In order to characterize the cellular component of the polymorphous low-grade adenocarcinoma (PLGA) of the salivary gland, a morphological and immunohistochemical study was carried out. Thirty cases of PLGA were studied by light microscopy and immunohistochemistry and five cases by transmission electron microscopy (TEM). The expression of cytokeratins (CKs) 7,8,10,13,14,18,19, vimentin and muscle-specific actin (MSA) was investigated through the streptavidin-biotin method. The majority of tumor cells stained for vimentin, CKs 8,18 and 7. CK 14 was positive in most cells of the papillary and trabecular sub-types. Although the expression of CKs 8,18 and 14 varied among the tumors sub-types, a straight relationship between each histologic pattern and the CK expression could not be delineated. MSA was reactive in only three tumors while CKs 10 and 13 were not detected in any tumor studied. The absence of MSA and the expression of CKs 8,18 and 7, in most of the tumor cells, lead to the hypothesis that myoepithelial cells are not the major cellular component of the PLGA. TEM revealed cells exhibiting microvilli and variable amounts of secretory granules, some of them suggesting an excretory activity. The presence of CKs 8, 18 and 7, added to the secretory granules, indicates that PLGA originates from cells located at the acinar-intercalated duct junction. (C) 1999 Elsevier B.V. Ltd. Calcium binding and charge distribution on highly branched and linear sulfated L-galactans from ascidians have been studied using a metallochromic indicator and conductimetric titrations. The distance between charged groups of the linear and highly branched galactans does not vary despite their marked differences in sulfate/total sugar molar ratios. These results indicate that the sulfated L-galactose units are concentrated in the central polysaccharide core and not intercalated among non-sulfated units. This inference is consistent with the chemical studies of these galactans. Surprisingly, calcium affinity increases with increasing amounts of non-sulfated sugar branches in the molecule. Thus, calcium binding in these polymers is not a simple function of availability of anion binding sites but a more complex calcium-polysaccharide interaction. (C) 1998 Elsevier B.V. Ltd. The golden-faced saki monkey Pithecia pithecia chrysocephala (Cebidae, Primates) was observed eating soil from termite nests during a long-term study of a family group in a Central Amazonian forest fragment. In this paper we describe the behaviour involved in the geophagy in these monkeys, and the results of geochemical and physical analyses of the termite nest material, as well as root mat and topsoil samples below the trees, in order to clarify the possible reasons for it. The sakis ate soil from nine arboreal termite nests on 26 soil feeding-bouts (in 853 observation hours); 25 soil feeding-bouts occurred in March 1987 (rainy season), during 19 days or 132 observation hours, and occupied 0.7% of the feeding time. Geophagy frequencies did not differ between sexes (17 feeding-bouts of four females and 8 for two males). Mineral composition was higher in arboreal termitaria than in the topsoil. Kaolinite was the major clay component. Tannin adsorptive capacity, tested through a modified radial diffusion method of Hagerman, was around 10-20%, similar to a control with kaolin (10-20%), but lower than bentonite or celite (30-45%). The observations reported here, although inconclusive as to the function of geophagy in this species, indicate that it is not a mineral supplement during times of scarcity or high consumption of leaves, as has been reported for other primates, nor that it is related to fruit consumption (redressing possible mineral imbalance), as has been suggested for some other frugivorous mammals. Our results do not rule out tannin adsorptive hypothesis for the ingestion of clays, but, being an irregular habit, we argue that it is most likely related to rare and occasional dietary components.

We have examined the binding processes of ethidium bromide interacting with calf thymus DNA using photoacoustic spectroscopy. These binding processes are generally investigated by a combination of absorption or fluorescence spectroscopies with hydrodynamic techniques. The employment of photoacoustic spectroscopy for the DNA-ethidium bromide system identified two binding manners for the dye. The presence of two isosbestic points (522 and 498 nm) during DNA titration was evidence of these binding modes. Analysis of the photoacoustic amplitude signal data was performed using the McGhee-von Hippel excluded site model. The binding constant obtained was 3.4 x 10(8) M(bp)(-1), and the number of base pairs excluded to another dye molecule by each bound dye molecule (n) was 2. A DNA drug dissociation process was applied using sodium dodecyl sulfate to elucidate the existence of a second and weaker binding mode. The dissociation constant determined was 0.43 mM, whose inverse value was less than the previously obtained binding constant, demonstrating the existence of the weaker binding mode. The calculated binding constant was adjusted by considering the dissociation constant and its new value was 1.2 x 10(9) M(bp)(-1) and the number of excluded sites was 2.6. Using the photoacoustic technique it is also possible to obtain results regarding the dependence of the quantum yield of the dye on its binding mode. While intercalated between two adjacent base pairs the quantum yield found was 0.87 and when associated with an external site it was 0.04. These results reinforce the presence of these two binding processes and show that photoacoustic spectroscopy is more extensive than commonly applied spectroscopies.

ANALYSIS OF CLAYS BY INDUCTIVELY COUPLED PLASMA OPTICAL EMISSION SPECTROMETRY AFTER CLOSED-VESSEL MICROWAVE-ASSISTED ACID DECOMPOSITION. In this work a closed-vessel microwave-assisted acid decomposition procedure for clays was developed. Aluminum, Ca, Fe, K, Mg, Na, Si, and Ti were determined in clay digestates by inductively coupled plasma optical emission spectrometry. The most critical parameter for total decomposition of clays was the composition of the reagent mixture. The applied power and the heating time exerted a less critical influence. Best decomposition conditions were attained using a reagent mixture containing 4 mL aqua regia plus 3 mL HF and the heating program was implemented in 12 min. The accuracy of the results was demonstrated using two standard reference materials and a paired t-test showed a good agreement between determined and certified values at a 95% confidence level.

We developed a procedure to take advantage of the magnetic-field-modulation-frequency effect on the line shape of conduction-electron-spin resonance of graphite intercalation compounds (GIC's) to extract the absolute value of the in-plane resistivity. We calculated the power absorbed in each slice of the sample normal to the wave penetration, multiplied by a factor to account for the magnetic-field-modulation-frequency effect. Room-temperature spectra of stage-I AlCl3-intercalated GIC in both H-0 perpendicular-to c and H-0 parallel-to c configurations were fitted to the theoretical line shapes and the value of in-plane resistivity (and also the value of c-axis resistivity) obtained from the fitting parameters are in reasonable agreement with those from the literature.

Diamictites interbedded with marine shales and turbidites onlap the eastern border of the Parana Basin (Southern Brazil). These poorly sorted sediments were deposited during the Permo-Carboniferous glaciation, and their matrix-supported clasts show no preferred orientation. These massive rocks have been studied using anisotropy of magnetic susceptibility (AMS) and grain shape fabric. Hysteresis loops and thermomagnetic measurements show that AMS depends mostly on the paramagnetic clays, but fine ferromagnetic particles also contribute to the anisotropy. The coarse silt to sand grain preferred orientation study supports the use of AMS in describing the diamictite fabric, at least regarding the orientation of the foliation. AMS and grain shape data reveal subhorizontal to weakly inclined magnetic and grain shape foliation parallel to the regional bedding. The magnetic lineations are normally scattered within the foliation plane in agreement with the oblate AMS ellipsoids found in these rocks. Both fabric patterns are consistent with deposition by subaqueous mudflows that were resedimented downslope, with elastic supply from continental sources. The off-vertical grain shape foliation poles suggest that the deposition of diamictites was controlled by the depocentre topography of the Rio do Sul sub-basin.
