For contain beneficial properties, aluminum alloys are gaining more importance in different industrial areas, becoming the subject of study in several academic fields. When related to welding these alloys have some peculiarities that may hinder the union, such as microscopic oxide layer present on the metal surface. The MIG welding process, also known as GMAW, has developed versions that can be effective for welding aluminum. Knowing this, for this paper, two versions of pulsed MIG (CC + and CA) were chosen to evaluate which best suits pass by filling bevel on AA5083 aluminum sheets with 8 and 12 mm thick respectively. Furthermore, two types of wire, ER5087 and ER5183 were evaluated. To evaluate the process and versions of the wires, the high-speed cameras and thermal were used to monitor the metal transfer and the thermal behavior respectively, and the metallographic analysis for macrographic view of the weld beads and non-destructive testing by radiography for observation of possible discontinuities. It was found that the technique of MIG-P CA showed better results ahead of another technique both welding conditions imposed. In the last decades, analogue modelling has been used in geology to improve the knowledge of how geological structures are nucleated, how they grow and what are the main important points in such processes. The use of this tool in the oil industry, to help seismic interpretations and mainly to search for structural traps contributed to disseminate the use of this tool in the literature. Nowadays, physical modelling has a large field of applications, since landslide to granite emplacement along shear zones. In this work, we deal with physical modelling to study the influence of mechanical stratifications in the nucleation and development of faults and fractures in a context of orthogonal and conjugated oblique basins. To simulate a mechanical stratigraphy we used different materials, with distinct physical proprieties, such as gypsum powder, glass beads, dry clay and quartz sand. Some experiments were run along with a PIV (Particle Image Velocimetry), an instrument that shows the movement of the particles to each deformation moment. Two series of experiments were studied: i) Series MO: We tested the development of normal faults in a context of an orthogonal (to the extension direction) basin. Experiments were run taking into account the change of materials and strata thickness. Some experiments were done with sintectonic sedimentation. We registered differences in the nucleation and growth of faults in layers with different rheological behavior. The gypsum powder layer behaves in a more competent mode, which generates a great number of high angle fractures. These fractures evolve to faults that exhibit a higher dip than when they cross less competent layers, like the one of quartz sand. This competent layer exhibits faulted blocks arranged in a typical domino-style. Cataclastic breccias developed along the faults affecting the competent layers and showed different evolutional history, depending on the deforming stratigraphic sequence; ii) Series MOS2: Normal faults were analyzed in conjugated sub-basins (oblique to the extension direction) developed in a sequence with and without rheological contrast. In experiments with rheological contrast, two important grabens developed along the faulted margins differing from the subbasins with mechanical stratigraphy. Both experiments developed oblique fault systems and, in the area of sub-basins intersection, faults traces became very curved.

Bionanocomposites systems clay base (montmorillonite and sepiolite), layered double hidroxides and biopolymers (carboxymethylcellulose and zein) were evaluated as topical delivery systems with antibacterial activity and as oral delivery systems. For this study, neomycin, a topical antibiotic, indicated mainly for open wound infections. The drug amoxicillin, an antibiotic indicated mainly for throat infections, were also used in this study. Both antibiotics were used as model drugs. Initially, drugs were incorporated directly into the biopolymer matrix, comprising the combination of carboxymethylcellulos and zein, being conformed as movies and balls and evaluated for their antibacterial activity and controlled release simulating gastrointestinal fluids. Moreover, hybrids materials have been prepared where the neomycin drug was incorporated into the lamellar inorganic solids, such as montmorillonite by ion exchange reaction, and the fibrous type, such as sepiolite by adsorption in aqueous solution. But the drug amoxicillin was incorporated into layered double hydroxides by anion exchange and montmorillonite by cation exchange. The resulting hybrids were in turn combined with the biopolymer matrix yielding bionanocomposites shaped materials such as films were tested for their antibacterial activity, and the shaped materials beads were tested for their release in the gastrointestinal fluids. Through the analysis of various physico-chemical techniques, we observed the interactions between the studied materials, the formation of hybrids materials, obtaining the bionanocomposites materials and material efficiency when applied in controlled release of drugs both topical and use oral mainly influenced by the presence of zein, are promising as topical delivery systems and oral drugs.

The maintenance of masticatory function is especially important for patients wearing complete dentures due to their limitations. Thus, the bilateral balanced occlusal concept is used to achieve greater masticatory efficiency. However, a critical review of the literature reveals that there is not sufficient scientific evidence to support bilateral balanced occlusion as the most appropriate occlusal concept in complete dentures. Therefore, the aim of this study was to evaluate the masticatory efficiency in complete dentures wearers with bilateral balanced occlusion and canine guidance. A double-blinded controlled crossover clinical trial was conducted. The sample was composed by 24 edentulous patients who wore sets of complete dentures with both occlusal concepts during equal periods of 3 months. Objective data were collected through the masticatory efficiency test performed by the colorimetric method with the beads, in which capsules of a synthetic material enclosing fuchsine-containing granules were used. Subjective data were recorded by patient's ratings of their chewing function. No significant statistical difference was found for masticatory efficiency (p=0.095) between the two occlusal concepts studied. The results suggest that bilateral balanced occlusion does not improve the masticatory efficiency in complete denture wearers.

Commercial exploitation and abrupt changes of the natural conditions may have severe impacts on the Arctic deep-sea ecosystem. The present recolonisation experiment mimicked a situation after a catastrophic disturbance (e.g. by turbidites caused by destabilized continental slopes after methane hydrate decomposition) and investigated if the recolonisation of a deep-sea habitat by meiobenthic organisms is fostered by variations innutrition and/or sediment structure. Two "Sediment Tray Free Vehicles" were deployed for one year in summer 2003 at 2500 m water depth in the Arctic deep-sea in the eastern Fram Strait. The recolonisation trays were filled with different artificial and natural sediment types (glass beads, sand, sediment mixture, pure deep-sea sediment) and were enriched with various types of food (algae, yeast, fish). After one year, meiobenthos abundances and various sediment related environmental parameters were investigated. Foraminifera were generally the most successful group: they dominated all treatments and accounted for about 87% of the total meiobenthos. Colonizing meiobenthos specimens were generally smaller compared to those in the surrounding deep-sea sediment, suggesting an active recolonisation by juveniles. Although experimental treatments with fine-grained, algae-enriched sediment showed abundances closest to natural conditions, the results suggest that food availability was the main determining factor for a successful recolonisation by meiobenthos and the structure of recolonised sediments was shown to have a subordinate influence.

Welding is one of the most employed process for joining steel pipes. Although, manual welding is still the most used one, mechanized version and even automatized one have increased its demand. Thus, this work deals with girth welding of API 5L X65 pipes with 8” of nominal diameter and 8.0 mm thickness, beveled with V-30º narrow gap. Torch is moved by a bug carrier (mechanized welding) and further the parameters are controlled as a function of angular position (automatized welding). Welding parameters are presented for filling the joint with two-passes (root and filling/capping passes). Parameters for the root pass were extracted from previous author´s work with weldments carried out in plates, but validated in this work for pipe welding. GMAW processes were assessed with short-circuit metal transfer in both conventional and derivative modes using different technologies (RMD, STT and CMT). After the parameter determination, mechanical testing was performed for welding qualification (uniaxial tension, face and root bending, nick break, Charpy V-notch impact, microhardness and macrograph). The initially obtained results for RMD and CMT were acceptable for all testing and, in a second moment, also for the STT. However, weld beads carried out by using the conventional process failed and revealed the existence of lack of fusion, which required further parametrization. Thus, a Parameter-Variation System for Girth Welding (SVP) was designed and built to allow varying the welding parameters as a function of angular position by using an inclinometer. The parameters were set for each of the three angular positions (flat, vertical downhill and overhead). By using such equipment and approach, the conventional process with parameter variation allowed reducing the welding time for joint accomplishment of the order of 38% for the root pass and 30% for the filling/capping pass.

This thesis demonstrates a new way to achieve sparse biological sample detection, which uses magnetic bead manipulation on a digital microfluidic device. Sparse sample detection was made possible through two steps: sparse sample capture and fluorescent signal detection. For the first step, the immunological reaction between antibody and antigen enables the binding between target cells and antibody-­‐‑ coated magnetic beads, hence achieving sample capture. For the second step, fluorescent detection is achieved via fluorescent signal measurement and magnetic bead manipulation. In those two steps, a total of three functions need to work together, namely magnetic beads manipulation, fluorescent signal measurement and immunological binding. The first function is magnetic bead manipulation, and it uses the structure of current-­‐‑carrying wires embedded in the actuation electrode of an electrowetting-­‐‑on-­‐‑dielectric (EWD) device. The current wire structure serves as a microelectromagnet, which is capable of segregating and separating magnetic beads. The device can achieve high segregation efficiency when the wire spacing is 50µμm, and it is also capable of separating two kinds of magnetic beads within a 65µμm distance. The device ensures that the magnetic bead manipulation and the EWD function can be operated simultaneously without introducing additional steps in the fabrication process. Half circle shaped current wires were designed in later devices to concentrate magnetic beads in order to increase the SNR of sample detection. The second function is immunological binding. Immunological reaction kits were selected in order to ensure the compatibility of target cells, magnetic bead function and EWD function. The magnetic bead choice ensures the binding efficiency and survivability of target cells. The magnetic bead selection and binding mechanism used in this work can be applied to a wide variety of samples with a simple switch of the type of antibody. The last function is fluorescent measurement. Fluorescent measurement of sparse samples is made possible of using fluorescent stains and a method to increase SNR. The improved SNR is achieved by target cell concentration and reduced sensing area. Theoretical limitations of the entire sparse sample detection system is as low as 1 Colony Forming Unit/mL (CFU/mL).

Historical archaeology, in its narrow temporal sense -as an archaeology of the emergence and subsequent evolution of the Modern world- is steadily taking pace in Spanish academia. This paper aims at provoking a more robust debate through understanding how Spanish historical archaeology is placed in the international scene and some of its more relevant particularities. In so doing, the paper also stresses the strong links that have united historical and prehistorical archaeology since its inception, both in relation to the ontological, epistemological and methodological definition of the first as to the influence of socio-political issues in the latter. Such reflection is partly a situated reflection from prehistory as one of the paper’s authors has been a prehistorian for most of her professional life.

O objetivo deste trabalho foi utilizar diferentes técnicas químicas (dimetilsulfóxido, ácido clorídrico, acético e lático), técnicas mecânicas (banho ultrassônico, abrasão com pérolas de vidro, maceração com terra diatomácea, ruptor ultrassônico e imersão em nitrogênio líquido) e técnica enzimática (preparado enzimático comercial Glucanex®) para a recuperação de carotenoides a partir da ruptura da parede celular das leveduras Sporidiobolus pararoseus e Rhodotorula mucilaginosa isoladas de amostras ambientais. Para isso a obtenção de biomassa foi realizada através de cultivos submersos no meio YM, a 25 °C, 180 rpm por 168 h. Para a ruptura celular, a operação de congelamento da biomassa (-18°C por 48 h) foi estudada. Os métodos de secagem convencional por ar forçado (35°C/48h) e liofilização (-80°C/48h, em ultrafreezer, seguido de liofilizador até alcançar 2% de umidade da amostra) também foram avaliados. Nas técnicas químicas aplicadas, o dimetilsulfóxido apresentou os melhores resultados para as duas leveduras, porém o seu uso é limitado devido a sua toxicidade. Para S. pararoseus, os maiores valores encontrados foram para o ácido clorídrico, seguido do acético e do lático, sendo detectada diferença entre eles quando aplicado o congelamento. Com R. mucilaginosa, os maiores valores foram encontrados para os ácidos acético e lático, seguido do ácido clorídrico, no qual o congelamento da biomassa também não influenciou a recuperação dos carotenoides. Dentre as técnicas mecânicas estudadas, para a levedura S. pararoseus, o banho ultrassônico e a abrasão com pérolas de vidro apresentaram os resultados mais promissores comparados ao DMSO (84,79±2,34 e 76,87±2,06 μg/g respectivamente), onde o processo de congelamento da biomassa não influenciou positivamente no percentual de extratibilidade e na concentração específica dos carotenoides quando utilizada estas técnicas. Com Rhodotorula mucilaginosa, o banho ultrassônico propiciou a recuperação da maior concentração específica de carotenoides (193,5±25,8 μg/g), sendo que o processo de congelamento também não influenciou positivamente no percentual de extratibilidade e na concentração específica dos carotenoides. Através do Delineamento Central Composto Rotacional (DCCR) 23 foi possível avaliar que a levedura S. pararoseus não demonstrou nenhum efeito sob as variáveis pH, temperatura e concentração de enzima. Assim, a melhor condição de trabalho escolhida foi pH 7,4, 30 ºC e concentração de enzima de 1,0 g/gcs, onde apresentou a concentração específica de 42,6 μg/g e volumétrica de 308 μg/L de carotenoides. Para R. mucilaginosa, a condição ótima foi definida como 1,0 g/gcs, pH 5,0 e temperatura de 30 ºC, onde foi encontrado 115,1±8,1 μg/g e 470,1±38,8 μg/L para a concentração específica e volumétrica de carotenoides, respectivamente. A utilização de técnicas combinadas empregando banho ultrassônico e lise enzimática não proporcionou melhorias nos resultados para ambas as leveduras. A liofilização provocou um ganho de 20% e 13,7% na concentração específica dos carotenoides das leveduras S. pararoseus e R. mucilaginosa, respectivamente, onde o congelamento da biomassa não influenciou significativamente (p<0,05) a recuperação de carotenoides provenientes das duas leveduras, podendo ser eliminada do processo. Assim, para S. pararoseus o banho ultrassônico e as pérolas de vidro apresentaram os melhores resultados na recuperação de carotenoides, e para R. mucilaginosa o melhor resultado foi alcançado com o banho ultrassônico.

O uso de corantes sintéticos na indústria de alimentos tem provocado transtornos à saúde humana e ao meio ambiente. A quitosana pode ser imobilizada em matrizes sólidas e aplicada na remoção de corantes em coluna de leito fixo. A análise da dinâmica de uma coluna de leito fixo é baseada na curva de ruptura, esta é dependente da geometria da coluna, das condições operacionais e dos dados de equilíbrio. Neste contexto, o objetivo deste trabalho foi estudar o recobrimento de esferas de vidro por quitosana e sua aplicação como adsorvente de corantes em coluna de leito fixo. No estudo do recobrimento avaliaram-se os efeitos da concentração de quitosana e dos métodos de cura. As esferas recobertas foram aplicadas em ensaios de adsorção estático e dinâmico. Inicialmente, avaliou-se o equilíbrio de adsorção através da construção de isotermas e ajuste de modelos, e após, avaliaram-se os efeitos do tipo de cura e do grau de desacetilação da quitosana. Em seguida, foram analisados os efeitos do tipo de corante e do pH, e o comportamento cinético da adsorção pela construção de curvas de ruptura e ajuste de modelos dinâmicos. A influência da altura do leito e da concentração inicial de corante sobre os parâmetros da adsorção em leito fixo foram analisados através da metodologia de superfície de resposta (MSR). Ao final, estudou-se a regeneração da coluna. Os resultados mostraram que os maiores percentuais de recobrimento foram obtidos pelos métodos físico e físico/químico, na concentração de quitosana de 0,5% (m/v). Nestas condições o percentual de recobrimento foi de 46%. Nas imagens da superfície das esferas (MEV) observou-se que as mesmas foram recobertas de forma homogênea pela quitosana. As isotermas de equilíbrio obtidas foram classificadas como do tipo V, sendo o modelo de Sips o mais adequado para representar os dados experimentais. As capacidades máximas de adsorção foram 337 mg g-1, 286 mg g-1 e 200 mg g-1 para os corantes amarelo tartrazina, amarelo crepúsculo e vermelho 40, respectivamente. A aplicação das esferas recobertas com quitosana em leito fixo mostrou-se mais adequada utilizando o método de cura físico/químico e quitosana com grau de desacetilação de 85%. A máxima capacidade de adsorção da coluna em função do corante e do pH variou de 13 a 108 mg g–1. Os modelos BDST (bed–depth–service–time), Thomas e Yoon–Nelson foram adequados para representar os dados experimentais. De acordo com a MSR, o melhor desempenho do leito foi com altura de 30 cm e concentração inicial de corante de 50 mg L-1. Nestas condições, obteve-se tempo de ruptura de 88 min, máxima capacidade da coluna de 108 mg g-1 e remoção de 86 %. Na regeneração da coluna observou-se que cerca de 75% da capacidade máxima da coluna foi mantida após cinco ciclos de adsorção–eluição. Diante do exposto, a coluna de leito fixo empacotada com esferas recobertas com quitosana mostrou-se promissora na remoção de corantes de soluções aquosas.

Microplastics are present in marine habitats worldwide and may be ingested by low trophic organisms such as fish larvae, with uncertain physiological consequences. The present study aims at assessing the impact of polyethylene (PE 10-45µM) microbeads ingestion in European sea bass (Dicentrarchus labrax) larvae. Fish were fed an inert diet including 0, 104 and 105 fluorescent microbeads per gram from 7 until 43 days post-hatching (dph). Microbeads were detected in the gastrointestinal tract in all fish fed diet incorporating PE. Our data revealed an efficient elimination of PE beads from the gut since no fluorescent was observed in the larvae after 48h depuration. While the mortality rate increased significantly with the amount of microbeads scored per larvae at 14 and 20 dph, only ingestion of the highest concentration slightly impacted mortality rates. Larval growth and inflammatory response through Interleukine-1-beta (IL-1) gene expression were not found to be affected while cytochrome-P450-1A1 (cyp1a1) expression level was significantly positively correlated with the number of microbeads scored per larva at 20 dph. Overall, these results suggest that ingestion of PE microbeads had limited impact on sea bass larvae possibly due to their high potential of egestion

Experimental characterization of molecular details is challenging, and although single molecule experiments have gained prominence, oligomer characterization remains largely unexplored. The ability to monitor the time evolution of individual molecules while they self assemble is essential in providing mechanistic insights about biological events. Molecular dynamics (MD) simulations can fill the gap in knowledge between single molecule experiments and ensemble studies like NMR, and are increasingly used to gain a better understanding of microscopic properties. Coarse-grained (CG) models aid in both exploring longer length and time scale molecular phenomena, and narrowing down the key interactions responsible for significant system characteristics. Over the past decade, CG techniques have made a significant impact in understanding physicochemical processes. However, the realm of peptide-lipid interfacial interactions, primarily binding, partitioning and folding of amphipathic peptides, remains largely unexplored compared to peptide folding in solution. The main drawback of existing CG models is the inability to capture environmentally sensitive changes in dipolar interactions, which are indigenous to protein folding, and lipid dynamics. We have used the Drude oscillator approach to incorporate structural polarization and dipolar interactions in CG beads to develop a minimalistic peptide model, WEPPROM (Water Explicit Polarizable PROtein Model), and a lipid model WEPMEM (Water Explicit Polarizable MEmbrane Model). The addition of backbone dipolar interactions in a CG model for peptides enabled us to achieve alpha-beta secondary structure content de novo, without any added bias. As a prelude to studying amphipathic peptide-lipid membrane interactions, the balance between hydrophobicity and backbone dipolar interactions in driving ordered peptide aggregation in water and at a hydrophobic-hydrophilic interface, was explored. We found that backbone dipole interactions play a crucial role in driving ordered peptide aggregation, both in water and at hydrophobic-hydrophilic interfaces; while hydrophobicity is more relevant for aggregation in water. A zwitterionic (POPC: 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine) and an anionic lipid (POPS: 1-palmitoyl-2-oleoyl-sn-glycero-3-phospho-L-serine) are used as model lipids for WEPMEM. The addition of head group dipolar interactions in lipids significantly improved structural, dynamic and dielectric properties of the model bilayer. Using WEPMEM and WEPPROM, we studied membrane-induced peptide folding of a cationic antimicrobial peptide with anticancer activity, SVS-1. We found that membrane-induced peptide folding is driven by both (a) cooperativity in peptide self interaction and (b) cooperativity in membrane-peptide interactions. The dipolar interactions between the peptide and the lipid head-groups contribute to stabilizing folded conformations. The role of monovalent ion size and peptide concentration in driving lipid domain formation in anionic/zwitterionic lipid mixtures was also investigated. Our study suggest monovalent ion size to be a crucial determinant of interaction with lipid head groups, and hence domain formation in lipid mixtures. This study reinforces the role of dipole interactions in protein folding, lipid membrane properties, membrane induced peptide folding and lipid domain formation. Therefore, the models developed in this thesis can be used to explore a multitude of biomolecular processes, both at longer time-scales and larger system sizes.

Muito interesse tem sido focado no potencial biotecnológico das microalgas, principalmente devido à identificação de diversas substâncias sintetizadas por estes organismos, dentre elas a anidrase carbônica e as ficobiliproteínas. A anidrase carbônica é uma metaloenzima que catalisa a hidratação reversível do CO2 em bicarbonato com alta eficiência, sendo utilizada para captação de CO2 através de sistemas biológicos. A C-ficocianina e a aloficocianina, corantes naturais, são os dois principais componentes das ficobiliproteínas em cianobactérias e apresentam diversas aplicações dentro da indústria alimentícia, cosmética e farmacêutica. O objetivo principal desta tese foi avaliar a produção e a extração da anidrase carbônica e das ficobiliproteínas a partir de diferentes microalgas. Para isso, primeiramente foi realizado uma investigação da produção da anidrase carbônica pela microalga Dunaliella tertiolecta, onde foi estudada a extração da enzima e sua aplicação em sistemas de captura enzimática de CO2. Posteriormente foi avaliada a produção da enzima ao longo do cultivo de diferentes microalgas marinhas e dulcícolas (Dunaliella tertiolecta, Tetraselmis sueccica, Phaeodactylum tricornutum, Nannochloropsis oculata, Isochysis galbana, Chlorella vulgaris e Scenedesmus obliquus). A produção da enzima e de ficobiliproteínas, também, foi estudada para as cianobactérias Spirulina platensis LEB 52, Spirulina sp. LEB 18 e Synechococcus nidulans. Todos os cultivos foram acompanhados em termos de biomassa e pH. Por último, foi realizado um estudo de extração da enzima de P. tricornutum e extração conjunta da anidrase carbônica e de ficobiliproteínas da cianobactéria S. sp. LEB 18. Os cultivos foram realizados em frascos erlenmeyer contendo os meios Conway (marinhas), BG-11 (dulcícolas) e Zarrouk 20% (cianobactérias). Na avaliação da ruptura celular foram testadas as técnicas de maceração em gral e pistilo, agitação em vórtex com pérolas de vidro, sonicação com pérolas de vidro, homogeneizador ultrassônico, secagem, congelamento e descongelamento e a combinação de tratamentos. Maiores rendimentos de extração da enzima a partir da microalga D. tertiolecta foram obtidos utilizando tratamento ultrassônico, juntamente com baixas concentrações de biomassa úmida (0,1 e 0,2 g/L), e a mesma apresentou potencial para aplicação em processos de captação enzimática do CO2. Durante os cultivos, a microalga C. vulgaris se destacou como maior produtora da enzima anidrase carbônica, atingindo valores de atividade enzimática de 44,0 U/L. As cianobactérias apresentaram valores de atividade entre 41,6 e 45,9 U/L, sendo que a S. sp. LEB 18 foi a que apresentou maiores produções de C-ficocianina e aloficocianina no ponto de máxima atividade volumétrica, 65,9 e 82,2 µg/mL, respectivamente. A enzima extraída da biomassa de S. platensis LEB 52 catalisou a hidratação do CO2 que precipitou na forma de CaCO3. Maiores rendimentos de extração da enzima a partir das microalgas P. tricornutum e S. sp. LEB 18 foram obtidos utilizando homogeneizador ultrassônico, que foram 31,3 U/g e 25,5 U/g, respectivamente. A biomassa de S. sp. LEB 18, também apresentou potencial para a extração de ficobiliproteínas, obtendo- se altas concentrações de C-ficocianina (100,5 mg/g) e aloficocianina (69,9 mg/g). Através dos resultados obtidos, pode-se verificar a potencialidade das microalgas e das cianobactérias para produção da enzima anidrase carbônica e das ficobiliproteínas, biomoléculas de alto valor industrial. Este trabalho apresenta processos eficientes para a extração da enzima e de ficobiliproteínas tanto para escala laboratorial como industrial.
