Antioxidant and inflammatory aspects of lipoprotein-associated phospholipase A2 (Lp-PLA2 ): a review

The association of cardiovascular events with Lp-PLA2 has been studied continuously today. The enzyme has been strongly associated with several cardiovascular risk markers and events. Its discovery was directly related to the hydrolysis of the platelet-activating factor and oxidized phospholipids, which are considered protective functions. However, the hydrolysis of bioactive lipids generates lysophospholipids, compounds that have a pro-inflammatory function. Therefore, the evaluation of the distribution of Lp-PLA2 in the lipid fractions emphasized the dual role of the enzyme in the inflammatory process, since the HDL-Lp-PLA2 enzyme contributes to the reduction of atherosclerosis, while LDL-Lp-PLA2 stimulates this process. Recently, it has been verified that diet components and drugs can influence the enzyme activity and concentration. Thus, the effects of these treatments on Lp-PLA2 may represent a new kind of prevention of cardiovascular disease. Therefore, the association of the enzyme with the traditional assessment of cardiovascular risk may help to predict more accurately these diseases.

Aims: The renin–angiotensin system (RAS) plays a major role in cardiovascular diseases in postmenopausal women, but little is known about its importance to lower urinary tract symptoms. In this study we have used the model of ovariectomized (OVX) estrogen-deficient rats to investigate the role of RAS in functional and molecular alterations in the urethra and bladder. Main methods: Responses to contractile and relaxant agents in isolated urethra and bladder, as well as cystometry were evaluated in 4-month OVX Sprague–Dawley rats. Angiotensin-converting enzyme activity and Western blotting for AT1/AT2 receptors were examined. Key findings: Cystometric evaluations in OVX rats showed increases in basal pressure, capacity and micturition frequency, as well as decreased voiding pressure. Angiotensin II and phenylephrine produced greater urethral contractions in OVX compared with Sham group. Carbachol-induced bladder contractions were significantly reduced in OVX group. Relaxations of urethra and bladder to sodium nitroprusside and BAY 41-2272 were unaffected by OVX. Angiotensin-converting enzyme activity was 2.6-fold greater (p < 0.05) in urethral tissue of OVX group,whereas enzyme activity in plasma and bladder remained unchanged. Expressions of AT1 and AT2 receptors in the urethra were markedly higher in OVX group. In bladder, AT1 receptors were not detected, whereas AT2 receptor expression was unchanged between groups. 17β-Estradiol replacement (0.1 mg/kg, weekly) or losartan (30 mg/kg/day) largely attenuated most of the alterations seen in OVX group. Significance: Prolonged estrogen deprivation leads to voiding dysfunction and urethral hypercontractility that are associated with increased ACE activity and up-regulation of angiotensin AT1/AT2 receptor in the urethral tissue.

BACKGROUND: Generation of active procoagulant cofactor factor Va (FVa) and its subsequent association with the enzyme activated factor X (FXa) to form the prothrombinase complex is a pivotal initial event in blood coagulation and has been the subject of investigative effort, speculation, and controversy. The current paradigm assumes that FV activation is initiated by limited proteolysis by traces of (meizo) thrombin. METHODS AND RESULTS: Recombinant tick salivary protein TIX-5 was produced and anticoagulant properties were studied with the use of plasma, whole blood, and purified systems. Here, we report that TIX-5 specifically inhibits FXa-mediated FV activation involving the B domain of FV and show that FXa activation of FV is pivotal for plasma and blood clotting. Accordingly, tick feeding is impaired on TIX-5 immune rabbits, displaying the in vivo importance of TIX-5. CONCLUSIONS: Our data elucidate a unique molecular mechanism by which ticks inhibit the host's coagulation system. From our data, we propose a revised blood coagulation scheme in which direct FXa-mediated FV activation occurs in the initiation phase during which thrombin-mediated FV activation is restrained by fibrinogen and inhibitors.

[EN] Many ecologically important chemical transformations in the ocean are controlled by biochemical enzyme reactions in plankton. Nitrogenase regulates the transformation of N2 to ammonium in some cyanobacteria and serves as the entryway for N2 into the ocean biosphere. Nitrate reductase controls the reduction of NO3 to NO2 and hence new production in phytoplankton. The respiratory electron transfer system in all organisms links the carbon oxidation reactions of intermediary metabolism with the reduction of oxygen in respiration. Rubisco controls the fixation of CO2 into organic matter in phytoplankton and thus is the major entry point of carbon into the oceanic biosphere. In addition to these, there are the enzymes that control CO2 production, NH4 excretion and the fluxes of phosphate. Some of these enzymes have been recognized and researched by marine scientists in the last thirty years. However, until recently the kinetic principles of enzyme control have not been exploited to formulate accurate mathematical equations of the controlling physiological expressions. Were such expressions available they would increase our power to predict the rates of chemical transformations in the extracellular environment of microbial populations whether this extracellular environment is culture media or the ocean. Here we formulate from the principles of bisubstrate enzyme kinetics, mathematical expressions for the processes of NO3 reduction, O2 consumption, N2 fixation, total nitrogen uptake.

Although nickel is a toxic metal for living organisms in its soluble form, its importance in many biological processes recently emerged. In this view, the investigation of the nickel-dependent enzymes urease and [NiFe]-hydrogenase, especially the mechanism of nickel insertion into their active sites, represent two intriguing case studies to understand other analogous systems and therefore to lead to a comprehension of the nickel trafficking inside the cell. Moreover, these two enzymes have been demonstrated to ensure survival and colonization of the human pathogen H. pylori, the only known microorganism able to proliferate in the gastric niche. The right nickel delivering into the urease active site requires the presence of at least four accessory proteins, UreD, UreE, UreF and UreG. Similarly, analogous process is principally mediated by HypA and HypB proteins in the [NiFe]-hydrogenase system. Indeed, HpHypA and HpHypB also have been proposed to act in the activation of the urease enzyme from H. pylori, probably mobilizing nickel ions from HpHypA to the HpUreE-HpUreG complex. A complete comprehension of the interaction mechanism between the accessory proteins and the crosstalk between urease and hydrogenase accessory systems requires the determination of the role of each protein chaperone that strictly depends on their structural and biochemical properties. The availability of HpUreE, HpUreG and HpHypA proteins in a pure form is a pre-requisite to perform all the subsequent protein characterizations, thus their purification was the first aim of this work. Subsequently, the structural and biochemical properties of HpUreE were investigated using multi-angle and quasi-elastic light scattering, as well as NMR and circular dichroism spectroscopy. The thermodynamic parameters of Ni2+ and Zn2+ binding to HpUreE were principally established using isothermal titration calorimetry and the importance of key histidine residues in the process of binding metal ions was studied using site-directed mutagenesis. The molecular details of the HpUreE-HpUreG and HpUreE-HpHypA protein-protein assemblies were also elucidated. The interaction between HpUreE and HpUreG was investigated using ITC and NMR spectroscopy, and the influence of Ni2+ and Zn2+ metal ions on the stabilization of this association was established using native gel electrophoresis, light scattering and thermal denaturation scanning followed by CD spectroscopy. Preliminary HpUreE-HpHypA interaction studies were conducted using ITC. Finally, the possible structural architectures of the two protein-protein assemblies were rationalized using homology modeling and docking computational approaches. All the obtained data were interpreted in order to achieve a more exhaustive picture of the urease activation process, and the correlation with the accessory system of the hydrogenase enzyme, considering the specific role and activity of the involved protein players. A possible function for Zn2+ in the chaperone network involved in Ni2+ trafficking and urease activation is also envisaged.

Zusammenfassung In der vorliegenden Arbeit wurden im Zuge derAjmalinbiosynthese in Rauvolfia serpentina die NADPH2abhängigen Reduktionsschritte des Alkaloids Vomilenin zu 17O Acetylnorajmalin genauer untersucht.Dabei konnte erstmals die exakte Reaktionsreihenfolgeaufgedeckt und die daran beteiligten Enzyme ausPflanzenzellsuspensionskulturen isoliert und aufgereinigtwerden. Die ausgearbeiteten, optimierten Reinigungsprotokolleführten in wenigen Stufen gezielt zu den voneinandergetrennten Reduktase Fraktionen. Durch die Trennung derReduktase Aktivitäten war der Grundstein gelegt, dasZwischenprodukt der Reaktion anzureichern und mitverschiedenen analytischen Verfahren als 2?-(R)-1.2Dihydrovomilenin zu identifizieren. Die daraufhin vergebenenBezeichnungen Vomilenin Reduktase (EC.1.5.1.32) und 1.2Dihydrovomilenin Reduktase (EC.1.3.1.73) zielen auf dasumzusetzende Substrat ab.Für die Vomilenin Reduktase konnte eine 4 stufige Reinigungüber (NH4)2SO4 Fällung, Anionen-austauschchromatographie mitSOURCE 30Q, Hydrophobe Interaktionschromatographie an SOURCE15Phe und Affinitätschromatographie mit 2,5 ADP Sepharoseausgearbeitet werden. Hierbei konnte die 1.2Dihydrovomilenin Reduktase schon nach dem erstensäulenchromatogra-phischen Schritt (S 30Q) abgetrenntwerden. Das am Ende der Proteinreinigung angefertigte SDSGel zeigte nur noch 3 Banden, von denen die beiden bei ca.40 und 43 kDa gelegenen Banden mit dem Aktivitätsverlaufder Vomilenin Reduktase korrelierten. Diese wurden einempartiellen Verdau mit der Endoproteinase LysC unterworfen,wobei jeweils 2 Spaltpeptide erhalten werden konnten.Die 1.2 Dihydrovomilenin Reduktase Reinigung umfaßte 6Reinigungsschritte mit (NH4)2SO4-Fällung, SOURCE 30QAnionenaustauschchromatographie, HydroxylapatitChromatographie, 2,5 ADP Sepharose-Chromatographie,Anionenaustausch an DEAE Sepharose und abschließen-denAnionenaustausch über MonoQ. Die resultierendeProteinfraktion wies eine ca. 200 fache Anreicherung an 1.2Dihydrovomilenin Reduktase auf. Auch hierbei wurde die nachSDS Gelelek-trophorese als 1.2 Dihydrovomilenin Reduktasebestimmte Proteinbande (bei ca. 48 kDa) sequen-ziert. Eskonnten vier Peptidfragmente erhalten werden, die ebenso wiedie sequenzierten Peptidstücke der 40 und 43 kDa Bande einehohe Homologie zu Oxidoreduktasen, im einzelnen zuCinnamoylalcohol- und Mannitol Dehydrogenasen, aufwiesen.Um die Identität der sequenzierten Proteinbanden zubestätigen, wurde über reverse genetics die jeweilscodierende cDNA eruiert. Dafür wurden - ausgehend von denPeptidstücken der Mikro-sequenzierung - degenerierte Primerentwickelt und über PCR Teilbereiche der cDNA amplifiziert.Diese konnten für eine radioaktive Durchmusterung einerRauvolfia cDNA Bank herangezogen werden. Alternativ botallein die Kenntnis der spezifischen Nukleotidabfolge dieMöglichkeit der Gewinnung von 5 und 3 Ende derVollängenklone durch RACE PCR.Nach Abschluß dieser Arbeiten konnten für die 40 und 48 kDaBande je ein Vollängenklon und für die 43 kDa Bande 2Vollängenklone (Isoformen) gefunden werden. SämtlicheVollängenklone besitzen einen offenen Leserahmen, der durchnicht zu translatierende Bereiche am 5 und 3 Endeeingefaßt wird. Um die entsprechenden Proteine produzierenzu können, mußten die dafür codierenden cDNA Bereiche dereinzelnen Klone in ein geeignetes Vektor Wirt-System(Expressionssystem) eingebracht werden.Nach erfolgreicher Umklonierung wurde die Expression durchIPTG Zugabe kontrolliert und Proteinrohextrakte aus denBakterienstämmen isoliert. Als Substrate wurden Vomilenin,das strukturisomere Alkaloid Perakin und aufgrund derHomologien zu Cinnamoylalcohol und Mannitol Dehydrogenasen Zimtaldehyd, Dihydrozimtaldehyd und D(-)Fructose getestet. In allen E. coli Stämmen konnte ein unspezifischesReduktionspotential nachgewiesen werden, ohne daß jedochVomilenin reduziert wurde. Die Testung der 1.2Dihydrovomilenin Reduktase Klone mußte wegen Substratmangelentfallen.Die weitere Charakterisierung der pflanzlichen Enzymeerbrachte eine enorm hohe Substratspezifität mit einer sichauf Rauvolfia beschränkenden taxonomischen Verbreitung.Die Molekulargewichtsbestimmung für die Vomilenin Reduktaseergab nach Größenausschluß-chromatographie an Superdex 75ein Gewicht von etwa 43 kDa. Das ebenfalls über Superdex 75ermittelte Molekulargewicht für die 1.2 DihydrovomileninReduktase lag bei ca. 49.8 kDa. Weiterhin wurde eine Metallionenabhängigkeit für dieVomilenin Reduktase aufgezeigt und die Cofaktorspezifitätsowie die pH und Temperatur Optima für beide Reduktasenbestimmt.

Alzheimer's disease (AD) is a fatal neurodegenerative condition characterized clinically by progressive memory loss and irreversible cognitive deterioration. It has been shown that there is a progressive degeneration of the brain cholinergic neurons which leads to the appearance of cognitive symptoms of the disease. The aim of this work was the formulation of multifunctional nanocarriers for nasal administration of tacrine-HCl (THA). This route has many advantages; in particular is possible to convey the drug directly to the Central Nervous System, through the olfactory bulb. In particular, were prepared Albumin nanoparticles carrying beta cyclodextrin and two different beta cyclodextrin derivatives (hydroxypropyl beta cyclodextrin and sulphobutylether beta cyclodextrin), and Multifunctional liposomes, prepared using traditional excipients (cholesterol and phosphatidylcholine), partly enriched with α-tocopherol (Toc) and/or polyunsaturated fatty acids (eicosapentaenoic acid and docosahexaenoic acid) (Ω3). Both nanosystems were characterized in terms of size, Zeta potential and encapsulation efficiency. Were also evaluated their functional properties such as mucoadhesion and permeability, using an ex-vivo assay based on nasal sheep mucosa. On Liposomes were also assessed drug neuronal uptake, cell toxicity, antioxidant and, cytoprotective activity in the human neuronal cell line SH-SY5Y and finally tocopherol trans-membrane diffusion. Both the nanocarriers produced presented excellent properties and a high potential as new systems for CNS-delivery of anti-Alzheimer drugs via the nasal route.

Die transmembrane Potenzialdifferenz Δφm ist direkt mit der katalytischen Aktivität der Cytochrom c Oxidase (CcO) verknüpft. Die CcO ist das terminale Enzym (Komplex IV) in der Atmungskette der Mitochondrien. Das Enzym katalysiert die Reduktion von O2 zu 2 H2O. Dabei werden Elektronen vom natürlichen Substrat Cytochrom c zur CcO übertragen. Der Eleltronentransfer innerhalb der CcO ist an die Protonentranslokation über die Membran gekoppelt. Folglich bildet sich über der inneren Membrane der Mitochondrien eine Differenz in der Protonenkonzentration. Zusätzlich wird eine Potenzialdifferenz Δφm generiert.rnrnDas Transmembranpotenzial Δφm kann mit Hilfe der Fluoreszenzspektroskopie unter Einsatz eines potenzialemfindlichen Farbstoffs gemessen werden. Um quantitative Aussagen aus solchen Untersuchungen ableiten zu können, müssen zuvor Kalibrierungsmessungen am Membransystem durchgeführt werden.rnrnIn dieser Arbeit werden Kalibrierungsmessungen von Δφm in einer Modellmembrane mit inkorporiertem CcO vorgestellt. Dazu wurde ein biomimetisches Membransystem, die Proteinverankerte Doppelschicht (protein-tethered Bilayer Lipid Membrane, ptBLM), auf einem transparenten, leitfähigem Substrat (Indiumzinnoxid, ITO) entwickelt. ITO ermöglicht den simultanen Einsatz von elektrochemischen und Fluoreszenz- oder optischen wellenleiterspektroskopischen Methoden. Das Δφm in der ptBLM wurde durch extern angelegte, definierte elektrische Spannungen induziert. rnrnEine dünne Hydrogelschicht wurde als "soft cushion" für die ptBLM auf ITO eingesetzt. Das Polymernetzwerk enthält die NTA Funktionsgruppen zur orientierten Immobilisierung der CcO auf der Oberfläche der Hydrogels mit Hilfe der Ni-NTA Technik. Die ptBLM wurde nach der Immobilisierung der CcO mittels in-situ Dialyse gebildet. Elektrochemische Impedanzmessungen zeigten einen hohen elektrischen Widerstand (≈ 1 MΩ) der ptBLM. Optische Wellenleiterspektren (SPR / OWS) zeigten eine erhöhte Anisotropie des Systems nach der Bildung der Doppellipidschicht. Cyklovoltammetriemessungen von reduziertem Cytochrom c bestätigten die Aktivität der CcO in der Hydrogel-gestützten ptBLM. Das Membranpotenzial in der Hydrogel-gestützten ptBLM, induziert durch definierte elektrische Spannungen, wurde mit Hilfe der ratiometrischen Fluoreszenzspektroskopie gemessen. Referenzmessungen mit einer einfach verankerten Dopplellipidschicht (tBLM) lieferten einen Umrechnungsfaktor zwischen dem ratiometrischen Parameter Rn und dem Membranpotenzial (0,05 / 100 mV). Die Nachweisgrenze für das Membranpotenzial in einer Hydrogel-gestützten ptBLM lag bei ≈ 80 mV. Diese Daten dienen als gute Grundlage für künftige Untersuchungen des selbstgenerierten Δφm der CcO in einer ptBLM.

Ischemic preconditioning is a complex cardioprotective phenomenon that involves adaptive changes in cells and molecules. This adaptation occurs in a biphasic pattern: an early phase which develops after 1-2 h, and a late phase that develops after 12-24 h. While it is widely accepted that reactive oxygen species (ROS) are strongly involved in triggering ischemic preconditiong, it is not clear if they play a major role in the early or late phase of preconditioning and which are the mechanisms involved. Methylglyoxal, a metabolic compound formed mainly from the glycolytic intermediate glyceraldehyde-3-phosphate., is a precursor of advanced glycation end product (AGEs) .It is more reactive than glucose and shows a stronger ability to cross-link with protein amino groups to form AGEs. Methylglyoxal induced cytotoxicity may be at least partially responsible for cardiovascular and Alzheimer diseases. Methylglyoxal omeostasis is controlled by the glyoxalase system that consists of two enzyme, glyoxalase 1 (GLO1) and glyoxalase 2. In a recent study it was demonstrated that the transcriptional levels of GLO1 are controlled by NF-E2-related factor 2 (Nrf2). The isothiocyanate sulforaphane, derived from the hydrolysis of glucoraphanin abundantly present in broccoli, represents one of the most potent inducers of phase II enzymes through the Keap1–Nrf2 pathway. The aim of this thesis was evaluated molecular mechanisms in cardio- and neuroprotection and the possibility of modulation by nutraceutical phytocomponents This thesis show to one side that the protection induced by H2O2 is mediated by detoxifying and antioxidant phase II enzymes induction, regulated, not only by transcriptional factor Nrf2, but also by Nrf1; on the other side our data represent an innovative result because for the first time it was demonstrated the possibility of inducing GLO1 by SF supplementation.

Die vorliegende Arbeit befasste sich mit der Charakterisierung molekularer Funktionen humaner Paraoxonase (PON) Enzyme, insbesondere die der Proteine PON2 und PON3 im Hinblick auf medizinisch-relevante Fragestellungen. Zum einen wurde die Rolle von PON3 in der Tumorgenese und zum anderen eine mögliche Schutzfunktion von PON2 und PON3 gegenüber P. aeruginosa Infektionen untersucht. Bereits seit dem Jahr 2000 ist die anti-oxidative Eigenschaft von PON3 bekannt, jedoch war der zugrundeliegende Mechanismus bisher ungeklärt. Im Rahmen dieser Arbeit wurde gezeigt, dass PON3 die Superoxid-Entstehung in den Mitochondrien abschwächt, wobei sie ihre anti-oxidative Eigenschaft vermutlich durch eine direkte Coenzym Q10-Interaktion in der inneren mitochondrialen Membran vermittelt. Dies führt zu weniger oxidativen Stress, zur Abschwächung mitochondrial-induzierter apoptotischer Signalwege und zur erhöhten Resistenz gegenüber Chemotherapeutika. Gleichzeitig wurde demonstriert, dass sich Tumorzellen diese anti-oxidative Eigenschaft zu Nutze machen. PON3 war in zahlreichen Tumorgeweben überexprimiert. Es konnte eine mögliche Funktion von PON3 als Tumormarker und Angriffspunkt in der Krebstherapie aufgezeigt werden. Die hier erlangten Daten liefern wertvolle Hinweise auf die Rolle von PON3 in Krebserkrankungen, welche eine Basis für zukünftige Analysen darstellen, die der Entwicklung neuer Krebstherapien dienen könnten. Ein weiterer Teil der Arbeit befasste sich mit der gegenseitigen Beeinflussung der Enzyme PON2 / PON3 und der für P.aeruginosa essentiellen Virulenzfaktoren Pyocyanin (PCN) und dem Lacton 3OC12. Erstmalig wurde gezeigt, dass PON3 zellschädigende PCN-Effekte abschwächen kann, nämlich die PCN-induzierte Superoxid-Produktion, NF-kB-Aktivierung und IL-8-Sekretion. PON2 schützt in gleicher Weise gegen PCN und hydrolysiert zugleich noch das Lacton 3OC12. Folglich sind PON2 und PON3 wichtige Bestandteile der angeborenen Immunität, werden jedoch durch eine 3OC12-induzierte Ca2+-Mobilisation inaktiviert. Weitere Analysen ergaben, dass die PON2-Inaktivierung wahrscheinlich über einen Ca2+ / Calcineurin / Calmodulin-abhängigen Signalweg erfolgt, welcher eine offenbar regulative Serin311-Dephosphorylierung in PON2 vermittelt. Ähnliches könnte für PON3 gelten und wird derzeit erforscht, da eine Stabilisierung der enzymatischen Aktivitäten von PON2 und PON3 der bakteriellen Virulenz entscheidend entgegen wirken könnte.
Targeting neuronal populations by AAV-mediated gene transfer for studying the endocannabinoid system

The cannabinoid type 1 (CB1) receptor is involved in a plethora of physiological functions and heterogeneously expressed on different neuronal populations. Several conditional loss-of-function studies revealed distinct effects of CB1 receptor signaling on glutamatergic and GABAergic neurons, respectively. To gain a comprehensive picture of CB1 receptor-mediated effects, the present study aimed at developing a gain-of-function approach, which complements conditional loss-of-function studies. Therefore, adeno-associated virus (AAV)-mediated gene delivery and Cre-mediated recombination were combined to recreate an innovative method, which ensures region- and cell type-specific transgene expression in the brain. This method was used to overexpress the CB1 receptor in glutamatergic pyramidal neurons of the mouse hippocampus. Enhanced CB1 receptor activity at glutamatergic terminals caused impairment in hippocampus-dependent memory performance. On the other hand, elevated CB1 receptor levels provoked an increased protection against kainic acid-induced seizures and against excitotoxic neuronal cell death. This finding indicates the protective role of CB1 receptor on hippocampal glutamatergic terminals as a molecular stout guard in controlling excessive neuronal network activity. Hence, CB1 receptor on glutamatergic hippocampal neurons may represent a target for novel agents to restrain excitotoxic events and to treat neurodegenerative diseases. Endocannabinoid synthesizing and degrading enzymes tightly regulate endocannabinoid signaling, and thus, represent a promising therapeutic target. To further elucidate the precise function of the 2-AG degrading enzyme monoacylglycerol lipase (MAGL), MAGL was overexpressed specifically in hippocampal pyramidal neurons. This genetic modification resulted in highly increased MAGL activity accompanied by a 50 % decrease in 2-AG levels without affecting the content of arachidonic acid and anandamide. Elevated MAGL protein levels at glutamatergic terminals eliminated depolarization-induced suppression of excitation (DSE), while depolarization-induced suppression of inhibition (DSI) was unchanged. This result indicates that the on-demand availability of the endocannabinoid 2-AG is crucial for short-term plasticity at glutamatergic synapses in the hippocampus. Mice overexpressing MAGL exhibited elevated corticosterone levels under basal conditions and an increase in anxiety-like behavior, but surprisingly, showed no changes in aversive memory formation and in seizure susceptibility. This finding suggests that 2 AG-mediated hippocampal DSE is essential for adapting to aversive situations, but is not required to form aversive memory and to protect against kainic acid-induced seizures. Thus, specific inhibition of MAGL expressed in hippocampal pyramidal neurons may represent a potential treatment strategy for anxiety and stress disorders. Finally, the method of AAV-mediated cell type-specific transgene expression was advanced to allow drug-inducible and reversible transgene expression. Therefore, elements of the tetracycline-controlled gene expression system were incorporated in our “conditional” AAV vector. This approach showed that transgene expression is switched on after drug application and that background activity in the uninduced state was only detectable in scattered cells of the hippocampus. Thus, this AAV vector will proof useful for future research applications and gene therapy approaches.

To elucidate the local formation of angiotensin II (Ang II) in the neurons of sensory dorsal root ganglia (DRG), we studied the expression of angiotensinogen (Ang-N)-, renin-, angiotensin converting enzyme (ACE)- and cathepsin D-mRNA, and the presence of protein renin, Ang II, Substance P and calcitonin gene-related peptide (CGRP) in the rat and human thoracic DRG. Quantitative real time PCR (qRT-PCR) studies revealed that rat DRG expressed substantial amounts of Ang-N- and ACE mRNA, while renin mRNA as well as the protein renin were untraceable. Cathepsin D-mRNA and cathepsin D-protein were detected in the rat DRG indicating the possibility of existence of pathways alternative to renin for Ang I formation. Angiotensin peptides were successfully detected with high performance liquid chromatography and radioimmunoassay in human DRG extracts. In situ hybridization in rat DRG confirmed additionally expression of Ang-N mRNA in the cytoplasm of numerous neurons. Intracellular Ang II staining could be shown in number of neurons and their processes in both the rat and human DRG. Interestingly we observed neuronal processes with angiotensinergic synapses en passant, colocalized with synaptophysin, within the DRG. In the DRG, we also identified by qRT-PCR, expression of Ang II receptor AT(1A) and AT(2)-mRNA while AT(1B)-mRNA was not traceable. In some neurons Substance P and CGRP were found colocalized with Ang II. The intracellular localization and colocalization of Ang II with Substance P and CGRP in the DRG neurons may indicate a participation and function of Ang II in the regulation of nociception. In conclusion, these results suggest that Ang II may be produced locally in the neurons of rat and human DRG and act as a neurotransmitter.

1.Biologists have long puzzled over the apparent conspicuousness of blue-green eggshell coloration in birds. One candidate explanation is the sexual signalling hypothesis that the blue-green colour of eggshells can reveal an intrinsic aspect of females' physiological quality, with only high-quality females having sufficient antioxidant capacity to pigment their eggs with large amounts of biliverdin. Subsequent work has argued instead that eggshell colour might signal condition-dependent traits based on diet. 2.Using Araucana chickens that lay blue-green eggs, we explored (i) whether high levels of dietary antioxidants yield eggshells with greater blue-green reflectance, (ii) whether females differ from one another in eggshell coloration despite standardized environments, diets and rearing conditions, and (iii) the relative strength with which diet vs. female identity affects eggshell coloration. 3.We reared birds to maturity and then placed them on either a high- or low-antioxidant diet, differing fourfold in Vitamin E acetate and Vitamin A retinol. After 8 weeks, the treatments were reversed, such that females laid eggs on both diets in an order-balanced design. We measured the reflectance spectra of 545 eggs from 25 females. 4.Diet had a very limited effect on eggshell spectral reflectance, but individual females differed strongly and consistently from one another, despite having been reared under uniform conditions. However, predictions from avian visual modelling suggest that most of the egg colour differences between females, and nearly all of the differences between diets, are unlikely to be visually discriminable. 5.Our data suggest that eggshell reflectance spectra may carry information on intrinsic properties of the female that laid the eggs, but the utility of this coloration as a signal to conspecifics in this species may be limited by the sensitivity of a receiver to detect it.

Numerous bacterial pathogens subvert cellular functions of eukaryotic host cells by the injection of effector proteins via dedicated secretion systems. The type IV secretion system (T4SS) effector protein BepA from Bartonella henselae is composed of an N-terminal Fic domain and a C-terminal Bartonella intracellular delivery domain, the latter being responsible for T4SS-mediated translocation into host cells. A proteolysis resistant fragment (residues 10-302) that includes the Fic domain shows autoadenylylation activity and adenylyl transfer onto Hela cell extract proteins as demonstrated by autoradiography on incubation with α-[(32)P]-ATP. Its crystal structure, determined to 2.9-Å resolution by the SeMet-SAD method, exhibits the canonical Fic fold including the HPFxxGNGRxxR signature motif with several elaborations in loop regions and an additional β-rich domain at the C-terminus. On crystal soaking with ATP/Mg(2+), additional electron density indicated the presence of a PP(i) /Mg(2+) moiety, the side product of the adenylylation reaction, in the anion binding nest of the signature motif. On the basis of this information and that of the recent structure of IbpA(Fic2) in complex with the eukaryotic target protein Cdc42, we present a detailed model for the ternary complex of Fic with the two substrates, ATP/Mg(2+) and target tyrosine. The model is consistent with an in-line nucleophilic attack of the deprotonated side-chain hydroxyl group onto the α-phosphorus of the nucleotide to accomplish AMP transfer. Furthermore, a general, sequence-independent mechanism of target positioning through antiparallel β-strand interactions between enzyme and target is suggested.

The objective was to analyze the outcome following prenatal exposure to angiotensin-converting enzyme inhibitors (ACE-Is) or angiotensin receptor antagonists (ARBs). For this purpose, a systematic review of published case reports and case series dealing with intrauterine exposure to ACE-Is or to ARBs using Medline as the source of data was performed. The publications retained for analysis included patients who were described individually, revealing, at minimum, the gestational age, substance used, period of medication intake, and the outcome. In total, 72 reports were included; 37 articles (118 well-documented cases) described the prenatal exposure to ACE-Is; and 35 articles (68 cases) described the prenatal exposure to ARBs. Overall, 52% of the newborns exposed to ACE-Is and 13% of the newborns exposed to ARBs did not exhibit any complications (P<0.0001). Neonatal complications were more frequent following exposure to ARBs and included renal failure, oligohydramnios, death, arterial hypotension, intrauterine growth retardation, respiratory distress syndrome, pulmonary hypoplasia, hypocalvaria, limb defects, persistent patent ductus arteriosus, or cerebral complications. The long-term outcome is described as positive in only 50% of the exposed children. Fetopathy caused by exposure to ACE-Is or ARBs has relevant neonatal and long-term complications. The outcome is poorer following exposure to ARBs. We propose the term "fetal renin-angiotensin system blockade syndrome" to describe the related clinical findings. Thirty years after the first description of ACE-I fetopathy, relevant complications are, at present, regularly described, indicating that the awareness of the deleterious effect of prenatal exposure to drugs inhibiting the renin-angiotensin system should be improved.
