This study aims to clinically and macroscopically evaluate the adjuvant therapy with platelet-rich plasma in the form of eyedrops or clot, for corneal ulcers in dogs treated at the Veterinary Ophthalmology Service. We analyzed 20 eyes diagnosed with ulcerative keratitis, divided into two experimental groups. The eyedrop group (GC) was composed of eyes treated topically with eyedrops of autologous plateletrich plasma (PRP), and the clot group (GT) was composed of eyes treated with a platelet-rich clot and covered with a third eyelid for retention of the clot. The groups were evaluated by clinical and macroscopic analysis and by the analysis of epithelial defect reduction, at different times, at three, five, ten, 15 and 30 days, except for the third day in GT. The coverage of the third eyelid was removed on the fifth day. In both groups the inflammation signs reduced, there was an improvement in ocular sensibility and proper repair of epithelial defect. All GT eyes and 70% GC eyes showed complete healing on the fifth day, the remainder of GC completed healing on the tenth day. PRP in the form of eyedrops and clot is an excellent adjuvant therapy to be instituted in the clinical treatment for corneal ulcer in dogs, because it decreases the inflammatory signs and the ocular pain and it potentially assists in healing epithelial defects.

The application technology shows many parameters related to the quality of the application, one is the droplet spectrum, which is influenced by the spray nozzles and the adjuvant used. Therefore, the objective of this work was estimate the behavior of the droplet spectrum generated with different nozzles and different adjuvants. The experiment was installed containing four solutions from different type adjuvant dilution, as vegetal oil, mineral oil, surfactant and drift reduction, which were applied with two nozzle, one pre-orifice flat fan (DG 8003 VS) and other of air induction flat fan (AI 8003 VS), totaling 8 treatments with 3 repetitions. The experiment was realized in ideal weather conditions for spraying. The treatments averages were compared using Confidence Interval at 95% probability and the correlations between variables were analyzed using Pearson at 5% of probability. The analysis of droplet spectrum showed different behavior for each adjuvant and nozzle. The surfactant treatment showed VMD superior for all treatments when sprayed with AI nozzles. For the %vol.<100 µm the lowest value found was for the AI nozzle in combination with the surfactant. The significant correlations found for the nozzles AI and DG were negative between VMD and %vol.<100 µm. It can be concluded that the values of DMV and %vol.<100 µm showed that the nozzle with pre-orifice have droplet spectrum more prone to drift. The surfactant showed to be the best drift reduction technique when combined with the AI nozzle.

The Asian rust currently is the main disease of soybean culture, having as characteristics the difficult control, by start at the bottom of plants where penetration of the droplets is harder. The fine droplets has been used with the intention of improve the penetration and increase efficiency of agrochemicals, but that are losses by drift easily. New products have been developed to increase deposition of the drops at targets. The aim of this work was evaluate the TA- 35 capacity to improving the deposition of fungicides spray solution with or without mineral oil by aerial and ground applications. Was used a factorial 3x2, three spray solutions composed by Priori Xtra (concentrated suspension of azoxystrobin 200 g L-1 + cyproconazole 80 g L-1 ) mixed with adjuvants, Nimbus (emulsifiable concentrate containing aliphatic hydrocarbons 428 g L-1 ) and TA-35 (soluble concentrate containing sodium lauryl ether sulfate, surfactants, sequestering agents and emulsifiers), in aerial and ground applications. In ground applications was used 50 L ha-1 , TXA 8002 VS spray nozzles and on aerials was used 15 L ha-1 , Turboaero atomizer, both applying fine droplets. Was utilized the Brilliant Blue (FD & C n. 1) tracer to determine the deposits. There were used glass slides as targets to collect spray droplets. After to extract the tracer of the targets using distilled water, the samples were analyzed by spectrophotometry, thereby was possible quantify the tracer deposited on each glass slide. A study to evaluate possible losses of the tracer by degradation or retention also was done. The comparative analysis of treatments was done by statistical method "Confidence Interval for Differences Between the Averages" with 95% of confidence degree (IC95%). There was degradation or retention of the tracer between the processes of application of the droplets and the processing of the samples. The deposition averages with the presence of TA-35 were greatest for both sprayers however, there were not significant differences among the treatments. The viability of TA-35 use may consider other parameters or complementary studies.

This study aimed to verify the influence of adjuvants on the droplet spectrum of an air induction nozzle. The experiment used nine spray solutions, one including only water and eight containing adjuvants: Nimbus® (mineral oil), Óleo vegetal Nortox (vegetal oil), Li-700® (a mixture of lecithin and propionic acid), Agral® (nonyl phenoxy poly ethanol), In-Tec® (nonyl phenol ethoxylate), Antideriva (nonyl phenol ethoxylate), Silwet® L-77 Ag (copolymer polyester and silicon) and TA 35 (sodium lauryl ether sulfate). A flat fan air induction nozzle Hypro® Guardian Air 110 03 was used for the droplet spectrum evaluation. The study was conducted at the Laboratory for Particle Size Analysis (Lapar), at FCAV/UNESP, Jaboticabal/SP - Brazil. The determination of the droplet spectrum characteristics (Volume Median Diameter/VMD, percentage of droplets smaller than 100 micrometers and span) was carried out by a particle size analyzer by laser diffraction Mastersizer S (Malvern Instruments). For statistical analysis the mean values were compared using Confidence Interval at 95% (CI 95%). The results showed that for the Hypro® GA air induction nozzle the oil based adjuvants (Óleo Vegetal Nortox e Nimbus®) increased the VMD. The percentage of droplets smaller than 100 micrometers was lower for the Agral®, Antideriva, In-Tec® e TA 35, in comparison with the Óleo Vegetal Nortox and Li-700®. The span was higher for the oil based adjuvants (Óleo Vegetal Nortox e Nimbus®) and lower for the TA 35 (sodium lauryl ether sulfate), showing that the TA 35 adjuvant has a potential to improve the quality of the droplet spectrum of the Hypro® GA 11003 nozzle.

The aim of this study was to build and validate a low cost reduced-scale wind tunnel for drift evaluation on pesticide application technology. The work was carried out at the NEMPA - Núcleo de Ensaio de Máquinas e Pneus Agroflorestais (NEMPA), FCA/UNESP, Departamento de Engenharia Rural, Botucatu/SP, Brazil. The wind tunnel main characteristics were an open circuit and a closed working section with a fan blowing air into the tunnel. Screens were fitted downstream after the fan in order to stabilize the air flow on the working section. The tunnel was built with 3.0 mm eucalyptus hardboard, with a total length of 4.8 m and a squared section of 0.56 m. The air flow was provided by a 180 W axial fan. The system was adjusted and calibrated to provide a laminar and stable flow at 2.0 m s-1. Validation studies were carried out by using a Teejet XR 8003 flat fan nozzle at 200 kPa (medium droplets) to apply a spray solutions containing water plus a food dye (Blue FDC) at 0,6% m v-1 mixed with two adjuvants: a polymer based anti drift formulation at 0,06% m v-1 and a sodium lauryl ether sulfate based surfactant at 0,2% v v-1. After a 10-second application the drift was collected on nylon strips transversally fixed along the tunnel at different distances from the nozzle and different high from the bottom part of the tunnel. Drift deposits were evaluated by spectrophotometry. The wind tunnel had low levels of turbulence and high repeatability of the data, which means that the flow was uniform and able to be used for carrying out measures to estimate drift. The validation results showed that the tunnel was effective to enable comparative drift measurements on the spray solution used in this work making possible the evaluation of drift risk potential under those spray technologies. The use of an adjuvant based on a polymer reduced the amount of drift from the nozzle compared to the surfactant.

The present study had as objectives to verify the effect of rainfastness of different flutriafol formulations, in laboratory conditions, applied on soybean plants with and without the oil adjuvant in the spray solution, as well as to verify the influence of the oil on the droplet spectrum. The experiment considered ten spray solutions related to five treatments containing flutriafol (four formulations of flutriafol and a flutriafol mixture with tebuconazole), all applied with and without mineral oil. Particles size analysis were based on the determination of the droplet spectrum, medium volumetric diameter and the amount of droplets below 100 μm. All the solutions were sprayed with Teejet XR 11001 (fine droplets). Droplet spectrum was determined in a direct way by diffraction of laser (Malvern Mastersizer S®, version 2.15). Confidence interval at 90% degree was used to compare the mean data. The results showed that the addition of mineral oil in the solutions provided tendencies of larger medium volumetric diameter, smaller amount of droplets below than 100 µm and better uniformity of the droplet spectrum. All of the solutions with the addition of mineral oil presented larger adhesion and/or absorption of the fungicide on the plants in comparison with the solutions without oil. The increase of the time between the application and the rain, caused reduction of the fungicide removal, independently of the rain intensity. The increase of the amount of rain didn't change the relative behavior among the solutions; however, this larger amount of rain caused larger fungicide removal along the time. It was observed significant removal of flutriafol by the rain up to 48 hours after the spray application.

O câncer destaca-se pela alta incidência e mortalidade. Os tratamentos atualmente usados são agressivos e não específicos, com isso cresce a busca por novas drogas. Uma substância que vem despertando muito interesse são as zeólitas, minerais com característica porosa e estrutura conhecida. Estas possuem ações como adjuvante de vacinas, imunomoduladores e imunoestimuladores, o que desperta o interesse em estuda-las no modelo antitumoral. O presente estudo avaliou o efeito antitumoral e imunomodulador da zeólita natural clinoptilolita e da zeólita comercial, utilizando um modelo de câncer mamário (Tumor de Ehrlich). Para tanto a zeólita natural foi caracterizada (Microscopia Eletrônica de Varredura e Difração de raio X), realizada avaliação da viabilidade celular (ensaio de MTT), determinada a produção de óxido nítrico por macrófagos peritoneais, quantificação de citocinas (ELISA) e avaliação do crescimento tumoral. As zeólitas natural e comercial apresentaram elevada ativação de macrófagos, e não produziram quantidades significativas de NO. A zeólita natural apresentou citotoxicidade frente ao Tumor de Ehrlich em duas concentrações testadas (5 e 25 mg/ml). Não houve liberação significativa da citocina IL-10, no entanto os grupos que foram reestimulados com zeólita natural apresentaram maior liberação de IL-1β e TNF-α. Nos testes in vivo, a zeólita comercial foi a única que apresentou inibição tumoral frente ao Tumor de Ehrlich, sendo necessários estudos mais aprofundados para definir a sua atividade antitumoral nesse tipo celular.

A busca por novos agentes antimicrobianos é muito importante, uma vez que as bactérias possuem um alto potencial de recombinação genética, levando a um aumento de cepas resistentes aos fármacos antimicrobianos já existentes. Nesse contexto, os cogumelos podem ser uma alternativa, uma vez que extratos e constituintes químicos destes apresentam atividade frente a diversos isolados patogênicos e utilizados na medicina popular. Dentre as espécies de maior interesse estão o Agaricus blazei (cogumelo do sol), Agaricus bisporus (champignon de Paris) e Lentinus edodes (shitake). O Helicobacter pylori é responsável por doenças gastrointestinais de extrema importância para a saúde pública, como úlceras, gastrites até tumores malignos. O tratamento mais utilizado nos controles deste micro-organismo não é eficaz. Os cogumelos aparecem como alternativa para novas pesquisas neste ramo podendo vir a ser utilizados no desenvolvimento de novo fitoterápico ou mesmo como adjuvante deste tratamento. Existem diversos métodos para determinar a atividade antimicrobiana, entre eles a microdiluição. Esta técnica vem sendo muito utilizada principalmente devido à sua sensibilidade e pequena quantidade de reagentes e amostras, o que possibilita um maior número de réplicas, aumentando a confiabilidade dos resultados. Em continuidade ao estudo do nosso grupo de pesquisa este trabalho objetiva determinar o potencial anti-H. pylori de espécies de cogumelos comestíveis instigados pela possibilidade de encontrar uma alternativa para o tratamento desse micro-organismos dada a importância do mesmo na saúde pública. Kaurenoic acid [ent-kaur-16-en-19-oic acid (1)] is a diterpene present in several plants including Sphagneticola trilobata. The only documented evidence for its antinociceptive effect is that it inhibits the writhing response induced by acetic acid in mice. Therefore, the analgesic effect of 1 in different models of pain and its mechanisms in mice were investigated further. Intraperitoneal and oral treatment with 1 dose-dependently inhibited inflammatory nociception induced by acetic acid. Oral treatment with 1 also inhibited overt nociception-like behavior induced by phenyl-p-benzoquinone, complete Freund's adjuvant (CFA), and both phases of the formalin test. Compound 1 also inhibited acute carrageenin- and PGE(2)-induced and chronic CFA-induced inflammatory mechanical hyperalgesia. Mechanistically, 1 inhibited the production of the hyperalgesic cytokines TNF-alpha and IL-1 beta. Furthermore, the analgesic effect of 1 was inhibited by L-NAME, ODQ, KT5823, and glybenclamide treatment, demonstrating that such activity also depends on activation of the NO-cyclic GMP-protein kinase G-ATP-sensitive potassium channel signaling pathway, respectively. These results demonstrate that 1 exhibits an analgesic effect in a consistent manner and that its mechanisms involve the inhibition of cytokine production and activation of the NO-cyclic GMP-protein lcinase G-ATP-sensitive potassium channel signaling pathway.

Background: Pain markedly activates the hypothalamic-pituitary-adrenal (HPA) axis and increases plasma corticosterone release interfering significantly with nociceptive behaviour as well as the mechanism of action of analgesic drugs. Aims/Methods: In the present study, we monitored the time course of circulating corticosterone in two mouse strains (C57Bl/6 and Balb/C) under different pain models. In addition, the stress response was investigated following animal handling, intrathecal (i.t.) manipulation and habituation to environmental conditions commonly used in nociceptive experimental assays. We also examined the influence of within-cage order of testing on plasma corticosterone. Results: Subcutaneous injection of capsaicin precipitated a prompt stress response whereas carrageenan and complete Freund's adjuvant induced an increased corticosterone release around the third hour post-injection. However, carrageenan induced a longer increased corticosterone in C57Bl/6 mice. In partial sciatic nerve ligation, neuropathic pain model corticosterone increased only in the first days whereas mechanical hypersensitivity remained much longer. Animal handling also represents an important stressor whereas the i.t. injection per se does not exacerbate the handling-induced stress response. Moreover, the order of testing animals from the same cage does not interfere with plasma corticosterone levels in the intrathecal procedure. Animal habituation to the testing apparatus also does not reduce the immediate corticosterone increase as compared with non-habituated mice. Conclusion: Our data indicate that HPA axis activation in acute and chronic pain models is time dependent and may be dissociated from evoked hyperalgesia. Therefore, HPA-axis activation represents an important variable to be considered when designing experimental assays of persistent pain as well as for interpretation of data.

The dengue virus non-structural 1 (NS1) protein contributes to evasion of host immune defenses and represents a target for immune responses. Evidences generated in experimental models, as well as the immune responses elicited by infected individuals, showed that induction of anti-NS1 immunity correlates with protective immunity but may also result in the generation of cross-reactive antibodies that recognize platelets and proteins involved in the coagulation cascade. In the present work, we evaluated the immune responses, protection to type 2 dengue virus (DENV2) challenges and safety parameters in BALB/c mice vaccinated with a recombinant NS1 protein in combination with three different adjuvants: aluminum hydroxide (alum), Freund's adjuvant (FA) or a genetically detoxified derivative of the heat-labile toxin (LTG33D), originally produced by some enterotoxigenic Escherichia coil (ETEC) strains. Mice were subcutaneously (s.c.) immunized with different vaccine formulations and the induced NS1-specific responses, including serum antibodies and T cell responses, were measured. Mice were also subjected to lethal challenges with the DENV2 NGC strain. The results showed that maximal protective immunity (50%) was achieved in mice vaccinated with NS1 in combination with LIG33D. Analyses of the NS1-specific immune responses showed that the anti-virus protection correlated mainly with the serum anti-NS1 antibody responses including higher avidity to the target antigen. Mice immunized with LTG33D elicited a prevailing IgG2a subclass response and generated antibodies with stronger affinity to the antigen than those generated in mice immunized with the other vaccine formulations. The vaccine formulations were also evaluated regarding induction of deleterious side effects and, in contrast to mice immunized with the FA-adjuvanted vaccine, no significant hepatic damage or enhanced C-reactive protein levels were detected in mice immunized with NS1 and LTG33D. Similarly, no detectable alterations in bleeding time and hematological parameters were detected in mice vaccinated with NS1 and LTG33D. Altogether, these results indicate that the combination of a purified recombinant NS1 and a nontoxic LT derivative is a promising alternative for the generation of safe and effective protein-based anti-dengue vaccine.
