This work reports the optimization and method validation for sulfonamides (sulfamethazine, sulfaquinoxaline, sulfadimethoxine and sulfathiazole) in shrimp muscle using HPLC-UV. The sulfonamides were extracted with acetonitrile and acetic acid, and the extract cleaned up with a Strata SCX SPE cartridge prior to analysis. The method presented linearity in the range of 20-120 µg kg-1, good linear correlation (r > 0.99), and limits of quantification in the range of 4.7-20.2 µg kg-1. The recovery for shrimp muscles spiked with 50-150 µg kg-1 ranged from 63.2-108.0%. Precision and accuracy analysis showed acceptable relative standard deviation. Commercial shrimps were analyzed and sulfonamides don't were found above of the method limit of quantification.

Prior to obtain a standardized dried extract from V. ferruginea, lupeol was first time isolated from leaves and used as chemical maker. An analytical method using HPLC-PDA for lupeol determination in V. ferruginea intermediate products was developed using a C8 reverse-phase column, acetonitrile-acetic acid (99.99:0.01, v/v) as mobile phase at 0.8 mL min-1, oven temperature at 23-25 ºC, sample injection volume at 30 µL and detection at 210 nm. The method presented linearity from 10 to 160 µg mL-1, accuracy, precision, robustness and suitable sensitivity proving to be a useful tool to the obtainment process of lupeol standardized dried extracts of V. ferruginea.

This work describes the phytochemical study of stem bark and leaves of Cenostigma macrophyllum Tul. (Leguminosae). Through usual chromatographic techniques were isolated bergenin as the primary compound of the stem bark of and from the leaves gallic acid, methyl gallate, ellagic acid, quercetin, quercetin-3-O-β-D-glucopyranoside, quercetin-3-O-(6"-O-galloyl)-β-D-glucopyranoside (tellimoside), quercetin-3-O-(6"-O-E-p-coumaroyl)-β-D-glucopyranoside (helichrysroside), agathisflavone and vitexin were obtained. The isolates were identified by spectroscopic data analysis, and bergenin showed dose-related antinociception when assessed in acetic acid-induced writhing in mice.

A method using HPLC-UV was developed and validated for the determination of etoposide incorporated into polycaprolactone implants. The method was carried out in isocratic mode using a C18 column (250 x 4.6 mm; 5 µm), at 25 ºC, with acetonitrile and acetic acid 4% (70:30) as mobile phase, a flow rate of 2 mL/min, and UV detection at 285 nm. The method was linear (r² > 0.99) over the range of 5 to 65 µg/mL, precise (RSD < 5%), accurate (recovery of 98.7%), robust, selective regarding excipient of the sample, and had a quantitation limit equal to 1.76 µg/mL. The validated method can be successfully employed for routine quality control analyses.

In this work, theoretical and experimental infrared spectra of fatty acid methyl esters (FAME) contained in soybean biodiesel were analyzed seeking the assignments of the relevant vibrational modes to characterize crude soybean oil and soybean biodiesel. The results showed the usefulness of infrared spectra for monitoring saturated and unsaturated compounds as well as impurities (mainly glycerol) in raw samples. This is the first step toward proposing an efficient molecular spectroscopy routine to certify biodiesel fuel.

Our solemn homage to the great Master Otto R. Gottlieb who knew how to teach the mystery of evolutionary relationships between chemistry and its natural sources. The micromolecular chemical study of the family Bignoniaceae shows a profile predominantly characterized by the occurrence of metabolites derived from acetic acid biosynthetic pathways such as terpenoids, quinones, flavonoids and special aromatic derivatives. Analysis of different chemosystematic parameters for the metabolite data collected, provided valuable information for the systematic characterization of the Bignoniaceae family within the Angiosperm derived taxa.

In this work, we present an efficient and inexpensive device for undergraduate chemistry classes aimed at teaching and learning the photolytic synthesis concepts. A photochemical reactor was tested for the synthesis of the organometallic compound enneacarbonyldiiron from iron pentacarbonyl in acetic acid, and its formation evidenced by FTIR analysis. Although similar devices have been described in other studies, none of these offered the simplicity, low cost, class-compatible reaction times and good yields afforded by the procedure reported herein.

The seed oils from four plants (Scheelea phalerata, Butia capitata, Syagrus romanzoffiana, Terminalia cattapa) found in Mato Grosso do Sul were extracted at good yields. Alkaline transesterification of these seed oils to esters using methanol and ethanol was studied and also produced good yields. Oleic acid (30.5/32.3%), lauric acid (30.7/32.9%) methyl and ethyl esters, were the main components of transesterification of the oils from Scheelea phalerata and Syagrus romanzoffiana. Lauric acid (42.2%), capric acid (15.9%) and caprylic acid (14.6%) methyl and ethyl esters were the main ester components of transesterification of the oil from Butia capitata. Oleic acid (37.8%), palmitic acid (33.5%) and linoleic acid (22.6%) methyl and ethyl esters were the main components of transesterification of oil from Terminalia catappa. Based on differential scanning calorimetry (DSC) studies, the first crystallization peak temperature of esters was observed. Esters derived from oils of the family Arecaceae (Scheelea phalerata, Butia capitata, Syagrus romanzoffiana) showed the lowest points of crystallization, despite having high levels of saturated fat. Esters of Terminalia cattapa oil, rich in unsaturated fat, showed the highest crystallization temperature. This difference in behavior is probably related to the high concentration of esters derived from lauric acid and palmitic acid.

The adsorption kinetics and equilibrium of methylene blue (MB) onto reticulated formic lignin (RFL) from sugar cane bagasse was studied. The adsorption process is pH, temperature and ionic strength (µ) dependent and obeys the Langmuir model. Conditions for higher adsorption rate and capacity were determined. The faster adsorption (12 hours) and higher adsorption capacity (34.20 mg.g-1) were observed at pH = 5.8 (acetic acid-sodium acetate aqueous buffer), 50 ºC and 0.1 ionic strength. Under temperature (50 ºC) control and occasional mechanical stirring it took from 1 to 10 days to reach the equilibrium.

Four simple titrimetric procedures are described for the determination of lisinopril (LNP) in bulk and in pharmaceuticals based on the neutralization of basic-amino and acidic carboxylic acid groups present in LNP. Method A is based on the neutralization of basic amino groups using perchloric acid as titrant in anhydrous acetic acid medium. Method B, method C and method D are based on neutralization of carboxylic acid group using NaOH, sodium methoxide and methanolic KOH, as titrants, respectively. Method A is applicable over 2.0-20.0 mg range and the calculations are based in the molar ratio of 1:2 (LNP:HClO4). Method B, method C and method D are applicable over 2.0-20.0 mg, 1.0-10.0 mg and 5.0-15.0 mg range, respectively, and their respective molar ratios are 1:1 (LNP:NaOH), 1:2 (LNP:CH3ONa) and 1:1 (LNP:KOH). Intraday and inter day accuracy and precision of the methods were evaluated and the results showed intra- and inter-day precision less than 2.7% (RSD), and accuracy of < 2.5 % (RE). The developed methods were applied to determine LNP in tablets and the results were validated statistically by comparing the results with those of the reference method by applying the Student's t-test and F-test. The accuracy was further ascertained by recovery studies via standard addition technique. No interferences from common tablet exipients was observed.

Työssä tutkittiin lämpötilan vaikutusta kalvon likaantumiseen mustalipeällä ja puuperäisellä hydrolysaatilla lämpötiloissa 20, 45 ja 70 °C:tta. Työn tavoitteena oli löytää lämpötila, jossa tutkimuksessa käytetyt puuperäiset liuokset likasivat kalvoja mahdollisimman vähän. Tutkimuksessa käytettiin Alfa Lavalin UFX5-ultrasuodatuskalvoa sekä Microdyn-Nadirin ultrasuodatuskalvoa UP010 ja nanosuodatuskalvoa NP010. Ennen puuperäisten liuosten adsorboimista kalvoihin säilöntäaineet poistettiin kalvoista sekä kalvojen toiminta stabiloitiin esikäsittelemällä kalvot. Esikäsittelyssä kalvoja pestiin alkalisella pesuaineella, puristettiin 8 bar:n paineessa sekä huuhdeltiin vedellä ja etikkahapolla. Esikäsitellyt kalvot karakterisoitiin mittaamalla malliaineliuoksen vuo ja malliaineiden retentiot suodattamalla polyetyleeniglykoliliuosta kolmikennosuodattimella paineissa 2, 4 ja 6 bar. Tämän jälkeen kalvot altistettiin lämpötiloissa 20, 45 ja 70 °C:tta mustalipeälle ja hydrolysaatille. Altistuksen jälkeen karakterisointi tehtiin uudelleen, jotta vuoarvoja ja retentioita vertaamalla voitiin arvioida altistuksen aikana tapahtunutta kalvojen likaantumista. Adsorboituneen lian puhdistusta kalvoista tutkittiin pesemällä kalvoja alkalisella pesuliuoksella 20 °C:ssa tehdyn altistuksen jälkeen. Mustalipeälle altistettujen kalvojen likaantuminen oli vähäisintä kokeissa, joissa lämpötila oli 20 °C. Alfa Lavalin UFX5-kalvolla 70 °C:ssa tehdyssä kokeessa kalvon likaantuminen oli myös erittäin vähäistä, sillä permeabiliteetti pieneni mustalipeäaltistuksen seurauksena alle 5 %. Hydrolysaatille altistetut kalvot likaantuivat adsorptiolla vähiten Alfa Laval UFX5-kalvoilla 20 °C:ssa tehdyissä kokeissa ja 45 °C:ssa Microdyn-Nadir NP010-kalvoilla tehdyissä kokeissa. Kokeiden perusteella tutkituista vaihtoehdoista korkein tutkimuksessa käytetty lämpötila 70 °C ja Alfa Lavalin UFX5-kalvo soveltuvatkin parhaiten mustalipeän kalvosuodatukseen adsorptiivisen likaantumisen kannalta.

The objective of this work was to establish an efficient protocol for in vitro multiplication and rooting, as well as ex vitroacclimatization of Aegiphila verticillata, a woody species found in Brazilian rocky fields. Aseptic cultures were established by seeds and two multiplication analyses were performed. In the first, we employed 6-benzylaminopurine (BAP – 0, 2.5, 5 and 7.5 μM) + α-naphthalene acetic acid (NAA – 0, 0.2, 0.4 and 0.6 μM) and, in the second, were studied adenine sulfate, kinetin and thidiazuron (0, 5, 7.5, 10 and 12.5 μM). After 90 days, we assessed the quantitative and qualitative shoot propagation. There were more than 90% seed germination and low contamination (2%). In multiplication phase, the culture medium that promoted the best quantitative and qualitative culture development was supplemented with 7.5 μM BAP + 0.4 μM NAA. In the rooting assay, were used NAA, indole-3-acetic acid (IAA) and indole-3-butyric acid (IBA) (0, 0.1, 0.2, 0.3 or 0.4 μM). After 90 days, the root number and rooting quality were evaluated. In this analysis, differences were not found between the control and the other treatments. Rooted plantlets were acclimatized in styrofoam trays for 30 days, after which they were transferred to pots in the greenhouse. Only 3% of the plants subjected to initial acclimatization died and 70% of the plants transferred to the field conditions survived and showed normal development. The results founded in this work are the first involving in vitro propagation and ex vitroacclimatization of Aegiphila verticillata and provide a continuous supply of this medicinal native species, endangered due anthropogenic activities.

Työssä tutkittiin lignoselluloosapohjaisten väkevähappohydrolysaattien (monosakkaridit, rikki- ja etikkahppo) jatkuvatoimista kromatografista fraktiointia käyttäen Japan Organo SMB -prosessia. Adsorbenttinä toimi happomuotoinen (H+) vahva kationinvaihtohartsi. Panostoimista fraktiointia käytettiin vertailukohtana. Jatkuvatoimisen prosessin optimoinnilla saavutettiin monosakkaridien suurimmaksi tuottavuudeksi 283 mol/(m3 h) (panosprosessille 145 mol/(m3 h). JO-prosessilla saavutettiin korkeat rikki- ja etikkahapon saannot: 97,3 % ja 93,5 %. Monosakkaridisaanto jäi hieman alhaisemmaksi (61,7 %) johtuen monosakkaridi- ja rikkihappo-profiilien hankalasta erottamisesta. Ulostulo virtojen puhtaudet olivat korkeat: 89 % monosakkarideille, 89 % rikkihapolle ja 100 % etikkahapolle.

Many attempts have been made to establish the control of foodborne pathogens through Lactobacillus isolates and their metabolism products with success being obtained in several situations. The aim of this study was to investigate the antagonistic effect of eight Lactobacillusisolates, including L. caseisubsp. pseudoplantarum,L. plantarum, L. reuteri and L. delbrueckii subsp. delbrueckii, on the pathogenic Escherichia colistrain O157:H7. The inhibitory effect of pure cultures and two pooled cultures supernatants of Lactobacillus on the growth of pathogenic bacteria was evaluated by the spot agar method and by monitoring turbidity. Antimicrobial activity was confirmed for L. reuteri and L. delbrueckii subsp. delbrueckii and for a pool of lactic acid bacteria. The neutralized supernatant of the pool exerted a higher antimicrobial activity than that of the individual strains. Furthermore, D-lactic acid and acetic acid were produced during growth of the Lactobacillus isolates studied.
