The development of alkali-activated binders with superior engineering properties and longer durability has emerged as an alternative to ordinary portland cement (OPC). It is possible to use alkali-activated natural pozzolans to prepare environmentally friendly geopolymer cement leading to the concept of sustainable development. This paper presents a summary of an experimental work that was conducted to determine mechanical strength, modulus of elasticity, ultrasonic pulse velocity, and shrinkage of different concrete mixtures prepared with alkali-activated Iranian natural pozzolans—namely Taftan andesite and Shahindej dacite, both with and without calcining. Test data were used for Taftan pozzolan to identify the effects of water-binder ratios (w/b) and curing conditions on the properties of the geopolymer concrete, whereas the influence of material composition was studied by activating Shahindej pozzolan both in the natural and calcined states. The results show that alkali-activated natural pozzolan (AANP) concretes develop moderate-to-high mechanical strength with a high modulus of elasticity and a shrinkage much lower than with OPC.

One of the important factors in the use of portland cement concrete is its durability, and most of the situations where durability is lacking have been identifi ed and strategies to manage durability have been implemented. Geopolymer concrete, made from an alkali-activated natural pozzolan (AANP), provides an important opportunity for the reduction of carbon dioxide (CO2) emissions associated with the manufacture of concrete but has a limited history of durability studies. Until its different properties are well understood there is no desire to adopt this new technology of unknown provenance by the concrete industry. This paper presents an experimental study of oxygen and chloride permeability of AANP concrete prepared by activating Taftan andesite and Shahindej dacite (Iranian natural pozzolans), with and without calcining, and the correlations between these properties and compressive strength. The results show that compared to ordinary portland cement (OPC) concrete, AANP concrete has lower oxygen permeability at later ages; but it shows moderate to high chloride ion penetrability.

In the near future, geopolymers or alkali-activated cementitious materials will be used as new high-performance construction materials of low environmental impact with a reasonable cost. This material is a good candidate to partially replace ordinary portland cement (OPC) in concrete as a major construction material that plays an outstanding role in the construction industry of different structures. Geopolymer materials are inorganic polymers based on alumina and silica units; they are synthesized from a wide range of dehydroxylated alumina-silicate powders condensed with alkaline silicate in a highly alkaline environment. Geopolymeric materials can be produced from a wide range of alumina-silica, including natural products--such as natural pozzolan and metakaolin--or coproducts--such as fly ash (coal and lignite), oil fuel ash, blast furnace or steel slag, and silica fume--and provide a route toward sustainable development. Using lesser amounts of calcium-based raw materials, lower manufacturing temperature, and lower amounts of fuel result in reduced carbon emissions for geopolymer cement manufacture up to 22 to 72% in comparison with portland cement. A study has been done by the authors to investigate the intrinsic nature of different types of Iranian natural pozzolans to determine the activators and methods that could be used to produce a geopolymer concrete based on alkali-activated natural pozzolan (AANP) and optimize mixture design. The mechanical behavior and durability of these types of geopolymer concrete were investigated and compared with normal OPC concrete mixtures cast by the authors and also reported in the literature. This paper summarizes the main conclusions of the research regarding pozzolanic activity, activator properties, engineering and durability properties, applications and evaluation of carbon footprint, and cost for AANP concrete.

Fliegende Insekten orientieren sich in ihrer Umwelt mit Hilfe ihres hoch entwickelten olfaktorischen Systems. Es ermöglicht ihnen das Auffinden geeigneter Futter- und Eiablageplätze und ist unverzichtbar bei der innerartlichen Kommunikation. Der Geruchssinn muss dabei gleichzeitig sehr schnell und sensitiv sein um selbst geringste Mengen, z.B. des arteigenen Sexualpheromons, wahrnehmen zu können. Spezifische olfaktorische Rezeptoren (ORs) zur Detektion dieser Duftstoffe werden zusammen mit einem hoch konservierten Co-Rezeptor (Orco) in olfaktorischen Rezeptorneuronen (ORNs) auf den Insektenantennen exprimiert. Sie gehören zu den 7 Transmembran Rezeptoren, zeigen jedoch eine invertierte Membrantopologie im Vergleich zu den ORs der Vertebraten. Darüber hinaus bildet der OR/Orco-Komplex einen spontanaktiven Kationenkanal, die Bindung an ein G Protein ist allerdings umstritten. Daher ist noch ungeklärt, ob die Duftstoffbindung zu einer ionotropen Aktivierung des OR/Orco Kanals führt oder ob metabotrope Mechanismen die Bildung von zyklischem Adenosinmonophosphat (cAMP) oder Inositol 1,4,5-trisphosphat (IP3) bewirken. Mit Hilfe von extrazellulären Ableitungen einzelner Trichoidsensillen (tip recordings) auf den Antennen männlicher Manduca sexta wurde die Rolle von Orco sowie die Beteiligung einer Phospholipase Cβ (PLCβ)-abhängigen Transduktionskaskade untersucht. Es konnte gezeigt werden, dass die durch VUAA1 induzierte Spontanaktivität der ORNs durch OLC15 inhibiert und Orco somit kompetitiv gehemmt wurde. Eine Inhibition von Orco sollte die Antwort auf kurze Pheromonpulse sofort reduzieren, sollte die Transduktion über die Aktivierung des OR/Orco Kanals erfolgen. Die Ergebnisse dieser Arbeit zeigten jedoch keine Beeinflussung der primären Pheromonantwort, vielmehr wurde die späte, langanhaltende Antwort reduziert. Die ebenfalls als Orco-Antagonisten charakterisierten Amiloride MIA und HMA beeinflussen offensichtlich weitere Ziele, da eine substanz- und zeitgeberzeitabhängige Reduzierung der primären Antwort auftrat. Zusätzlich wurde die primäre Pheromonantwort durch die Inhibierung der PLCβ und der Proteinkinase C (PKC), sowie durch die Verwendung zweier Diacylglycerol (DAG)- Derivate signifikant beeinflusst. Hierbei zeigte die Inhibierung von PLCβ und PKC zeitgeberzeitabhängige Unterschiede in der Stärke der Antwortreduktion. Auch die Applikation des DAG-Derivates DOG reduzierte die Pheromonantwort, während die Zugabe von OAG die ORN Aktivität steigern oder reduzieren konnte, abhängig von der verwendeten Derivatkonzentration und der Pheromonkonzentration. Die Ergebnisse dieser Arbeit deuten somit auf einen metabotropen, sehr wahrscheinlich PLCβ-abhängigen Mechanismus für die Pheromontransduktion bei Manduca sexta.

Nell’ambito delle telecomunicazioni risulta di notevole rilievo la tecnologia ad onde convogliate, definita Power-Line Communication (PLC), che permette la trasmissione di dati sfruttando la rete elettrica come mezzo di trasmissione. Essa rappresenta una delle tecnologie promettenti per quanto concerne la fornitura di servizi come l’accesso ad Internet, alla rete telefonica e al telecontrollo di apparecchi sempre più sofisticati. Al fine di rendere possibile sia la trasmissione di potenza (alla frequenza di 50 Hz per l’Italia) che di altri segnali dati (su bande a frequenza maggiore) saranno necessarie tecniche di modulazione avanzata, che però non rappresentano un argomento trattato a fondo in questa tesi. Lo sviluppo tecnologico degli ultimi anni ha permesso una notevole diffusione delle PLC soprattutto in applicazioni domestiche, grazie ai prezzi molto contenuti in parallelo alle ottime prestazioni, soprattutto nell’automazione. Obbiettivo dell’elaborato risulta la rilevazione della risposta impulsiva in ambienti Power-Line di tipo Narrowband (a banda stretta), immettendo in rete sequenze di tipo casuale o pseudo-casuale, queste ultime definite Pseudo-Noise (PN); esse infatti hanno particolari proprietà che renderanno possibile il raggiungimento dello scopo prefissato. Lo strumento fondamentale per le misurazioni sul campo effettuate risulta l’Analog Discovery (Digilent ne è il produttore), utilizzato non solo come generatore di forme d’onda in ingresso ma anche come oscilloscopio per l’analisi del segnale ricevuto.

Nella seguente tesi è descritto il principio di sviluppo di una macchina industriale di alimentazione. Il suddetto sistema dovrà essere installato fra due macchine industriali. L’apparato dovrà mettere al passo e sincronizzare con la macchina a valle i prodotti che arriveranno in input. La macchina ordina gli oggetti usando una serie di nastri trasportatori a velocità regolabile. Lo sviluppo è stato effettuato al Laboratorio Liam dopo la richiesta dell’azienda Sitma. Sitma produceva già un tipo di sistema come quello descritto in questa tesi. Il deisderio di Sitma è quindi quello di modernizzare la precedente applicazione poiché il dispositivo che le permetteva di effettuare la messa al passo di prodotti era un PLC Siemens che non è più commercializzato. La tesi verterà sullo studio dell’applicazione e la modellazione tramite Matlab-Simulink per poi proseguire ad una applicazione, seppure non risolutiva, in TwinCAT 3.

Abstract : The use of social media tools to support small and medium-sized enterprises (SME) to support their business activities throughout the product life cycle (PLC) phases represents an interesting opportunity. SMEs operate in very competitive environments, and face significant challenges primarily caused by their size disadvantage. By nature, social media tools and platforms can enable them to overcome some of these challenges, as they are often very inexpensive, familiar and easy to use, allowing them to reach large audiences they would not be able to reach with traditional and expensive marketing initiatives. To provide solutions to this problem, this research identified three main objectives. The first objective was to draw a picture of the existing academic literature on the use of social media tools in the PLC context to better understand how these tools were studied and used in businesses, and for what purpose. Second, this research aimed at understanding how SMEs actually use social media tools to support their different business activities to identify the gap between academic research and actual business practices. Finally, based on the findings highlighted from the previous objectives, this research aimed at developing theory on this topic by proposing a conceptual framework of customer engagement enabled by social media. The conceptual framework aimed at answering general questions that emerged from the initial two objectives: Why do some SMEs use social media to support customer engagement, while others do not? Why do firms use different social media tools to support their customer engagement initiatives? Why does the scope of customer engagement initiatives (i.e., across different PLC phases) vary between SMEs? What are the potential outcomes of conducting customer engagement initiatives for the organizing firms? In order to achieve these research objectives, the methodology employed for this research is threefold. First, a systematic literature review was performed in order to properly understand how the use of social media tools in the PLC context had been studied. The final results consisted of 78 academic articles which were analyzed based on their bibliometric information and their content. Second, in order to draw the contrast between the academic publications and managerial reality of SMEs, six semi-structured interviews were conducted to understand how these firms actually use social media to support different activities in each of the PLC phases. Third, five additional semi-structured interviews were performed to gather a deeper understanding of this phenomenon and generate theory to support the proposed conceptual framework. The conceptual framework focuses on the degree of customer engagement, which is comprised of the scope (PLC phases) of customer engagement and the technology (social media tools) employed to support these initiatives. Two sets of antecedents were examined, firm motivators and firm impediments, as they could both potentially affect the scope and the social media tools used to support customer engagement initiatives. Finally, potential customer engagement outcomes for SMEs developing these initiatives were also examined. The semi-structured interviews lasted approximately 25-35 minutes, and were performed using an interview grid consisting of 24 open-ended questions. The interview grid was developed based on the findings of the systematic literature review, and this qualitative approach allowed for a rich understanding of the interviewed SMEs’ use of social media tools to support and engage customers in their different PLC activities. The main results highlighted by this project demonstrate that this field is relatively recent and sees constant increase in research interest since 2008. However, most of the academic research focuses on the use of social media tools to support innovation activities during the new product development process, while the interviewed firms almost exclusively used the tools to engage customers in the later phases of the PLC, primarily for promotion, customer service support, and business development activities. Interestingly, the interviewed firms highlighted several benefits of using social media tools to engage customers, some of which could help them overcome certain size disadvantages previously mentioned. These firms are in need of further guidelines to properly implement such initiatives and reap the expected benefits. Results suggest that SMEs are far behind both large companies and academic research in their use of social media to engage customers in different business activities. The proposed conceptual framework serves as a great tool to better understand their reality and eventually better support them in their social media and customer engagement efforts. However, this framework needs to be further developed and improved. This research project provides a 360-degree view of the phenomenon of the use of social media to support customer engagement for SMEs, by providing both a thorough systematic review of the academic research and an understanding of the managerial reality of SMEs behind this phenomenon. From this analysis, a conceptual framework is then proposed and serves as a stepping stone for future researchers who are interested in developing theory in this field.

L’endométriose est une maladie gynécologique, touchant les femmes en âge de procréer. Cette pathologie est caractérisée par la présence de tissu endométrial ectopique, c’est-à-dire en dehors de la cavité utérine. Des dysfonctions du système immunitaire sont de plus en plus souvent suspectées comme étant un des éléments responsables de la pathogenèse de cette maladie. L’objectif général de ce projet a donc été d’étudier les mécanismes cellulaires de molécules pro-inflammatoires aux propriétés variées et à l’expression anormalement élevée dans cette pathologie, que sont MIF et les prostaglandines PGE2 et PGF2α, dans les anomalies inflammatoires et invasives en cause dans cette pathologie. La première partie de nos travaux a porté sur l’étude d’un modèle murin de l’endométriose déficient du gène MIF. Le nombre et le volume des lésions collectées à partir des souris déficientes pour le gène MIF sont significativement inférieurs à ceux mesurés dans des souris sauvages utilisées comme contrôle. L’analyse par PCR des cellules isolées des lésions de souris déficientes du gène MIF a révélé une expression réprimée des protéines d’adhésion, d’inflammation et d’angiogenèse. Ces données démontrent pour la première fois que le MIF agit directement sur la croissance et la progression de lésions d’endométriose in vivo. Une partie de nos travaux a porté sur les molécules nécessaires au métabolisme de PGE2 et PGF2α dans l’endomètre eutopique des femmes normales et l’endomètre eutopique et ectopique des femmes atteintes d’endométriose. Selon nos données, l’expression de certains de ces facteurs est perturbée durant cette maladie, ce qui peut avoir des effets délétères sur la physiologie de la procréation. La stimulation des cellules ectopiques par PGF2α entraîne une libération accrue de VEGF et CXCL-8, ceci via l’induction de COX-2 et des deux variants d’épissage du récepteur FP. De plus, la PKC joue un rôle dans ce phénomène, dépendamment et indépendamment de la PLC. Par son effet inducteur sur la libération de VEGF et CXCL-8, PGF2α pourrait favoriser l’aspect inflammatoire et le développement ectopique des lésions d’endométriose, notamment par des phénomènes d’angiogenèse et de prolifération cellulaire accrus. L’effet de PGF2α sur la libération de VEGF et CXCL-8 par les cellules endométriales ectopiques pourrait également expliquer les quantités élevées de ces cytokines dans le liquide péritonéal des femmes atteintes d’endométriose, un phénomène suspecté dans l’infertilité et les douleurs associées à cette maladie. Nos derniers résultats obtenus à partir du liquide péritonéal montrent un profil cytokinique en faveur de l’angiogenèse et la prolifération des lésions d’endométriose, avec une forte augmentation des facteurs suivants : EGF, FGF-2, IL-1α, MIP-1β, TGFα, PDGF-AA, PDGF-BB, MCP-3, sCD40L, Gro Pan, IL-17α, MDC et Rantes, confortant nos observations préalables redéfinissant la maladie comme étant d’origine angio-inflammatoire. L’endométriose et ses symptômes sont des phénomènes complexes ayant probablement plus qu’une seule origine. Parmi les nombreux facteurs à l’expression altérée dans l’endométriose, notre étude montre que MIF, PGE2 et PGF2α, ainsi qu’une pléthore de facteurs pro-angiogéniques pourraient être de ceux jouant un rôle dans l’infertilité et les douleurs reliées à cette maladie.

A crescente urbanização global tem como consequência o aumento dos níveis de poluentes na atmosfera e a respetiva deterioração da qualidade do ar. O controlo da poluição atmosférica e monitorização da qualidade do ar são passos fundamentais para implementar estratégias de redução e estimular a consciência ambiental dos cidadãos. Com este intuito, existem várias técnicas e tecnologias que podem ser usadas para monitorizar a qualidade do ar. A utilização de microsensores surge como uma ferramenta inovadora para a monitorização da qualidade do ar. E, apesar dos desempenhos dos microsensores permitirem uma nova estratégia, resultando em respostas rápidas, baixos custos operacionais e eficiências elevadas, que não podem ser alcançados apenas com abordagens convencionais, ainda é necessário aprofundar o conhecimento a fim de integrar estas novas tecnologias, particularmente quanto à verificação do desempenho dos sensores comparativamente aos métodos de referência em campanhas experimentais. Esta dissertação, desenvolvida no Instituto do Ambidente e Desenvolvimento em forma de estágio, teve como objetivo a avaliação do desempenho de sensores de baixo custo comparativamente com os métodos de referência, tendo como base uma campanha de monitorização da qualidade do ar realizada no centro de Aveiro durante 2 semanas de outubro de 2014. De forma mais específica pretende-se perceber até que ponto se podem utilizar sensores de baixo custo que cumpram os requisitos especificados na legislação e as especificidades das normas, estabelecendo assim um protocolo de avaliação de microsensores. O trabalho realizado passou ainda pela caracterização da qualidade do ar no centro de Aveiro para o período da campanha de monitorização. A aplicação de microsensores eletroquímicos, MOS e OPC em paralelo com equipamento de referência neste estudo de campo permitiu avaliar a fiabilidade e a incerteza destas novas tecnologias de monitorização. Com este trabalho verificou-se que os microsensores eletroquímicos são mais precisos comparativamente aos microsensores baseados em óxidos metálicos, apresentando correlações fortes com os métodos de referência para diversos poluentes. Por sua vez, os resultados obtidos pelos contadores óticos de partículas foram satisfatórios, contudo poderiam ser melhorados quer pelo modo de amostragem, quer pelo método de tratamento de dados aplicado. Idealmente, os microsensores deveriam apresentar fortes correlações com o método de referência e elevada eficiência de recolha de dados. No entanto, foram identificados alguns problemas na eficiência de recolha de dados dos sensores que podem estar relacionados com a humidade relativa e temperaturas elevadas durante a campanha, falhas de comunicação intermitentes e, também, a instabilidade e reatividade causada por gases interferentes. Quando as limitações das tecnologias de sensores forem superadas e os procedimentos adequados de garantia e controlo de qualidade possam ser cumpridos, os sensores de baixo custo têm um grande potencial para permitir a monitorização da qualidade do ar com uma elevada cobertura espacial, sendo principalmente benéfico em áreas urbanas.

Esta Dissertação aborda a temática da eficiência energética em sistemas de iluminação fabris. Desde os primórdios da história da humanidade que a iluminação tem representado um papel importante. Não só garante condições de segurança, como começou por ser o intuito primordial das fogueiras pré-históricas, traduzindo-se num elemento de conforto. Atualmente seria impensável conceber as mais diversas atividades económicas e produtivas de um país sem iluminação. Nesse sentido a iluminação representa também um elemento fomentador de produtividade. Não é bem conhecido o impacto que a iluminação tem nos custos operacionais de uma unidade fabril. Principalmente por ser difícil de quantificar separadamente na fatura de eletricidade qual a percentagem correspondente à iluminação. Tipicamente estes custos podem ascender a 20% do orçamento global da autarquia em países desenvolvidos. Por esse motivo, o sector da iluminação tem investido esforço no sentido de promover melhores soluções de iluminação capazes de por um lado de satisfazer os requisitos do espaço a iluminar, e por outro propiciar soluções eficientes do ponto de vista energético. Estas soluções assentam essencialmente no uso de dispositivos mais eficientes (como por exemplo os LED´s ou as lâmpadas compactas fluorescentes) bem como em estratégias de controlo eficientes baseadas em dados sensoriais (como por exemplo deteção de movimento/presença). A combinação destas estratégias permite reduzir os consumos em cerca de 80% quando comparadas com soluções passivas. A empresa Amorim Cork Research Lda. é uma empresa prestadora de serviços ao universo das empresas da Corticeira Amorim, uma das maiores multinacionais de origem portuguesa. Teve origem no negócio da cortiça, em 1870, sendo hoje líder destacada no sector a nível mundial. Dada a dimensão da empresa e a sua subdivisão em várias Unidades Industriais com requisitos individuais, será expectável que a iluminação represente uma parte significativa da fatura de eletricidade. Atualmente, os vários setores encontram-se equipados com soluções de iluminação diferentes, na sua maioria empregando tecnologias passivas, noutros casos com dispositivos eficientes. Há algum interesse em migrar para soluções mais eficientes, em parte suportadas em DALI (com interface de comunicação PLC ou Zigbee).

New devices were designed to generate a localized mechanical vibration of flexible gels where human umbilical vein endothelial cells (HUVECs) were cultured. The stimulation setups were able to apply relatively large strains (30%~50%) at high temporal frequencies (140~207 Hz) in a localized subcellular region. One of the advantages of this technique was to be less invasive to the innate cellular functions because there was no direct contact between the stimulating probe and the cell body. A mechanical vibration induced by the device in the substrate gel where cells were seeded could mainly cause global calcium responses of the cells. This global response was initiated by the influx of calcium across the stretch-activated channels in the plasma membrane. The subsequent production of inositol triphosphate (IP3) via phospholipase C (PLC) activation triggered the calcium release from the endoplasmic reticulum (ER) to cause a global intracellular calcium fluctuation over the whole cell body. This global calcium response was also shown to depend on actomyosin contractility and F-actin integrity, probably controlling the membrane stretch-activated channels. The localized nature of the stimulation is one of the most important features of these new designs as it allowed the observation of the calcium signaling propagation by ER calcium release. The next step was to focus on the calcium influx, more specifically the TRPM7 channels. As TRPM7 expression may modulate cell adhesion, an adhesion assay was developed and tested on HUVECs seeded on gel substrates with different treatments: normal treatment on gels showed highest attachment rate, followed by the partially treated gels (only 5% of usual fibronectin amount) and untreated gels, with the lowest attachment rate. The trend of the attachment rates correlated to the magnitude of the calcium signaling observed after mechanical stimulation. TRPM7 expression inhibition by siRNA caused an increased attachment rate when compared to both control and non-targeting siRNA-treated cells, but resulted in an actual weaker response in terms of calcium signaling. It suggests that TRPM7 channels are indeed important for the calcium signaling in response to mechanical stimulation. A complementary study was also conducted consisting in the mechanical stimulation of a dissected Drosophila embryo. Although ionomycin treatment showed calcium influx in the tissue, the mechanical stimulation delivered as a vertical vibration did not elicited calcium signaling in response. One possible reason is the dissection procedure causing desensitization of the tissue due to the scrapings and manipulations to open the embryo.

Oxidative refolding is one of the key challenges hampering the development of peptide based compounds as therapeutics. The correct refolding for three disulfide peptide like w-Conotoxi n MVIIA is difficult and crucial for biological activity. This work advanced knowledge of chemical and biological for improve oxidative refolding of synthetic w-Conotoxi n MVIIA in base of Conus magus venom. The present study aimed to set up an appropriate and effective protocols for refolding of disulfide-rich w-Conotoxin MVIIA. In this study, the crude peptide was protected with Acm group, according to the right amino acid sequences (Synthesized by Australian Company). The crude peptide was purified by H PLC. To prepare the peptide to refolding, innovative deprotection applied molar ratio (AMR) method was performed based on mercury. Accuracy of deprotection was approved by reverse phase chromatography. The deprotected target peptide (omega-conotoxin) was determined by SDS-PAGE. Then the Oxidative refolding of target peptide was performed in six protocol based on Guanidinium chloride and oxidized and reduced Glutathione. Analgesic effect of refolded peptide was surveyed with formalin test in mice Balb/c. Non neurotoxic effects of target peptides were survey with ICV injection in mice model (C57/BL6). The innovative deprotection protocol performed based on the best ratio of mercury/2-mercaptoethanol adjusted to 1mg/10p1 in 90 minute. The results showed the yield and purity of omega-conotoxin MVIIA as 93 and 95%, respectively. Refolding of 40 mg omega Conotoxin with GSSG and GSH on ratio of 10:1 and 20 mM ammonium acetate showed the best analgesic effect compared with the other methods. The result showed 95.5% yield and 98% purity of omega-conotoxin MVIIA in this refolding method. Related refolding method reduced 85% pain in experimented mice using 7 ng of the peptide. That was 71.5 fold stronger than morphine and 2 times than standard Prialt®. And it was not neurotoxic in mice. In this study, refolding method for omega-conotoxin MVIIA was optimized in the fourth factor including: reducing the time, amount and number of reagent and increase the efficiency. We introduced new method for deprotection of omega-conotoxin MVIIA. Effective, economic and applied refolding and deprotecti on method was performed in this research may al so be applied to similar omega conotoxin peptides.

Spinal cord injury (SCI) is a devastating condition, which results from trauma to the cord, resulting in a primary injury response which leads to a secondary injury cascade, causing damage to both glial and neuronal cells. Following trauma, the central nervous system (CNS) fails to regenerate due to a plethora of both intrinsic and extrinsic factors. Unfortunately, these events lead to loss of both motor and sensory function and lifelong disability and care for sufferers of SCI. There have been tremendous advancements made in our understanding of the mechanisms behind axonal regeneration and remyelination of the damaged cord. These have provided many promising therapeutic targets. However, very few have made it to clinical application, which could potentially be due to inadequate understanding of compound mechanism of action and reliance on poor SCI models. This thesis describes the use of an established neural cell co-culture model of SCI as a medium throughput screen for compounds with potential therapeutic properties. A number of compounds were screened which resulted in a family of compounds, modified heparins, being taken forward for more intense investigation. Modified heparins (mHeps) are made up of the core heparin disaccharide unit with variable sulphation groups on the iduronic acid and glucosamine residues; 2-O-sulphate (C2), 6-O-sulphate (C6) and N-sulphate (N). 2-O-sulphated (mHep6) and N-sulphated (mHep7) heparin isomers were shown to promote both neurite outgrowth and myelination in the SCI model. It was found that both mHeps decreased oligodendrocyte precursor cell (OPC) proliferation and increased oligodendrocyte (OL) number adjacent to the lesion. However, there is a difference in the direct effects on the OL from each of the mHeps; mHep6 increased myelin internode length and mHep7 increased the overall cell size. It was further elucidated that these isoforms interact with and mediate both Wnt and FGF signalling. In OPC monoculture experiments FGF2 treated OPCs displayed increased proliferation but this effect was removed when co-treated with the mHeps. Therefore, suggesting that the mHeps interact with the ligand and inhibit FGF2 signalling. Additionally, it was shown that both mHeps could be partially mediating their effects through the Wnt pathway. mHep effects on both myelination and neurite outgrowth were removed when co-treated with a Wnt signalling inhibitor, suggesting cell signalling mediation by ligand immobilisation and signalling activation as a mechanistic action for the mHeps. However, the initial methods employed in this thesis were not sufficient to provide a more detailed study into the effects the mHeps have on neurite outgrowth. This led to the design and development of a novel microfluidic device (MFD), which provides a platform to study of axonal injury. This novel device is a three chamber device with two chambers converging onto a central open access chamber. This design allows axons from two points of origin to enter a chamber which can be subjected to injury, thus providing a platform in which targeted axonal injury and the regenerative capacity of a compound study can be performed. In conclusion, this thesis contributes to and advances the study of SCI in two ways; 1) identification and investigation of a novel set of compounds with potential therapeutic potential i.e. desulphated modified heparins. These compounds have multiple therapeutic properties and could revolutionise both the understanding of the basic pathological mechanisms underlying SCI but also be a powered therapeutic option. 2) Development of a novel microfluidic device to study in greater detail axonal biology, specifically, targeted axonal injury and treatment, providing a more representative model of SCI than standard in vitro models. Therefore, the MFD could lead to advancements and the identification of factors and compounds relating to axonal regeneration.

The aims of this thesis were evaluation the type of wave channel, wave current, and effect of some parameters on them and identification and comparison between types of wave maker in laboratory situations. In this study, designing and making of two dimension channels (flume) and wave maker for experiment son the marine buoy, marine building and energy conversion systems were also investigated. In current research, the physical relation between pump and pumpage and the designing of current making in flume were evaluated. The related calculation for steel building, channels beside glasses and also equations of wave maker plate movement, power of motor and absorb wave(co astal slope) were calculated. In continue of this study, the servo motor was designed and applied for moving of wave maker’s plate. One Ball Screw Leaner was used for having better movement mechanisms of equipment and convert of the around movement to linear movement. The Programmable Logic Controller (PLC) was also used for control of wave maker system. The studies were explained type of ocean energies and energy conversion systems. In another part of this research, the systems of energy resistance in special way of Oscillating Water Column (OWC) were explained and one sample model was designed and applied in hydrolic channel at the Sheikh Bahaii building in Azad University, Science and Research Branch. The dimensions of designed flume was considered at 16 1.98 0. 57 m which had ability to provide regular waves as well as irregular waves with little changing on the control system. The ability of making waves was evaluated in our designed channel and the results were showed that all of the calculation in designed flume was correct. The mean of error between our results and theory calculation was conducted 7%, which was showed the well result in this situation. With evaluating of designed OWC model and considering of changes in the some part of system, one bigger sample of this model can be used for designing the energy conversion system model. The obtained results showed that the best form for chamber in exit position of system, were zero degree (0) in angle for moving below part, forty and five (45) degree in front wall of system and the moving forward of front wall keep in two times of height of wave.
