Among the various possible embodiements of Advanced Therapies and in particular of Tissue Engineering the use of temporary scaffolds to regenerate tissue defects is one of the key issues. The scaffolds should be specifically designed to create environments that promote tissue development and not merely to support the maintenance of communities of cells. To achieve that goal, highly functional scaffolds may combine specific morphologies and surface chemistry with the local release of bioactive agents. Many biomaterials have been proposed to produce scaffolds aiming the regeneration of a wealth of human tissues. We have a particular interest in developing systems based in nanofibrous biodegradable polymers1,2. Those demanding applications require a combination of mechanical properties, processability, cell-friendly surfaces and tunable biodegradability that need to be tailored for the specific application envisioned. Those biomaterials are usually processed by different routes into devices with wide range of morphologies such as biodegradable fibers and meshes, films or particles and adaptable to different biomedical applications. In our approach, we combine the temporary scaffolds populated with therapeutically relevant communities of cells to generate a hybrid implant. For that we have explored different sources of adult and also embryonic stem cells. We are exploring the use of adult MSCs3, namely obtained from the bone marrow for the development autologous-based therapies. We also develop strategies based in extra-embryonic tissues, such as amniotic fluid (AF) and the perivascular region of the umbilical cord4 (Whartonâ s Jelly, WJ). Those tissues offer many advantages over both embryonic and other adult stem cell sourcess. These tissues are frequently discarded at parturition and its extracorporeal nature facilitates tissue donation by the patients. The comparatively large volume of tissue and ease of physical manipulation facilitates the isolation of larger numbers of stem cells. The fetal stem cells appear to have more pronounced immunomodulatory properties than adult MSCs. This allogeneic escape mechanism may be of therapeutic value, because the transplantation of readily available allogeneic human MSCs would be preferable as opposed to the required expansion stage (involving both time and logistic effort) of autologous cells. Topics to be covered: This talk will review our latest developments of nanostructured-based biomaterials and scaffolds in combination with stem cells for bone and cartilage tissue engineering.

La síntesis y estudios de hidrogeles está siendo muy desarrollada desde que se conoce su potencial aplicación como carriers para medicamentos, membranas hidrofílicas para separación, en agricultura, para usarse como complemento de suelos áridos por su capacidad de retener agua y en liberación controlada de herbicidas, fertilizantes, abonos, etc. Para su obtención, se suelen usar reacciones de copolimerización a radicales libres entre monómeros hidrofílicos y agentes entrecruzantes, variando proporciones relativas de cada compuesto y demás condiciones experimentales (temperatura, tiempo, agente porógeno, etc.). Así, se pueden lograr productos con propiedades mecánicas y de difusión distintas, como por ejemplo, polímeros con fuerza y elongación, o rigidez y resistencia y que sean capaces de absorber su propio peso de agua. En base a la experiencia adquirida en nuestro grupo de trabajo en síntesis de geles que forman soportes poliméricos con propiedades concretas de aplicación en el área de afinidad cromatográfica y, conociendo algunas variables (ciclización por reacciones intramoleculares, problemas de impedimento esterico) que afectan la obtención de buenos rendimientos de productos reticulados y las propiedades finales de los productos, proponemos realizar síntesis de hidrogeles que pueden ser usados como matriz polimérica en aplicaciones específicas. Para ello, como objetivo general se plantea: * Realizar el estudio de las variables que afectan los procesos de gelación y la influencia que tienen los mismos sobre las propiedades de los productos. Como objetivo específico se plantea: * Llevar a cabo la obtención de geles hidrofílicos realizando estudios de copolimerización radicalaría de comonomeros como acrilamida (AAm), ácido acrílico (AAm), hidroxietilmetacrilato (HEMA) y agentes entrecruzantes como etilenglicoldimetacrilato (EGDM). Así, una vez conocidas las condiciones óptimas y la metodología apropiada para los procesos de gelación, se comenzará con los estudios para la obtención de hidrogeles a partir de nuevos monómeros polifuncionales sintetizados en nuestros laboratorios o a partir de monómeros tradicionales modificados químicamente.

Muchos esfuerzos se están realizando en el diseño de nuevos métodos para la eliminación de las células tumorales y así inhibir el crecimiento neoplásico. Entre los métodos no convencionales se encuentran la Terapia Fotodinámica.La Terapia Fotodinámica (TFD) es un tratamiento experimental de algunos tipos de cáncer, basado en el efecto citotóxico inducido en el tejido tumoral, por la acción combinada de una droga (fotosensibilizador) y la luz visible. El fotosensibilizador posee la propiedad de absorber la luz y reaccionar con el oxígeno molecular, produciendo una forma activa del oxígeno: el oxígeno singlete (1O2) que oxida diversas moléculas biológicas, induciendo un efecto citotóxico que se traduce en la regresión tumoral. Los nuevos avances en la dosimetría de la luz, así como la búsqueda de una segunda generación de nuevos fotosensibilizadores más eficaces que los actualmente utilizados, han permitido incluir protocolos de Terapia Fotodinámica en numerosos centros hospitalarios principalmente para el tratamiento de cánceres de pulmón, vejiga, esófago y piel. Plantas fototóxicas, sus metabolitos fotosensibilizantes y sus posibles usos; En general, dentro de las especies vegetales tóxicas existen aquellas denominadas plantas alergénicas, que son las que pueden producir sus efectos indeseables por vía dérmica. También existen aquellas que pueden producir efectos tóxicos por vía sistémica. Sin embargo, coexiste en la naturaleza otro grupo de plantas tóxicas que desencadenan sus efectos nocivos bajo la acción de la luz, por lo que son llamadas plantas fototóxicas, cuyos principios activos son comúnmente denominados agentes fotosensibilizantes La apoptosis como blanco terapéutico contra el cáncer: Los conocimientos moleculares sobre la apoptosis adquiridos en los últimos años están siendo aplicados al desarrollo de nuevos fármacos que puedan modular selectivamente las señales involucradas en la muerte de las células. Una de las razones que justifica el interés en el estudio de este tipo de moléculas, es que una de las características más tempranas en la transformación de la células neoplásicas esta relacionada con la incapacidad de responder a los estímulos de muerte. Esto lleva a una desregulación del proceso de apoptosis desencadenando una proliferación descontrolada. Los otros eventos que desencadenan el cáncer son, la invasión vascular y la metástasis a distanciaLa adquisición de resistencia a los efectos citotóxicos de los tratamientos anticancerígenos ha emergido como un significante impedimento para el efectivo tratamiento de la enfermedad. Por ello, en el presente proyecto se investigará si la adquisición de resistencia a TFD inducida en la línea celular estudiada es conferida por el aumento de la proteína MDRP1 a través de la vía de señalización PI3K/Akt. Además, se estudiará la correlación entre la posible resistencia a drogas y la inducción de apoptosis, analizando los mecanismos involucrados. Los resultados obtenidos contribuirán a dilucidar y entender los mecanismos moleculares implicados en la resistencia y sensibilidad tumoral a la TFD, y de esta manera mejorar la eficacia de dicha terapia antitumoral para sensibilizar a las células a la apoptosis. OBJETIVOS Estudiar el efecto de agentes fotosensibilizadores de origen sintético (ftalocianinas), comercialmente ya aprobadas por la FDA (Me-ALA), de origen natural (antraquinonas), y obtenidas en procesos nanotecnologicos (nanofibras) respecto a su capacidad de inducir la muerte celular en sistemas experimentales in vivo, para el desarrollo de nuevas drogas de aplicación en Terapia Fotodinámica (PDT). Estudiar las señales de apoptosis que se desencadenan, combinando la PDT con iRNA (antisurvivina) con la finalidad de aumentar la eficiencia de la muerte tumoral. Estudiar los mecanismos de resistencia a la Terapia Fotodinámica en carcinoma de células escamosas con fotosensibilizadores permitidos (Me-ALA).

El presente estudio establece una línea de investigación que permite iniciar el planteamiento de las bases de un programa de acción para determinar una serie de políticas y estrategias de integración de la población inmigrante, orientadas a su inserción en zonas rurales que necesiten conservar o mejorar su situación socioeconómica actual. Dicha situación está principalmente amenazada por la despoblación y, en algunos casos, también por la disminución –o al menos el cambio de condiciones– de la percepción de los Fondos Estructurales europeos. Con esta investigación se pretende conceptualizar el tema y tratar de detectar la posibilidad y la disposición para absorber contingentes de población inmigrante por parte de algunas zonas rurales de Cataluña. En una segunda fase, se procederá a la determinación de zonas concretas con capacidad y disponibilidad de infraestructuras suficientes como para ser destino de inmigrantes que posean una formación específica y la predisposición necesaria para garantizar su adaptación al medio rural de este país.

Methicillin resistant Staphylococcus aureus (MRSA) bacteria have emerged in the early 1980's in numerous health care institutions around the world. The main transmission mechanism within hospitals and healthcare facilities is through the hands of health care workers. Resistant to several antibiotics, the MRSA is one of the most feared pathogens in the hospital setting since it is very difficult to eradicate with the standard treatments. There are still a limited number of anti-MRSA antibiotics but the first cases of resistance to these compounds have already been reported and their frequency is likely to increase in the coming years. Every year, the MRSA infections result in major human and financial costs, due to the high associated mortality and expenses related to the required care. Measures towards a faster detection of resistant bacteria and establishment of appropriate antibiotic treatment parameters are fundamental. Also as part as infection prevention, diminution of bacteria present on the commonly touched surfaces could also limit the spread and selection of antibiotic resistant bacteria. During my thesis, projects were developed around MRSA and antibiotic resistance investigation using innovative technologies. The thesis was subdivided in three main parts with the use of atomic force microscopy AFM for antibiotic resistance detection in part 1, the importance of the bacterial inoculum size in the selection of antibiotic resistance in part 2 and the testing of antimicrobial surfaces creating by sputtering copper onto polyester in part 3. In part 1 the AFM was used two different ways, first for the measurement of stiffness (elasticity) of bacteria and second as a nanosensor for antibiotic susceptibility testing. The stiffness of MRSA with different susceptibility profiles to vancomycin was investigated using the stiffness tomography mode of the AFM and results have demonstrated and increased stiffness in the vancomycin resistant strains that also paralleled with increased thickness of the bacterial cell wall. Parts of the AFM were also used to build a new antibiotic susceptibility-testing device. This nano sensor was able to measure vibrations emitted from living bacteria that ceased definitively upon antibiotic exposure to which they were susceptible but restarted after antibiotic removal to which they were resistant, allowing in a matter of minute the assessment of antibiotic susceptibility determination. In part 2 the inoculum effect (IE) of vancomycin, daptomycin and linezolid and its importance in antibiotic resistance selection was investigated with MRSA during a 15 days of cycling experiment. Results indicated that a high bacterial inoculum and a prolonged antibiotic exposure were two key factors in the in vitro antibiotic resistance selection in MRSA and should be taken into consideration when choosing the drug treatment. Finally in part 3 bactericidal textile surfaces were investigated against MRSA. Polyesters coated after 160 seconds of copper sputtering have demonstrated a high bactericidal activity reducing the bacterial load of at least 3 logio after one hour of contact. -- Au cours des dernières décennies, des bactéries multirésistantes aux antibiotiques (BMR) ont émergé dans les hôpitaux du monde entier. Depuis lors, le nombre de BMR et la prévalence des infections liées aux soins (IAS) continuent de croître et sont associés à une augmentation des taux de morbidité et de mortalité ainsi qu'à des coûts élevés. De plus, le nombre de résistance à différentes classes d'antibiotiques a également augmenté parmi les BMR, limitant ainsi les options thérapeutiques disponibles lorsqu'elles ont liées a des infections. Des mesures visant une détection plus rapide des bactéries résistantes ainsi que l'établissement des paramètres de traitement antibiotiques adéquats sont primordiales lors d'infections déjà présentes. Dans une optique de prévention, la diminution des bactéries présentes sur les surfaces communément touchées pourrait aussi freiner la dissémination et l'évolution des bactéries résistantes. Durant ma thèse, différents projets incluant des nouvelles technologies et évoluant autour de la résistance antibiotique ont été traités. Des nouvelles technologies telles que le microscope à force atomique (AFM) et la pulvérisation cathodique de cuivre (PCC) ont été utilisées, et le Staphylococcus aureus résistant à la méticilline (SARM) a été la principale BMR étudiée. Deux grandes lignes de recherche ont été développées; la première visant à détecter la résistance antibiotique plus rapidement avec l'AFM et la seconde visant à prévenir la dissémination des BMR avec des surfaces crées grâce à la PCC. L'AFM a tout d'abord été utilisé en tant que microscope à sonde locale afin d'investiguer la résistance à la vancomycine chez les SARMs. Les résultats ont démontré que la rigidité de la paroi augmentait avec la résistance à la vancomycine et que celle-ci corrélait aussi avec une augmentation de l'épaisseur des parois, vérifiée grâce à la microscopie électronique. Des parties d'un AFM ont été ensuite utilisées afin de créer un nouveau dispositif de test de sensibilité aux antibiotiques, un nanocapteur. Ce nanocapteur mesure des vibrations produites par les bactéries vivantes. Après l'ajout d'antibiotique, les vibrations cessent définitivement chez les bactéries sensibles à l'antibiotique. En revanche pour les bactéries résistantes, les vibrations reprennent après le retrait de l'antibiotique dans le milieu permettant ainsi, en l'espace de minutes de détecter la sensibilité de la bactérie à un antibiotique. La PCC a été utilisée afin de créer des surfaces bactéricides pour la prévention de la viabilité des BMR sur des surfaces inertes. Des polyesters finement recouverts de cuivre (Cu), connu pour ses propriétés bactéricides, ont été produits et testés contre des SARMs. Une méthode de détection de viabilité des bactéries sur ces surfaces a été mise au point, et les polyesters obtenus après 160 secondes de pulvérisation au Cu ont démontré une excellente activité bactéricide, diminuant la charge bactérienne d'au moins 3 logio après une heure de contact. En conclusion, l'utilisation de nouvelles technologies nous a permis d'évoluer vers de méthodes de détection de la résistance antibiotique plus rapides ainsi que vers le développement d'un nouveau type de surface bactéricide, dans le but d'améliorer le diagnostic et la gestion des BMR.

Sir James Lighthill proposed in 1992 that acoustic streaming occurs in the inner ear, as part of the cochlear amplifier mechanism. Here we hypothesize that some of the most ancient organisms use acoustic streaming not only for self-propulsion but also to enhance their nutrient uptake. We focus on a motile strain of Synechococcus, a yanobacteria whose mechanism for self-propulsion is not known. Molecular motors could work like piezoelectric transducers acting on the crystalline structure surrounding the outer cell membrane. Our calculations show that a traveling surface acoustic wave (SAW)could account for the observed velocities. These SAW waves will also produce a non-negligible Stokes layer surrounding the cell: motion within this region being essentially chaotic. Therefore, an AS mechanism would be biologically advantageous, enhancing localized diffusion processes and consequently, chemical reactions. We believe that acoustic streaming, produced by nanometer scale membrane vibrations could be widespread in cell biology. Other possible instances are yeast cells and erythrocytes. Flows generated by acoustic streaming may also be produced by silica coated diatoms along their raphe. We note that microelectromechanical (MEMS) acoustic streaming devices were first introduced in the 1990’s. Nature may have preceded this invention by 2.7 Gyr.

Past multisensory experiences can influence current unisensory processing and memory performance. Repeated images are better discriminated if initially presented as auditory-visual pairs, rather than only visually. An experience's context thus plays a role in how well repetitions of certain aspects are later recognized. Here, we investigated factors during the initial multisensory experience that are essential for generating improved memory performance. Subjects discriminated repeated versus initial image presentations intermixed within a continuous recognition task. Half of initial presentations were multisensory, and all repetitions were only visual. Experiment 1 examined whether purely episodic multisensory information suffices for enhancing later discrimination performance by pairing visual objects with either tones or vibrations. We could therefore also assess whether effects can be elicited with different sensory pairings. Experiment 2 examined semantic context by manipulating the congruence between auditory and visual object stimuli within blocks of trials. Relative to images only encountered visually, accuracy in discriminating image repetitions was significantly impaired by auditory-visual, yet unaffected by somatosensory-visual multisensory memory traces. By contrast, this accuracy was selectively enhanced for visual stimuli with semantically congruent multisensory pasts and unchanged for those with semantically incongruent multisensory pasts. The collective results reveal opposing effects of purely episodic versus semantic information from auditory-visual multisensory events. Nonetheless, both types of multisensory memory traces are accessible for processing incoming stimuli and indeed result in distinct visual object processing, leading to either impaired or enhanced performance relative to unisensory memory traces. We discuss these results as supporting a model of object-based multisensory interactions.

How much water we really need depends on water functions and the mechanisms of daily water balance regulation. The aim of this review is to describe the physiology of water balance and consequently to highlight the new recommendations with regard to water requirements. Water has numerous roles in the human body. It acts as a building material; as a solvent, reaction medium and reactant; as a carrier for nutrients and waste products; in thermoregulation; and as a lubricant and shock absorber. The regulation of water balance is very precise, as a loss of 1% of body water is usually compensated within 24 h. Both water intake and water losses are controlled to reach water balance. Minute changes in plasma osmolarity are the main factors that trigger these homeostatic mechanisms. Healthy adults regulate water balance with precision, but young infants and elderly people are at greater risk of dehydration. Dehydration can affect consciousness and can induce speech incoherence, extremity weakness, hypotonia of ocular globes, orthostatic hypotension and tachycardia. Human water requirements are not based on a minimal intake because it might lead to a water deficit due to numerous factors that modify water needs (climate, physical activity, diet and so on). Water needs are based on experimentally derived intake levels that are expected to meet the nutritional adequacy of a healthy population. The regulation of water balance is essential for the maintenance of health and life. On an average, a sedentary adult should drink 1.5 l of water per day, as water is the only liquid nutrient that is really essential for body hydration.

Different interferometric techniques were developed last decade to obtain full field, quantitative, and absolute phase imaging, such as phase-shifting, Fourier phase microscopy, Hilbert phase microscopy or digital holographic microscopy (DHM). Although, these techniques are very similar, DHM combines several advantages. In contrast, to phase shifting, DHM is indeed capable of single-shot hologram recording allowing a real-time absolute phase imaging. On the other hand, unlike to Fourier phase or Hilbert phase microscopy, DHM does not require to record in focus images of the specimen on the digital detector (CCD or CMOS camera), because a numerical focalization adjustment can be performed by a numerical wavefront propagation. Consequently, the depth of view of high NA microscope objectives is numerically extended. For example, two different biological cells, floating at different depths in a liquid, can be focalized numerically from the same digital hologram. Moreover, the numerical propagation associated to digital optics and automatic fitting procedures, permits vibrations insensitive full- field phase imaging and the complete compensation for a priori any image distortion or/and phase aberrations introduced for example by imperfections of holders or perfusion chamber. A blind speech watermarking scheme that meets hard real-time deadlines is presented and implemented. In addition, one of the key issues in these block-oriented watermarking techniques is to preserve the synchronization. Namely, to recover the exact position of each block in the mark extract process. In fact, the presented scheme can be split up into two distinguished parts, the synchronization and the information mark methods. The former is embedded into the time domain and it is fast enough to be run meeting real-time requirements. The latter contains the authentication information and it is embedded into the wavelet domain. The synchronization and information mark techniques are both tunable in order to allow a con gurable method. Thus, capacity, transparency and robustness can be con gured depending on the needs. It makes the scheme useful for professional applications, such telephony authentication or even sending information throw radio applications.

The impact of the social support and the psychic morbidity on the quality of life of patients with antiretroviral therapy. The aim of this study is to analyse the existing relation between the psychic morbidity and social support and the quality of life. Besides this, the paper analyses the buffer rol that social support plays on the psychic morbidity in these patients. We studied 320 HIV+ patients in truatment with antiretrovirals, who attended the infectious disease services of four hospitals of the Autonomous Andalusian Community. Being associated a better quality of life to an absence of psychic morbidity and to the presence of social support, it is observed the relevant buffer role that the social support like shock absorber of the psychic morbidity in this one type of patients. These results show the importance that the psycho-social factors have during the course of chronic diseases.

In this paper, we address this problem through the design of a semiactive controller based on the mixed H2/H∞ control theory. The vibrations caused by the seismic motions are mitigated by a semiactive damper installed in the bottom of the structure. It is meant by semiactive damper, a device that absorbs but cannot inject energy into the system. Sufficient conditions for the design of a desired control are given in terms of linear matrix inequalities (LMIs). A controller that guarantees asymptotic stability and a mixed H2/H∞ performance is then developed. An algorithm is proposed to handle the semiactive nature of the actuator. The performance of the controller is experimentally evaluated in a real-time hybrid testing facility that consists of a physical specimen (a small-scale magnetorheological damper) and a numerical model (a large-scale three-story building)
