Commercial longline fishing data were analyzed and experiments were conducted with gear equipped with hook timers and timedepth recorders in the Réunion Island fishery (21°5ʹS lat., 53°28ʹE long.) to elucidate direct and indirect effects of the lunar cycle and other operational factors that affect catch rates, catch composition, fish behavior, capture time, and fish survival. Logbook data from 1998 through 2000, comprising 2009 sets, indicated that swordfish (Xiphias gladius) catch-per unit of effort (CPUE) increased during the first and last quarter of the lunar phase, whereas albacore (Thunnus alalunga) CPUE was highest during the full moon. Swordfish were caught rapidly after the longline was set and, like bigeye tuna (Thunnus obesus), they were caught during days characterized by a weak lunar illumination—mainly during low tide. We found a significant but very low influence of chemical lightsticks on CPUE and catch composition. At the time the longline was retrieved, six of the 11 species in the study had >40% survival. Hook timers indicated that only 8.4% of the swordfish were alive after 8 hours of capture, and two shark species (blue shark [Prionace glauca] and oceanic whitetip shark [Carcharhinus longimanus]) showed a greater resilience to capture: 29.3% and 23.5% were alive after 8 hours, respectively. Our results have implications for current fishing practices and we comment on the possibilities of modifying fishing strategies in order to reduce operational costs, bycatch, loss of target fish at sea, and detrimental impacts on the environment.

Short-duration (5- or 10-day) deployments of pop-up satellite archival tags were used to estimate survival of white marlin (Tetrapturus albidus) released from the western North Atlantic recreational fishery. Forty-one tags, each recording temperature, pressure, and light level readings approximately every two minutes for 5-day tags (n= 5) or four minutes for 10-day tags (n= 36), were attached to white marlin caught with dead baits rigged on straight-shank (“J”) hooks (n =21) or circle hooks (n=20) in offshore waters of the U.S. Mid-Atlantic region, the Dominican Republic, Mexico, and Venezuela. Forty tags (97.8%) transmitted data to the satellites of the Argos system, and 33 tags (82.5%) transmitted data consistent with survival of tagged animals over the deployment duration. Approximately 61% (range: 19−95%) of all archived data were successfully recovered from each tag. Survival was significantly (P<0.01) higher for white marlin caught on circle hooks (100%) than for those caught on straight-shank (“J”) hooks (65%). Time-to-death ranged from 10 minutes to 64 hours following release for the seven documented mortalities, and five animals died within the first six hours after release. Reflecting this emphasis the six elements of this approach are a commitment to people and place, participatory action research, gender transformative research, learning and networking, partnerships, and capacity building. It is through the careful pursuit of these six elements that we believe that the program will achieve the development outcomes we aspire to, and do so at scale.

Following a static bioassay techniques the acute toxicity of cadmium to six species of intertidal invertebrates was determined. The sensitivity of the animals to cadmium was of the following order: Emerita sp. (burrowing crustacean) Donax spiculum (burrowing bivalve) Perna viridis (sedentary bivalve) Sabellaria clandestinus (tube-dwelling polychaete) Modiolus carvalhoi and Modiolus sp. (sedentary bivalves). The above observation was based on the median lethal concentrations recorded for the different species, Emerita sp. 1.35 p.p.m., Donax spiculum 1.8 p.p.m., Perna viridis 2.5 p.p.m., Sabellaria clandestinus 2.8 p.p.m., Modiolus carvalhoi 5.6 p.p.m. and Modiolus sp. 9.6 p.p.m. The findings throw insight into the toxicity of cadmium to the common intertidal animals which are either suspension or detritus feeders.

The culture of Penaeus monodon has explicitly defined the need for diet formulations or supplementary feeds that would promote optimum growth and survival of the animal. A total of 28 feed combinations were developed for P. monodon. Fish meal, shrimp head meal, squid head meal, Ascetes spp. rice bran, and soybean cake were used as primary ingredients in these feeds. The commercial vitamin mix No. 22 was added to the dry ingredients. Gelatinized corn starch and wheat flour were used as binders. The pellets were extruded using a portable kitchen grinder with a diameter of 4 mm. The products were either sun-dried for 8 hours or oven-dried overnight at 50 degree C to stabilize moisture at 8-10%. The pellets were then kept in covered glass bottles and stored in the laboratory at room temperature. The cost of the feeds excluding labour were also computed. The pellets were analyzed for protein, fat, carbohydrate, crude fiber, ash, and moisture contents using standard procedures. They were also analyzed for water stability. To test the stability of pellets in water, 2-g samples were placed in plankton nets (mesh #40) and suspended in water for two, and six hours. The undissolved samples were then vacuum-dried and the moisture determined. Cost of the feeds ranged from P1.10 to P2.60 per kg depending on the feed ingredient. Squid and Ascetes spp. were rather expensive for use as basic ingredients. Proximate analysis of dry weight showed percentage protein content ranged from 20-63 g; fat, 8-20 g; carbohydrate (by difference), 11-36 g; ash, 8-28 g; moisture, 6-11 g; and crude fiber, 5 . 13 g. Stability tests showed that after two hours, 35-88% of solids remained intact and after 6 hours, 20-55% of the pellets remained undissolved. When a pellet disintegrates easily, pollution of the water occurs. Chances for the shrimp to feed on the pellet is minimized when the pellet is unstable. Thus, the search for a more compact feed pellet has to be continued.

Rice bran is widely used by fish farmers as supplementary feed while soybean cake is used both as feed and as fertilizer in fishponds. Both fish meal and shrimp head have been found acceptable as feed ingredients. However, not much is known of the acceptability and efficiency of a mixture of these ingredients as feed for Penaeus monodon larvae. Ninety 127-day old P. monodon were measured for length and weight and were randomly divided into nine aquaria each containing 20 liters of water. These were fed 'lampirong' for two months previous to the study. There were three replications for each treatment. Length, weight, and survival rates were used to compare the efficiency of the diets. Weighed amounts of pellets equivalent to 100% of the body weight were fed during the first three days and reduced to 50% thereafter. A stopwatch was used to determine the length of time that elapsed before the shrimps would approach the pellet. Ten shrimps approximately 4 months in age were placed in 10 liters of water in a 25-liter aquarium. Two grams of each pellet type were placed simultaneously on opposite sides of the aquarium. The time that elapsed from the moment the pellets sunk to the bottom up to the time that any one shrimp approached the pellets was recorded. The group fed the imported pellets gained the most. Those fed FP-2s-77 elongated faster than those fed FP-1s-77. Survival rate of those fed FP-2s-77 was 37% while those fed imported pellets was 73%. Both 1s and 2s pellets disintegrated in water easily but the imported pellets were stable even after six hours in water. The attractability test for the pellets showed that the prawns were more readily attracted to the pellets 1s and 2s than to the imported pellets.

The species are: Arabella iricolor (Montagu), Capitella capitata (Fabricius), Mesochaetopterus sagittarius (Claparede), Phyllochaetopterus herdmani Willey, Lysidice natalensis Kinberg and Marphysa depressa (Schmarda).

Commercial cultivation of the dioecious brown macroalga Hizikia fusiformis (Harvey) Okamura in East Asia depends on the supply of young seedlings from regenerated holdfasts or from wild population. Recent development of synchronized release of male and female gametes in tumble culture provides a possibility of mass production of young seedlings via sexual reproduction. In this paper, we demonstrate that controlled fertilization can be efficiently realized in ambient light and temperature in a specially designed raceway tank in which the sperm-containing water has been recirculated. The effective fertilization time of eggs by sperm was found to be within six hours. Fast growth and development of the young seedlings relied on the presence of water currents. Velocity tests demonstrated that young seedlings of 2-3 mm in length could withstand a water current of 190 cm s(-1) stop without detachment. Culture experiments at 24 h postfertilization showed that elongation of both the seedlings and their rhizoids were not hampered by high irradiance up to 600 mu mol photons m(-2) stop s(-1) stop. However, growth was slightly retarded if cultured at a temperature of 16 degrees C compared to other culture temperatures of 22, 25 and 29 degrees C. No seedling detachment was observed after transfer of the young seedlings to raft cultivation in the sea after one and 1.5 months post-fertilization, indicating the feasibility of obtaining large quantity of seedlings in such a system.

Neural network models of working memory, called Sustained Temporal Order REcurrent (STORE) models, are described. They encode the invariant temporal order of sequential events in short term memory (STM) in a way that mimics cognitive data about working memory, including primacy, recency, and bowed order and error gradients. As new items are presented, the pattern of previously stored items is invariant in the sense that, relative activations remain constant through time. This invariant temporal order code enables all possible groupings of sequential events to be stably learned and remembered in real time, even as new events perturb the system. Such a competence is needed to design self-organizing temporal recognition and planning systems in which any subsequence of events may need to be categorized in order to to control and predict future behavior or external events. STORE models show how arbitrary event sequences may be invariantly stored, including repeated events. A preprocessor interacts with the working memory to represent event repeats in spatially separate locations. It is shown why at least two processing levels are needed to invariantly store events presented with variable durations and interstimulus intervals. It is also shown how network parameters control the type and shape of primacy, recency, or bowed temporal order gradients that will be stored.

This research aimed to investigate the main concern facing nurses in minimising risk within the perioperative setting and to generate an explanatory substantive theory of how they resolve this through anticipatory vigilance. In the context of the perioperative setting, nurses encounter challenges in minimising risks for their patients on a continuous basis. Current explanations of minimising risk in the perioperative setting offers insights into how perioperative nurses undertake their work. Currently research in minimising risk is broadly related to dealing with errors as opposed to preventing them. To date, little is known about how perioperative nurses practice and maintain safety. This study was guided by the principles of classic grounded theory as described by Glaser (1978, 1998, 2001). Data was collected through individual unstructured interviews with thirty seven perioperative nurses (with varying lengths of experiences of working in the area) and thirty three hours of non-participant observation within eight different perioperative settings in the Republic of Ireland. Data was simultaneously collected and analysed. The theory of anticipatory vigilance emerged as the pattern of behaviour through which nurse’s deal with their main concern of minimising risk in a high risk setting. Anticipatory vigilance is enacted through orchestrating, routinising and momentary adapting within a spirit of trusting relations within the substantive area of the perioperative setting. This theory of offers an explanation on how nurses resolve their main concern of minimising risk within the perioperative setting. The theory of anticipatory vigilance will be useful to nurses in providing a comprehensive framework of explanation and understanding on how nurses deal with minimising risk in the perioperative setting. The theory links perioperative nursing, risk and vigilance together. Clinical improvements through understanding and awareness of the theory of anticipatory vigilance will result in an improved quality environment, leading to safe patient outcomes.

This PhD thesis describes work carried out on investigation of various interventions with the aim to optimise the anaesthetic management of patients scheduled to undergo operative fixation of hip fractures. We analysed the perioperative effects of continuous femoral nerve block, single preoperative dose of i.v. dexamethasone, the intention to deposit local anaesthetic in different locations around the femoral nerve during ultrasound guided femoral nerve block, continuous spinal anaesthesia and peri-surgical site infiltration with local anaesthetic after surgical fixation of hip fractures. Continuous femoral nerve block provided more effective preoperative analgesia six hours after the insertion of the perineural catheter compared to a standard opiate-based regimen in patients undergoing operative fixation of fractured hip. A single low dose of preoperative dexamethasone in the intervention group decreased pain scores by 75% six hours after the surgery. Both interventions had no major effect on the functional recovery in the first year after the surgical fixation of fractured hip. The results of the ultrasound guided femoral nerve block trial showed no clinical advantage of intending to deposit local anaesthetic circumferentially during performing femoral nerve block. Using the Dixon and Massey’s “up- and-down” method, we demonstrated that intrathecal 0.26 ml of 0.5% bupivacaine provided adequate surgical anaesthesia within 15 minutes in 50% of patients undergoing operative fixation of hip fracture. Finally, we demonstrated that local anaesthetic infiltration had no effect on pain scores 12 hours after the surgical fixation of fractured neck of femur. In addition to this original body of work, a review article was published on femoral nerve block highlighting the use of ultrasound guidance. In conclusion, the results of this thesis offer an insight into interventions aimed at optimising perioperative analgesia in patients scheduled to undergo operative fixation of hip fractures.

Light is a critical environmental signal that regulates every phase of the plant life cycle, from germination to floral initiation. Of the many light receptors in the model plant Arabidopsis thaliana, the red- and far-red light-sensing phytochromes (phys) are arguably the best studied, but the earliest events in the phy signaling pathway remain poorly understood. One of the earliest phy signaling events is the translocation of photoactivated phys from the cytoplasm to the nucleus, where they localize to subnuclear foci termed photobodies; in continuous light, photobody localization correlates closely with the light-dependent inhibition of embryonic stem growth. Despite a growing body of evidence supporting the biological significance of photobodies in light signaling, photobodies have also been shown to be dispensable for seedling growth inhibition in continuous light, so their physiological importance remains controversial; additionally, the molecular components that are required for phy localization to photobodies are largely unknown. The overall goal of my dissertation research was to gain insight into the early steps of phy signaling by further defining the role of photobodies in this process and identifying additional intragenic and extragenic requirements for phy localization to photobodies.

Even though the domain structure of phys has been extensively studied, not all of the intramolecular requirements for phy localization to photobodies are known. Previous studies have shown that the entire C-terminus of phys is both necessary and sufficient for their localization to photobodies. However, the importance of the individual subdomains of the C-terminus is still unclear. For example a truncation lacking part of the most C-terminal domain, the histidine kinase-related domain (HKRD), can still localize to small photobodies in the light and behaves like a weak allele. However, a point mutation within the HKRD renders the entire molecule completely inactive. To resolve this discrepancy, I explored the hypothesis that this point mutation might impair the dimerization of the HKRD; dimerization has been shown to occur via the C-terminus of phy and is required for more efficient signaling. I show that this point mutation impairs nuclear localization of phy as well as its subnuclear localization to photobodies. Additionally, yeast-two-hybrid analysis shows that the wild-type HKRD can homodimerize but that the HKRD containing the point mutation fails to dimerize with both itself and with wild-type HKRD. These results demonstrate that dimerization of the HKRD is required for both nuclear and photobody localization of phy.

Studies of seedlings grown in diurnal conditions show that photoactivated phy can persist into darkness to repress seedling growth; a seedling's growth rate is therefore fastest at the end of the night. To test the idea that photobodies could be involved in regulating seedling growth in the dark, I compared the growth of two transgenic Arabidopsis lines, one in which phy can localize to photobodies (PBG), and one in which it cannot (NGB). Despite these differences in photobody morphology, both lines are capable of transducing light signals and inhibiting seedling growth in continuous light. After the transition from red light to darkness, the PBG line was able to repress seedling growth, as well as the accumulation of the growth-promoting, light-labile transcription factor PHYTOCHROME INTERACTING FACTOR 3 (PIF3), for eighteen hours, and this correlated perfectly with the presence of photobodies. Reducing the amount of active phy by either reducing the light intensity or adding a phy-inactivating far-red pulse prior to darkness led to faster accumulation of PIF3 and earlier seedling growth. In contrast, the NGB line accumulated PIF3 even in the light, and seedling growth was only repressed for six hours; this behavior was similar in NGB regardless of the light treatment. These results suggest that photobodies are required for the degradation of PIF3 and for the prolonged stabilization of active phy in darkness. They also support the hypothesis that photobody localization of phys could serve as an instructive cue during the light-to-dark transition, thereby fine-tuning light-dependent responses in darkness.

In addition to determining an intragenic requirement for photobody localization and further exploring the significance of photobodies in phy signaling, I wanted to identify extragenic regulators of photobody localization. A recent study identified one such factor, HEMERA (HMR); hmr mutants do not form large photobodies, and they are tall and albino in the light. To identify other components in the HMR-mediated branch of the phy signaling pathway, I performed a forward genetic screen for suppressors of a weak hmr allele. Surprisingly, the first three mutants isolated from the screen were alleles of the same novel gene, SON OF HEMERA (SOH). The soh mutations rescue all of the phenotypes associated with the weak hmr allele, and they do so in an allele-specific manner, suggesting a direct interaction between SOH and HMR. Null soh alleles, which were isolated in an independent, tall, albino screen, are defective in photobody localization, demonstrating that SOH is an extragenic regulator of phy localization to photobodies that works in the same genetic pathway as HMR.

In this work, I show that dimerization of the HKRD is required for both the nuclear and photobody localization of phy. I also demonstrate a tight correlation between photobody localization and PIF3 degradation, further establishing the significance of photobodies in phy signaling. Finally, I identify a novel gene, SON OF HEMERA, whose product is necessary for phy localization to photobodies in the light, thereby isolating a new extragenic determinant of photobody localization. These results are among the first to focus exclusively on one of the earliest cellular responses to light - photobody localization of phys - and they promise to open up new avenues into the study of a poorly understood facet of the phy signaling pathway.

The paper presents a dynamic study of the Spanish labour market which tries to determine if it matches the characteristics of transitional labour markets from a fl exicurity approach. Employment trajectories of Spanish workers during the years 2007-2010 are studied using the Continuous Sample of Working Lives. This period covers the end of the expansion of the Spanish economy and the beginning of the current employment crisis. From the combination of the chosen topic, the approach, and the database used, this is a novel perspective in our country. The article shows evidence of the evolution of the employment and unemployment spells, the Spanish labour market turnover degree, and the diffi culties of some groups for carrying out transition between employment and unemployment. The results obtained show a labour market in which a) transitions have come to a halt, and b) there is high job insecurity.

Self-compacting concrete (SCC) flows into place and around obstructions under its own weight to fill the formwork completely and self-compact without any segregation and blocking. Elimination of the need for compaction leads to better quality concrete and substantial improvement of working conditions. This investigation aimed to show possible applicability of genetic programming (GP) to model and formulate the fresh and hardened properties of self-compacting concrete (SCC) containing pulverised fuel ash (PFA) based on experimental data. Twenty-six mixes were made with 0.38 to 0.72 water-to-binder ratio (W/B), 183–317 kg/m3 of cement content, 29–261 kg/m3 of PFA, and 0 to 1% of superplasticizer, by mass of powder. Parameters of SCC mixes modelled by genetic programming were the slump flow, JRing combined to the Orimet, JRing combined to cone, and the compressive strength at 7, 28 and 90 days. GP is constructed of training and testing data using the experimental results obtained in this study. The results of genetic programming models are compared with experimental results and are found to be quite accurate. GP has showed a strong potential as a feasible tool for modelling the fresh properties and the compressive strength of SCC containing PFA and produced analytical prediction of these properties as a function as the mix ingredients. Results showed that the GP model thus developed is not only capable of accurately predicting the slump flow, JRing combined to the Orimet, JRing combined to cone, and the compressive strength used in the training process, but it can also effectively predict the above properties for new mixes designed within the practical range with the variation of mix ingredients.

A surface plasmon resonance biosensor method was developed to measure zilpaterol residues in sheep urine. A CM-5 sensor chip previously reacted with ethylenediamine to produce an aminoethyl group was coupled with 4-carboxybutyl zilpaterol activated using EDC/NHS. Five polyclonal and four monoclonal antibodies were screened for their suitability to detect low levels of zilpaterol using the biosensor technology. Total binding was greater for polyclonal than monoclonal antibodies, but a less diluted antibody solution was required for polyclonal antibodies. A fixed antibody concentration and various concentrations of zilpaterol were injected to obtain a standard curve for each antibody to allow for B-0 and IC50 determination. The stability of the assay was assessed by the consistency of B0 in repeated experiments extending at least six hours. A measure of non-specific binding allowed the assessment of the specificity of the antibody-immobilized ligand interaction.
