, administrar e monitorizar a terapia. Apesar da baixa taxa de erro e das poucas ou nenhumas consequências clínicas para os doentes associadas, o verdadeiro risco reside na possibilidade de o erro não ser detectado, perpetuando-se no tempo ou afetando vários doentes. Os avanços na tecnologia que proporcionam técnicas mais sofisticadas e precisas representam também uma necessidade crescente de treino exaustivo, formação contínua e atenção por parte dos técnicos de RT que planeiam e administram a terapia. Estes integram uma equipa de especialistas, constituída por médicos, físicos e enfermeiros, que sob a supervisão do médico radioterapeuta se articulam de forma a assegurar um tratamento consistente, preciso e eficiente. Os técnicos de radioterapia trabalham com equipamentos e softwares cada vez mais sofisticados, monitorizam os tratamentos e contactam diariamente com o doente durante o seu tratamento, assumindo a autonomia e responsabilidade de interpretar todo o processo de tratamento e quando necessário articula-se com a restante equipa. A interação profunda entre funções automatizadas, equipamentos tecnologicamente avançados, decisões e atividades humanas que acompanham todo o processo da RT, aliados à pressão crescente para a aplicação das técnicas de tratamento mais avançadas a mais doentes e cumprimento de metas temporais, confere um potencial de risco à atividade do técnico de RT que é por vezes subestimado. O reconhecimento deste potencial passa pelo enquadramento da sua atividade no processo da RT e caracterização do seu papel no mesmo, revelando as suas competências e limitações.

The relentless discovery of cancer biomarkers demands improved methods for their detection. In this work, we developed protein imprinted polymer on three-dimensional gold nanoelectrode ensemble (GNEE) to detect epithelial ovarian cancer antigen-125 (CA 125), a protein biomarker associated with ovarian cancer. CA 125 is the standard tumor marker used to follow women during or after treatment for epithelial ovarian cancer. The template protein CA 125 was initially incorporated into the thin-film coating and, upon extraction of protein from the accessible surfaces on the thin film, imprints for CA 125 were formed. The fabrication and analysis of the CA 125 imprinted GNEE was done by using cyclic voltammetry (CV), differential pulse voltammetry (DPV) and electrochemical impedance spectroscopy (EIS) techniques. The surfaces of the very thin, protein imprinted sites on GNEE are utilized for immunospecific capture of CA 125 molecules, and the mass of bound on the electrode surface can be detected as a reduction in the faradic current from the redox marker. Under optimal conditions, the developed sensor showed good increments at the studied concentration range of 0.5–400 U mL−1. The lowest detection limit was found to be 0.5 U mL−1. Spiked human blood serum and unknown real serum samples were analyzed. The presence of non-specific proteins in the serum did not significantly affect the sensitivity of our assay. Molecular imprinting using synthetic polymers and nanomaterials provides an alternative approach to the trace detection of biomarker proteins.

Mucin-16 (MUC16) is the established ovarian cancer marker used to follow the disease during or after treatment for epithelial ovarian cancer. The emerging science of cancer markers also demands for the new sensitive detection methods. In this work, we have developed an electrochemical immunosensor for antigen MUC16 using gold nanoelectrode ensemble (GNEE) and ferrocene carboxylic acid encapsulated liposomes tethered with monoclonal anti-Mucin-16 antibodies ( MUC16). GNEEs were fabricated by electroless deposition of the gold within the pores of polycarbonate track-etched membranes. Afterwards, MUC16 were immobilized on preformed self-assembled monolayer of cysteamine on the GNEE via cross-linking with EDC-Sulfo-NHS. A sandwich immunoassay was performed on MUC16 functionalized GNEE with MUC16 and immunoliposomes. The differential pulse voltammetry was employed to quantify the faradic redox response of ferrocene carboxylic acid released from immunoliposomes. The dose–response curve for MUC16 concentration was found between the range of 0.001–300 U mL−1. The lowest detection limit was found to be 5 × 10−4 U mL−1 (S/N = 3). We evaluated the performance of this developed immunosensor with commercial ELISA assay by comparing results obtained from spiked serum samples and real blood serum samples from volunteers.

OBJECTIVE: To analyze the association between dietary patterns and oral cancer. METHODS: The study, part of a Latin American multicenter hospital-based case-control study, was conducted in São Paulo, Southeastern Brazil, between November 1998 and March 2002 and included 366 incident cases of oral cancer and 469 controls, frequency-matched with cases by sex and age. Dietary data were collected using a food frequency questionnaire. The risk associated with the intake of food groups defined a posteriori, through factor analysis (called factors), was assessed. The first factor, labeled "prudent," was characterized by the intake of vegetables, fruit, cheese, and poultry. The second factor, "traditional," consisted of the intake of rice, pasta, pulses, and meat. The third factor, "snacks," was characterized as the intake of bread, butter, salami, cheese, cakes, and desserts. The fourth, "monotonous," was inversely associated with the intake of fruit, vegetables and most other food items. Factor scores for each component retained were calculated for cases and controls. After categorization of factor scores into tertiles according to the distribution of controls, odds ratios and 95% confidence intervals were calculated using unconditional multiple logistic regression. RESULTS: "Traditional" factor showed an inverse association with cancer (OR=0.51; 95% CI: 0.32; 0.81, p-value for trend 0.14), whereas "monotonous" was positively associated with the outcome (OR=1.78; 95% CI: 1.78; 2.85, p-value for trend <0.001). CONCLUSIONS: The study data suggest that the traditional Brazilian diet, consisting of rice and beans plus moderate amounts of meat, may confer protection against oral cancer, independently of any other risk factors such as alcohol intake and smoking.
