Työn tarkoituksena oli etsiä mahdollisia kohteita sellutehtaan veden käytön vähen-tämiseksi. Analyysien avulla selvitettiin voidaanko havaittujen kohteiden tällä hetkellä menetettyjä vesivirtoja käyttää uudelleen. Lopuksi arvioitiin miten kierrä-tettävät vedet vaikuttaisivat raakaveden laatuun, mikäli ne yhdistettäisiin tämän joukkoon. Veden käytön vähentämiskohteita etsittiin tutkimalla tehtaan prosessi-kaavioita osastokohtaisesti sekä selvittämällä näin havaittuja kohteita osastoilla. Työn kokeellinen osa koostui vesianalyyseistä. Vesianalyyseissä määritettiin seu-raavat arvot sellutehtaalle tulevasta vedestä: sameus (FTU), kiintoaine, sähkönjoh-tavuus, pH, CODCr, BOD7ATU, kokonaisfosfori P, Mn, Cl, K, Ca, Mg ja AOX. Nämä tehtiin valituista kohteista havaituista tällä hetkellä tehtaalta pois johdetuista mutta mahdollisesti kierrätettävistä vesistä. Työn tuloksien perusteella havaittiin, että seuraavissa tarkasteluun valituissa koh-teissa vedet olivat hyvin puhtaita ja talteen otettavia kierrätykseen. Näitä olivat savukaasupesurin vedet, sekundäärilauhdesäiliön vedet sekä tiivistevedet. Haih-duttamon lämminvesisäiliön sekä kuivauskone 4 kiertovesitornin vedet olivat hieman likaantuneet mutta hyvin kierrätettävissä. Koivulinjan D0-vaiheen, havu-linjan D1-vaiheen sekä kuivauskone 4 lajittamon vesivirtoja ei likaantumisen vuoksi kannattanut kerätä hyötykäyttöön.

This study was designed to assess the relationship between antibodies against bovine viral diarrhea virus (BVDV) determined in the bulk tank milk (BTM) and the within-herd seroprevalence. We also assessed the efficiency of measuring antibody levels in BTM samples to monitor BVDV infection status in a herd. In the 81 farms included in the study, BTM samples were obtained and blood samples withdrawn from all cattle older than one year. The infection status was then determined in serum and milk using a commercial blocking ELISA based on the detection of anti-p80 antibodies. Apart from these baseline serum and milk samples, another BTM sample was collected from each herd 9 months later, and a third BTM sample obtained 9 months after this. In these second and third milk samples, anti-BVDV antibodies were determined using the same ELISA kit. Statistical tests revealed good agreement between herd seroprevalences (% seropositive animals in the herd) and the antibody levels detected in the BTM samples. During the 18 months of follow-up, the farms with persistently infected cattle at the study outset (14.8% of the herds) showed a significant decrease in BTM antibody titers after virus clearance. Conversely, a significant increase in BTM antibody levels was observed in the herds infected with BVDV during the follow-up period. Our findings indicate that monitoring antibody levels in the BTM is a useful method of identifying changes in the BVDV infection status of a herd.

This study aimed to determine both the lethal and sublethal concentrations of Cypermethrin in young Silver Catfish (Brazilian "Jundiá", Rhamdia quelen) on aquatic environment during 96 hours, as well as to determine the Cypermethrin and Deltamethrin sublethal concentrations during the initial embryonic development period of Rhamdia quelen, and to verify their respective rates of fertilization, hatching and survival. Pyrethroid nowadays is a widely used insecticide, which presents a high toxicity to fish. In order to determine lethal and sublethal concentrations, 120 silver catfish were used; each one had an average weight of 59.58±4.50g and an average size of 20.33±2.34cm. Concentrations used were 0, 1.0, 1.5, 2.0, 2.5, 3.0, 5.0, 10.0, 15.0 and 20.0mg of Cypermethrin per liter of water (mg/L). Fish were exposed to the product in 30-liter fish tanks. In each fish tank there were four fishes and the product was applied three times, i.e., a total of twelve fish were exposed to the product at each application, and a total of 120 fish during the entire experiment (n=120). In order to determine the Cypermethrin and Deltamethrin sublethal concentrations during the initial embryonic development, ovulation induction was performed on female fishes using hormones, and then and egg collection was performed. The eggs were then hydrated and fertilized in Cypermethrin and Deltamethrin in different concentrations: 0.001, 0.01, 0.1, 1.0 and 10.0mg/L of Cypermethrin and 0.001, 0.01, 0.1, 0.5 and 1.0mg/L of Deltamethrin, in addition to the control group (0mg/L). After fertilization, the eggs were kept in containers with the respective pesticides of Cypermethrin and Deltamethrin until hatching, when hatching rate was verified. Then the alevins, from the hatching, were kept on their respective concentrations of Cypermethrin and Deltamethrin so that the survival rate could be analyzed regarding the tested insecticides, during both 12-hour and 24-hour periods. Concerning the sublethal and lethal concentrations, Silver Catfish was sensitive to the tested concentrations of Cypermethrin, showing symptoms of poisoning, such as loss of balance, swimming alteration, dyspnea (they kept their mouths and opercula open), upright swimming and sudden spiral swimming movements. The intensity of such symptoms varied in proportion to the concentration used. The concentrations above 3.0mg/L were considered lethal to the species, since every animal exposed to concentrations between 3.0 and 20.0mg/L had died, while concentrations between 1.0 and 2.5mg/L were considered sublethal. Lethal concentration of Cypermethrin to Silver catfish, in 96 hours, was 1.71 milligram per liter of water. Concerning the sublethal concentration of Cypermethrin and Deltamethrin during the initial embryonic development, the results show that both pyrethroids had significantly decreased the analyzed parameters when comparing them with the control group. It was concluded that, even with the fish being more resistant to pyrethroids in comparison with other species, both the young animals and the ones in stage of embryonic development were susceptible to the effects of these pesticides.

The objectives of this study were to isolate Klebsiella pneumoniae from different sources in three dairy cattle herds, to use the pulsed-field gel electrophoresis (PFGE) to measure genotypic similarities between isolates within a dairy herd, to verify the production of extended-spectrum β-lactamases (ESBLs) by the double-disk synergy test (DDST), and to use the PCR to detect the main ESBLs subgroups genes. Three dairy farms were selected based on previous mastitis outbreaks caused by K. pneumoniae. Milk samples were collected from lactating cows and from the bulk tank. Swabs were performed in different locations, including milking parlors, waiting room, soil, animal's hind limbs and rectum. K. pneumoniae was isolated from 27 cases of intramammary infections (IMI) and from 41 swabs. For farm A isolates from IMI and bulk tank were considered of the same PGFE subtype. One isolate from a bulk tank, three from IMI cases and four from environmental samples were positive in the DDST test. All eight DDST positive isolates harbored the bla shv gene, one harbored the bla tem gene, and three harbored the bla ctx-m gene, including the bulk tank isolate. Our study confirms that ESBL producing bacteria is present in different locations in dairy farms, and may be responsible for IMI. The detection of ESBLs on dairy herds could be a major concern for both public and animal health.

The goal of the Master’s thesis is to design a test stand for a centrifugal compressor. Different theoretical aspects of flow parameters measurements and test rigs built for the similar purposes in other research units are described in the theoretical part of the work. The process of components selection and the description of chosen components are given in the second part of the thesis. Besides measuring and control stages, the designed test stand has a closed-loop piping, an aftercooler and a surge tank. Overview and layout of the test rig is presented in the last chapter of the work.

The interference imposed the by weeds on corn decreases practically all vegetative characteristics. As consequence, the green ear and grain yield are also reduced. Losses due to the fall armyworm (Spodoptera frugiperda) attack can reduce corn grain yield up to 34%. In general, weed and insect control issues are addressed separately in research papers. Nevertheless, interaction between weeds and insects may exist. This study aimed to evaluate green ear and corn grain yield response to weed and fall armyworm control. A completely randomized block design with split-plots and five replicates was adopted. Corn cultivar AG 1051 was grown under weedy conditions or with control by hand hoeings performed at 20 and 40 days after planting. Fall armyworm control (applied to subplots) was performed with sprays of water (control), deltamethrin (5g active ingredient ha-1); neem oil, at 0.5% (diluted in water), and neem leaf extract at 5%. Each product was sprayed three times, at seven-day intervals, starting at the 7th day after planting, using 150 L ha-1 of the tank solution. Dry mass of the above-ground part, internode diameter, leaf length, leaf width, leaf area, green ear yield and grain yield of corn were reduced due to the lack of weed control. Fall armyworm control in the weeded plots did not influence green ear yield and grain yield, except green mass of marketable, husked ears, which was reduced when the caterpillar was not controlled. Without weed control, neem extracts and deltamethrin sprays provided highest yields of number and total weight of green ears with husks, number and weight of marketable ears with husks and number of marketable ears without husks. The best results for husked ear mass and for grain yield were obtained with neem extract and deltamethrin, respectively.

In pot experiments, two adjuvants were evaluated for their efficacy in enhancing activity of five herbicides applied at reduced rates (75% of the recommended rates) on Emex spinosa at the cotyledon-leaf and at the two- to four- leaf stage. Herbicides (at recommended rates) including fluroxypyr+MCPA at 450 g a.i. ha-1, carfentrazone-ethyl at 20 g a.i. ha-1, bromoxynil+MCPA at 450 g a.i. ha-1, thifensulfuron-methyl at 75 g a.i. ha-1 and tribenuronmethyl at 75 g a.i. ha-1 alone and tank mixed at reduced rates with adjuvants, namely, alkyl ether sulphate sodium salt at 625 mL ha-1 or fatty alcohol ethoxylate at 375 mL ha-1. Addition of the adjuvants to reduced rates of fluroxypyr+MCPA and carfentrazone-ethyl, increased their efficacy with 100% mortality and biomass reduction of E. spinosa at the cotyledon- leaf stage and at the two- to four- leaf stage which was similar to their recommended rates without the adjuvants except for carfentrazone-ethyl at a reduced rate without adjuvants at the two- to four- leaf stage. Bromoxynil+MCPA at reduced rates with alkyl ether sulphate sodium salt also gave 100% control of E. spinosa over weedy check at the two- to four- leaf stage. Both the adjuvants generally increased the efficacy of tribenuron-methyl at reduced rates when sprayed at both leaf stages. These findings suggest that the use of adjuvants may increase the efficacy of the above mentioned herbicides against E. spinosa and it may be incorporated in an integrated weed management program.

Tässä diplomityössä selvitettiin UPM Kaukaan tehtailla esiintyviin lakisääteisiin hitsauskohteisiin liittyvää lainsäädäntöä ja lainsäädännön vaikutusta hitsaukselle. Lakisääteisiä hitsauskohteita ovat räjähdysvaaralliset tilat, painelaitteet, kemikaaliputkistot, kemikaalisäiliöt, maakaasulaitteet sekä kantavat teräsrakenteet. Diplomityössä selvitettiin lisäksi hitsauksen laadunhallintaa huomioiden korjaushitsaustyöt sekä uuden rakentaminen. Lakisääteet tuovat hitsauksen suorittamiselle usein tarkasti määritellyt velvoitteet koskien esimerkiksi dokumentointia, hitsaushenkilöstön pätevyyksiä, materiaaleja ja niin edelleen. Lakisääteet lisääntyvät jatkuvasti ja soveltamisalueet laajenevat, jolloin asioiden ylläpitäminen vaatii jatkuvaa seurantaa. Työssä luotiin ohjeistus koskien yleisesti hitsausta ja lisäksi käyttäjän ja omistajan velvoitteita koskevat ohjeet painelaitteista, maakaasulaitteista ja kemikaalilaitteista. Hitsauksen yleisohje koskee jokaista hitsaustapahtumaa korjaushitsauksesta uuden rakentamiseen. Ohje soveltuu lisäksi sopimusliitteeksi hitsaustyön tilauksissa.

Diplomityössä tutkittiin erilaisilla loppuasennusmenetelmillä toimitettujen terästank-kien kustannus- ja aikataulurakennetta. Usein terästankit valmistetaan valmiiksi muotoon valssatuista levyistä työmaalla kasaanhitsaamalla. Tämä ei ole ainoa terästankin valmistusvaihtoehto, myös ruuvaus, niittaus, kiilaliitos ja konepajalla suuremmiksi kappaleiksi hitsaus ovat yleisesti käytössä. Terästankkien valmistuskustannuksiin vaikuttavat materiaali, esivalmisteiden työstäminen, kuljetus ja loppuasennus. Tankin valmistusmenetelmä ei merkittävästi vaikuta tankin materiaalikustannuksiin. Materiaalikustannuksiin vaikuttavat eniten käytettävä materiaali ja tankin geometriset ominaisuudet. Tankin esivalmisteiden valmistusaika määräytyy loppuasennusmenetelmästä. Pidempi esivalmistusaika lyhentää loppu-asennusaikaa. Kokonaisena toimitetussa tankissa esivalmistuksen osuus on suurempi kuin hitsatussa tankissa, mutta loppuasennusaika on vain murto-osa hitsattavan tankin loppuasennusajasta. Ruuvattavassa tankissa laippojen hitsaus levyihin lisää esivalmistusaikaa, kun rullattavassa tankissa aikaa vievät vaippalevyjen kasaanhitsaaminen ja rullaaminen. Tankin loppuasennustapa voidaan, etenkin halkaisijaltaan noin 10 m:llä tankeilla, valita kohdemaan mukaan, koska erot valmistuskustannuksissa ovat pieniä eri toimitusmuotojen välillä. Tosin kokonaisen 10 m halkaisijaltaan olevan tankin kuljettaminen ei aina ole mahdollista eikä kannattavaa. Sen sijaan halkaisijaltaan 5 m:llä tankeilla kokonaisena toimittaminen on varteenotettava vaihtoehto. Sopivin tankkivaihtoehto, kokonainen, hitsattava, ruuvattava tai rullattava, tulee miettiä aina tapauskohtaisesti, kohdemaan, hitsausosaamisen, kuljetusmahdollisuuksien ja loppuasennuspaikan mukaan.

Among the herbicides recommended for the dry season and registered to sugarcane crop, amicarbazone, isoxaflutole and the association diuron + hexazinone + sulfomethuron-methyl can be highlighted. These are pre-emergence herbicides efficient against broad-leaved weeds. Morning glory causes large losses in infested sugarcane fields by bending the stalks and interfering in harvesting. In this study the effectiveness of pre-emergence herbicides for two species of morning glory (Ipomoea hederifolia and Ipomoea grandifolia) was evaluated. Treatments were arranged in completely randomized factorial design (4 x 7). There were four periods of water restriction (0, 30, 60 and 90 days), seven chemical treatments [diuron + hexazinone + sulfometuron-methyl (1387 + 391 + 33.35 g a.i. ha-1), diuron + hexazinone + sulfometuron-methyl (1507.5 + 425 + 36.25 g a.i. ha-1), diuron + hexazinone + sulfometuron-methyl (1658.25 + 467.5 + 39.87 g a.i. ha-1), diuron + hexazinone + sulfometuronmethyl (1809 + 510 + 43.5 g a.i. ha-1), amicarbazone (1190 g a.i. ha-1), amicarbazone + isoxaflutole (840 + 82.5 g a.i. ha-1)] and a control with no application. At 7, 14, 21 and 28 days after the restoration of moisture, control was visually evaluated. After the final evaluation, the dry mass of morning glories was measured. At 90 days of water restriction, diuron + hexazinone + sulfometuron-methyl was more effective to control I. hederifolia than the amicarbazone + isoxaflutole tank mixture. The four diuron + hexazinone + sulfometuronmethyl doses have reduced morning glory dry mass to zero; whereas treatments with amicarbazone have not. The most effective treatment for morning glory control was diuron + hexazinone + sulfometuron-methyl. This result may be due to a possible synergistic interaction.

The effect of five adjuvants (non-ionic surfactant, paraffinic oil, vegetable oil, mixture of fatty acids methyl esters plus surfactant blend, and organosilicone) on diquat efficacy was assessed on poverty brome, sterile oat, and Italian ryegrass in field and pot experiments. All tank mixtures with diquat increased diquat efficacy from 50-54% to 77-98% as for fresh weight reduction, indicating significant enhancement of diquat efficacy on grasses. The increased efficacy was most likely attributed to better droplet retention and diffusion on the leaf surfaces. When combined with non-ionic surfactant, diquat showed slightly more rapid control of grass weeds (i.e. symptoms were visible within a few hours after application).

The loss of grains during the harvest of glyphosate tolerant corn may generate volunteer plants, which can interfere in the conventional or glyphosate crop in succession. The current work aim to evaluate the control of the volunteer corn glyphosate tolerant under two weed stages. Aimed to evaluate the control of volunteer glyphosate tolerant corn in two stages of development. There were conducted two experiments with hybrid 2B688 HR (lepidoptera and glyphosate tolerant), the application were at V5 and V8 stage. The experiment was randomized block design with four replicates, using the treatments: haloxyfop at 25, 50 and 62 g ha-1 alone and associated with 2,4-D at 670 g ha-1 or fluroxypyr at 200 g ha-1. The standard was clethodim at 84 g ha-1 with 2,4-D and fluroxypyr at same rates. The applications of haloxyfop and clethodim both isolated or in a mixture with 2,4-D and fluroxypyr at V5 stage showed total control (100%) at 32 and 39 days after the application, except for haloxyfop + 2,4-D (25 + 670 g ha-1) mixture, which did not provided adequate control. At V8 stage, haloxyfop + 2,4-D (50 + 670 g ha-1) and haloxyfop + 2,4-D (62 + 670 g ha-1) mixtures took up to 6 and 10 days or longer to reach adequate to excellent control, when compared to haloxyfop isolated applications in the same doses, respectively. Either isolated clethodim or mixed with 2, 4-D and fluroxypyr did not show adequate control. The treatments showed efficient control on volunteer corn plants at V5 stage, except for haloxyfop + 2, 4-D (25 + 670 g ha-1) mixture. At V8 stage applications, haloxyfop either isolated or mixture with fluroxypyr demonstrated excellent control on every evaluated dose. The mixture with 2, 4-D can reduce haloxyfop efficiency at low doses. Clethodim alone or mixed with 2,4-D or furoxypyr did not provide acceptable level of control.

Cotton is highly susceptible to the interference imposed by weed community, being therefore essential to adopt control measures ensuring the crop yield. Herbicides are the primary method of weed control in large-scale areas of production, and usually more than one herbicide application is necessary due to the extensive crop cycle. This study aimed to evaluate the selectivity of different chemical weed control systems for conventional cotton. The experiment took place in the field in a randomized block design, with twenty nine treatments and four replications in a split plot layout (adjacent double check). Results showed that triple mixtures in pre-emergence increased the chance of observing reductions in the cotton yield. To avoid reductions in crop yield, users should proceed to a maximum mixture of two herbicides in pre-emergence, followed by S-metolachlor over the top, followed by one post-emergence mixture application of pyrithiobac-sodium + trifloxysulfuron-sodium.

ABSTRACTInadequate herbicide application can result in failures in weed control and/or poisoning of the crops, resulting in yield losses. In this research were assessed the effects of the sprayer nozzle boom height in the distribution of the spray solution for weed control, influencing intoxication of beans and crop yield. Experiments were conducted in laboratory and field conditions. In laboratory, the performance of flat spray tip TT 11002 was assessed at heights 0.20, 0.30, 0.40 and 0.50 meters with respect to the target surface. In the field the same heights were assessed in applications of herbicides fomesafen, fluazifop-P-butyl and fomesafen + fluazifop-P-butyl. There was an inverse relationship between the height of the spray boom and the coefficients of variation of the patterns. The mixture better efficiency in a tank of fluazifop-P-butyl + fomesafen was obtained with the height of 0.50 m from the target. This treatment resulted in better weed control, lower poisoning of the bean plants and better crop yield rates.

ABSTRACT The aim of this work was to evaluate the selectivity of fomesafen alone or in a tank mixture with other preemergent herbicides, with or without S-metolachlor application in early postemergence in cotton plant, cultivar DP 555 BG RR(r). The design utilized was a randomized complete block, organized in a split-plot arrangement, with four replications. For that, 24 herbicides were evaluated with fomesafen (0.45 and 0.625 kg ha-1), prometryn (1.25 kg ha-1), diuron (1.25 kg ha-1), trifluralin (1.8 kg ha-1), and S-metolachlor (0.77 kg ha-1), applied as preemergent, with or without S-metolachlor (0.77 kg ha-1) applied in early postemergence. The variables evaluated were: phytotoxicity, insertion height of the first reproductive branch, plant height, stand, number of reproductive branches per plant, number of bolls per plant, bolls weight, and productivity of cotton seed. Fomesafen alone or in a tank mixture with preemergent prometryn, diuron, trifluralin and S-metolachlor was selective to cotton plant. Preemergent fomesafen isolated application followed by the application of S-metolachlor in early postemergence was also selective. However, on average, preemergent tank mixtures applied in association with preemergent S-metolachlor early application was not selective to cotton crop.
