The objective was to determine the concentration of total selenium (Se) and the proportion of total Se comprised as selenomethionine (SeMet) and selenocysteine (SeCys) in post mortem tissues of lambs in the six weeks period following the withdrawal of a diet containing high dose selenized yeast (SY), derived from a specific strain of Saccharomyces cerevisae CNCM (Collection Nationale de Culture de Micro-organism) I-3060. Thirty Texel x Suffolk lambs used in this study had previously received diets (91 days) containing either high dose SY (HSY; 6.30 mg Se/kg DM) or an unsupplemented control (C; 0.13 mg Se/kg DM). Following the period of supplementation all lambs were then offered a complete pelleted diet, without additional Se (0.15 mg Se/kg DM), for 42 days. At enrollment and 21 and 42 days later, five lambs from each treatment were blood sampled, euthanased and samples of heart, liver, kidney and skeletal muscle (Longissimus Dorsi and Psoas Major) tissue were retained. Total Se concentration in whole blood and tissues was significantly (P < 0.001) higher in HSY lambs at all time points that had previously received long term exposure to high dietary concentrations of SY. The distribution of total Se and the proportions of total Se comprised as SeMet and SeCys differed between tissues, treatment and time points. Total Se was greatest in HSY liver and kidney (22.64 and 18.96 mg Se/kg DM, respectively) and SeCys comprised the greatest proportion of total Se. Conversely, cardiac and skeletal muscle (Longissimus Dorsi and Psoas Major) tissues had lower total Se concentration (10.80, 7.02 and 7.82 mg Se/kg DM, respectively) and SeMet was the predominant selenized amino acid. Rates of Se clearance in HSY liver (307 µg Se/day) and kidney (238 µg Se/day) were higher compared with HSY cardiac tissue (120 µg Se/day) and skeletal muscle (20 µg Se/day). In conclusion differences in Se clearance rates were different between tissue types, reflecting the relative metabolic activity of each tissue, and appear to be dependant upon the proportions of total Se comprised as either SeMet or SeCys.

The objective of the study was to determine if there were adverse effects on animal health and performance when a range of ruminant animals species were fed at least 10 times the maximum permitted European Union (EU) selenium (Se) dietary inclusion rate (0.568 mg Se/kg DM) in the form of selenium enriched yeast (SY) derived from a specific strain of Saccharomyces cerevisiae CNCM I-3060. In a series of studies, dairy cows, beef cattle, calves and lambs were offered either a control diet which contained no Se supplement or a treatment diet which contained the same basal feed ingredients plus a SY supplement which increased total dietary Se from 0.15 to 6.25, 0.20 to 6.74, 0.15 to 5.86 and 0.14 to 6.63 mg Se/kg DM, respectively. The inclusion of the SY supplement (P < 0.001) increased whole blood Se concentrations, reaching maximum mean values of 716, 1,505, 1,377, and 724 ng Se/mL for dairy cattle, beef cattle, calves and lambs, respectively. Selenomethionine accounted for 10% of total whole blood Se in control animals whereas the proportion in SY animals ranged between 40 and 75%. Glutathione peroxidase (EC 1.11.1.9) activity was higher (P < 0.05) in SY animals when compared with controls. A range of other biochemical and hematological parameters were assessed, but few differences of biological significance were established between treatments groups. There were no differences between treatment groups within each species with regard to animal physical performance or overall animal health. It was concluded that there were no adverse effects on animal health, performance and voluntary feed intake to the administration of at least ten times the EU maximum, or approximately twenty times the US FDA permitted concentration of dietary Se in the form of SY derived from a specific strain of Saccharomyces cerevisiae CNCM I-3060.

The objective was to determine the concentration of total selenium (Se) and the proportion of total Se comprised as selenomethionine (SeMet) and selenocysteine (SeCys) in post mortem tissues of beef cattle offered diets containing graded additions of selenized enriched yeast (SY) [Saccharomyces cerevisae CNCM I-3060]), or sodium selenite (SS). Oxidative stability and tissue glutathione peroxidase (GSH-Px) activity of edible muscle tissue were assessed 10 d post-mortem. Thirty two beef cattle were offered, for a period of 112 d, a total mixed ration which had either been supplemented with SY (0, 0.15 or 0.35 mg Se/kg DM) or SS (0.15 mg Se/kg DM). At enrollment (0 d) and at 28, 56, 84 and 112 d following enrollment, blood samples were taken for Se and Se species determination, as well as whole blood GSH-Px activity. At the end of the study beef cattle were euthanized and samples of heart, liver, kidney, and skeletal muscle (LM and psoas major) were retained for Se and Se species determination. Tissue GSH-Px activity and thiobarbituric acid reactive substances (TBARS) were determined in skeletal muscle tissue (LM only). The incorporation into the diet of ascending concentrations of Se as SY increased whole blood total Se and the proportion of total Se comprised as SeMet, as well as GSH-Px activity. There was also a dose dependant response to the graded addition of SY on total Se and proportion of total Se as SeMet in all tissues and GSH-Px activity in skeletal muscle tissue. Furthermore, total Se concentration of whole blood and tissues was greater in those animals offered SY when compared with those receiving a comparable dose of SS, indicating an improvement in Se availability and tissue Se retention. Likewise, GSH-Px activity in whole blood and LM was greater in those animals offered SY when compared with those receiving a comparable dose of SS. However, these increases in tissue total Se and GSH-Px activity appeared to have little or no effect in meat oxidative stability.

Forty-multiparous Holstein cows were used in a 16-wk continuous design study to determine the effects of either selenium (Se) source, selenized yeast (SY) (derived from a specific strain of Saccharomyces cerevisiae CNCM I-3060 Sel-Plex®) or sodium selenite (SS), or inclusion rate of SY on Se concentration and speciation in blood, milk and cheese. Cows received ad libitum a TMR with 1:1 forage:concentrate ratio on a dry matter (DM) basis. There were four diets (T1-T4) which differed only in either source or dose of Se additive. Estimated total dietary Se for T1 (no supplement), T2 (SS), T3 (SY) and T4 (SY) was 0.16, 0.30, 0.30 and 0.45 mg/kg DM, respectively. Blood and milk samples were taken at 28 day intervals and at each time point there were positive linear effects of SY on Se concentration in blood and milk. At day 112 blood and milk Se values for T1-T4 were 177, 208, 248, 279 ± 6.6 and 24, 38, 57, 72 ± 3.7 ng/g fresh material, respectively and indicate improved uptake and incorporation of Se from SY. While selenocysteine (SeCys) was the main selenised amino acid in blood its concentration was not markedly affected by treatment, but the proportion of total Se as selenomethionine (SeMet) increased with increasing inclusion rate of SY. In milk, there were no marked treatment effects on SeCys content, but Se source had a marked effect on the proportion of total Se as SeMet. At day 112 replacing SS (T2) with SY (T3) increased the SeMet concentration of milk from 36 to 111 ng Se/g and its concentration increased further to 157 ng Se/g as the inclusion rate of SY increased further (T4) to provide 0.45 mg Se/kg TMR. Neither Se source nor inclusion rate effected the keeping quality of milk. At day 112, milk from T1, T2, and T3 was made into a hard cheese and Se source had a marked effect on total Se and the proportion of total Se comprised as either SeMet or SeCys. Replacing SS (T2) with SY (T3) increased total Se, SeMet and SeCys content from 180 to 340 ng Se/g, 57 to 153 ng Se/g and 52 to 92 ng Se/g, respectively. The objective was to determine the concentration of total selenium (Se) and the proportion of total Se comprised as selenomethionine (SeMet) and selenocysteine (SeCys), as well as meat quality in terms of oxidative stability in post mortem tissues of lambs offered diets with an increasing dose rate of selenized enriched yeast (SY), or sodium selenite (SS). Fifty lambs were offered, for a period of 112 d, a total mixed ration which had either been supplemented with SY (0, 0.11, 0.21 or 0.31 mg/kg DM to give total Se contents of 0.19, 0.3, 0.4 and 0.5 mg Se/kg DM for treatments T1, T2, T3 and T4, respectively) or SS (0.11 mg/kg DM to give 0.3 mg Se/kg DM total Se [T5]). At enrolment and at 28, 56, 84 and 112 d following enrolment, blood samples were taken for Se and Se species determination, as well as glutathione peroxidase (GSH-Px) activity. At the end of the study lambs were euthanased and samples of heart, liver, kidney, and skeletal muscle were retained for Se and Se species determination. Tissue GSH-Px activity and thiobarbituric acid reactive substances (TBARS) were determined in Longissimus Thoracis. The incorporation into the diet of ascending concentrations of Se as SY increased whole blood total Se and the proportion of total Se comprised as SeMet, and erythrocyte GSH-Px activity. Comparable doses of SS supplementation did not result in significant differences between these parameters. With the exception of kidney tissue, all other tissues showed a dose dependant response to increasing concentrations of dietary SY, such that total Se and SeMet increased. Selenium content of Psoas Major was higher in animals fed SY when compared to a similar dose of SS, indicating improvements in Se availability and retention. There were no significant treatment effects on meat quality assessments GHS-Px and TBARS, reflecting the lack of difference in the proportion of total Se that was comprised as SeCys. However, oxidative stability improved marginally with ascending tissue Se content, providing an indication of a linear dose response whereby TBARS improved with ascending SY inclusion.

Here, we analyze the complete coding sequences of all recognized tick-borne flavivirus species, including Gadgets Gully, Royal Farm and Karshi virus, seabird-associated flaviviruses, Kadam virus and previously uncharacterized isolates of Kyasanur Forest disease virus and Omsk hemorrhagic fever virus. Significant taxonomic improvements are proposed, e.g. the identification of three major groups (mammalian, seabird and Kadam tick-borne flavivirus groups), the creation of a new species (Karshi virus) and the assignment of Tick-borne encephalitis and Louping ill viruses to a unique species (Tick-borne encephalitis virus) including four viral types (i.e. Western Tick-borne encephalitis virus, Eastern Tick-borne encephalitis virus, Turkish sheep Tick-borne encephalitis virus and Louping ill Tick-borne encephalitis virus). The analyses also suggest a complex relationship between viruses infecting birds and those infecting mammals. Ticks that feed on both categories of vertebrates may constitute the evolutionary bridge between the three distinct identified lineages.

Faba bean (Vicia faba L.) is a globally important nitrogen-fixing legume, which is widely grown in a diverse range of environments. In this work, we mine and validate a set of 845 SNPs from the aligned transcriptomes of two contrasting inbred lines. Each V. faba SNP is assigned by BLAST analysis to a single Medicago orthologue. This set of syntenically anchored polymorphisms were then validated as individual KASP assays, classified according to their informativeness and performance on a panel of 37 inbred lines, and the best performing 757 markers used to genotype six mapping populations. The six resulting linkage maps were merged into a single consensus map on which 687 SNPs were placed on six linkage groups, each presumed to correspond to one of the six V. faba chromosomes. This sequence-based consensus map was used to explore synteny with the most closely-related crop species, lentil, and the most closely related fully sequenced genome, Medicago. Large tracts of uninterrupted colinearity were found between faba bean and Medicago, making it relatively straightforward to predict gene content and order in mapped genetic interval. As a demonstration of this, we mapped a flower colour gene to a 2 cM interval of Vf chromosome 2 which was highly collinear with Mt3. The obvious candidate gene from 77 gene models in the collinear Medicago chromosome segment was the previously characterized MtWD40-1 gene (Mt3g092830, Mt3g092840) controlling anthocyanin production in Medicago and re-sequencing of the Vf orthologue showed a putative causative deletion of the entire 5’ end of the gene.

The aim of the present study is to analyze the representation of the mythic timelessness and the religious subversion in the novel Pedro Páramo (1953) by the Mexican writer Juan Rulfo. The novel is analyzed from a narratological perspective based on the concept of focalization proposed by the French theoretician Gérard Genette. According to Genette it is possible to identify different levels of focalization depending on the position of the narrative voices.The key question in this investigation is if any salvation is possible in the universe of Pedro Páramo and how the mythic timelessness and the religious subversion are represented in the textual intentionality through the narrative voices. The main conclusion is that there is no possible salvation in the fictional universe of Pedro Páramo. The collective sin is so vast that the intermediation between God and the inhabitants of Comala does not work anymore. Thus, the only possible ”salvation” consists in the separation of the soul from the body the people of Comala experience after death. In Pedro Páramo the dead body is liberated from remorses, but - contrary to the Catholic concepts - becomes a conscious materia sentenced to remember eternally.Keywords: Juan Rulfo, Pedro Páramo, narratological analysis, Gérard Genette, textual intentionality, focalization, mythic timelessness, religious subversion, narrative voices.

Con esta tesina se pretende poner en relieve la importancia de escribir textos narrativos de modo que el lector pueda concebir lo que está leyendo como real. Por este motivo, el objetivo es ilustrar de qué modo un texto puede faltar a las pautas de credibilidad, haciendo que el lector ponga en cuestión la verosimilitud de lo relatado. Como objeto de estudio se ha elegido la novela Castillos de cartón de Almudena Grandes. Se ha realizado una interpretación hermenéutica del texto centrada en la relación que mantienen los tres protagonistas. Con el apoyo de la teoría aristotélica de la amistad y de la teoría triangular del amor de Stenberg principalmente, se muestra por qué el lector no puede aceptar como creíble que dicha relación sea de amor y amistad como afirma la narradora. Asimismo, se estudia la credibilidad de la narradora a través del análisis del discurso narrativo partiendo de los conceptos de narratología de Gérard Genette. La conclusión a la que se llega es que no se consigue crear una realidad fictiva creíble debido, por una parte, a que los sentimientos que la narradora afirma existen (amor y amistad) no están respaldados por lo que se relata y, por otra, a que el uso lingüístico de la narradora y sus razonamientos contrastan drásticamente con las expectativas del lector.

Ultimement nous répondons à la question suivante :En quoi la narration dans le roman L’Élégance du hérisson permet-elle de bien rendrela métaphore du « bocal à poissons »?

The purpose of the study is, based on a narratological perspective, to analyze and interpret the novel written by Astrid Lindgren: Mio, min Mio. It is based on the legacy of the folktale tradition, which is also the basis for carnival concept according to Bachtin. The study is based on a narrative model of Gérard Genette. Mio, min Mio is told as a first person narrator and can be interpreted as Bo tells it to himself. It is a frame story in which the outer story is the reality, while the inner story is the structure of the folktale where Mio embarks on a predetermined mission to fight against evil. Carnival concept involves a timed up-and-down-turn that is sanctioned and controlled, this fits well with this story seen from this interpretation. In the fantasy world everything has an idealized counterpart and Bo is, via Mio, in this world for a limited time. Children in our society are powerless, but are allowed to be mighty in children's literature. The story gets a subversive effect as it shows that adult standards are not absolute. Even if Bo turns back to his foster parents, he goes through this inner battle processing his thoughts and feelings. The love Bo will experience in imagination gives him the power to manage his real situation.

Este estudo sobre o romance Le Désert mauve, da escritora quebequense Nicole Brossard, foi desenvolvido tendo, como instrumentos de análise, a tradução e a identidade da América ou a americanidade. Esses temas foram determinados levando-se em conta a estrutura e o tema da obra, focalizados na figura do(a) tradutor(a), figura essa representativa da era pós-moderna. Os aspectos relacionados às questões feministas foram, até certo ponto, também abordados, principalmente pelo fato de a autora ocupar posição de destaque no meio feminista. Tendo-se em vista as especificidades do corpus, apresentamos, em um primeiro capítulo, uma contextualização do Quebec, abordando a literatura, o pós-modernismo e o feminismo quebequenses. O segundo capítulo é dedicado à obra brossardiana e seu impacto sobre o meio litérario, a partir do Quebec. Nossa análise propriamente dita do romance é apresentada no terceiro capítulo. Este trabalho tem como objetivo refletir sobre a problemática da tradução partindo-se de uma abordagem transcultural, isto é, dando-se ênfase à transformação que ela provoca. Esses traços do « novo » serão colocados em diálogo com os elementos próprios da América, como as referências culturais, a mescla de gêneros, a linguagem dos espaços abertos, as utopias, as buscas. Nosso estudo fundamenta-se basicamente nas teorias de Julia Kristeva, no que diz respeito à linguagem, ao texto; nas teorias de Henri Meschonnic e Sherry Simon, para os estudos da tradução e de Gérard Bouchard, quanto às questões que se referem à americanidade.

Esta dissertação mapeia a rede de relações intertextuais em Half a Life (2001) e sua continuação Magic Seeds (2004), os romances mais recentes do Prêmio Nobel de Literatura de 2001, V. S. Naipaul, como contribuição para o estudo da obra do autor. A noção de intertextualidade permeia os estudos literários, e o termo tem sido largamente empregado desde que foi cunhado por Julia Kristeva nos anos sessenta. Desde então as mais variadas, e muitas vezes divergentes, teorias sobre intertextualidade compartilham a idéia de que um texto só adquire significado pleno na interação com outros textos. A abordagem metodológica proposta é baseada na teoria da transtextualidade de Gérard Genette. Esta escolha implica o estudo de intertextos, paratextos, metatextos, arquitextos e hipertextos que constituem a interface entre os dois romances e outros escritos. O nome do protagonista "William Somerset Chandran" constitui o fio que guia o estudo das várias relações transtextuais nos dois romances. A partir do prenome do protagonista – William – este estudo situa os romances no contexto da tradição do Bildungsroman, e argumenta que estes estabelecem uma paródia arquitextual do gênero na medida em que subvertem seu cerne, ou seja, a formação do caráter do protagonista. O nome do meio do protagonista – Somerset – remete à ficcionalização do escritor Somerset Maugham na narrativa, ao mesmo tempo em que esta desmistifica a ótica ocidental sobre o hinduísmo popularizada por Maugham em The Razor's Edge. O sobrenome do protagonista – Chandran – leva ao estudo do conjunto de referências à origem indiana de Naipaul e o papel desta na produção do autor. Este nome se reporta ao romance de Narayan The Bachelor of Arts, cujo protagonista também é nomeado Chandran. Narayan é um escritor de destaque na literatura anglo-indiana e referência recorrente na obra de Naipaul. Os temas de migração e choque cultural apresentados nos dois romances têm sido presença constante na obra de Naipaul. Esta pesquisa mapeia a relação de continuidade entre os dois romances em questão e o conjunto da obra de Naipaul, salientando o papel da ambientação geográfica da narrativa, marcada pela jornada do protagonista através de três continentes. A teoria da transtextualidade é uma ferramenta operacional para a pesquisa, a qual examina a densidade das referências geográficas, históricas e literárias em Half a Life e Magic Seeds, visando aportar elementos para o estudo da produção literária de Naipaul, na medida em que estes romances recentes condensam e revisitam a visão de mundo deste autor.

A avaliação de risco sísmico, fundamental para as decisões sobre as estruturas de obras de engenharia e mitigação de perdas, envolve fundamentalmente a análise de ameaça sísmica. Calcular a ameaça sísmica é o mesmo que calcular a probabilidade de que certo nível de determinada medida de intensidade em certo local durante um certo tempo seja excedido. Dependendo da complexidade da atividade geológica essas estimativas podem ser bas- tante sofisticadas. Em locais com baixa sismicidade, como é o caso do Brasil, o pouco tempo (geológico) de observação e a pouca quantidade de informação são fontes de muitas incer- tezas e dificuldade de análise pelos métodos mais clássicos e conhecidos que geralmente consideram, através de opiniões de especialistas, determinadas zonas sísmicas. Serão discutidas algumas técnicas de suavização e seus fundamentos como métodos al- ternativos ao zoneamento, em seguida se exemplifica suas aplicações no caso brasileiro.

A formação de emulsão de água-em-óleo gera um significativo incremento na viscosidade, o que afeta diretamente a produção do poço, pois aumenta a perda de carga ao longo da linha de produção, dificultando o escoamento e diminuindo a produção de óleo. A presença e natureza da emulsão, e seu impacto na reologia do petróleo, podem determinar a viabilidade econômica e técnica dos processos envolvidos. A medida que a fração de água aumenta e a temperatura é reduzida, o comportamento das emulsões se torna cada vez mais não-Newtoniano. A decorrência disso, é que a temperatura e a taxa de cisalhamento passam a ter maior impacto na variação da viscosidade das emulsões. Nesse estudo são propostos novos métodos que levam em conta essas variáveis. Os dados reológicos experimentais de 15 petróleos leves foram utilizados para avaliar o desempenho dos modelos existentes na literatura e compará-los com os novos métodos propostos nesse estudo.
