The viability and interpretation of techniques for the evaluation of immunocompetence of animals in their natural environment has been largely debated. One of these methods is based on testing the antimicrobial capacity of the blood and/or plasma in vitro, which could rapidly and effectively assess the immunological conditions of natural populations. We tested the applicability of the antimicrobial capacity of plasma (ACP) assay in anuran amphibians from the Atlantic Forest. The assay was performed by measuring both the turbidity (in a spectrophotometer) and the colony forming units (CFU) of the remaining bacteria (Escherichia coli) following exposure to amphibian plasma. Although both assays were correlated, the ACP assay by spectrophotometry showed 10 times lower intra-assay variation. We also found interspecific variation in ACP, as well as the maintenance of ACP values in males from the same population, collected in different breeding seasons. Thus, the estimation of ACP by spectrophotometry provides a convenient and accurate method for evaluating innate immunocompetence in comparative and ecophysiological studies of anuran amphibians.

Hemiklonale Vererbung im hybridogenetische Rana grafi-Komplex (Anura: Ranidae)Im Rahmen der vorliegenden Studie wurde eine großflächige Untersuchung an südfranzösischen Wasserfröschen durchgeführt. Es wurden 31 Populationen mit 918 Tieren beprobt und mit Referenzproben genetisch verglichen. Die Genotypen der Tiere wurden mittels Allozymelektrophorese an sieben diagnostischen Loci bestimmt. Für Teilproben wurde das Alter, die Fläche der Erythrocyten und der DNA-Gehalt bestimmt. Die wichtigsten Ergebnisse der unterschiedlichen methodischen Ansätze lassen sich wie folgt zusammenfassen:(1) Es wurden neben einem großen Anteil von R. grafi (452 Tiere), R. perezi (200 Tiere) und R. ridibunda (254 Tiere) auch ein R. esculenta und zwei R. lessonae in der Camargue nachgewiesen.(2) Die Geschlechterverhältnisse der einzelnen Taxa waren stark gestört. Dies gilt besonders für den Hybriden R. grafi mit einem Männchenanteil von nur 7,3%. Das Geschlechterverhältnis von R. ridibunda ist besonders in PGR-Population zu Gunsten von Weibchen verschoben. Eine Modellierung von Populationen zeigt, daß wahrscheinlich R. grafi-Männchen durch eine gestörte Gametogenese für die gestörten Geschlechtsverhältnisse seiner Parentalformen verantwortlich ist.(3) Die genetische Analyse zeigt eine unerwartet hohe genetische Variabilität von R. ridibunda im Vergleich zu R. perezi und eine geographische Varianz der genetischen Variabilität in allen untersuchten Taxa. Die genetische Variabilität zeigte bei allen Wasserfroschformen in der Camargue ein Maximum.(4) Die hohe genetische Variabilität geht hauptsächlich, neben Einführung von verschiedenen genetischen R. ridibunda-Linien, auf Introgression zurück.

Species richness varies greatly across geographical regions. Eastern Arc Mountains (EAM) of Kenya and Tanzania is one of the global biodiversity hotspots. Despite this, high species diversity the explanatory factors have remained largely unexplored. Herein, this study first investigated amphibian species richness patterns in the EAM and particularly the reasons for the low richness in Taita Hills. It tested the hypothesis that the low richness is due to past forest loss or other factors. The results demonstrated that the regional species richness pattern was influenced largely by mean annual rainfall and not forest area. Secondly, using the 26 currently recorded amphibians in the Taita Hills, it investigated the relationship between amphibian species composition along anthropogenic habitat disturbance and elevation gradients. It tested the hypothesis that sites with similar environmental characteristics (temperature, rainfall and elevation), in close proximity and with similar disturbance levels (habitat types) harbour similar species composition. It was found that amphibian species composition differed in terms of elevation and was explained by both temperature and rainfall. Therefore sites with similar environmental characteristics, disturbance levels and in close proximity geographically have similar amphibian composition. Thirdly, diagnostic characters, distribution, basic life history characteristics and conservation status of all currently known amphibians in the Taita Hills were provided. Finally, first long term life history and ecological characteristics of a brevicipitid frog (Callulina sp) was provided. The results showed that this frog abundance and distribution is influenced mainly by mean monthly temperature, breeds during the long dry season and exhibit parental care. Results of this study strongly recommend increasing indigenous forest cover in order to enhance the conservation of the endemic indigenous forest associated amphibians such as Callulina sp, Boulengerula taitana and Boulengerula niedeni.

Many global amphibian declines have been linked to the fungal pathogen Batrachochytrium dendrobatidis (Bd). The knowledge on Bd distribution provides a fundamental basis for amphibian conservation planning. Yet, such Bd distribution information is currently insufficient, in particular at a regional scale. The college classroom provides an excellent opportunity to expand the knowledge of Bd distribution. Here we provide an example of such research projects to detect Bd prevalence among local amphibians in a college course setting and present the results of work conducted in central Pennsylvania, USA. We collected toe clips and conducted PCR assays of six species, Plethodon cinereus, Desmognathus fuscus, Notophthalmus viridescens, Lithobates catesbeianus, L. clamitans, and L. sylvaticus (59 individuals). Four groups of students independently conducted entire projects, orally presented their findings, and submitted manuscripts to the professor at the end of the semester. This example demonstrates that it is feasible for an undergraduate class to complete a Bd-detection project within a single semester. Such a project not only contributes to Bd research but also promotes conservation education among students through hands-on research experiences. We found Bd infection in only one sample of N. viridescens, but no sign of infection in the rest of the samples. As a relatively high prevalence of Bd has been reported in surrounding areas, our results suggest spatial heterogeneity in Bd occurrence at a regional scale and thus, the need for continued efforts to monitor Bd prevalence.

The primary goal of this project was to launch a pilot population study in the spring-fed wetland area southwest of Montana Tech to establish baseline data on density, distribution, abundance, and diversity of amphibians in the area. The current confirmed species at the select site is the Columbia spotted frog (Rana luteiventris). Based on preliminary habitat assessment and existing literature, other species possibilities included the long-toed salamander (Ambrystoma macrodactylum) boreal chorus frog (Pseudacris maculata), the Rocky Mountain tailed frog (Ascaphus montanus), and the leopard frog (Rana pipiens), (Werner et al. 2004) though the latter species is considered unlikely based on the specie’s declining status (Werner 2003; Werner et al. 2004). The project’s secondary goal was to collect basic habitat and environmental data: vegetation, precipitation, temperature. The third goal was to explore correlations between species prevalence and environmental data to expand the scientific understanding of population dynamics in the field of amphibian studies (see: Ferner, 2007; Dodd, 2010).

Roads and highways present a unique challenge to wildlife as they exhibit substantial impacts on the surrounding ecosystem through the interruption of a number of ecological processes. With new roads added to the national highway system every year, an understanding of these impacts is required for effective mitigation of potential environmental impacts. A major contributor to these negative effects is the deposition of chemicals used in winter deicing activities to nearby surface waters. These chemicals often vary in composition and may affect freshwater species differently. The negative impacts of widespread deposition of sodium chloride (NaCl) have prompted a search for an `environmentally friendly' alternative. However, little research has investigated the potential environmental effects of widespread use of these alternatives. Herein, I detail the results of laboratory tests and field surveys designed to determine the impacts of road salt (NaCl) and other chemical deicers on amphibian communities in Michigan's Upper Peninsula. Using larval amphibians I demonstrate the lethal impacts of a suite of chemical deicers on this sensitive, freshwater species. Larval wood frogs (Lithobates sylvatica) were tolerant of short-term (96 hours) exposure to urea (CH4N2O), sodium chloride (NaCl), and magnesium chloride (MgCl2). However, these larvae were very sensitive to acetate products (C8H12CaMgO8, CH3COOK) and calcium chloride (CaCl2). These differences in tolerance suggest that certain deicers may be more harmful to amphibians than others. Secondly, I expanded this analysis to include an experiment designed to determine the sublethal effects of chronic exposure to environmentally realistic concentrations of NaCl on two unique amphibian species, L. sylvatica and green frogs (L. clamitans). L. sylvatica tend to breed in small, ephemeral wetlands and metamorphose within a single season. However, L. clamitans breed primarily in more permanent wetlands and often remain as tadpoles for one year or more. These species employ different life history strategies in this region which may influence their response to chronic NaCl exposure. Both species demonstrated potentially harmful effects on individual fitness. L. sylvatica larvae had a high incidence of edema suggesting the NaCl exposure was a significant physiologic stressor to these larvae. L. clamitans larvae reduced tail length during their exposure which may affect adult fitness of these individuals. In order to determine the risk local amphibians face when using these roadside pools, I conducted a survey of the spatial distribution of chloride in the three northernmost counties of Michigan. This area receives a relatively low amount of NaCl which is confined to state and federal highways. The chloride concentrations in this region were much lower than those in urban systems; however, amphibians breeding in the local area may encounter harmful chloride levels arising from temporal variations in hydroperiods. Spatial variation of chloride levels suggests the road-effect zone for amphibians may be as large as 1000 m from a salt-treated highway. Lastly, I performed an analysis of the use of specific conductance to predict chloride concentrations in natural surface water bodies. A number of studies have used this regression to predict chloride concentrations from measurements of specific conductance. This method is often chosen in the place of ion chromatography due to budget and time constraints. However, using a regression method to characterize this relationship does not result in accurate chloride ion concentration estimates.

In order to maintain pond-breeding amphibian species richness, it is important to understand how both natural and anthropogenic disturbances affect species assemblages and individual species distributions both at the scale of individual ponds and at a larger landscape scale. The goal of this project was to investigate what characteristics of ponds and the surrounding wetland landscape were most effective in predicting pond-breeding species richness and the individual occurrence of wood frog (Rana sylvatica), bullfrog (Rana catesbeiana) and pickerel frog (Rana palustris) breeding sites in a beaver-modified landscape and how this landscape has changed over time. The wetland landscape of Acadia National Park was historically modified by the natural disturbance cycles of beaver (Castor cazadensis), and since their reintroduction to the island in 1921, beaver have played a large role in creating and maintaining palustrine wetlands. In 2000 and 2001, I studied pond-breeding amphibian assemblages at 71 palustrine wetlands in Acadia National Park, Mount Desert Island, Maine. I determined breeding presence of 7 amphibian species and quantified 15 variables describing local pond conditions and characteristics of the wetland landscape. I developed a priori models to predict sites with high amphibian species and used model selection with Akaike's Information Criterion (AIC) to identify important variables. Single species models were also developed to predict wood frog, bullfrog and pickerel frogs breeding presence. The variables for wetland connectivity by stream corridors and the presence of beaver disturbance were the most effective variables to predict sites with high amphibian richness. Wood frog breeding was best predicted by local scale variables describing temporary, fishless wetlands and the absence of active beaver disturbance. Abandoned beaver sites provided wood frog breeding habitat (70%) in a similar proportion to that found in non beaver-influenced sites (79%). In contrast, bullfrog breeding presence was limited to active beaver wetlands with fish and permanent water, and 80% of breeding sites were large (≥2ha in size). Pickerel frog breeding site selection was predicted best by the connectivity of sites in the landscape by stream corridors. Models including the presence of beaver disturbance, greater wetland perimeter and greater depth were included in the confidence set of pickerel frog models but showed considerably less support. Analysis of historic aerial photographs showed an 89% increase in the total number of ponded wetlands available in the landscape between the years of 1944 and 1997. Beaver colonization generally converted forested wetlands and riparian areas to open water and emergent wetlands. Temporal colonization of beaver wetlands favored large sites low in the watersheds and sites that were impounded later were generally smaller, higher in the watershed, and more likely to be abandoned. These results suggest that beaver have not only increased the number of available breeding sites in the landscape for pond-breeding amphibians, but the resulting mosaic of active and abandoned beaver wetlands also provides suitable breeding habitat for species with differing habitat requirements.

This report summarizes the results of a herpetofauna (reptile and amphibian) survey at Naval Station Rota (NAVSTA Rota), Spain conducted during June and October 2008, March 2009 and July 2010. The main objective of this investigation was to develop an inventory of herpetofauna, including their base-wide distribution and habitat use. Data from these surveys was used to supplement information in the 2010 Cultural/Natural Resources Management User's Guide and can also be used for environmental planning, natural resource management and conservation. Prior to this survey, only cursory field work had been conducted on the station for herpetofauna with the exception of the common chameleon (Chamaleo chamaeleon). A comprehensive population count and habitat assessment for the common chameleon was conducted in September 2001.

N-methyl-d-aspartate receptor (NMDAR) activation has been implicated in forms of synaptic plasticity involving long-term changes in neuronal structure, function, or protein expression. Transcriptional alterations have been correlated with NMDAR-mediated synaptic plasticity, but the problem of rapidly targeting new proteins to particular synapses is unsolved. One potential solution is synapse-specific protein translation, which is suggested by dendritic localization of numerous transcripts and subsynaptic polyribosomes. We report here a mechanism by which NMDAR activation at synapses may control this protein synthetic machinery. In intact tadpole tecta, NMDAR activation leads to phosphorylation of a subset of proteins, one of which we now identify as the eukaryotic translation elongation factor 2 (eEF2). Phosphorylation of eEF2 halts protein synthesis and may prepare cells to translate a new set of mRNAs. We show that NMDAR activation-induced eEF2 phosphorylation is widespread in tadpole tecta. In contrast, in adult tecta, where synaptic plasticity is reduced, this phosphorylation is restricted to short dendritic regions that process binocular information. Biochemical and anatomical evidence shows that this NMDAR activation-induced eEF2 phosphorylation is localized to subsynaptic sites. Moreover, eEF2 phosphorylation is induced by visual stimulation, and NMDAR blockade before stimulation eliminates this effect. Thus, NMDAR activation, which is known to mediate synaptic changes in the developing frog, could produce local postsynaptic alterations in protein synthesis by inducing eEF2 phosphorylation.

Amphibian metamorphosis is marked by dramatic, thyroid hormone (TH)-induced changes involving gene regulation by TH receptor (TR). It has been postulated that TR-mediated gene regulation involves chromatin remodeling. In the absence of ligand, TR can repress gene expression by recruiting a histone deacetylase complex, whereas liganded TR recruits a histone acetylase complex for gene activation. Earlier studies have led us to propose a dual function model for TR during development. In premetamorphic tadpoles, unliganded TR represses transcription involving histone deacetylation. During metamorphosis, endogenous TH allows TR to activate gene expression through histone acetylation. Here using chromatin immunoprecipitation assay, we directly demonstrate TR binding to TH response genes constitutively in vivo in premetamorphic tadpoles. We further show that TH treatment leads to histone deacetylase release from TH response gene promoters. Interestingly, in whole animals, changes in histone acetylation show little correlation with the expression of TH response genes. On the other hand, in the intestine and tail, where TH response genes are known to be up-regulated more dramatically by TH than in most other organs, we demonstrate that TH treatment induces gene activation and histone H4 acetylation. These data argue for a role of histone acetylation in transcriptional regulation by TRs during amphibian development in some tissues, whereas in others changes in histone acetylation levels may play no or only a minor role, supporting the existence of important alternative mechanisms in gene regulation by TR.

There has been a great deal of recent attention on the suspected increase in amphibian deformities. However, most reports of amphibian deformities have been anecdotal, and no experiments in the field under natural conditions have been performed to investigate this phenomenon. Under laboratory conditions, a variety of agents can induce deformities in amphibians. We investigated one of these agents, UV-B radiation, in field experiments, as a cause for amphibian deformities. We monitored hatching success and development in long-toed salamanders under UV-B shields and in regimes that allowed UV-B radiation. Embryos under UV-B shields had a significantly higher hatching rate and fewer deformities, and developed more quickly than those exposed to UV-B. Deformities may contribute directly to embryo mortality, and they may affect an individual’s subsequent survival after hatching.

The human immunodeficiency virus type 1 (HIV-1) Rev protein is required for nuclear export of late HIV-1 mRNAs. This function is dependent on the mutationally defined Rev activation domain, which also forms a potent nuclear export signal. Transcription factor IIIA (TFIIIA) binds to 5S rRNA transcripts and this interaction has been proposed to play a role in the efficient nuclear export of 5S rRNA in amphibian oocytes. Here it is reported that amphibian TFIIIA proteins contain a sequence element with homology to the Rev activation domain that effectively substitutes for this domain in inducing the nuclear export of late HIV-1 mRNAs. It is further demonstrated that this TFIIIA sequence element functions as a protein nuclear export signal in both human cells and frog oocytes. Thus, this shared protein motif may play an analogous role in mediating the nuclear export of both late HIV-1 RNAs and 5S rRNA transcripts.
