Matrix sublimation has demonstrated to be a powerful approach for high-resolution matrix-assisted laser desorption ionization (MALDI) imaging of lipids, providing very homogeneous solvent-free deposition. This work presents a comprehensive study aiming to evaluate current and novel matrix candidates for high spatial resolution MALDI imaging mass spectrometry of lipids from tissue section after deposition by sublimation. For this purpose, 12 matrices including 2,5-dihydroxybenzoic acid (DHB), sinapinic acid (SA), α-cyano-4-hydroxycinnamic acid (CHCA), 2,6-dihydroxyacetphenone (DHA), 2',4',6'-trihydroxyacetophenone (THAP), 3-hydroxypicolinic acid (3-HPA), 1,8-bis(dimethylamino)naphthalene (DMAN), 1,8,9-anthracentriol (DIT), 1,5-diaminonapthalene (DAN), p-nitroaniline (NIT), 9-aminoacridine (9-AA), and 2-mercaptobenzothiazole (MBT) were investigated for lipid detection efficiency in both positive and negative ionization modes, matrix interferences, and stability under vacuum. For the most relevant matrices, ion maps of the different lipid species were obtained from tissue sections at high spatial resolution and the detected peaks were characterized by matrix-assisted laser desorption ionization time-of-flight/time-of-flight (MALDI-TOF/TOF) mass spectrometry. First proposed for imaging mass spectrometry (IMS) after sublimation, DAN has demonstrated to be of high efficiency providing rich lipid signatures in both positive and negative polarities with high vacuum stability and sub-20 μm resolution capacity. Ion images from adult mouse brain were generated with a 10 μm scanning resolution. Furthermore, ion images from adult mouse brain and whole-body fish tissue sections were also acquired in both polarity modes from the same tissue section at 100 μm spatial resolution. Sublimation of DAN represents an interesting approach to improve information with respect to currently employed matrices providing a deeper analysis of the lipidome by IMS.

Background: Nursing home short stays (NHSS) in the canton of Vaud have been introduced for respite care purpose. However, a growing number of older patients are urgently admitted from home (within 24h) or directly after hospital discharge (58% of all admissions in 2010). NHSS appears therefore as an increasingly important component of the health care system, but the characteristics of admitted patients have not been previously described. A better knowledge would contribute to identify specific care needs and enhance their care. Objectives: 1) To describe the characteristics of patients admitted in unplanned NHSS ( after hospital stay or urgently from home); 2) To determine living disposition 3-month after NHSS discharge. Method: Over a 18-month period, elderly patients with unplanned NHSS admission to 2 facilities in Lausanne were identified. Demographic, social, health, and functional data, as well as main reason for admission were collected. Death and place of living at 3-months were collected using the administrative database. Results: Overall, 114 patients (mean age 83.1 ± 6.2 years, 77% women, 84% living alone) were assessed, 80% being admitted from hospital. Mean score in Lawton's instrumental ADL before NHSS admission was 4.6 ± 2.5 and 69% of the patients were home care recipients (median number of weekly visits: 5 ± 3). Patients reported going out 4.2 ± 1.3 times/week and 56% reported at least one fall over the past year. Among the 91 patients coming from the hospital, main reason for admission was injury/limb immobilization (58%), recuperation (13%) and functional impairment in basic ADL (10%). Mean score at Katz's Basic ADL at admission was 3.7 ± 1.9. Overall, 90% of patients were identified with gait and balance impairment, 78% with cognitive impairment and 70% with polypharmacy (>6 different drugs). At 3-month after NHSS discharge (N = 92), 72% patients were living at home, 16% had been admitted to long term care, and 6% died. Among patients living at home at follow-up, 11% had been readmitted to hospital during the follow-up period. Conclusion: Older patients with unplanned NHSS admission show a high prevalence of functional, mobility, and cognitive impairments, as well as other geriatric syndromes. Specific measures should be considered during these stays to prevent further functional decline and, possibly, hospital readmission. Patients admitted with basic ADLs impairment might be candidate for higher levels of care (rehabilitation).

O objetivo deste trabalho foi avaliar o efeito de modalidades de poda verde da videira (Vitis vinifera L.) na composição do vinho Merlot. Um experimento foi conduzido de 1993/1994 a 1996/1997, em Bento Gonçalves, RS, com uma testemunha e 11 modalidades de poda verde em vinhedo conduzido em latada. A análise de componentes principais mostrou que o efeito da poda verde variou conforme o ano. Considerando a média dos quatro anos avaliados, os três principais componentes foram responsáveis por 72% da variação total. Adesbrota, desponta e desfolha realizada no início da floração, com eliminação de todas as folhas abaixo dos cachos foi a modalidade de poda que apresentou os valores mais elevados de intensidade de cor, absorbâncias a 280 e 520 nm, antocianinas, taninos, álcool, 3-metil-1-butanol/2-metil-1-propanol, álcool em peso/extrato seco reduzido e fósforo, e foi a mais indicada para a produção de vinho Merlot.

The development of nuclear hormone receptor antagonists that directly inhibit the association of the receptor with its essential coactivators would allow useful manipulation of nuclear hormone receptor signaling. We previously identified 3-(dibutylamino)-1-(4-hexylphenyl)-propan-1-one (DHPPA), an aromatic β-amino ketone that inhibits coactivator recruitment to thyroid hormone receptor β (TRβ), in a high-throughput screen. Initial evidence suggested that the aromatic β-enone 1-(4-hexylphenyl)-prop-2-en-1-one (HPPE), which alkylates a specific cysteine residue on the TRβ surface, is liberated from DHPPA. Nevertheless, aspects of the mechanism and specificity of action of DHPPA remained unclear. Here, we report an x-ray structure of TRβ with the inhibitor HPPE at 2.3-Å resolution. Unreacted HPPE is located at the interface that normally mediates binding between TRβ and its coactivator. Several lines of evidence, including experiments with TRβ mutants and mass spectroscopic analysis, showed that HPPE specifically alkylates cysteine residue 298 of TRβ, which is located near the activation function-2 pocket. We propose that this covalent adduct formation proceeds through a two-step mechanism: 1) β-elimination to form HPPE; and 2) a covalent bond slowly forms between HPPE and TRβ. DHPPA represents a novel class of potent TRβ antagonist, and its crystal structure suggests new ways to design antagonists that target the assembly of nuclear hormone receptor gene-regulatory complexes and block transcription.

BACKGROUND: Patients with type 1 diabetes and nephropathy maintain an excess cardiovascular mortality compared with diabetic patients with normoalbuminuria. We sought to evaluate coronary and aortic atherosclerosis in a cohort of asymptomatic type 1 diabetic patients with and without diabetic nephropathy using cardiovascular magnetic resonance imaging. METHODS AND RESULTS: In a cross-sectional study, 136 subjects with long-standing type 1 diabetes without symptoms or history of cardiovascular disease, including 63 patients (46%) with nephropathy and 73 patients with normoalbuminuria, underwent cardiovascular magnetic resonance imaging. All subjects underwent cardiac exercise testing and noninvasive tests for peripheral artery disease and autonomic neuropathy. Coronary artery stenoses were identified in 10% of subjects with nephropathy (versus 0% with normoalbuminuria; P=0.007). Coronary plaque burden, expressed as right coronary artery mean wall thickness (1.7+/-0.3 versus 1.3+/-0.2 mm; P<0.001) and maximum right coronary artery wall thickness (2.2+/-0.5 versus 1.6+/-0.3 mm; P<0.001), was greater in subjects with nephropathy. The prevalence of thoracic (3% versus 0%; P=0.28) and abdominal aortic plaque (22% versus 16%; P=0.7) was similar in both groups. Subjects with and without abdominal aortic plaques had similar coronary plaque burden. CONCLUSIONS: In asymptomatic type 1 diabetes, cardiovascular magnetic resonance imaging reveals greater coronary plaque burden in subjects with nephropathy compared with those with normoalbuminuria.

OBJECTIVE: To assess the seroprevalence of herpes simplex virus type 1 (HSV-1) and type 2 (HSV-2) IgG antibodies and the seroincidence of HSV-1 and HSV-2 infections in pregnant women attending the maternity clinic of the University Hospital Lausanne. STUDY DESIGN: Blood samples from 1030 women were taken at the usual pregnancy visit in the first trimester to assess the prevalence rate of IgG antibodies against HSV-1 and HSV-2 using a type-specific assay. A second blood sample was taken 6-8 weeks postpartum from returning women who were seronegative for HSV-2 or HSV-1 to assess the incidence of seroconversion (primary infection). RESULTS: The seroprevalence rates were 79.4% (95% CI: 76.9-81.9) for HSV-1 and 21.2% (18.7-23.7) for HSV-2 in women 14-46 years old. Type-specific serostatus patterns were as follows: 17.3% HSV-1/-2: +/+, 62.1% HSV-1/-2: +/-, 3.9% HSV-1/-2: -/+, 16.7% HSV-1/-2: -/-. Two hundred and sixty five women (59 of the 212 seronegative for HSV-1 (27.8%) and 265 of the 812 seronegative for HSV-2 (32.6%)) returned to the outpatient clinic for the post-delivery check and a second blood sample was obtained. One HSV-1 seroconversion was detected (HSV-1 seroconversion rate 2.4%/100 patient×year (95% CI: 0.06-13.4)) in a patient who had symptoms compatible with primary genital herpes. No HSV-2 seroconversion was detected (HSV-2 seroconversion rate: 0/100 patient×year (97.5% one-sided CI: 0-2)). CONCLUSION: Compared to a previous population-based study, our study results suggest a rise in the prevalence of HSV-2 among pregnant women in Switzerland. The low incidence of seroconversion detected during pregnancy is consistent with the very low reported incidence of neonatal herpes in Switzerland. CONDENSATION: This study in a public hospital in Western Switzerland suggests an increasing prevalence of HSV-2, but a low incidence of primary infections in women of childbearing age.

Resumo:O objetivo deste trabalho foi avaliar o efeito da aplicação de 1-metilciclopropeno (1-MCP) em manga 'Palmer' (Mangifera indica), nos estádios de maturação 2 e 3, para a conservação pós-colheita do fruto durante o período de armazenamento. Foram realizados dois experimentos em delineamento inteiramente casualizado, com quatro repetições. No primeiro, mangas no estádio de maturação 2 foram submetidas a diferentes doses de 1-MCP (controle, 300, 600 e 1.000 nL L-1), por 12 horas, e tempos de armazenamento refrigerado de 0, 8 e 15 dias a 14,5±2°C e 85±6% UR, seguidos de 3, 5, 7 e 9 dias a 24±2°C e 60±6% UR. No segundo experimento, mangas no estádio de maturação 3 foram submetidas aos mesmos tratamentos do primeiro, porém com aplicação de 1-MCP durante 14 horas e com armazenamento a 13±0,6°C e 87±2% UR, e 24,4±1,9°C e 47±8% UR. O uso de 1-MCP nas doses de 300 e 600 nL L-1, no estádio de maturação 2, melhora a aparência dos frutos, embora se restrinja a efeitos temporários sobre a firmeza de polpa e a degradação de amido. Nas doses de 600 e 1.000 nL L-1, no estádio de maturação 3, o 1-MCP limita a perda de firmeza e mantém a aparência comercial do fruto até o vigésimo quarto dia.

Maçãs cv. Fuji foram tratadas com 42 mimol·m-3 de 1-metilciclopropeno (1-MCP) por 24 h a 20°C, um dia após a colheita, e então armazenadas a 0; 10 ou 20ºC por 70 dias. Tratamento com 1-MCP efetivamente retardou a maturação de maçãs 'Fuji'. 1-MCP reduziu a taxa respiratória dos frutos mantidos a 10 e 20ºC e inibiu a produção de etileno dos frutos mantidos nas três temperaturas de armazenagem. Frutos tratados com 1-MCP e armazenados a 20ºC exibiram taxas respiratórias similares ou inferiores àquelas de frutos-controle armazenados a 10ºC. Quando armazenados a 10 ou 20ºC, frutos tratados com 1-MCP preservaram mais a firmeza da polpa e a acidez titulável e exibiram menor amarelecimento da epiderme que frutos-controle. Entretanto, não houve benefícios significativos do tratamento 1-MCP sobre a conservação da qualidade dos frutos armazenados a 0ºC no período de 70 dias após a colheita. Os resultados indicam que o tratamento com 1-MCP pode ser uma estratégia para o aumento da conservação de maçãs cv. Fuji durante o transporte e a distribuição sob 10 ou 20ºC. O prolongamento da armazenagem a 20ºC por período superior a 40 dias pode ser limitado pelo murchamento dos frutos e desenvolvimento de podridões.

Graviolas 'Morada', provenientes de pomar comercial localizado em Limoeiro do Norte, Ceará, foram colhidas na maturidade fisiológica com o objetivo de avaliar o efeito da aplicação pós-colheita de 1-meticiclopropeno (1-MCP) e cera na conservação, durante o armazenamento refrigerado. Os frutos foram armazenados por 0; 4; 8; 11; 13 e 15 dias, a 15,4±1,1ºC e 86,0±7,3% UR. Os tratamentos pós-colheita foram os seguintes: controle, 200 nL L-1 de 1-MCP (SmartFresh™), pulverização com a cera Fruit wax® e pulverização com Fruit wax® seguida de aplicação de 200 nL L-1 de 1-MCP. O delineamento experimental foi o inteiramente casualizado, em fatorial 4x6 (tratamentos pós-colheita x tempo de armazenamento), com quatro repetições. As variáveis analisadas foram: cor da casca e da polpa, a partir da luminosidade (L), cromaticidade (C) e ângulo de cor (Hº); perda de massa; pH; acidez titulável (AT); sólidos solúveis totais (SST); açúcares solúveis totais (AST) e açúcares redutores (AR). Os tratamentos cera e cera+1-MCP mantiveram estáveis os valores de L da casca e da polpa até o 8º e o 4º dia de armazenamento, respectivamente. O aumento em SST foi temporariamente atrasado pelos tratamentos pós-colheita. A AT, pH, AST e AR não foram influenciados pela cera e pelo 1-MCP. O uso da cera diminuiu a perda de massa em 23%, quando comparado ao controle.

O 1-metilciclopropeno (1-MCP) é um gás que atua inibindo o sítio de ação do etileno, aumentando o tempo de armazenamento de frutos. O objetivo deste trabalho foi avaliar o efeito deste gás na conservação pós-colheita de kiwi 'Monty', à temperatura ambiente. Ele consistiu na exposição dos frutos a concentrações de 0; 0,5 e 1,0 mg dm-3 de 1-MCP, durante 24 horas, e posterior estocagem a 20 ± 4 ºC, por até 20 dias, com avaliações a cada 4 dias. Avaliaram-se: firmeza, translucidez da polpa, sólidos solúveis (SS), pH, acidez titulável (AT), pectina, % de frutos firmes para transporte e consumo. A translucidez da polpa mostrou-se um parâmetro adequado para monitoramento do amadurecimento, visto que houve correlação com a firmeza. A aplicação de 1-MCP preservou a firmeza e preveniu a translucidez da polpa, prolongando a qualidade para transporte e consumo por 4 e 12 dias, respectivamente, e não teve efeito sobre o pH, SS e pectinas. Nos tratamentos com 1-MCP, ocorreu um aumento inicial na acidez, mas com o passar do tempo, ocorreu diminuição da mesma, da firmeza e aumento dos sólidos solúveis e da translucidez da polpa, caracterizando o amadurecimento dos frutos.

Effects of water activity and 1-propanol concentration on synthesis of propyl oleate from oleic acid using Aspergillus niger cell-bound lipases in isooctane are described. A. niger produces lipases (EC 3.1.1.3) which partly bind to the mycelium during growth. Ester production was monitored for 72 hours at different 1-propanol concentrations and water activities. Aliquots were sequentially withdrawn and propyl esters were quantified using GC and methyl palmitate as an internal standard. In all assayed conditions A. niger cell-bound lipases catalysed propyl oleate synthesis, but at different degrees.

RESUMO O armazenamento refrigerado pode induzir o desenvolvimento de danos por frio (caracterizado pelo escurecimento de polpa) em maçãs de algumas cultivares, mesmo quando mantidas em temperatura superior ao ponto de congelamento. O presente estudo avaliou o aumento da temperatura de armazenagem de maçãs clones de ‘Gala’ sob atmosfera controlada (AC), tratadas com 1-metilciclopropeno (1-MCP), como método para redução do desenvolvimento de escurecimento da polpa e do consumo de energia durante o armazenamento. Experimentos foram conduzidos em 2011, com maçãs produzidas em Fraiburgo (armazenadas em câmaras experimentais a -0,3 e 1,2 °C, e em câmaras comerciais a 0,7 e 2 °C) e em São Joaquim (armazenadas em câmaras comerciais a 0,8; 1,4 e 1,9 °C). Frutos de todos os tratamentos foram mantidos sob AC (1,8±0,2 kPa de O2 e 2,0±0,2 kPa de CO2/UR de 91±4%), sendo que metade dos frutos de cada temperatura de armazenamento foi tratada com 1-MCP. Os frutos foram analisados após o armazenamento em AC quanto à firmeza da polpa e incidência de escurecimento da polpa, rachaduras e podridões. De maneira geral, maçãs armazenadas em temperaturas mais elevadas mantiveram melhor firmeza da polpa e apresentaram menor incidência de escurecimento da polpa rachaduras e podridões, quando não tratadas com 1-MCP. O tratamento com 1-MCP retardou a perda de firmeza de polpa e reduziu a ocorrência de escurecimento de polpa e rachaduras. O aumento da temperatura de armazenagem em câmaras comerciais de AC em Fraiburgo, de 0,7 °C para 2,0 °C resultou em economia no consumo de energia em aproximadamente 21% para ventilação do ar da câmara e 50% para refrigeração.

HIV-infected women are at increased risk of cervical intraepithelial neoplasia (CIN) and invasive cervical cancer (ICC), but it has been difficult to disentangle the influences of heavy exposure to HPV infection, inadequate screening and immunodeficiency. A case-control study including 364 CIN2/3 and 20 ICC cases matched to 1,147 controls was nested in the Swiss HIV Cohort Study (1985-2013). CIN2/3 risk was significantly associated with low CD4+ cell counts, whether measured as nadir [odds ratio (OR) per 100-cell/μL decrease = 1.15, 95% CI: 1.08, 1.22], or at CIN2/3 diagnosis (1.10, 95% CI: 1.04, 1.16). An association was evident even for nadir CD4+ 200-349 versus ≥350 cells/μL (OR = 1.57, 95% CI: 1.09, 2.25). After adjustment for nadir CD4+, a protective effect of >2-year cART use was seen against CIN2/3 (OR versus never cART use = 0.64, 95% CI: 0.42, 0.98). Despite low study power, similar associations were seen for ICC, notably with nadir CD4+ (OR for 50 vs. >350 cells/μL= 11.10, 95% CI: 1.24, 100). HPV16-L1 antibodies were significantly associated with CIN2/3, but HPV16-E6 antibodies were nearly exclusively detected in ICC. In conclusion, worsening immunodeficiency, even at only moderately decreased CD4+ cell counts, is a significant risk factor for CIN2/3 and cervical cancer.

The solution fluorescence of N-alkyl-2,3-naphthalimides (1-4) in polar protic and aprotic solvents was compared to the emission from solid samples resulting from the imide complexation with b-cyclodextrin or adsorption on the surface of microcrystalline cellulose. Solid samples of the inclusion complex 2,3-naphthalimides/b-cyclodextrin show maximum for fluorescence emission significantly different to the observed in methanolic solution. Beside this, a clear effect on the alkyl chain length could be observed for these samples which is probably due to differences in probe location inside the cyclodextrin cavity. The constancy for fluorescence quantum yield and fluorescence lifetime for the imides 1 - 4 adsorbed on microcrystalline cellulose suggests that, independently of the polarity of the solvent used for sample preparation, the probe is preferentially located on the cellulose surface. An increase of fluorescence quantum yield and fluorescence lifetime for solid samples, when compared to the values obtained in solution for the different solvents employed in this study (acetonitrile, methanol and water), is fully in accordance with a decrease of the probe mobility due to inclusion in b-cyclodextrin or to adsorption on cellulose.
