Lychee (Litchi chinensis Sonn.) has a high commercial value; however, it has a short shelf-life because of its rapid pericarp browning. The objective of this study was to evaluate the shelf-life of 'Bengal' lychee fruits stored after treatment with hydrochloric acid and citric acid, associated with cassava starch and plastic packaging. Uniformly red pericarp fruits were submitted to treatments: 1-(immersion in citric acid 100 mM for 5 minutes + cassava starch 30 g L-1 for 5 minutes), 2-(immersion in hydrochloric acid 1 M for 2 minutes + starch cassava 30 g L-1 for 5 minutes), 3-(immersion in citric acid 100 mM for 5 minutes + polyvinyl chloride film (PVC, 14 µm thick)) and 4-(immersion in hydrochloric acid 1 M for 2 minutes + PVC film). During 20 days, the fruits were evaluated for mass loss, pericarp color, pH, soluble solids and titratable acidity, vitamin C of the pulp and pericarp and activities of polyphenol oxidase and peroxidase of the pericarp. The treatment with hydrochloric acid associated with PVC was the most effective in maintaining the red color of the pericarp for a period of 20 days and best preservation of the fruit. The cassava starch associated with citric acid, and hydrochloric acid did not reduce the mass loss and did not prevent the browning of lychee fruit pericarp.

As espécies reativas de oxigénio (ROS) estão envolvidas no desenvolvimento de dor neuropática. No entanto, a aplicação clínica de moléculas antioxidantes no tratamento desta patologia tem demonstrado pouca eficácia. A inibição da NADPH oxidase (NOX), uma das principais fontes de ROS, poderá ser uma boa estratégia terapêutica. O nosso grupo verificou que a apocinina (inibidor da NOX) melhora parcialmente os sintomas de dor neuropática e a disfunção redox espinhal no modelo SNI (spared nerve injury). De forma a melhorar este efeito terapêutico, o presente estudo insere-se num projeto maior, que visa identificar as isoformas da NOX envolvidas na fisiopatologia da doença e avaliar o efeito da administração de inibidores específicos para essas isoformas. Assim, propusemo-nos a avaliar a disfunção redox espinhal em fases precoces dador neuropática periférica induzida pelo modelo SNI no Rato, relacionando-a com os comportamentos de dor demonstrados pelos animais. Foram constituídos três grupos experimentais: SNI, sham e naïve, com subgrupos testados e sacrificados aos dias 1, 3, 7 e 14 após a cirurgia. Avaliou-se a sensibilidade mecânica (vonFrey e pinprick) e ao frio (acetona) dos animais, sacrificaram-se e recolheram-se as medulas espinhais para análise imunohistoquímica, com marcadores de dano oxidativo no DNA e de dano nitrosativo. Ao contrário dos animais sham, que demonstraram um comportamento muito próximo dos naïve, os animais SNI desenvolveram alodínia mecânica e ao frio e hiperalgesia mecânica na pata ipsilateral. No entanto, o dano oxidativo no corno dorsal ipsilateral da medula espinhal apresentou-se idêntico nos grupos SNI e sham ao longo dos 14 dias de estudo, não havendo também diferenças entre os cornos ipsi e contralateral à lesão nervosa. É possível que o desenvolvimento de dor neuropática nos animais SNI não se faça acompanhar de disfunção redox espinhal, pelo menos até aos 14 dias pós indução. O facto de a lesão nervosa no modelo SNI se localizar numa porção distal do ciático, ao contrário de outros modelos em que o stresse oxidativo espinhal foi já descrito, poderia explicar essas diferenças. Em todo o caso, considerando que os resultados comportamentais obtidos indicam que as cirurgias SNI e sham causam diferentes níveis de sensibilização nos animais, parece-nos fulcral prolongar os tempos de neuropatia, e executar uma avaliação do estado redox com outros marcadores, de forma a elucidar se, de facto, existem ROS envolvidas nesta sensibilização e, em caso positivo, poder identificar essas espécies, bem como as suas fontes.

Aflatoxins were first isolated about 40 years ago afier outbreaks of disease and death in turkeys and cancer in rainbow trout fed with rations formulated from peanut and cottonseed meals. These toxins are secondary metabolites produced under certain conditions of temperature, p14 and humidity predominantiy by Aspergilius flavus and Aspergilius parasiticus fungi species. Among 18 different types of aflatoxins identified, major members are aflatoxin B1, B2, G1 and G2. Aflatoxin B1 (AFB1) is normaily predominant in cultures as well as in food products. AFB1 was shown to be genotoxic and a potent hepatocarcinogen. This mycotoxin is metabolized by the mixed function oxidase system to a number of hydroxylated metabolites including the 8,9-epoxide. The latter is considered to be the ultimate carcinogen that reacts with cellular deoxyribonucleic acid (DNA) and proteins to form covalent adducts.

The development of a FIA system for the determination of total choline content in several types of milk is described. The samples were submitted to hydrochloric acid digestion before injection into the system and passed through an enzymatic reactor containing choline oxidase immobilised on glass beads. This enzymatic reaction releases hydrogen peroxide which then reacts with a solution of iodide. The decrease in the concentration of iodide ion is quantified using an iodide ion selective tubular electrode based on a homogeneous crystalline membrane. Validation of the results obtained with this system was performed by comparison with results from a method described in the literature and applied to the determination of total choline in milks. The relative deviation was always < 5%. The repeatability of the method developed was assessed by calculation of the relative standard deviation (RSD) for 12 consecutive injections of one sample. The RSD obtained was < 0.6%.

Iron plays a central role in host-parasite interactions, since both intervenients need iron for survival and growth, but are sensitive to iron-mediated toxicity. The host’s iron overload is often associated with susceptibility to infection. However, it has been previously reported that iron overload prevented the growth of Leishmania major, an agent of cutaneous leishmaniasis, in BALB/c mice. In order to further clarify the impact of iron modulation on the growth of Leishmania in vivo, we studied the effects of iron supplementation or deprivation on the growth of L. infantum, the causative agent of Mediterranean visceral leishmaniasis, in the mouse model. We found that dietary iron deficiency did not affect the protozoan growth, whereas iron overload decreased its replication in the liver and spleen of a susceptible mouse strain. The fact that the iron-induced inhibitory effect could not be seen in mice deficient in NADPH dependent oxidase or nitric oxide synthase 2 suggests that iron eliminates L. infantum in vivo through the interaction with reactive oxygen and nitrogen species. Iron overload did not significantly alter the mouse adaptive immune response against L. infantum. Furthermore, the inhibitory action of iron towards L. infantum was also observed, in a dose dependent manner, in axenic cultures of promastigotes and amastigotes. Importantly, high iron concentrations were needed to achieve such effects. In conclusion, externally added iron synergizes with the host’s oxidative mechanisms of defense in eliminating L. infantum from mouse tissues. Additionally, the direct toxicity of iron against Leishmania suggests a potential use of this metal as a therapeutic tool or the further exploration of iron anti-parasitic mechanisms for the design of new drugs.

40 Echinococcus isolates from sheep and cattle in Southern Brazil were genetically analysed in order to obtain further data on the presence of different taxa of the Echinococcus granulosus complex. Differentiation was done using a PCR technique and sequencing of mitochondrial cytochrome c oxidase subunit 1 (CO1). Most samples (38) could be allocated to the sheep strain (G1) of E. granulosus, while two samples belonged to E. ortleppi, previously known as cattle strain (G5) of E. granulosus. Due to the shorter prepatent period in dogs of the latter taxon, this records have important implications for the design of control measures in this endemic region.

Introdução: Na criança e no adolescente os episódios depressivos podem apresentar-se como tristeza generalizada, anedonia (característica bastante típica da 1ªInfância), tédio ou irritabilidade (que substitui muitas vezes o humor depressivo) e sentimento de indiferença perante actividades que anteriormente causavam prazer. É a presença de comprometimento funcional que distingue a depressão dos “altos e baixos” normais da infância e adolescência. Estima-se que a prevalência de depressão na 2ª Infância seja de 1-2%. Esta prevalência aumenta proporcionalmente com a idade, podendo atingir, no fim da adolescência, os 20%. O diagnóstico pode ser difícil: por vezes a irritabilidade e as alterações do comportamento dominam o quadro clínico. Objectivos: Este trabalho tem como principais objectivos fazer uma revisão das teorias biólogicas existentes e abordar, sucintamente, o tratamento farmacológico da depressão, dando ênfase aos fármacos mais utilizados em Psiquiatria da Infância e da Adolescência. Resumo: Trata-se de um trabalho de revisão bibliográfica onde serão abordados conceitos farmacológicos chave como as definições de Resposta, Remissão, Recuperação, Recaída, Recorrência, bem como o funcionamento dos neurónios e circuitos monoaminérgicos (noradrenérgicos, serotoninérgicos e dopaminérgicos). Nas bases biológicas da depressão serão enfatizadas as teorias da monoamina e dos receptores de neurotransmissores. Abordar-se-á de modo sucinto a influência dos receptores de monoaminas na transdução de sinal e regulação da expressão de genes. O vasto grupo farmacológico dos antidepressivos engloba, entre outros, os inibidores da monoamina oxidase, os antidepressivos tricíclicos e os inibidores selectivos da recaptação da serotonina. Não se pretendendo uma revisão exaustiva, tentar-se-á salientar os grupos mais importantes e os novos antidepressivos. Sendo a Infância e a Adolescência uma faixa etária para a qual existem poucos estudos de eficácia vs efeitos secundários dos antidepressivos, tentaremos salientar os fármacos melhor estudados e aqueles que estão recomendados.

Cystic echinococcosis (CE) is a globally parasitic zoonosis caused by larval stages of Echinococcus granulosus. This study investigated E. granulosus genotypes isolated from livestock and humans in the Golestan province, northern Iran, southeast of the Caspian sea, using partial sequencing data of the cytochrome c oxidase subunit 1 (cox1) and NADH dehydrogenase 1 (nad1) mitochondrial genes. Seventy E. granulosus isolates were collected from animals in slaughterhouses: 18 isolates from sheep, 40 from cattle, nine from camels, two from buffaloes and one from a goat, along with four human isolates (formalin-fixed, paraffin-embedded tissues) from CE patients of provincial hospitals. All isolates were successfully analysed by PCR amplification and sequencing. The sequence analysis found four E. granulosus genotypes among the 74 CE isolates: G1 (78.3%), G2 (2.7%), G3 (15%) and G6 (4%). The G1-G3 complex genotype was found in all of the sheep, goat, cattle and buffalo isolates. Among the nine camel isolates, the frequency of G1-G3 and G6 genotypes were 66.7% and 33.3%, respectively. All four human CE isolates belonged to E. granulosus sensu stricto. This study reports the first occurrence of the G2 genotype in cattle from Iran and confirms the previously reported G3 genotype in camels in the same country.

In visceral leishmaniasis, the detection of the agent is of paramount importance to identify reservoirs of infection. Here, we evaluated the diagnostic attributes of PCRs based on primers directed to cytochrome-B (cytB), cytochrome-oxidase-subunit II (coxII), cytochrome-C (cytC), and the minicircle-kDNA. Although PCRs directed to cytB, coxII, cytC were able to detect different species of Leishmania, and the nucleotide sequence of their amplicons allowed the unequivocal differentiation of species, the analytical and diagnostic sensitivity of these PCRs were much lower than the analytical and diagnostic sensitivity of the kDNA-PCR. Among the 73 seropositive animals, the asymptomatic dogs had spleen and bone marrow samples collected and tested; only two animals were positive by PCRs based on cytB, coxII, and cytC, whereas 18 were positive by the kDNA-PCR. Considering the kDNA-PCR results, six dogs had positive spleen and bone marrow samples, eight dogs had positive bone marrow results but negative results in spleen samples and, in four dogs, the reverse situation occurred. We concluded that PCRs based on cytB, coxII, and cytC can be useful tools to identify Leishmania species when used in combination with automated sequencing. The discordance between the results of the kDNA-PCR in bone marrow and spleen samples may indicate that conventional PCR lacks sensitivity for the detection of infected dogs. Thus, primers based on the kDNA should be preferred for the screening of infected dogs.

Descrevemos um lactente com doença neurológica grave caracterizada por convulsões mioclónicas e tónicas, com início no período neonatal, refractárias a vários anticonvulsivantes, assim como, tetraparésia espástica. A tomografia computorizada e a ressonância magnética cerebrais evidenciaram imagens de leucomalácia periventricular e, posteriormente, de atrofia cerebral progressiva e encefalomalácia quística. Os exames bioquímicos e o estudo da actividade enzimática permitiram o diagnóstico de défice do cofactor molibdénio. O défice do cofactor molibdénio é uma doença rara, autossómica recessiva, que se comporta como um défice combinado da sulfito oxidase e da xantina desidrogenase (ou xantina oxidase) alterando o metabolismo das purinas e da cisteína. A terapêutica é controversa e o prognóstico reservado. O nosso objectivo é relembrar esta patologia no diagnóstico diferencial das convulsões neonatais e da encefalopatia hipóxico- -isquémica, sobretudo quando os exames imagiológicos sugerem lesões de leucomalácia no recém-nascido de termo. Salientamos a importância deste diagnóstico diferencial, apesar do prognóstico pobre, devido à possibilidade de aconselhamento genético adequado e diagnóstico pré-natal.
