This article describes a quantitative study of the diffusion rate of ferrocene(Fc) dissolved in ploy(ethylene glycol)(PEG) medium containing MClO(4)(M = Li+, Na+, Bu(4)N(+), Hx(4)N(+)). The apparent diffusion coefficient D-app and the active concentration c(a) of Fc were simultaneously measured by using non-steady-state chronoamperometry. The D-app and c(a) of Fc have been estimated in PEG containing different concentrations and sizes of supporting electrolyte, and the dependence of D-app on ferrocene concentrations has been observed. The values of D-app decrease with increasing concentrations of Fc, increasing concentrations of LiClO4 or the ratio (O:Li) and also with 4 decreasing cation radius of the electrolyte. The temperature dependencies conform to a simple free volume model. The concentration and size of the counterion dependencies of the diffusion rate are similar to the behavior of their dependencies of ionic conductivity in polyelectrolyte.

Non-steady-state chronoamperometry of ultramicroelectrodes is a powerful method for the study of mass transport in polymer films. This method has many advantages over the conventional methods at a macroelectrode and the steady state method at an ultramicroelectrode, which yield the most information. The apparent diffusion coefficient, D(app), and the concentration of reactant in the film, c(f), can be determined from a single experiment without knowing the thickness of the film. We studied the transport of several species such as Ru(NH3)63+, Ru(bpy)3(2+), NR and MV2+ in Eastman-AQ polymer film coated ultramicroelectrodes by using this method.

A perturbation method is used to study effective response of nonlinear Kerr composites, which are subject to the constitutive relation of electric displacement and electric field, D-alpha = epsilon(alpha)E + chi(alpha)vertical bar E vertical bar(2)E. Under the external AC and DC electric field E-app = E-a (1 + sinwt), the effective nonlinear responses and local potentials are induced by the cubic nonlinearity of Kerr materials at all harmonics. As an example in three dimensions, we have investigated this kind of nonlinear composites with spherical inclusions embedded in a host. At all harmonic frequencies, the potentials in inclusion and host regions are derived. Furthermore, the formulae of the effective linear and nonlinear responses are given in the dilute limit.

The perturbation method is developed to investigate the effective nonlinear dielectric response of Kerr composites when the external ac and dc electric field is applied. Under the external ac and dc electric field E-app=E-a(1+sin omegat), the effective coupling nonlinear response can be induced by the cubic nonlinearity of Kerr nonlinear materials at the zero frequency, the finite basic frequency omega, the second and the third harmonics, 2omega and 3omega, and so on. As an example, we have investigated the cylindrical inclusions randomly embedded in a host and derived the formulas of the effective nonlinear dielectric response at harmonics in dilute limit. For a higher concentration of inclusions, we have proposed a nonlinear effective-medium approximation by introducing the general effective nonlinear response. With the relationships between the effective nonlinear response at harmonics and the general effective nonlinear response, we have derived a set of formulas of the effective nonlinear dielectric responses at harmonics for a larger volume fraction. For higher concentration of inclusions, an effective medium approximation (EMA) method is used to investigate the bulk effective response of weakly nonlinear composites, which are subject to the constitutive relation of electric displacement and electric field, D-alpha = epsilon E-alpha + chi(alpha)|E|(2) E. As an example of three dimensions, under the external AC and DC electric fields E-app = E-a (1 + sin omega t), we have derived the general effective nonlinear response of composites by the EMA method for higher concentration of spherical inclusions. Furthermore, the effective nonlinear responses at harmonics are predicted.

Gene regulation is required for activity-dependent changes in synaptic plasticity and remodeling. The metabotropic glutamate receptors (mGluRs) contribute to different brain functions, including learning/memory, mental disorders, drug addiction, and persistent pain in the CNS. We found that Gp I mGluRs activate PLCß through Gq and then lead to activation of several calcium-dependent signaling pathways, including ERK, which play an important role in gene transcription. These findings support a calcium-dependent role for Gq in release of Calcium and activation of calcium-stimulated adenylyl cyclases I in activity-dependent transcription in response to application of group I metabotropic glutamate receptors agonist and may provide insights into group I mGluRs-dependent synaptic plasticity through MAP kinases signaling. Moreover, the present study investigated the transcription-dependent changes of Arc in response to the activation of group I mGluRs and suggested the central role of ERK1/2 in group I mGluR-mediated Arc transcription. Further, we selected APP-interaction protein FE65 to investigate the mechanism of transcription-related process in synaptic plasticity. FE65 is expressed predominantly in the brain, and interacts with the C-terminal domain of β-amyloid precursor protein (APP). We examined hippocampus-dependent memory and in vivo long-term potentiation (LTP) at the CA1 synapses with the isoform-specific FE65 knock-out (p97FE65-/-) mice. p97FE65 knock-out mice showed impaired short-term memory for both TDPA and CFC when tested 10min after training, which is transcription-independent. Consistently, at the Schaffer collateral-CA1 synapses, p97FE65 knock-out mice showed defective early phase LTP. These results demonstrate novel roles of FE65 in synaptic plasticity, acquisition, and retention for certain forms of memory formation.

O Código Florestal brasileiro, juntamente com a resolução Conama no 303/2002, definiu áreas do território nacional classificadas como de preservação permanente (APP). Essas áreas englobam margens dos cursos e corpos d'água, terrenos com declividade acentuada, bordas de chapadas, topos de morro, entre outras feições. No entanto, a escassez de dados cartográficos em escala adequada e de abrangência nacional dificultam ou até impossibilitam as estimativas do alcance territorial da legislação ambiental brasileira. Um exemplo é a delimitação das APPs nas margens dos rios, que, para identificação correta, requer, além da localização, informações sobre a largura dos cursos d'água na época de cheias, dados raramente disponíveis. Este trabalho utilizou dados provenientes da Agência Nacional de Águas (ANA) para estimar as APPs das margens dos rios pertencentes à bacia do Rio Ji-Paraná, RO, identificados na escala 1:1.000.000, levando-se em consideração as larguras dos cursos d'água. Dados de todas as dez estações fluviométricas presentes na bacia com medidas de cota e perfil transversal da calha foram utilizados para estimar a largura máxima do canal de duas maneiras distintas. A primeira delimitou as sub-bacias definidas pelas estações fluviométricas e considerou que todos os cursos d'água pertencentes à sub-bacia apresentavam a mesma largura observada em seu ponto final. A segunda utilizou a relação empírica obtida entre a área de drenagem e a largura da calha nas nove estações para definir a largura de todos os trechos dos cursos d'água da bacia. Por fim, as APPs nas margens dos rios foram delimitadas seguindo os critérios estabelecidos na resolução Conama no 303/2002. A título de comparação, foram também delimitadas as APPs utilizando larguras constantes de 100 m, 200 m e 500 m de faixa marginal para toda a bacia. Entre os diferentes métodos utilizados, a APP nas margens dos rios variou de 1.541 km2 a 15.876 km2 (2,04% a 21,04% da área da bacia, respectivamente). A grande variação na delimitação das áreas marginais deixa claro que estimativas efetuadas para grandes regiões estarão sempre sujeitas a incertezas, de acordo com os métodos utilizados, e que, portanto, é imprescindível o detalhamento dos procedimentos efetuados para esclarecer as vantagens e limitações a que tais estimativas estão sujeitas.

A proteção de Áreas de Preservação Permanente (APP) tem sido foco de muita discussão na concepção da nova Lei 12.651/2012 que substitui a Lei 4.771/1965 que instituía o Código Florestal Brasileiro, com divergências entre ruralistas e ambientalistas no que tange à conservação ambiental. Quantificar e identificar o estado de degradação destas áreas é de fundamental importância para a orientação de políticas públicas e ações voltadas à conservação dos recursos florestais, com implicações na qualidade do solo e da água, e fauna. Desta forma, o presente estudo se propôs a traçar um perfil comparativo do déficit de vegetação natural em APPs - matas ciliares e nascentes - em duas bacias hidrográficas sob o bioma Mata Atlântica - RJ, Guapi-Macacu e Caceribu. Para tal, foram utilizadas ferramentas implementadas em SIG para processamento e organização de dados cartográficos e delimitação das APPs, bem como para identificação de áreas de vegetação natural presentes em APPs de matas ciliares e nascentes e cálculo de áreas. Dados secundários como o mapa de uso e cobertura da terra (obtido pelo processamento de imagens de 2007 do satélite Landsat) foram utilizados para a obtenção das áreas de vegetação. Foi obtido um mapa final com as áreas de vegetação natural em APPs de matas ciliares diferenciadas de áreas de vegetação natural em APPs de nascentes, apresentando também a vegetação natural presente em ambas as bacias que não em áreas de APPs de matas ciliares e nascentes. Os resultados apontaram que a bacia do Caceribu possui um déficit de vegetação natural em APP de matas ciliares e nascentes da ordem de 91%, e no caso da bacia Guapi-Macacu esse déficit é de aproximadamente 38%. Estes dados, apesar de terem sido obtidos na escala 1:50.000, a partir de imagens de média resolução espacial - 30 metros, chamam a atenção para o fato de que ações de revegetação destas áreas são prioritárias perante sua importância para a prestação de diversos serviços ambientais, essenciais para o bem estar das populações que vivem nestas bacias ou em bacias próximas.

The importance of γ-secretase protease activities in development, neurogenesis and the immune system are highlighted by the diversity of its substrates and phenotypic characterization of Presenilin (PS)-deficient transgenic animals. Since the discovery of Amyloid precursor protein (APP) and it’s cleavage by γ-secretase complexes, over 90 other type I membrane proteins have been identified as γ-secretase substrates. We have identified interleukin-1 (IL-1) receptor type I (IL-1R1), toll-like receptor 4 (TLR4) and tumour necrosis factor-α (TNFα) receptor-1 (TNFR1) as novel substrates for - secretase cleavage, which play an important role in innate immunity. In this study, using PS-deficient cells and PS-knockout animal models we examined the role of PS proteins, PS1 and PS2, in IL-1R1-, TLR4- and TNFR1- mediated inflammatory responses. Data presented show that in response to IL- 1β, lipopolysaccharide (LPS) or TNFα, immortalised fibroblasts from PS2- deficient animals have diminished production of specific cytokines and chemokine, with differential reduction in nuclear factor-κB (NF-κB) and (mitogen activated protein kinase) MAPK activities. In contrast, no defect in the response to IL-1β, LPS or TNFα was observed in PS1-deficient immortalised fibroblasts. These observations were confirmed using bone marrow-derived macrophages from PS2-null mice, which also display impaired responsiveness to IL-1β- and LPS, with decreased production of inflammatory cytokines. Furthermore, in whole animal in vivo responses, we show that PS2-deficient animals display ligand (IL-1β, LPS and TNFα)-dependent alterations in the production of specific pro-inflammatory cytokines or chemokines. Importantly, this reduced responsiveness to IL-1β, LPS or TNFα is independent of γ- secretase protease activity and γ-secretase cleavage of TNFR1, IL-1R1 or TLR4. These observations suggest a novel γ-secretase-independent role of PS2 in the regulation of innate immune responsiveness and challenge current concepts regarding the regulation of IL-1β-, LPS- and TNFα-mediated immune signalling.

We provide evidence that college graduation plays a direct role in revealing ability to the labor market. Using the NLSY79, our results suggest that ability is observed nearly perfectly for college graduates, but is revealed to the labor market more gradually for high school graduates. Consequently, from the beginning of their careers, college graduates are paid in accordance with their own ability, while the wages of high school graduates are initially unrelated to their own ability. This view of ability revelation in the labor market has considerable power in explaining racial differences in wages, education, and returns to ability.

A computer-based numerical modelling of the adsorption process of gas phase metallic particles on the surface of a graphite substrate has been performed via the application of molecular dynamics simulation method. The simulation relates to an extensive STM-based experiment performed in this field, and reproduces part of the experimental results. Both two-body and many-body inter-atomic potentials have been employed. A Morse-type potential describing the metal-carbon interactions at the interface was specifically formulated for this modelling. Intercalation of silver in graphite has been observed as well as the correct alignments of monomers, dimers and two-dimensional islands on the surface. The pseudo-spectral solution method offers a flexible and fast alternative to the more usual finite element and volume methods, particularly when the long-time transient behaviour of a system is of interest. The exact solution is obtained at grid collocation points leading to superior accuracy on modest grids. Furthermore, the grid can be freely adapted in time and space to particular flow conditions or geometric variations, especially useful where strongly coupled, time-dependent, multi-physics solutions are investigated. Examples include metallurgical applications involving the interaction of electromagnetic fields and conducting liquids with a free surface. The electromagnetic field determines the instantaneous liquid volume shape, which then affects the electromagnetic field. A general methodology of the pseudo-spectral approach is presented, with several instructive example applications: the aluminium electrolysis MHD problem, induction melting in a cold crucible and the dynamics of AC/DC magnetically levitated droplets. Finally, comparisons with available analytical solutions and to experimental measurements are discussed.
