The objective of this work was to propose a new selection strategy for the initial stages of sugarcane improvement, based on the methodology 'simulated individual BLUP (BLUPIS)', which promotes a dynamic allocation of individuals selected in each full-sib family, using BLUP as a base for both the genotypic effects of the referred families and plot effects. The method proposed applies to single full-sib families or those obtained from unbalanced or balanced diallel crosses, half-sib families and self-pollinated families. BLUPIS indicates the number of individuals to be selected within each family, the total number of clones to be advanced, and the number of families to contribute with selected individuals. Correlation between BLUPIS and true BLUP was 0.96, by method validation. Additionally, BLUPIS allows the identification of which replication contains the best individuals of each family.

We face the problem of characterizing the periodic cases in parametric families of (real or complex) rational diffeomorphisms having a fixed point. Our approach relies on the Normal Form Theory, to obtain necessary conditions for the existence of a formal linearization of the map, and on the introduction of a suitable rational parametrization of the parameters of the family. Using these tools we can find a finite set of values p for which the map can be p-periodic, reducing the problem of finding the parameters for which the periodic cases appear to simple computations. We apply our results to several two and three dimensional classes of polynomial or rational maps. In particular we find the global periodic cases for several Lyness type recurrences

O objetivo deste trabalho foi avaliar o efeito da aplicação de zinco em sementes de sorgo (Sorghum bicolor (L.) Moench) na nutrição, na germinação e no desenvolvimento inicial das plantas, em casa de vegetação. Utilizou-se delineamento inteiramente casualizado dos tratamentos, em esquema fatorial 2x5, duas cultivares de sorgo (BR 304 e BR 310) e cinco doses de Zn (0, 3,57, 7,14, 14,28 e 28,56 g de Zn por kg de sementes), com três repetições. Foram avaliadas as variáveis de germinação (tempo médio, média diária e a velocidade), e 28 dias depois da semeadura, determinou-se a massa da matéria seca e os teores de Zn na parte aérea e nas raízes. A aplicação de Zn nas sementes de sorgo não afetou o acúmulo de matéria seca da parte aérea, entretanto, diminuiu a germinação e o acúmulo de matéria seca das raízes e da planta inteira. O Zn aplicado nas sementes de sorgo acumulou-se principalmente nas raízes. A cultivar BR 304 apresentou maior absorção de Zn e acúmulo de matéria seca, em relação à cultivar BR 310.

Es tracta d’una obra fruit d’un conveni de col·laboració signat l’any 2004 entre la URV, l’ICAC i Repsol YPF, i resultat d’un projecte de recerca de l’ICAC inclòs en la línia de recerca Arqueologia del paisatge, poblament i territori, que ha comptat amb el cofinançament de Repsol YPF i la col·laboració de Codex. Aquesta obra se centra en les excavacions arqueològiques efectuades en una vil·la romana situada entre la via Tarraco-Ilerda i la llera del riu Francolí, a l’actual terme municipal del Morell. És un assentament datat entre els segles II aC i V dC que reflecteix els patrons d’ocupació romana a l’àrea central de l’ager Tarraconensis i complementa, a partir d’una exhaustiva anàlisi, les dades històriques que, fins a l’actualitat, oferien les vil·les de Centcelles a Constantí o de Paret Delgada a la Selva del Camp. L’estudi parteix de les excavacions que l’empresa Codex hi va efectuar l’any 1996 arran d’una actuació d’urgència motivada per la construcció d’una nova planta industrial de l’aleshores Repsol Química SA. L’obra és fruit de l’estudi de la documentació, fins ara inèdita, elaborada per l’empresa Codex i que ha estat cedida per a aquest projecte de recerca. La publicació ha estat coordinada per Josep M. Macias Solé i Joan J. Menchon Bes i els textos són obra del Dr. Domènec Campillo, M. Milagros Cuesta i Laura Devenat (Laboratori de Paleopatologia i Paleoantropologia, Museu d’Arqueologia de Catalunya), Montserrat García Noguera (Codex – Arqueologia i Patrimoni), Miguel Á. L’objectiu del projecte és realitzar la instal•lació d’enllumenat per a un tram del’autovia de Palafrugell a Calella de Palafrugell, aquesta instal•lació també disposaràd’una instal•lació de microgeneració. On a cada farola s’hi acoblarà un aerogeneradord’eix vertical de 4 kW per a la producció d’energia elèctrica.Com no es disposa de faroles prefabricades que compleixin aquesta funció desuport per a lluminàries i un aerogenerador, en aquest projecte també es faràal disseny de la farola.L’actuació prevista és la instal•lació de 12 faroles de nou dissenyamb un aerogenerador d’eix vertical acoblat a cada farola, en l’ espai de la mitjana de l’autovia

Inflammation participates in tissue repair through multiple mechanisms including directly regulating the cell fate of resident progenitor cells critical for successful regeneration. Upon surveying target cell types of the TNF ligand TWEAK, we observed that TWEAK binds to all progenitor cells of the mesenchymal lineage and induces NF-kappaB activation and the expression of pro-survival, pro-proliferative and homing receptor genes in the mesenchymal stem cells, suggesting that this pro-inflammatory cytokine may play an important role in controlling progenitor cell biology. We explored this potential using both the established C2C12 cell line and primary mouse muscle myoblasts, and demonstrated that TWEAK promoted their proliferation and inhibited their terminal differentiation. By generating mice deficient in the TWEAK receptor Fn14, we further showed that Fn14-deficient primary myoblasts displayed significantly reduced proliferative capacity and altered myotube formation. Following cardiotoxin injection, a known trigger for satellite cell-driven skeletal muscle regeneration, Fn14-deficient mice exhibited reduced inflammatory response and delayed muscle fiber regeneration compared with wild-type mice. These results indicate that the TWEAK/Fn14 pathway is a novel regulator of skeletal muscle precursor cells and illustrate an important mechanism by which inflammatory cytokines influence tissue regeneration and repair. Coupled with our recent demonstration that TWEAK potentiates liver progenitor cell proliferation, the expression of Fn14 on all mesenchymal lineage progenitor cells supports a broad involvement of this pathway in other tissue injury and disease settings.

Partial nucleotide sequences of five tomato infecting Begomovirus isolates were determined from DNA-A fragments, corresponding to the 5' region of the replication associated protein gene, the intergenic region and the 5' region of the coat protein gene. Isolate DFM shared 95% identity with Tomato mottle leaf curl virus (TMoLCV), isolates 34, PA-05, and Ta4 were 88% identical to Tomato yellow vein streak virus and isolate DF-BR3 shared 77% identity with TMoLCV. Recombination analysis indicated that isolate DF-BR3 was a chimaera, and it provided evidence that there is a complex and actively recombining population of tomato infecting begomoviruses in Brazil.

La medicina al llarg de la història ha anat evolucionant a mesura que també ho anava fent la societat. És per això que en el darrer segle existeix una medicina que no està orientada a la curació de la patologia, sinó que tracta d’aconseguir la major qualitat de vida del pacient ancià o jove, amb patologies cròniques, en estadi de final de la malaltia i també en pacients oncològics i no oncològics, és a dir, s’intenta prestar una atenció sanitària, no curativa, als que tenen una malaltia avançada, progressiva i incurable que precisen una sèrie d’intervencions que no només es basen en procediments mèdics, sinó també en socials i psicològics. És per la necessitat que la medicina s’adaptés a les demandes de la societat que al 1990 es crea a Catalunya el Pla de Cures Pal·liatives de Catalunya com a programa pilot de l’Organització Mundial de la Salut (OMS). Aquest programa ha esdevingut tot un referent de les cures pal·liatives (CP) a España, és el Programa d’Atenció Domiciliària i Equips de Suport (PADES) on un equip interdisciplinari (metge, infermer, treballador social, fisioterapeuta i psicòleg) treballen per tenir una cura holística del pacient al seu domicili. Així, doncs, l’objectiu del PADES és alleugerir el patiment i millorar, tant com es pugui, la qualitat de vida i acompanyar-los en el procés de mort al seu domicili.

Plasmodium sporozoites traverse several host cells before infecting hepatocytes. In the process, the plasma membranes of the cells are ruptured, resulting in the release of cytosolic factors into the microenvironment. This released endogenous material is highly stimulatory/immunogenic and can serve as a danger signal initiating distinct responses in various cells. Thus, our study aimed at characterizing the effect of cell material leakage during Plasmodium infection on cultured mouse primary hepatocytes and HepG2 cells. We observed that wounded cell-derived cytosolic factors activate NF-kappaB, a main regulator of host inflammatory responses, in cells bordering wounded cells, which are potential host cells for final parasite infection. This activation of NF-kappaB occurred shortly after infection and led to a reduction of infection load in a time-dependent manner in vitro and in vivo, an effect that could be reverted by addition of the specific NF-kappaB inhibitor BAY11-7082. Furthermore, no NF-kappaB activation was observed when Spect(-/-) parasites, which are devoid of hepatocyte traversing properties, were used. We provide further evidence that NF-kappaB activation causes the induction of inducible NO synthase expression in hepatocytes, and this is, in turn, responsible for a decrease in Plasmodium-infected hepatocytes. Furthermore, primary hepatocytes from MyD88(-/-) mice showed no NF-kappaB activation and inducible NO synthase expression upon infection, suggesting a role of the Toll/IL-1 receptor family members in sensing cytosolic factors. Indeed, lack of MyD88 significantly increased infection in vitro and in vivo. Thus, host cell wounding due to parasite migration induces inflammation which limits the extent of parasite infection

Chikungunya virus (CHIKV) is the causative agent of an outbreak that began in La Réunion in 2005 and remains a major public health concern in India, Southeast Asia, and southern Europe. CHIKV is transmitted to humans by mosquitoes and the associated disease is characterized by fever, myalgia, arthralgia, and rash. As viral load in infected patients declines before the appearance of neutralizing antibodies, we studied the role of type I interferon (IFN) in CHIKV pathogenesis. Based on human studies and mouse experimentation, we show that CHIKV does not directly stimulate type I IFN production in immune cells. Instead, infected nonhematopoietic cells sense viral RNA in a Cardif-dependent manner and participate in the control of infection through their production of type I IFNs. Although the Cardif signaling pathway contributes to the immune response, we also find evidence for a MyD88-dependent sensor that is critical for preventing viral dissemination. Moreover, we demonstrate that IFN-alpha/beta receptor (IFNAR) expression is required in the periphery but not on immune cells, as IFNAR(-/-)-->WT bone marrow chimeras are capable of clearing the infection, whereas WT-->IFNAR(-/-) chimeras succumb. This study defines an essential role for type I IFN, produced via cooperation between multiple host sensors and acting directly on nonhematopoietic cells, in the control of CHIKV.

O objetivo deste trabalho foi avaliar uma alternativa para a quantificação simultânea de sulfatiazol, sulfametazina e sulfadimetoxina no leite, na forma de azoderivados, por meio de cromatografia líquida de alta eficiência (CLAE). O isolamento e concentração das sulfas foram conduzidos por extração em fase sólida (C18), com metanol como eluente. Após a extração, os analitos foram transformados em sais de diazônio e submetidos ao acoplamento com resorcinol. Os derivados foram separados e quantificados por CLAE (coluna C8, λ = 430 nm, MeOH/KH2PO4 aq.). Foram obtidas recuperações entre 65 e 78%.

Rapid amplification of cDNA ends (RACE) is a widely used approach for transcript identification. Random clone selection from the RACE mixture, however, is an ineffective sampling strategy if the dynamic range of transcript abundances is large. To improve sampling efficiency of human transcripts, we hybridized the products of the RACE reaction onto tiling arrays and used the detected exons to delineate a series of reverse-transcriptase (RT)-PCRs, through which the original RACE transcript population was segregated into simpler transcript populations. We independently cloned the products and sequenced randomly selected clones. This approach, RACEarray, is superior to direct cloning and sequencing of RACE products because it specifically targets new transcripts and often results in overall normalization of transcript abundance. We show theoretically and experimentally that this strategy leads indeed to efficient sampling of new transcripts, and we investigated multiplexing the strategy by pooling RACE reactions from multiple interrogated loci before hybridization.

Cyclooxygenase-derived prostaglandin E(2) (PGE(2)) is the predominant prostanoid found in most colorectal cancers (CRC) and is known to promote colon carcinoma growth and invasion. However, the key downstream signaling pathways necessary for PGE(2)-induced intestinal carcinogenesis are unclear. Here we report that PGE(2) indirectly transactivates PPARdelta through PI3K/Akt signaling, which promotes cell survival and intestinal adenoma formation. We also found that PGE(2) treatment of Apc(min) mice dramatically increased intestinal adenoma burden, which was negated in Apc(min) mice lacking PPARdelta. We demonstrate that PPARdelta is a focal point of crosstalk between the prostaglandin and Wnt signaling pathways which results in a shift from cell death to cell survival, leading to increased tumor growth.

The objective of this study was to produce citrus somatic asymmetric hybrids by fusing gamma-irradiated protoplasts with iodoacetamide-treated protoplasts. Protoplasts were isolated from embryogenic suspension cells of grapefruit (Citrus paradisi Macfad.) cultivars Ruby Red and Flame, sweet oranges (C. sinensis Osbeck) 'Itaboraí', 'Natal', Valencia', and 'Succari', from 'Satsuma' (C. unshiu Marcow.) and 'Changsha' mandarin (C. reticulata Blanco) and 'Murcott' tangor (C. reticulata x C. sinensis). Donor protoplasts were exposed to gamma rays and receptor protoplasts were treated with 3 mmol L-1 iodoacetamide (IOA), and then they were fused for asymmetric hybridization. Asymmetric embryos were germinated, and the resulting shoots were either grafted onto sour orange, rough lemon or 'Swingle' (C. paradisi x Poncirus trifoliata) x 'Sunki' mandarin rootstock seedlings, or rooted after dipping their bases in indol-butyric acid (IBA) solution. The products were later acclimatized to greenhouse conditions. Ploidy was analyzed by flow cytometry, and hybridity was confirmed by amplified fragment length polymorphism (AFLP) analysis of plantlet DNAsamples. The best treatment was the donor-recipient fusion combination of 80 Gy-irradiated 'Ruby Red' protoplasts with 20 min IOA-treated 'Succari' protoplasts. Tetraploid and aneuploid plants were produced. Rooting recalcitrance was solved by dipping shoots' stems in 3,000 mg L-1 IBA solution for 10 min.

O objetivo deste trabalho foi avaliar uma metodologia de análise de adaptabilidade e estabilidade fenotípica de genótipos de café baseada em regressão não paramétrica. A técnica utilizada difere das demais, pois reduz a influência na estimação do parâmetro de adaptabilidade de algum ponto extremo, ocasionado pela presença de genótipos com respostas demasiadamente diferenciadas a determinado ambiente. Foram utilizados dados provenientes de um experimento sobre produtividade média de grãos de 40 genótipos de café (Coffea canephora), com delineamento em blocos ao acaso, com seis repetições. Os genótipos foram avaliados em cinco anos (1996, 1998, 1999, 2000 e 2001), em dois locais (Sooretama e Marilândia, ES) no total de dez ambientes. A metodologia proposta demonstrou ser adequada e eficiente, pois extingue os efeitos impróprios induzidos pela presença de pontos extremos e evita a recomendação incorreta de genótipos quanto à adaptabilidade.
