Streptomyces clavuligerus é uma importante espécie industrial que produz múltiplos compostos de interesse clínico, destacando-se o antibiótico cefamicina C (CefC) e o ácido clavulânico (AC), um potente inibidor de b-lactamases produzidas por bactérias resistentes a penicilinas e cefalosporinas. Sabe-se que situações limite como subculturas sucessivas, forma inadequada de armazenamento das células, variações bruscas de condições de cultura, podem causar deleções de mais de 25% dos genes de Streptomyces sp que, embora muitas vezes não comprometam a sobrevivência do micro-organismo, acarretam perdas na capacidade produtiva. Neste trabalho, investigou-se a estabilidade da linhagem selvagem S. clavuligerus ATCC 27064 quanto à produção de CefC variando-se condições do inóculo utilizado nos cultivos submersos de obtenção do antibiótico. As fermentações foram realizadas em frascos agitados (28ºC, 260 rpm) e as amostras coletadas em 48 e 72 horas do caldo fermentativo foram analisadas quanto ao pH, biomassa (massa seca a 105ºC por 24 h) e CefC foi determinada por bioensaio de difusão em ágar (bactéria-teste Escherichia coli ESS 2235). Foi selecionado o micro-organismo melhor produtor por meio de screening, e o que apresentou a melhor produção de CefC e os menores desvios com relação à média foi utilizado para preparar novos lotes de células: ampolas de micélios liofilizados e criotubos contendo 108 esporos.mL-1 . Observou-se que a capacidade produtiva foi afetada quando foram utilizadas subculturas do micro-organismo original. A realização do screening mostrou-se eficiente na seleção de colônias superiores em termos de produção volumétrica, sendo recomendada a realização de tal procedimento periodicamente para manutenção de uma linhagem com maior potencial produtivo... This study was addressed to investigate the composition and antifungal activity of essential oils from leaves of Piperaceae species (Piper aduncum, Piper amalago, Piper cernuum, Piper diospyrifolium, Piper crassinervium, Piper gaudichaudianum, Piper solmsianum, Piper regnellii, Piper tuberculatum, Piper umbelata and Peperomia obtusifolia) against Candida albicans, C. parapsilosis, C. krusei and Cryptococcus neoformans. The essential oils from P. aduncum, P. gaudichaudianum and P. solmsianum showed the highest antifungal activity against Cryptococcus neoformans with the MIC of 62.5, 62.5 and 3.9 mg.mL-1, respectively. The oil from P. gaudichaudianum showed activity against C. krusei with MIC of 31.25 mg.mL-1.

Background: Prostate cancer is the second most common cancer diagnosed in men; however its etiology remains unknown. Previous studies have shown that environmental adverse factors, such as maternal nutritional status during pregnancy, can influence fetal development and predispose people to diseases in adult life. The feeding of low-protein diets to pregnant rats result in fetal growth disturbance, androgen/estrogen unbalance and changes in the expression and sensibility of hormone receptors in male offspring. These alterations can promote permanent changes in androgen dependent organs, such as in the prostate. In this sense, we hypothesized that the hormonal unbalance that occurs during aging can lead to an increase in the susceptibility to prostatic disorders. Aim: To evaluate our hypothesis, malnourished male rat offspring were submitted to simultaneous estrogen and testosterone exposure in adulthood, to drive lesions in the rat ventral prostate gland (VP). Methods: 17 week-old Wistar rats (n=48) that received in utero normal protein diet (NP group, AIN93G=17% protein) or low protein diet (RP group, AIN93G modified=6% protein) were given implants with 17β-estradiol plus testosterone administration (NPH and RPH groups) for 17 weeks. The animals were killed at the age of 34 weeks and the VP were excised, weighted and processed for histopathological, immunohistochemical (Ki67, AR, p63, e-caderin, laminin, c-myc and GSTP), biochemical and ultrastructural analysis. Results: Both absolute and relative VP weight from NPH animals were about 30% higher than RPH. Serological data showed that estradiol levels were similar in both groups, but testosterone levels were lower in the RPH male offspring. The steroid hormone exposure in adult life promoted prostate lesions in both RPH and NPH offspring associated with reactive stroma. VP from RPH group exhibited heightened susceptibility to prostatic intraepithelial neoplasia (mainly cribriform and signet ring-cell patterns) and increased the incidence and aggressiveness of prostatitis. In this group, a higher proportion of basal cells, increased proliferation index, lower expression ofthe androgen receptor and increased focus of collagenous micronodules closely associated to epithelial neoplasias were also observed. Conclusion:These observations suggest that maternal protein restriction alters adult prostate response to androgen/estrogen handling and increases susceptibility to prostate diseases. In order to assess the occurrence and distribution of spores and toxins of Clostridium botulinum types C and D in three farms in Cocalinho, at the Araguaia River valley, State of Mato Grosso, Brazil, we analyzed sediment samples from 40 water holes, soil and cattle feces, collected around water holes. Sediments were analyzed by direct method, whilst feces, soil and also sediment samples were individually analyzed by indirect method. The detection of spores and botulinum toxins in the filtered material was performed by bioassay in Swiss Webster mice strain, as well as the serum-neutralization of the positive materials for typing. Samples of cattle feces showed the largest positive rate for C. botulinum, with 25/40 (62.5%), followed by soil, 12/40 (30%), and by sediment, 13/40 (32.5%). From the 40 cattle feces samples, 25 (62.00%) were positive for Clostridium botulinum; six samples were identified as type C, other six as type D, and 13 samples were classified as CD complex. From the equal number (40) of soil samples, 12 (30%) were positive for C. botulinum; two samples were identified as type C, other three as type D, and seven samples were classified as CD complex. Regarding the 40 sediment samples, 13 (32.5%) were positive for C. botulinum; two samples were identified as type C, other three as type D, and eight samples were classified as CD complex. No botulism toxin was detected by indirect method.

Necrotic Enteritis (NE) caused by Clostridium perfringens (CP) in poultry is probably the most important bacterial disease in terms of economic implications. The disease is multi-factorial and is invariably associated with predisposing factors. The present study investigated the effect of a commercially available Lactobacillus-based probiotic (FM-B11) for the control of necrotic enteritis in broiler chickens. In experiment 1, one-day-of-hatch broiler chicks were randomly allocated to the following treatment groups: 1) Non-challenged (NC); 2) Challenged (C); 3) Challenged + probiotic (C+ FM-B11). Prior to placement, chicks in groups 2 and 3 received 0.25 mL of Salmonella typhimurium (ST) containing 105 cfu of viable cells by oral gavage. At 14, 15 and 16 days of age, all chicks in group 3 were treated with FM-B11 in the drinking water at a concentration of 106 cfu/ml. At 21d of age, all chicks in groups 2 and 3, were individually challenged with 5 × 104 sporulated oocysts of E. maxima by oral gavage. At 26d of age, all chicks in groups 2 and 3, were individually challenged with 108 cfu CP; body weight (BW) was recorded prior to challenge. The experiment was terminated at 29 days of age and the following parameters were evaluated: NE-associated mortality, CP lesion scores, CP concentrations in ileum, BW, and body weight gain (BWG). Chicks treated with FM-B11 had significantly (P < 0.05) higher body weight gain after challenge when compared to control challenge chickens. Total mortality was higher in the C group (48.8%) when compared to the C + FM-B11 (12.7%). Even though there was no significant (P > 0.05) difference in lesion score between C and C + FM-B11, group C + FM-B11 had significantly (P < 0.05) lower total number of cfu of CP recovered from the ileal mucosa and content samples when compared to group C. Experiment 2 was a unique and remarkable case report of a field outbreak of NE in a commercial broiler farm in Argentina. A reduction and control of the mortality associated with NE following 3 days of administration of FM-B11 was observed as compared with the control non treated house. These results imply that the commercially available Lactobacillus-based probiotic FM-B11 was able to reduce the severities of NE, as a secondary bacterial infection, in an experimental NE challenge model; as well as, in a commercial field outbreak of NE.

We report an improved measurement of (nu) over bar (mu) disappearance over a distance of 735 km using the MINOS detectors and the Fermilab Main Injector neutrino beam in a (nu) over bar (mu)-enhanced configuration. From a total exposure of 2.95 x 10(20) protons on target, of which 42% have not been previously analyzed, we make the most precise measurement of Delta(m) over bar (2) = [2.62(-0.28)(+0.31)(stat) +/- 0.09(syst)] x 10(-3) eV(2) and constrain the (nu) over bar (mu) mixing angle sin(2)(2 (theta) over bar) > 0.75 (90% C.L.). These values are in agreement with Delta m(2) and sin(2)(2 theta) measured for nu(mu), removing the tension reported in [P. Adamson et al. (MINOS), Phys. Rev. Lett. 107, 021801 (2011).].

In order to provide adequate medical assistance to neonates, the extent of vitality impairment has to be investigated through complementary exams, as well as clinical assessment. This investigation aimed to identify the physiological changes that occur during neonatal adaptation and to develop a clinical approach that can be performed during the first hour of life in neonatal lambs born through vaginal eutocic labor. The neonatal vitality of 14 Santa Ines lambs was verified using the Apgar system and rectal temperature at birth and after 5 and 60 min after birth. From the total number of neonates, 7 lambs were randomly selected for blood gas analysis and glucose immediately at birth and 1 h after birth. The lambs had hypoglycemia immediately after birth, as well as acidosis due to metabolic and respiratory causes. Given their hypoxemia at birth, lambs immediately exhibit tachycardia and tachypnea. However, neonatal lambs reached Apgar score superior than 7 after 5 min of birth. Ovine neonates are relatively mature at birth, with adequate thermoregulation and active mechanisms to compensate for physiological acid-base imbalances. In conclusion, a systematic clinical examination of newborn sheep should include the implementation of the Apgar score coupled with the confirmation of any acid-base imbalances. Further research should evaluate neonatal adaptation to this critical period over a longer period of time. (C) 2012 Elsevier B.V. All rights reserved.

Vector mesons may be photoproduced in relativistic heavy-ion collisions when a virtual photon emitted by one nucleus scatters from the other nucleus, emerging as a vector meson. The STAR Collaboration has previously presented measurements of coherent rho(0) photoproduction at center of mass energies of 130 GeV and 200 GeV in AuAu collisions. Here, we present a measurement of the cross section at 62.4 GeV; we find that the cross section for coherent rho(0) photoproduction with nuclear breakup is 10.5 +/- 1.5 +/- 1.6mb at 62.4 GeV. The cross-section ratio between 200 GeV and 62.4 GeV is 4.4 +/- 0.6, less than is predicted by most theoretical models. It is, however, proportionally much larger than the previously observed 15% +/- 55% increase between 130 GeV and 200 GeV.
