Der demographische Wandel mit seinen Auswirkungen und damit verbundenen Befürchtungen in Bezug auf das soziale Sicherungssystem und das Wirtschaftswachstum in der Bundesrepublik Deutschland ist in den Medien in der Vergangenheit häufig vertreten gewesen und hat auch gegenwärtig nicht an Aktualität verloren. Im Rahmen dieser Bachelorarbeit steht allerdings der Wohnungsmarkt im Fokus der Betrachtung, eben unter dem Aspekt der Demographie. Dabei ist das Erkennen von Tendenzen hinsichtlich der künftigen demographischen Entwicklung und deren Ausprägungen Voraussetzung um zu klären, ob eine Notwendigkeit für den Wohnungsmarkt tatsächlich gegeben ist. Es wird hierbei auf den Wohnungsmarkt der Stadt Leipzig Bezug genommen. Angebot und Nachfrage werden gegenübergestellt und mit dem Wissen des demographischen Wandels die zukünftige Entwicklung der Nachfrage abgeschätzt. Daraus ergibt sich die Möglichkeit, Defizite auf dem Wohnungsmarkt aufzudecken. Es hat sich dabei herausgestellt, dass das Seniorenwohnen bzw. barrierefreie Wohnen unzureichend auf dem Wohnungsmarkt der Stadt Leipzig angeboten wird. Daher verdient diese Nachfragegruppe im letzten Teil der Arbeit eine genauere Betrachtung.

Com a finalidade de determinar as doses de nitrogênio, fósforo e potássio mais adequadas para obtenção de plantas aptas para enxertia em viveiro de seringueira, instalou-se um experimento em Latossolo Amarelo textura média na Ilha do Mosqueiro-PA. O delineamento experimental foi de blocos ao acaso com duas repetições obedecendo ao arranjo fatorial 33 Foram utilizadas as doses de 0-2,1-4,2 g/planta de N; 0-3,5-7,0 g/planta de P2O5; 0-1,4-2,8 g/planta de K2O e dose constante de 0,8 g/planta de MgO, empregando-se como fontes, respectivamente, sulfato de amônio, superfosfato triplo, cloreto de potássio e sulfato de magnésio. Os resultados foram obtidos duzentos e vinte dias após a instalação do experimento, sendo realizadas as seguintes avaliações: análises químicas do solo e folhas, altura das plantas, diâmetro do caule, peso da matéria seca da parte aérea e plantas aptas para a enxertia. Pelos resultados obtidos chega-se à conclusão que as doses mais adequadas foram 330 kg/ha (4,6 g/p) de N, 340 kg/ha (4,8 g/p) de P2O5 e 190 kg/ha (2,7 g/p) de K2O, aliadas a dose constante de 60 kg/ha (0,8 g/p) de MgO, propiciando um índice de aproveitamento de oitenta e sete porcento de plantas aptas para enxertia.

De uma plantação de beterraba (Beta vulgaris var. Early wanter) situada em Latossolo Vermelho Orto, série "Luiz de Queiroz" de ótima fertilidade natural, foram coletadas plantas aos 40 , 60 e 80 dias após a semeadura. Determina-se o peso da matéria seca do material previamente dividido em parte aérea e raízes. As partes foram analisadas para N, P, K, Ca, Mg, Cu, Fe, Mn e Zn. Os autores observaram que a beterraba aumenta o seu peso de matéria seca atpe ao final do ciclo aos 80 dias de idade. Constataram uma concentração elevada dos nutrientes tanto em parte aérea como nas raízes. Uma população teórica de 330.000 plantas hectare, extrai: 77,88 kg de N; 17,77 kg de P; 202,85 kg de K; 19,59 kg de Ca; 29,1 kg de Mg; 22,9 g de Cu; 735,9 g de Fe; 583,6 g de Mn e 387,6 g de Zn.

Rigorous organization and quality control (QC) are necessary to facilitate successful genome-wide association meta-analyses (GWAMAs) of statistics aggregated across multiple genome-wide association studies. This protocol provides guidelines for (i) organizational aspects of GWAMAs, and for (ii) QC at the study file level, the meta-level across studies and the meta-analysis output level. Real-world examples highlight issues experienced and solutions developed by the GIANT Consortium that has conducted meta-analyses including data from 125 studies comprising more than 330,000 individuals. We provide a general protocol for conducting GWAMAs and carrying out QC to minimize errors and to guarantee maximum use of the data. We also include details for the use of a powerful and flexible software package called EasyQC. Precise timings will be greatly influenced by consortium size. For consortia of comparable size to the GIANT Consortium, this protocol takes a minimum of about 10 months to complete.

Les maladies chroniques représentent un fardeau important pour la société, en termes de morbidité, dépendance, qualité de vie, mortalité et coûts de la santé. La prise en charge des maladies chroniques constitue ainsi une priorité des systèmes de santé. Tandis que des programmes de prévention et prise en charge des maladies chroniques («prevention and chronic disease management») ont été mis sur pieds depuis plus d'une décennie d'abord en Amérique du Nord puis en Europe, leur développement en Suisse est récent. Ces programmes sont explicitement structurés et organisés, centrés sur les patients et comportent constamment un élément d'éducation thérapeutique. De plus, ils impliquent un travail en équipe ainsi que des prises en charge stratifiées en fonction de la sévérité de la maladie et des besoins des patients, et basées sur les preuves de leur efficacité. Il est important que les médecins et autres professionnels de la santé, ainsi que tous les autres acteurs du système sanitaire reconnaissent la nécessité de mettre en place des modalités adéquates et efficaces de prise en charge des patients présentant des maladies chroniques, participent à leur développement et les soutiennent.

We report the case of a young break-dancer presenting with hammer syndrome. This syndrome has been correlated with many professional and recreational activities but this is, to our knowledge, the first description of hammer syndrome caused by break-dancing. The etiology, diagnosis and treatment modalities of this rare syndrome are considered.

A structural and functional analysis of the 5'-end region of the Xenopus laevis vitellogenin gene A1 revealed two transcription initiation sites located 1.8 kilobases apart. A RNA polymerase II binding assay indicates that both promoters form initiation complexes efficiently. In vitro, using a transcription assay derived from a HeLa whole-cell extract, the upstream promoter is more than 10-fold stronger than the downstream one. In contrast, both promoters have a similar strength in a HeLa nuclear extract. In vivo, that is in estrogen-stimulated hepatocytes, it is the downstream promoter homologous to the one used by the other members of the vitellogenin gene family, which is 50-fold stronger than the upstream promoter. Thus, if functional vitellogenin mRNA results from this latter activity, it would contribute less than 1% to the synthesis of vitellogenin by fully induced Xenopus hepatocytes expressing the four vitellogenin genes. In contrast, both gene A1 promoters are silent in uninduced hepatocytes. Transfection experiments using the Xenopus cell line B3.2 in which estrogen-responsiveness has been introduced reveal that the strong downstream promoter is controlled by an estrogen responsive element (ERE) located 330 bp upstream of it. The upstream promoter can also be controlled by the same ERE. Since the region comprising the upstream promoter is flanked by a 200 base pair long inverted repeat with stretches of homology to other regions of the X. laevis genome, we speculate that it might have been inserted upstream of the vitellogenin gene A1 by a recombination event and consequently brought under control of the ERE lying 1.5 kilobases downstream.

The Na,K-ATPase is a major ion-motive ATPase of the P-type family responsible for many aspects of cellular homeostasis. To determine the structure of the pathway for cations across the transmembrane portion of the Na,K-ATPase, we mutated 24 residues of the fourth transmembrane segment into cysteine and studied their function and accessibility by exposure to the sulfhydryl reagent 2-aminoethyl-methanethiosulfonate. Accessibility was also examined after treatment with palytoxin, which transforms the Na,K-pump into a cation channel. Of the 24 tested cysteine mutants, seven had no or a much reduced transport function. In particular cysteine mutants of the highly conserved "PEG" motif had a strongly reduced activity. However, most of the non-functional mutants could still be transformed by palytoxin as well as all of the functional mutants. Accessibility, determined as a 2-aminoethyl-methanethiosulfonate-induced reduction of the transport activity or as inhibition of the membrane conductance after palytoxin treatment, was observed for the following positions: Phe(323), Ile(322), Gly(326), Ala(330), Pro(333), Glu(334), and Gly(335). In accordance with a structural model of the Na,K-ATPase obtained by homology modeling with the two published structures of sarcoplasmic and endoplasmic reticulum calcium ATPase (Protein Data Bank codes 1EUL and 1IWO), the results suggest the presence of a cation pathway along the side of the fourth transmembrane segment that faces the space between transmembrane segments 5 and 6. The phenylalanine residue in position 323 has a critical position at the outer mouth of the cation pathway. The residues thought to form the cation binding site II ((333)PEGL) are also part of the accessible wall of the cation pathway opened by palytoxin through the Na,K-pump.

El proyecto desarrollado tiene como finalidad el diseño de una planta de producción de ácido láctico (88% en peso) con un montante anual de 3300 toneladas en 330 días de producción en continuo (incluyendo los períodos de parada de planta y mantenimiento) a partir de la vía sintética. Una vez analizadas todas las características del proceso, así como cada uno de los equipos que formarán parte de éste, el principal objetivo es la viabilidad económica de su implantación. Para ello se debe cerciorar que todo esté enmarcado dentro de la normativa urbanística (tanto a nivel local, como autonómico o estatal), la legislación medioambiental y las medidas de seguridad y contraincendios de la planta.

Background: Mucosal healing in ulcerative colitis (UC) is reported to be associated with favourable clinical outcomes such as reduced hospitalization and surgery rates. Activity monitoring by endoscopy has its shortcomings due to invasiveness, costs, and potential patient discomfort. Data on the correlation of noninvasive biomarkers with endoscopic severity in UC are scarce. Aim: to evaluate the correlation between endoscopic activity according to the modified Baron Index and fecal calprotectin, C-reactive protein (CRP), blood leukocytes, and the Lichtiger Index (clinical score). Methods: UC patients with leftsided and extensive colitis undergoing complete colonoscopy were prospectively enrolled and scored clinically and endoscopically. Fecal and blood samples were analyzed in UC patients (in a blinded fashion) and controls. The modified Baron score describes the following 5 endoscopic conditions: 0 = normal, 1 = granular mucosa, edema, 2 = friable mucosa but no spontaneous bleeding, 3 = microulcerations with spontaneous bleeding, 4 = gross ulceration, denuded mucosa. Results: We enrolled 228 UC patients (mean age 41 ± 13 years, 39 female) and 52 healthy controls. Disease was located in 40% in the left colon, 21% had an extensive and 39% a pancolitis. Endoscopic disease activity correlated best with fecal calprotectin (Spearman's rank correlation coefficient r = 0.821), followed by the Lichtiger Index (r = 0.682), CRP (r = 0.556), and blood leukocytes (r = 0.401). Fecal calprotectin was the only marker that could discriminate between different grades of endoscopic activity (grade 0, 25 ± 11 μg/g; grade 1, 44 ± 34 μg/g; grade 2, 111 ± 74 μg/g; grade 3, 330 ± 332 μg/g; grade 4, 659 ± 319 μg/g; P = 0.002 for discriminating grade 0 vs. 1, and P < 0.001 for discriminating grade 1 vs. 2, grade 2 vs. 3, and grade 3 vs. 4). Fecal calprotectin had the highest overall accuracy (91%) to detect endoscopically active disease (modified Baron Index ≥ 2), followed by the Lichtiger Index score of ≥ 4 (77%), CRP > 5 mg/L (69%) and blood leukocytosis (58%). Conclusions: Fecal calprotectin better correlated with endoscopic disease activity than clinical activity, CRP, and blood leukocytes. The strong correlation with endoscopic disease activity suggests that FC represents a useful biomarker for noninvasive monitoring of disease activity in UC patients.

Given the suspected role of mycobacteria in the establishment of disorders with an autoimmune background and joint damage, a study was conducted to analize whether rheumatic symptoms were likely to be present in tuberculosis (TB) patients. To this end, 330 patients with a bacteriologic confirmation of tuberculosis were investigated for the presence of arthritic complaints. The latter were recorded in five of them with rheumatic symptoms mostly involving interphalangeal and metacarpophalanged joints, and preceding the clinical manifestations of the TB illness. Three out of these five patients remained arthritic by the time of the bacteriologic conversion and fulfilled the criteria for the diagnosis of rheumatoid arthritis. In the two remaining patients sputum negativization was accompanied by a disappearance of rheumatic manifestations. These patients were also assessed for their peripheral levels of major T cell subsets as well as for the presence of autoantibodies. Comparisons with a series of non-arthritic TB cases, rheumatoid arthritis patients, and controls revealed that presence of rheumatic manifestations was associated with a different profile of autoantibody formation and T cell subset changes. Evidence recorded in the present study indicates that joint affectation in TB is a rare event, being rather the exception than the rule.
