A study on the nematode parasites of nine species of freshwater fishes from Peixe River São Paulo, State, Brazil. was conducted. Fish were collected between February 2010 and March 2011 and the following species were found: Procamallanus (Spirocamallanus) inopinatus and Contracaecum sp. (larvae) in Astyanax altiparanae; Contracaecum sp. (larvae), Dioctophyma renale (larvae), Philometroides caudata, P. (Spirocamallanus) inopinatus, P. (Spirocamallanus) neocaballeroi (larvae) and P. (Spirocamallanus) saofranciscensis in Acestrorhynchus lacustris; Contracaecum sp. (larvae), Guyanema sp., Hysterothylacium sp. (larvae) and Icthyouris sp. in Cyphocharax modestus; Contracaecum sp. (larvae), Cosmoxynemoides aguirrei and Pharyngodonidae gen. sp. in C. nagelii; Dioctophyma renale (larvae), Hysterothylacium sp. (larvae) and Rhabdochona sp. in Gymnotus sylvius; Capillariidae gen. sp. in Hoplosternum littorale; Cosmoxynema vianai, Guyanema sp., Ichthyouris sp. and Travnema travnema in Steindachnerina insculpta; Contracaecum sp. (larvae), Procamallanus (Spirocamallanus) rebecae (larvae) in Triportheus angulatus and Rhabdochona acuminata in Triportheus nematurus. This is first study of nematode parasites from the Peixe River, therefore all the species found are new geographical records and 19 are new host records.

NAPc2, an anticoagulant protein from the hematophagous nematode Ancylostoma caninum evaluated in phase-II/IIa clinical trials, inhibits the extrinsic blood coagulation pathway by a two step mechanism, initially interacting with the hitherto uncharacterized factor Xa exosite involved in macromolecular recognition and subsequently inhibiting factor VIIa (K-i = 8.4 pM) of the factor VIIa/tissue factor complex. NAPc2 is highly flexible, becoming partially ordered and undergoing significant structural changes in the C terminus upon binding to the factor Xa exosite. In the crystal structure of the ternary factor Xa/NAPc2/selectide complex, the binding interface consists of an intermolecular antiparallel beta-sheet formed by the segment of the polypeptide chain consisting of residues 74-80 of NAPc2 with the residues 86-93 of factor Xa that is additional maintained by contacts between the short helical segment (residues 67-73) and a turn (residues 26-29) of NAPc2 with the short C-terminal helix of factor Xa (residues 233-243). This exosite is physiologically highly relevant for the recognition and inhibition of factor X/Xa by macromolecular substrates and provides a structural motif for the development of a new class of inhibitors for the treatment of deep vein thrombosis and angioplasty. This investigation was carried out at the Madeira River basin, located in the state of Rondonia, Brazilian Amazon. Fish from Madeira, Jaciparana, and Jamari rivers between 7 and 11 degrees parallels south and between 62 and 65 degrees meridians west in Rondonia state, Brazil, were sampled and chemically analyzed for mercury in order to evaluate if the inputs of this metal into the food-chain is occurring in levels reaching values above those recommended by the World Health Organization. This is because such an element is very dangerous when ingested by humans and its presence was extensively identified some years ago in the area, since it was utilized as an amalgam in processes for recovering alluvial gold.

Samples of water, suspended solids, and bottom sediments from the Madeira River, Rondonia state, Brazil, were physically and chemically analyzed to investigate the actual Hg mobilization in the aquatic environment and compare it with that of other heavy metals and elements in the area. Two dimensionless Hg preference ratios were defined, expressing (1) the ratio of Hg and other elements in the liquid phase divided by the ratio of Hg and other elements in bottom sediments (P(l.phase)) and (2) the ratio of Hg and other elements in the particulate matter divided by the ratio of Hg and other elements in bottom sediments (P(s.solids)). These preference ratios are useful for comparing Hg transport in three different phases (liquid, particulate matter, and bottom sediments). They also were applicable to any analyzed elementin the area studied, because they generated an almost constant value when the maximum calculated was divided by the minimum (P(l.phase) = 2931; P(s.solids) = 84) and because of their sensitivity to the dominance of sorption processes by Fe oxides and hydroxides. Mercury could be transported preferentially to other analyzed elements in the particulate phase only if its concentration reached values at least 10(4)-fold higher than those expected or quantified in the area. O presente estudo traz informações sobre a utilização da madeira de duas variedades de Sclerolobium paniculatum (var. subvelutinum e rubiginosum) em propriedades rurais do sul do Maranhão, a análise anatômica do lenho e a correlação destas características com o uso da madeira. Foram realizadas entrevistas com 17 agricultores residentes na zona rural dessa região, com a finalidade de se conhecer a utilização destas variedades nas propriedades rurais. As variedades subvelutinum Benth. e rubiginosum (Mart. ex Tul. ) Benth. são conhecidas na região como cachamorra-preta e cachamorra-branca, respectivamente, sendo a primeira mais utilizada na confecção de cercados. Para o estudo anatômico do lenho, foram coletados discos à altura do peito de três indivíduos de cada variedade e, destes, obtidas amostras do cerne, na região de transição com o alburno. As amostras foram processadas de acordo com a metodologia usual para anatomia de madeira. Houve diferença estatística significativa (p<0,05) para seis parâmetros anatômicos entre as duas variedades. A utilização diferencial das variedades pode ser decorrente, principalmente, das características das fibras e dos elementos de vasos. Sugere-se que sejam realizados estudos que abordem variações climáticas e características do solo para melhor entendimento das diferenças anatômicas quantitativas encontradas no lenho das variedades.

Hookworms are hematophagous nematodes capable of growth, development and subsistence in living host systems such as humans and other mammals. Approximately one billion, or one in six, people worldwide are infected by hookworms causing gastrointestinal blood loss and iron deficiency anemia. The hematophagous hookworm Ancylostoma caninum produces a family of small, disulfide-linked protein anticoagulants (75-84 amino acid residues). One of these nematode anticoagulant proteins, NAP5, inhibits the amidolytic activity of factor Xa (fXa) with K-i = 43 pM, and is the most potent natural fXa inhibitor identified thus far. The crystal structure of NAP5 bound at the active site of gamma-carboxyglutamic acid domainless factor Xa (des-fXa) has been determined at 3.1 angstrom resolution, which indicates that Asp189 (fXa, S1 subsite) binds to Arg40 (NAP5, P1 site) in a mode similar to that of the BPTI/trypsin interaction. However, the hydroxyl group of Ser39 of NAP5 additionally forms a hydrogen bond (2.5 angstrom) with His57 NE2 of the catalytic triad, replacing the hydrogen bond of Ser195 OG to the latter in the native structure, resulting in an interaction that has not been observed before. Furthermore, the C-terminal extension of NAP5 surprisingly interacts with the fXa exosite of a symmetry-equivalent molecule forming a short intermolecular beta-strand as observed in the structure of the NAPc2/fXa complex. This indicates that NAP5 can bind to fXa at the active site, or the exosite, and to fX at the exosite. However, unlike NAPc2, NAP5 does not inhibit fVIIa of the fVIIa/TF complex.
