Arylsulfatase activity and growth were estimated in Escherichia coli, isolated from marine sediment. Maximum activity was observed at pH 6.6 whereas the maximum growth was at pH 5.6. 2x10ˉ³ M is the optimum substrate concentration for the highest level of enzyme activity/synthesis as well as for its growth. In general higher substrate concentration tended to inhibit enzyme activity and also the growth of the bacterium. Maximum growth and highest enzyme activity occurred at 29°C and above this temperature decreased both of them. Besides these, glucose, sodium sulfate, sodium chloride, sodium dihydrogen phosphate, sodium acetate and ammonium chloride at higher concentrations were inhibiting the enzyme activity and growth. Above 0.2% of glucose, 3% of sodium chloride, 10x10ˉ³ M concentrations of sodium sulfate, sodium dihydrogen phosphate, sodium acetate and ammonium chloride inhibited the activity and growth also. These observations indicate that, to generalize a compound as inhibitor or activator it is difficult since this depends not only on its concentration but also on the source of the enzyme when more than one type is encountered in nature.

The distribution of total hectrotrophic bacteria (THB) and lipolytic bacteria in various regions (body surface, gill, intestine and flesh) of fish Etroplus suratensis (Bloch) during storage at 28 ± 2°C and 4°C was studied. Pseudomonas dominated at reduced temperature whereas at 28 ± 2°C and in fresh condition Vibrio, Aeromonas, and Acinetobacter dominated. Lipolytic activity was elaborated by the members of various genera and their activity varied in different lipid compounds (tributyrin, tween 80, tween 60, tween 40 and tween 20). Tributyrin was utilized by majority of the isolates. All the selected isolates preferred a temperature of 35°C and pH 6.0 for their maximum growth. Aeromonas and Vibrio showed maximum growth at 0.5% NaCl concentration while 3% NaCl was found to be optimum for Pseudomonas.

Photosynthetic characteristics of a purple sulfur bacterium, Chromatium buderi, cultured under different ranges of pH, temperature, light intensities and ammonium chloride concentrations were examined. Maximum bacteriochlorophyll a synthesis was observed at pH 6.5 whereas the optimum growth was at pH 8.0. In general, higher temperature tended to inhibit the chlorophyll a synthesis and growth. 30°C is the optimum temperature both for chlorophyll a synthesis and growth. At 25µE mˉ²Sˉ¹ the bacteriochlorophyll a content and growth attained maximum level. The response to this low light intensity is an adaptation that ensures a high photosynthetic rate for the purple sulfur bacterium that usually occurs in dimly lit environment. Besides these, ammonium chloride at low concentration enhances both chlorophyll a synthesis and growth. Above 0.5% of it causes the nitrogen-chlorosis and also retards the growth of the bacterium. Possible chemical and structural mechanisms involved are discussed.

This study was made as an attempt to investigate the optimum packing density and the ice quantity suitable for the transport of Penaeus monodon juveniles. The results revealed that prawns of 40 mg size can be packed to as much as 3,000 per bag. While packing densities above 3,000 per bag containing 8 L seawater and 16 L oxygen can be used only for short transport periods. On the other hand in the ice-quantity experiment, mortality rate was less than 1% in all the bags containing 300 g, 600 g, 900 g and 1200 g of ice. A packing temperature of 20~’C must be maintained hence, 50 g of ice per hour should be allowed per box, counting from the moment the box is sealed to the time it is estimated to be opened.

Lagenidium scyllae , a marine mastigomycete from the ova and larvae of the mangrove crab, is described and illustrated as new. The fungus grew at a temperature range of 16-42 C, with an optimum at 22.5-31.8 C. It grew well in peptone-yeast-glucose (PYG) broth containing 2-3% NaCl. In PYG-sea water medium, it grew at a pH range of 5-11.

Results of mesh selectivity experiments on B. tor are presented in this paper. Selectivity curve on the basis of maximum girth of fish in relation to perimeter of mesh was worked out. The optimum girth/mesh perimeter ratio was found to be 1.31. A linear regression of G+0.445L=12.8 was fitted for conversion of length to girth.

A feeding trial of 8 weeks was conducted in a static indoor rearing system to investigate the optimum carbohydrate to lipid ratio (CHO:L ratio) in stinging catfish, Heteropneustes fossilis. Five iso-nitrogenous (35% crude protein) and iso-energetic (17.06 kJ gˉ¹ gross energy (GE)) fish meal based diets with varying carbohydrate to lipid (CHO:L g/g) ratios of 0.60, 0.98, 1.53, 2.29 and 3.44 for diets 1-5, were tested, respectively. The diets containing a fixed protein to energy ratio (P:E ratio) of 20.50-mg protein kJˉ¹ GE were fed to triplicate groups of 40 fish (per 70-L tank). Fish were fed 5% of their body weight per day adjusted fortnightly. Diet 1, containing 10% carbohydrate and 17% lipids with a CHO:L ratio of 0.60 produced the poorest (p<0.05) growth rates, feed and protein efficiency. Increasing carbohydrate content in the diets to 26% concomitant with a reduction in lipid content to 11% with a CHO:L ration of 2.29 of diet 5 significantly improved (p<0.05) growth rates, feed and protein efficiency. But did not differ with diet 4, containing CHO:L ratio 2.29. A further increase in dietary carbohydrate up to 31% and a decrease in lipids levels to 9% with a CHO:L ratio ranging from 2.29 to 3.44 (diet 4-5) did not significantly improve the fish performance. Apparent net protein utilisation (ANPU) of fish fed diet 5 was higher (p<0.05) than for diets 1 and 2 but did not differ from diets 3 and 4. Higher lipid deposition (p<0.05) in whole body was observed with decreasing dietary CHO:L ratios as increasing lipid levels. Whole body protein of fish fed varying CHO:L diets did not show any discernible changes among the dietary treatments. This study revealed that H. fossilis can perform equally well on diets containing carbohydrate ranging from 26 to 31%, with 9 to 11% lipid or at CHO:L g/g ratio of 2.29-3.44.

A 3x3 factorial experiment was conducted to determine the optimum protein to energy (P/E) ratio for rainbow trout in brackish water. Three crud protein levels and three energy levels at each protein level were utilized. Diets were made in semi-purified that in all of them fish meal, casein and gelatin as the sources of protein and dextrin, starch and oil as the sources of energy were used. Each of experimental diets was fed to triplicate groups of 20 fish with an average individual weight of 81.5 g in 9 2000-1 flow trough fiberglass tanks. During this experiment water temperature, dissolved oxygen, PH and EC were 15±2°C, 6.5-8.1 mg/1, 7.7-8.6 and 25400 grills respectively. The diets were fed at a rate between 1.6-2 wet body weight% per day depended to water temperature in three equal rations and adjusted two weekly for 84 days. At each of protein levels, weight gain percent (%WG), average daily growth percent (%ADG), protein efficiency ratio (PER), apparent net protein utilization percent (%ANPU), or percent of protein deposited, specific growth rate (SGR) and condition factor (CF) were found to increase and food conversion ratio (FCR) was found to decrease with an increasing energy levels from 370 to 430 Kcal/100g. Fish fed a 35% protein, 430 Kcal/100g energy diet with a P/E ratio of 81.4 mg protein/ Kcal PFV energy, attained the best growth performance. Fat and moisture of carcass were affected by protein and energy levels of test diets while protein and ash of carcass were relatively constant in different treatments.

In the design of high-speed low-power electrical generators for unmanned aircraft and spacecraft, maximization of specific output (power/weight) is of prime importance. Several magnetic circuit configurations (radial-field, axial-field, flux-squeezing, homopolar) have been proposed, and in this paper the relative merits of these configurations are subjected to a quantitative investigation over the speed range 10 000–100000 rev/min and power range 250 W-10 kW. The advantages of incorporating new high energy-density magnetic materials are described. Part I deals with establishing an equivalent circuit for permanent-magnet generators. For each configuration the equivalent circuit parameters are related to the physical dimensions of the generator components and an optimization procedure produces a minimum volume design at discrete output powers and operating speeds. The technique is illustrated by a quantitative comparison of the specific outputs of conventional radial-field generators with samarium cobalt and alnico magnets. In Part II the specific outputs of conventional, flux-squeezing, and claw-rotor magnetic circuit configurations are compared. The flux-squeezing configuration is shown to produce the highest specific output for small sizes whereas the conventional configuration is best at large sizes. For all sizes the claw-rotor configuration is significantly inferior. In Part III the power densities available from axial-field and flux-switching magnetic circuit configurations are maximized, over the power range 0.25-10 kW and speed range 10 000–100000 rpm, and compared to the results of Parts I & II. For the axial-field configuration the power density is always less than that of the conventional and flux-squeezing radial-field configurations. For the flux-switching generator, which is able to withstand relatively high mechanical forces in the rotor, the power density is again inferior to the radial-field types, but the difference is less apparent for small (low power, high speed) generator sizes. From the combined results it can be concluded that the flux-squeezing and conventional radial-field magnetic circuit configurations yield designs with minimum volume over the power and speed ranges considered. © 1985, IEEE. Portland cement has been widely used for stabilisation/solidification (S/S) treatment of contaminated soils. However, there is a dearth of literature on pH-dependent leaching of contaminants from cement-treated soils. This study investigates the leachability of Cu, Pb, Ni, Zn and total petroleum hydrocarbons (TPH) from a mixed contaminated soil. A sandy soil was spiked with 3000 mg/kg each of Cd, Cu, Pb, Ni and Zn, and 10,000 mg/kg of diesel, and treated with ordinary Portland cement (CEM I). Four different binder dosages, 5%, 10%, 15% and 20% (m/m) and different water contents ranging from 13%-19% dry weight were used in order to find a safe operating envelope for the treatment process. The pH-dependent leaching behaviour of the treated soil was monitored over an 84-day period using a 3-point acid neutralisation capacity (ANC) test. The monolithic leaching test was also conducted. Geotechnical properties such as unconfined compressive strength (UCS), hydraulic conductivity and porosity were assessed over time. The treated soils recorded lower leachate concentrations of Ni and Zn compared to the untreated soil at the same pH depending on binder dosage. The binder had problems with Pb stabilisation and TPH leachability was independent of pH and binder dosage. The hydraulic conductivity of the mixes was generally of the order, 10-8 m/sec, while the porosity ranged from 26%-44%. 采用恒温摇床培养方法,研究了不同营养源(处理1:尿素+乙酸钠;处理2:亚硝酸钠+乙酸钠;处理3:硝酸钾+乙酸钠;处理4:碳酸氢铵;处理5:硫酸铵+碳酸氢钠;处理6:磷酸二氢钾+碳酸氢钠)对复合垂直流人工湿地基质生物膜培养液pH值的影响,探讨了pH值变化过程中生物膜脱氢酶活性和多糖含量的变化规律。结果表明,处理1、4以及5中培养液pH值先下降然后再上升,但下降和上升的幅度不同。处理2、3以及6中培养液pH值添加碳源前在7.30~7.40之间缓慢变化,添加碳源后均上升至9.00左右。在培养液pH值变化的过程中

为了解培养温度和pH值对嗜水气单胞菌的毒力基因表达以及致病性的影响,利用反转录PCR(RT-PCR)对4种基因的表达进行了研究。研究结果显示嗜水气单胞菌毒力基因的表达受温度和pH值影响,其中溶血素(AHH)、气溶素(AerA)、外膜蛋白(OMP)和粘附素(Aha)毒力基因在15℃、25℃和37℃下均能高效表达,在4℃AHH和AerA两基因也能持续表达,但OMP和Aha两基因的表达停止。在中性或偏酸性的条件下(pH5.0和pH7.0)4种毒力基因都得以表达,在碱性条件下(pH9.0)4种基因中只有AHH得以
