Hypo-osmolality influences tissue metabolism, but research on protein turnover in skeletal muscle is limited. The purpose of this investigation was to examine the effects of hypo-osmotic stress on protein turnover in rat skeletal muscle. We hypothesized increased protein synthesis and reduced degradation following hypo-osmotic exposure. EDL muscles (n=8/group) were incubated in iso-osmotic (290 Osm/kg) or hypo-osmotic (190 Osm/kg) modified medium 199 (95% O2, 5% CO2, pH 7.4, 30±2 °C) for 60 min, followed by 75 min incubations with L-U[14C]phenylalanine or cycloheximide to determine protein synthesis and degradation. Immunoblotting was performed to assess signalling pathways involved. Phenylalanine uptake and incorporation were increased by 199% and 169% respectively in HYPO from ISO (p < 0.05). This was supported by elevated phosphorylation of mTOR Ser2448 (+12.5%) and increased Thr389 phosphorylation on p70s6 kinase (+23.6%) (p < 0.05). Hypo-osmotic stress increased protein synthesis and potentially amino acid uptake. Future studies should examine the upstream mechanisms involved.

The purpose of this study was to examine the effect of hyper-osmotic stress on protein turnover in skeletal muscle tissue using an established in-vitro model. Rat EDL muscles were incubated in either hyper-osmotic (400 ± 10 Osm) or isoosmotic (290 ± 10 Osm) custom-modified media (Gibco). L-[14C]-U-phenylalanine (n=8) and cycloheximide (n=8) were used to quantify protein synthesis and degradation, respectively. Western blotting analyses was performed to determine the activation of protein synthesis and degradation pathways. During hyperosmotic stress, protein degradation increased (p<0.05), while protein synthesis was decreased (p<0.05) as compared to the iso-osmotic condition. The decline in protein synthesis was accompanied by a decrease (p<0.05) in p70s6 kinase phosphorylation, while the increase in protein degradation was associated with an increase (p<0.05) in autolyzed calpain. Therefore, hyper-osmotic extracellular stress results in an intracellular catabolic environment in mammalian skeletal muscle tissue.

La plupart des conditions détectées par le dépistage néonatal sont reliées à l'une des enzymes qui dégradent les acyls-CoA mitochondriaux. Le rôle physiopathologique des acyls-CoA dans ces maladies est peu connue, en partie parce que les esters liés au CoA sont intracellulaires et les échantillons tissulaires de patients humains ne sont généralement pas disponibles. Nous avons créé une modèle animal murin de l'une de ces maladies, la déficience en 3-hydroxy-3-methylglutaryl-CoA lyase (HL), dans le foie (souris HLLKO). HL est la dernière enzyme de la cétogenèse et de la dégradation de la leucine. Une déficience chronique en HL et les crises métaboliques aigües, produisent chacune un portrait anormal et distinct d'acyls-CoA hépatiques. Ces profils ne sont pas prévisibles à partir des niveaux d'acides organiques urinaires et d'acylcarnitines plasmatiques. La cétogenèse est indétectable dans les hépatocytes HLLKO. Dans les mitochondries HLLKO isolées, le dégagement de 14CO2 à partir du [2-14C]pyruvate a diminué en présence de 2-ketoisocaproate (KIC), un métabolite de la leucine. Au test de tolérance au pyruvate, une mesure de la gluconéogenèse, les souris HLLKO ne présentent pas la réponse hyperglycémique normale. L'hyperammoniémie et l'hypoglycémie, des signes classiques de plusieurs erreurs innées du métabolisme (EIM) des acyls-CoA, surviennent de façon spontanée chez des souris HLLKO et sont inductibles par l'administration de KIC. Une charge en KIC augmente le niveau d'acyls-CoA reliés à la leucine et diminue le niveau d'acétyl-CoA. Les mitochondries des hépatocytes des souris HLLKO traitées avec KIC présentent un gonflement marqué. L'hyperammoniémie des souris HLLKO répond au traitement par l'acide N-carbamyl-L-glutamique. Ce composé permet de contourner une enzyme acétyl-CoA-dépendante essentielle pour l’uréogenèse, le N-acétylglutamate synthase. Ceci démontre un mécanisme d’hyperammoniémie lié aux acyls-CoA. Dans une deuxième EIM des acyls-CoA, la souris SCADD, déficiente en déshydrogénase des acyls-CoA à chaînes courtes. Le profil des acyls-CoA hépatiques montre un niveau élevé du butyryl-CoA particulièrement après un jeûne et après une charge en triglycérides à chaîne moyenne précurseurs du butyryl-CoA.

This thesis deals with the results of investigations on primary production and related aspects conducted in the Indian seas since 1957 and includes the regional and seasonal variations in the rate of production factors controlling the same and the magnitude of potential fishery resources derived from it. Data collected for various periods using oxygen and 14c techniques from the Gulf of Mannar, palk bay, the south west coast of India including laccadive sea together with other available data form the basis of these studies.

All over the world, several Quaternary proxy data have been used to reconstruct past sea levels, mainly radiocarbon or OSL dating of exposures of marine facies or shore line indicators (e.g. Carr et al., 2010) as well as paleoenvironmental indicators in lagoon or estuary sediments (e.g. Baxter and Meadows, 1999). Estuaries and deltas develop at river mouths during transgressive and regressive phases, respectively (Boyd et al., 1992). In particular, the postglacial Holocene sea-level rise has contributed importantly to the estuary-to-delta transition (Hori et al. 2004). By analyzing radiocarbon ages of the basal or near-basal sediments of the world’s deltas, Stanley and Warne (1994) showed that delta initiation occurred on a worldwide scale after about 8500–6500 years BP and concluded that the initiation was controlled principally by the declining rate of the Holocene sea-level rise. Worldwide there were different regional sea-level changes since the last glacial maximum (LGM) (Irion et al., 2012). Along the northern Canadian coast, for example, sea level has been falling throughout the Holocene due to the glacial rebound of the crust after the last glaciation (Peltier, 1988). This is comparable to the development in Scandinavia (Steffen and Kaufmann, 2005) where sea level drops today. From about Virginia/USA to Mexico there is a constant sea-level rise similar to the Holocene sea-level development of the southern North Sea (e.g. Vink et al., 2007). From the border of Ceará/Rio Grande do Norte down to Patagonia, indicators of Holocene sea level point to a level that was up to 5 m higher than today's mean sea level (Angulo et al., 1999; Martin et al., 2003; Caldas et al., 2006a, b)

Neuropeptid Y (NPY) ist ein potenter Neurotransmitter im zentralen und peripheren Nervensystem der Mammalia. Es ist an der Regulation einer Vielzahl von physiologischen Prozessen beteiligt und scheint auch im retino-tectalen Transfer des visuellen Systems von Anuren eine zentrale Funktion einzunehmen. Die Retina bildet die erste Funktionseinheit bei der Verarbeitung visuellen Inputs. Für die Weiterverarbeitung sind primär das Tectum opticum (TO) und das Praetectum verantwortlich. Es gilt als wahrscheinlich, dass der praetecto-tectale Transfer durch NPY inhibitorisch moduliert wird und damit wesentlichen Einfluss auf die visuelle Mustererkennung und die Dämpfung der tectalen Erregungsausbreitung hat. Die Applikation von NPY auf die Tectumoberfläche schwächt die anfängliche Erregungswelle visuell evozierter Feldpotenziale stark ab und NPY könnte somit Einfluss auf die Axonendknoten retinaler Ganglienzellen des Typs R2, R3 und auch R4 haben. Es können jedoch keine detaillierten Aussagen gemacht werden welche Neuronen in welchem Umfang daran beteiligt sind. Im Rahmen meiner Arbeit, sollte der Einfluss von NPY auf die Spike-Amplitude und die Spike-Rate retinaler Ganglienzellen R2 und R3 bei Bombina orientalis analysiert werden, da diese den größten Input bei der visuellen Mustererkennung liefern und unterschiedliche Funktionen in diesem neuronalen Netzwerk haben. Hierzu wurden visuell evozierte Aktionspotenziale von R2 und R3 Neuronen im TO von Bombina orientalis abgeleitet und mit Hilfe der Analysesoftware Spike 2 bearbeitet und analysiert. Es konnte nachgewiesen werden, dass die Spike-Amplituden der R2 Neuronen 20 min nach NPY Applikation auf die Tectumoberfläche reduziert werden. Nach einer Erholungsphase 10 min nach Beenden der NPY-Applikation konnte ein Wiederanstieg der Spike-Amplituden gemessen werden, 20 min nach Beenden der NPY-Applikation kam es zu einem Abfall der Spike-Amplituden dessen Ursache unbekannt ist. Ob es ein Artefakt ist oder ob es sich hierbei um einen spezifischen Effekt von R2 Neuronen handelt muss noch geklärt werden. Die Spike-Amplituden der R3 Neuronen waren bereits 10 min nach NPY-Applikation reduziert, ein weitere Abfall der Spike-Amplituden konnte nicht verzeichnet werden. 10 min nach Beenden der NPY-Applikation konnte ein Anstieg der Spike-Amplituden verzeichnet werden, der sich stetig fortsetzte. Bei beiden Neuronentypen wurden 20 min nach Beenden der NPY-Applikation Spike-Amplituden nahe der Ausgangsamplitudenhöhe gemessen. Aufgrund des Verlaufes der R3 Neuronen ist davon auszugehen, dass die Feldpotenziale eher durch R3 Neuronen als durch R2 Neuronen beeinflusst werden, da er dem der Feldpotenziale gleicht. Auch bei der Untersuchung der Spike-Raten konnte eine Beeinflussung durch NPY nachgewiesen werden. Die R2 Neuronen zeigten 10 min nach NPY-Applikation einen Abfall der Spike-Raten der sich nach 20 min weiter fortsetzte. 10 min nach Beenden der NPY-Applikation konnte ein Wiederanstieg der Spike-Raten verzeichnet werden der sich stetig fortsetzte, die Werte blieben jedoch deutlich unter den gemessenen Ausgangswerten ohne eine NPY-Beeinflussung. Bei den R3 Neuronen konnte ein Abfall der Spike-Raten deutlich zeitverzögert nachgewiesen werden. 20 min nach Beenden der NPY-Applikation konnte ein Anstieg der Spike-Rate verzeichnet werden, jedoch gab es keine signifikanten Unterschiede der Spike-Raten zu den Werten ohne NPY-Beeinflussung. Der Vergleich der R2 und R3 Neuronen zeigt, dass bei den der R2 Neuronen ein schnellerer Effekt von NPY nachweisbar ist als die den R3 Neuronen. Aufgrund der von mir nachgewiesene NPY-induzierte Spike-Amplitudenabnahme retinaler R2 und R3 Neuronen muss davon ausgegangen werden, dass die Reduktion der Feldpotential durch NPY eher auf den Einfluss anderer Neuronen als R2 und R3 Neuronen zurückzuführen ist. Weder bei den R2 noch bei den R3 Neuronen konnte eine so schnelle und so starke Beeinflussung der Spike- Amplituden verzeichnet werden. Weiterhin zeigen meine Ergebnisse neuronale Bestätigung der von Funke 2005 beschrieben geringeren Strahlungsintensität sowie der geringeren Glukosemetabolisierung bei der 14C-2-Desoxyglukose Technik. Dies ist in der Form nur auf den Einfluss von R2 und R3 Neuronen zurückzuführen. Die von mir erzielten Ergebnisse stützen die Hypothese, dass NPY den retino-tectalen Signaltransfer inhibitorisch steuert einhergehend mit einer reduzierten Ausschüttung des praetectotectalen Transmitters Glutamat und weisen darauf hin, dass NPY über zwei verschiedene second-messenger vermittelte Prozesse diesen Signaltransfer steuert. Interessant ist in diesem Zusammenhang, dass diese nachhaltige Beeinflussung der visuellen Informationsverarbeitung durch NPY bei Bombina orientalis einem phylogenetisch basalen Vertreter der Anuren nachgewiesen werden konnte. Dies lässt den Schluss zu, dass solche grundlegenden neurochemischen Effekte des retino-tectalen Informationsgefüges evolutionär konserviert sind.

Soil organic matter (SOM) vitally impacts all soil functions and plays a key role in the global carbon (C) cycle. More than 70% of the terrestric C stocks that participate in the active C cycle are stored in the soil. Therefore, quantitative knowledge of the rates of C incorporation into SOM fractions of different residence time is crucial to understand and predict the sequestration and stabilization of soil organic carbon (SOC). Consequently, there is a need of fractionation procedures that are capable of isolating functionally SOM fractions, i.e. fractions that are defined by their stability. The literature generally refers to three main mechanisms of SOM stabilization: protection of SOM from decomposition by (i) its structural composition, i.e. recalcitrance, (ii) spatial inaccessibility and/or (iii) interaction with soil minerals and metal ions. One of the difficulties in developing fractionation procedures for the isolation of functional SOM fractions is the marked heterogeneity of the soil environment with its various stabilization mechanisms – often several mechanisms operating simultaneously – in soils and soil horizons of different texture and mineralogy. The overall objective of the present thesis was to evaluate present fractionation techniques and to get a better understanding of the factors of SOM sequestration and stabilization. The first part of this study is attended to the structural composition of SOM. Using 13C cross-polarization magic-angle spinning (CPMAS) nuclear magnetic resonance (NMR) spectroscopy, (i) the effect of land use on SOM composition was investigated and (ii) examined whether SOM composition contributes to the different stability of SOM in density and aggregate fractions. The second part of the present work deals with the mineral-associated SOM fraction. The aim was (iii) to evaluate the suitability of chemical fractionation procedures used in the literature for the isolation of stable SOM pools (stepwise hydrolysis, treatments using oxidizing agents like Na2S2O8, H2O2, and NaOCl as well as demineralization of the residue obtained by the NaOCl treatment using HF (NaOCl+HF)) by pool sizes, 13C and 14C data. Further, (iv) the isolated SOM fractions were compared to the inert organic matter (IOM) pool obtained for the investigated soils using the Rothamsted Carbon Model and isotope data in order to see whether the tested chemical fractionation methods produce SOM fractions capable to represent this pool. Besides chemical fractionation, (v) the suitability of thermal oxidation at different temperatures for obtaining stable SOC pools was evaluated. Finally, (vi) the short-term aggregate dynamics and the factors that impact macroaggregate formation and C stabilization were investigated by means of an incubation study using treatments with and without application of 15N labeled maize straw of different degradability (leaves and coarse roots). All treatments were conducted with and without the addition of fungicide. Two study sites with different soil properties and land managements were chosen for these investigations. The first one, located at Rotthalmünster, is a Stagnic Luvisol (silty loam) under different land use regimes. The Ah horizons of a spruce forest and continuous grassland and the Ap and E horizons of two plots with arable crops (continuous maize and wheat cropping) were examined. The soil of the second study site, located at Halle, is a Haplic Phaeozem (loamy sand) where the Ap horizons of two plots with arable crops (continuous maize and rye cropping) were investigated. Both study sites had a C3-/C4-vegetational change on the maize plot for the purpose of tracing the incorporation of the younger, maize-derived C into different SOM fractions and the calculation of apparent C turnover times of these. The Halle site is located near a train station and industrial areas, which caused a contamination with high amounts of fossil C. The investigation of aggregate and density fractions by 13C CPMAS NMR spectroscopy revealed that density fractionation isolated SOM fractions of different composition. The consumption of a considerable part (10–20%) of the easily available O-alkyl-C and the selective preservation of the more recalcitrant alkyl-C when passing from litter to the different particulate organic matter (POM) fractions suggest that density fractionation was able to isolate SOM fractions with different degrees of decomposition. The spectra of the aggregate fractions resembled those of the mineral-associated SOM fraction obtained by density fractionation and no considerable differences were observed between aggregate size classes. Comparison of plant litter, density and aggregate size fractions from soil under different land use showed that the type of land use markedly influenced the composition of SOM. While SOM of the acid forest soil was characterized by a large content (> 50%) of POM, which contained high amounts of spruce-litter derived alkyl-C, the organic matter in the biologically more active grassland and arable soils was dominated by mineral-associated SOM (> 95%). This SOM fraction comprised greater proportions of aryl- and carbonyl-C and is considered to contain a higher amount of microbially-derived organic substances. Land use can alter both, structure and stability of SOM fractions. All applied chemical treatments induced considerable SOC losses (> 70–95% of mineral-associated SOM) in the investigated soils. The proportion of residual C after chemical fractionation was largest in the arable Ap and E horizons and increased with decreasing C content in the initial SOC after stepwise hydrolysis as well as after the oxidative treatments with H2O2 and Na2S2O8. This can be expected for a functional stable pool of SOM, because it is assumed that the more easily available part of SOC is consumed first if C inputs decrease. All chemical treatments led to a preferential loss of the younger, maize-derived SOC, but this was most pronounced after the treatments with Na2S2O8 and H2O2. After all chemical fractionations, the mean 14C ages of SOC were higher than in the mineral-associated SOM fraction for both study sites and increased in the order: NaOCl < NaOCl+HF ≤ stepwise hydrolysis << H2O2 ≈ Na2S2O8. The results suggest that all treatments were capable of isolating a more stable SOM fraction, but the treatments with H2O2 and Na2S2O8 were the most efficient ones. However, none of the chemical fractionation methods was able to fit the IOM pool calculated using the Rothamsted Carbon Model and isotope data. In the evaluation of thermal oxidation for obtaining stable C fractions, SOC losses increased with temperature from 24–48% (200°C) to 100% (500°C). In the Halle maize Ap horizon, losses of the young, maize-derived C were considerably higher than losses of the older C3-derived C, leading to an increase in the apparent C turnover time from 220 years in mineral-associated SOC to 1158 years after thermal oxidation at 300°C. Most likely, the preferential loss of maize-derived C in the Halle soil was caused by the presence of the high amounts of fossil C mentioned above, which make up a relatively large thermally stable C3-C pool in this soil. This agrees with lower overall SOC losses for the Halle Ap horizon compared to the Rotthalmünster Ap horizon. In the Rotthalmünster soil only slightly more maize-derived than C3-derived SOC was removed by thermal oxidation. Apparent C turnover times increased slightly from 58 years in mineral-associated SOC to 77 years after thermal oxidation at 300°C in the Rotthalmünster Ap and from 151 to 247 years in the Rotthalmünster E horizon. This led to the conclusion that thermal oxidation of SOM was not capable of isolating SOM fractions of considerably higher stability. The incubation experiment showed that macroaggregates develop rapidly after the addition of easily available plant residues. Within the first four weeks of incubation, the maximum aggregation was reached in all treatments without addition of fungicide. The formation of water-stable macroaggregates was related to the size of the microbial biomass pool and its activity. Furthermore, fungi were found to be crucial for the development of soil macroaggregates as the formation of water-stable macroaggregates was significantly delayed in the fungicide treated soils. The C concentration in the obtained aggregate fractions decreased with decreasing aggregate size class, which is in line with the aggregate hierarchy postulated by several authors for soils with SOM as the major binding agent. Macroaggregation involved incorporation of large amounts maize-derived organic matter, but macroaggregates did not play the most important role in the stabilization of maize-derived SOM, because of their relatively low amount (less than 10% of the soil mass). Furthermore, the maize-derived organic matter was quickly incorporated into all aggregate size classes. The microaggregate fraction stored the largest quantities of maize-derived C and N – up to 70% of the residual maize-C and -N were stored in this fraction.

Gisbert es uno de los principales autores de Literatura infantil y juvenil con más de 20 libros publicados. La obra consta de una bibliografía comentada, que incluye unos datos bibliográficos, un breve argumento, un comentario, el nivel de lectura y las propuestas educativas. Se publican unos cuentos y, por último, unos artículos de prensa referidos al autor..

The paludal deposits presented here belong to a geomorphological unit identified between Palafrugell Corridor and the littoral plains of Ter River (Baix Ter), associated to a carbonate-rich spring system. The sections show a distintic clay level, mixed with black silts, very rich in snail remains. The malacological assemblage contained stillwaters species (Bithynia leachii, Galba truncatula, Pisidium milium, Radix peregra) along with damp species (Monacha cartusiana, Oxyloma elegans, Succinea putris, Succinella oblonga, Vallonia pulchella, Vertigo sp.). Some open-dry species are also present (Cernuella virgata, Eobania vermiculata, Pomatias elegans, Rumina decollata) revailing a mosaic-like landscape. Radiocarbon dates place this level ca. 7.000BPwithin the Holocene sequence of the Baix Ter basin. Colluvial deposits on top of these paludal levels have a typical open-dry malacological assemblage (Cernuella virgata, Cochlicella acuta, Eobania vermiculata, Pomatias elegans, Rumina decollata, Trochoidea elegans) the same assemblage that inhabits the Empordà littoral plains nowadays

S'ha analitzat el pas del paleolític mitjà al paleolític superior a Catalunya en base a l'estudi dels jaciments de la cova dels Ermitons (Sales de Llierca, Garrotxa) i de les coves del Reclau -Arbreda, Mollet i Reclau Viver-(Serinyà, Pla de l'Estany). En concret s'ha estudiat la cultura material i la fauna dels nivells mosterians tardans (paleolítc mitjà final) de les coves de l'Arbreda i dels Ermitons i dels aurinyacians (paleolític superior inicial) de les coves de l'Arbreda i de Mollet. Igualment s'han analitzat els territoris d'explotació de la cova dels Ermitons i de les coves del Reclau. D'aquesta anàlisi es desprén que els dos jaciments se situen en medis absolutament diferents. S'han analitzat els contextos geològics dels dos llocs, s'ha fet les descripció morfològica de cada un dels dos jaciments, així com l'estratigràfica i sedimentològica de cada cova. S'han datat els nivells mosterians i aurinyacians esmentats pel mètode del Carboni 14 AMS, així com també els aurinyacians de la cova del Reclau Viver. S'acompanyen aquestes dades amb les anàlisis paleoecològiques corresponents que s'han pogut efectuar: palinologia, antracologia, ictiologia, herpetologia, aus, quiròpters, insectívors i rosegadors. Es comparen les datacions obtingudes ambles d'altres jaciments i troballes catalans del mateix període: l'abric Romaní (Capellades, Anoia), la Roca dels Bous (Camarasa, la Noguera) i la mandíbula de Banyoles (Pla de l'Estany). Cim a conclusions s'estableixen les diferències a Catalunya entre el paleolític mitjà (cultura de l'home Neandertal) i el paleolític superior (cultura de l'home anatòmicament modern). En el segon apareix la tècnica de talla laminar, la indústria òssia ben elaborada, l'ornamentació, la decoració, es generalitza la importació del sílex com a matèria primera, es diversifica l'utillatge lític i hi ha una major utilització de les coves. S'interpreta aquest pas dins del context geogràfic franco-ibèric i es conclou que a Catalunya es caracteritza fonamentalment per dos fenòments: el reemplaçament abrupte i la contemporaneïtat. Aquesta abastaria entre 40.000 BP i el 33.000 BP segons cronologia radiocarbònica. Durant aquests temps la influència de la cultura del paleolític superior sobre la del paleolític mitjà seria escassa, encara que identificada per la presencia d'alguns útils de tipus paleolític superior en el tardomosterià de la cova dels Ermitons

Identifying the signature of global warming in the world's oceans is challenging because low frequency circulation changes can dominate local temperature changes. The IPCC fourth assessment reported an average ocean heating rate of 0.21 ± 0.04 Wm−2 over the period 1961–2003, with considerable spatial, interannual and inter-decadal variability. We present a new analysis of millions of ocean temperature profiles designed to filter out local dynamical changes to give a more consistent view of the underlying warming. Time series of temperature anomaly for all waters warmer than 14°C show large reductions in interannual to inter-decadal variability and a more spatially uniform upper ocean warming trend (0.12 Wm−2 on average) than previous results. This new measure of ocean warming is also more robust to some sources of error in the ocean observing system. Our new analysis provides a useful addition for evaluation of coupled climate models, to the traditional fixed depth analyses.

Traits used by bacteria to enhance ecological performance in natural environments are not well understood. Recognizing that the saprophytic plant-colonizing bacterium Pseudomonas fluorescens SBW25 experiences temperatures in its natural environment significantly cooler than the 28°C routinely used in the laboratory, we identified proteins differentially expressed between 28°C and the more environmentally relevant temperature of 14°C. Of 2102 protein isoforms, 32 were temperature responsive and identified by mass spectrometry. Seven of these (OmpR, MucD, GuaD, OsmY and three of unknown function, Tee1, Tee2 and Tee3) were selected for genetic and ecological analyses. In each instance, changes in protein expression with temperature were mirrored by parallel transcriptional changes. The fitness contribution of the genes encoding each of the seven proteins was larger at 14°C than 28°C and included two cases of trade-offs (enhanced fitness at one temperature and reduced fitness at the other – mucD and tee2 deletions). The relationship between the fitness effects of genes in vitro and in vivo was variable, but two temperature-responsive genes – osmY and mucD – contribute substantially to the ability of P. fluorescens to colonize the plant environment.

Male rats were treated with triiodothyronine in the drinking water for 12 days. In vitro rates of isoprenaline stimulated lipolysis were significantly greater in brown but not white adipose tissue. Rates of [14C]glucose incorporation into triacylglycerols were significantly reduced in BAT (brown adipose tissue) and WAT (white adipose tissue) under basal and isoprenaline stimulated conditions. In a second experiment, hyperthyroid animals showed impaired weight gain, despite increased food intake during 19 days' treatment. Energy expenditure on days 5 and 12, and BAT core temperature differences (TBAT - TCORE) on day 19, were significantly greater than in control animals. Epididymal white fat pad weight was reduced and interscapular brown fat pad weight increased by triiodothyronine treatment.

The broad picture of the cultural and chronological succession from the Epipalaeolithic to the Neolithic in the southern Levant is generally well understood. However, at a more detailed, local level, many questions remain unanswered. In this paper we examine the archaeological record of cultural developments in southern Jordan and the Negev. Focusing on a series of 14C dates from the early occupation of the PPNA site of WF16, we provide a critical review of dating evidence for the region. This review suggests that while the 14C chronology is ambiguous and problematic there is good evidence for a local historical development from the Harifian variant of the Natufian to the early PPNA, well to the south of any core Mediterranean woodland zone. This stresses the importance of considering developments at local scales of analysis, and that the Neolithic transition occurred within a framework of many interacting sub-regional provinces.

Subfossil pollen and plant macrofossil data derived from 14C-dated sediment profiles can provide quantitative information on glacial and interglacial climates. The data allow climate variables related to growing season warmth, winter cold, and plant-available moisture to be reconstructed. Continental-scale reconstructions have been made for the mid-Holocene (MH, around 6 ka) and Last Glacial Maximum (LGM, around 21 ka), allowing comparison with palaeoclimate simulations currently being carried out as part of the fifth Assessment Report (AR5) of the Intergovernmental Panel on Climate Change. The synthesis of the available MH and LGM climate reconstructions and their uncertainties, obtained using modern analogue, regression and model-inversion techniques, is presented for four temperature variables and two moisture variables. Reconstructions of the same variables based on surface-pollen assemblages are shown to be accurate and unbiased. Reconstructed LGM and MH climate anomaly patterns are coherent, consistent between variables, and robust with respect to the choice of technique. They support a conceptual model of the controls of Late Quaternary climate change whereby the first-order effects of orbital variations and greenhouse forcing on the seasonal cycle of temperature are predictably modified by responses of the atmospheric circulation and surface energy balance.
