Knowing the structure and distribution of nutrients in plant tissues can clarify some mechanisms of pathogen attack in plants and plant defense against infection, thus helping management strategies. The aim of this study was verify differences in distribution of mineral nutrients in coffee leaf tissues around foliar lesions of bacterial blight of coffee, blister spot, cercospora leaf, phoma leaf spot and coffee leaf rust. Fragments of leaf tissue surrounding the lesions were dehydrated in silica gel, carbon covered and subjected to X-ray microanalysis (MAX). Thirty-three chemical elements were detected in leaf tissue; however, there was variation in potassium and calcium contents surrounding the lesions. The highest potassium content was found in asymptomatic tissues surrounding the lesions, decreasing toward the transition zone and reaching minimum content in symptomatic tissues. The highest calcium content was found in symptomatic tissues, decreasing toward the transition zone and reaching minimum content in asymptomatic tissues. Therefore, MAX can be used to analyze the composition and distribution of nutrients in plant tissues and, if associated with mineral nutrition, it may help understand host-pathogen relationships and plant disease management.

This study compared the effect of physicochemical surface conditioning methods on the adhesion of bis-GMA-based resin cement to particulate filler composite (PFC) used for indirect dental restorations. PFC blocks (N (block)=54, n (block)=9 per group) were polymerized and randomly subjected to one of the following surface conditioning methods: a) No conditioning (Control-C), b) Hydrofluoric acid (HF)etching for 60s (AE60), c) HF for 90s (AE90), d) HF for 120s (AE120), e) HF for 180s (AE180), and f) air-abrasion with 30 mu m silica-coated alumina particles (AB). The conditioned surfaces were silanized with an MPS silane, and an adhesive resin was applied. Resin composite blocks were bonded to PFC using resin cement and photo-polymerized. PFC-cement-resin composite blocks were cut under coolant water to obtain bar specimens (1mmx0.8mm). Microtensile bond strength test (mu TBS)was performed in a universal testing machine (1mm/min). After debonding, failure modes were classified using stereomicroscopy. Surface characterization was performed on a set of separate specimen surfaces using Scanning Electron Microscopy (SEM), X-Ray Dispersive Spectroscopy (XDS), X-Ray Photoelectron Spectroscopy (XPS), and Fourier Transform-Raman Spectroscopy (FT-RS). Mean mu TBS (MPa) of C (35.6 +/- 4.9) was significantly lower than those of other groups (40.2 +/- 5.6-47.4 +/- 6.1) (p<0.05). The highest mu TBS was obtained in Group AB (47.4 +/- 6.1). Prolonged duration of HF etching increased the results (AE180: 41.9 +/- 7), but was not significantly different than that of AB (p>0.05). Failure types were predominantly cohesive in PFC (34 out of 54) followed by cohesive failure in the cement (16 out of 54). Degree of conversion (DC) of the PFC was 63 +/- 10%. SEM analysis showed increased irregularities on PFC surfaces with the increased etching time. Chemical surface analyses with XPS and FT-RS indicated 11-70% silane on the PFC surfaces that contributed to improved bond strength compared to Group C that presented 5% silane, which seemed to be a threshold. Group AB displayed 83% SiO2 and 17% silane on the surfaces.

Although many studies have shown that soil solution chemistry can be a reliable indicator of biogeochemical cycling in forest ecosystems, the effects of litter manipulations on the fluxes of dissolved elements in gravitational soil solutions have rarely been investigated. We estimated the fluxes of NH4-N, NO3-N, K, Ca, Mg, Na, Cl, dissolved organic nitrogen (DON) and dissolved organic carbon (DOC) over the first two years after re-planting Eucalyptus trees in the coastal area of Congo. Two treatments were replicated in two blocks after clear-cutting 7-year-old stands: in treatment R, all the litter above the mineral soil was removed before planting, and in a double slash (DS) treatment, the amount of harvest residues was doubled. The soil solutions were sampled down to a depth of 4 m and the water fluxes were estimated using the Hydrus 1D model parameterized from soil moisture measurements in 4 plots. Isotopic and spectroscopic analytical techniques were used to assess the changes in dissolved organic matter (DOM) properties throughout the transfer in the soil. The first year after planting, the fluxes of NH4-N, K, Ca, Mg, Na, Cl and DOC in the topsoil of the DS treatment were 2-5 times higher than in R, which showed that litter was a major source of dissolved nutrients. Nutrient fluxes in gravitational solutions decreased sharply in the second year after planting, irrespective of the soil depth, as a result of intense nutrient uptake by Eucalyptus trees. Losses of dissolved nutrients were noticeably low in these Eucalyptus plantations despite a low cation exchange capacity, a coarse soil texture and large amounts of harvest residues left on-site at the clear cut in the DS treatment. All together, these results clarified the strong effect of litter manipulation observed on eucalypt growth in Congolese sandy soils. DOM fluxes, as well as changes in delta C-13, C:N and aromaticity of DOM throughout the soil profile showed that the organic compounds produced in the litter layer were mainly consumed by microorganisms or retained in the topsoil. Below a depth of 15 cm, most of the DOC and the DON originated from the first 2 cm of the soil and the exchanges between soil solutions and soil organic matter were low. (C) 2014 Elsevier B.V. In this study, the physicochemical characteristics of calcium phosphate based bioactive ceramics of different compositions and blends presenting similar micro/nanoporosity and micrometer scale surface texture were characterized and evaluated in an in vivo model. Prior to the animal experiment, the porosity, surface area, particle size distribution, phase quantification, and dissolution of the materials tested were evaluated. The bone regenerative properties of the materials were evaluated using a rabbit calvaria model. After 2, 4, and 8 weeks, the animals were sacrificed and all samples were subjected to histologic observation and histomorphometric analysis. The material characterization showed that all materials tested presented variation in particle size, porosity and composition with different degrees of HA/TCP/lower stoichiometry phase ratios. Histologically, the calvarial defects presented temporal bone filling suggesting that all material groups were biocompatible and osteoconductive. Among the different materials tested, there were significant differences found in the amount of bone formation as a function of time. At 8 weeks, the micro/nanoporous material presenting similar to 55,TCP:45%,HA composition ratio presented higher amounts of new bone regeneration relative to other blends and a decrease in the amount of soft tissue infiltration. This study investigated the effect of inexpensive carbon and nitrogen sources on enzyme production by Myceliophthora thermophila M.7.7 in solid-state fermentation. Three kinds of lignocellulosic waste (corn straw, sugarcane bagasse and sugarcane straw) and six nitrogen sources (urea, calcium nitrate, analytical ammonium sulphate, yeast extract, agricultural fertilizer NPK 20-05-20 and fertilizing grade ammonium sulphate) were tested. Some physical-chermical parameters of the fermentation, such as temperature, initial pH and moisture content of the substrate on enzyme production, were evoluated. The maximum activities of xylanase (446.9 U/ml) endoglucanase (94.7 U/ml) and beta-glucosidase (2.8 U/ml) were observed in a mixture of corn straw and wheat bran (1:1 w/w) as the carbon source using fertilizer grade ammonium sulphate as the nitrogen source. This production occurred for an incubation period of 96 h, at 40°C, with initial moisture content of 70% and pH 5.0. These results have significant interest since they could be used for the future production of enzymes in a low-cost industrial process.
