The diatom flora in a 164 cm long sediment core obtained from Jiaozhou Bay (Yellow Sea, China) was analyzed in order to trace the response of diatoms to environmental changes over the past 100 years. The sediment core was dated by Pb-210 and Cs-137 and represented approximately 100 years (1899-2001 A.D.). The flora was mainly composed of centric diatoms (59-96%). The concentration of diatoms declined sharply above 30 cm (after similar to 1981 A.D.), while the dominant species changed from Thalassiosira anguste-lineatus, Thalassiosira eccentria, Coscinodiscus excentricus, Coscinodiscus concinnus and Diploneis gorjanovici to Cyclotella stylorum and Paralia sulcata. Species richness decreased slightly, and the cell abundance of warm-water species increased. We argue that these floral changes were probably caused by climate change in combination with eutrophication resulting from aquaculture and sewage discharge. (c) 2007 Elsevier B.V. Background and Aims The systematic position of the genus Metagentiana and its phylogenetic relationships with Crawfurdia, Gentiana and Tripterospermum have not been explicitly addressed. These four genera belong to one of two subtribes (Gentianinae) of Gentianeae. The aim of this paper is to examine the systematic position of Crawfurdia, Metagentiana and Tripterospermum and to clarify their phylogenetic affinities more clearly using ITS and trnL intron sequences.Methods Nucleotide sequences from the internal transcribed spacers (ITS) of nuclear ribosomal DNA and the plastid DNA trnL (UAA) intron were analysed phylogenetically. Ten of fourteen Metagentiana species were sampled, together with 40 species of other genera in the subtribe Gentianinae.Key Results The data support several previously published conclusions relating to the separation of Metagentiana from Gentiana and its closer relationships to Crawfurdia and Tripterospermum based on studies of gross morphology, floral anatomy, chromosomes, palynology, embryology and previous molecular data. The molecular clock hypothesis for the tested sequences in subtribe Gentianinae was not supported by the data (P < 0.05), so the clock-independent non-parametric rate smoothing method was used to estimate divergence time. This indicates that the separation of Crawfurdia, Metagentiana and Tripterospermum from Gentiana occurred about 11.4-21.4 Mya (million years ago), and the current species of these three genera diverged at times ranging from 0.4 to 6.2 Mya.Conclusions The molecular analyses revealed that Crawfurdia, Metagentiana and Tripterospermum do not merit status as three separate genera, because sampled species of Crawfurdia and Tripterospermum are embedded within Metagentiana. The speciation and rapid radiation of these three genera is likely to have occurred in western China as a result of upthrust of the Himalayas during the late Miocene and the Pleistocene.

The combined occurrence of both herkogamy and dichogamy in a hermaphrodite species has been considered to strongly favour outcrossing. In this study, we investigated in detail the reproductive ecology of Gentiana straminea Maxim. (Gentianaceae), a hermaphrodite perennial endemic to the Qinghai-Tibet Plateau. In a series of observations and experiments over four consecutive years, we examined whether the combination of dichogamy and herkogamy in individual flowers completely prevents geitonogamous pollen transfer in this species. The mode of floral development clearly indicates that autonomous self-pollination is completely avoided through herkogamy and dichogamy in individual flowers. This implication was confirmed by the breeding experiments, since no seed was produced when flowers were isolated. However, this gentian proved to be highly self-compatible when geitonogamous selfing was artificially induced. Many flowers opened simultaneously on individual plants, the ratio of male to female phase flowers was close to 2:1 in each inflorescence, at the full anthesis phase, and they were randomly distributed amongst the upper, middle and lower parts of each stem's inflorescence. On average, Bombus sushikini Skorikov, the most frequent visitor and only legitimate pollinator of G. straminea, visited nearly two flowers per inflorescence, and four flowers per plant. Among the pollinators' foraging bouts, the proportions of geitonogamous visits to inflorescences or flowers within an individual plant were 29% and 37%, respectively. Therefore, despite the strict dichogamous and herkogamous characteristics of the individual flowers, geitonogamous selfing might still prevail in G. straininea because of the size of its floral displays and the continuous visiting behavior of B. sushkini. The evidence from cross morphology, floral anatomy, chromosomes, palynology, and embryology all indicates that sect. Stenogyne is discordant within the genus Gentiana and is as distinct from the other sections of Gentiana as are other genera, such as Tripterospermum and Crawfurdia. In light of these characters, sect. Stenogyne is removed from Gentiana and given generic rank as the new genus Metagentiana. It is more related to Tripterospermum and Crawfurdia than to Gentiana, though it is more primitive than the first two genera. Together with Tripterospermum and Crawfurdia the new genus forms a monophyletic group, which is the sister group to the genus Gentiana. Fourteen new combinations required at specific rank are proposed.

To better understand the floral diversity and the phylogeny of the Swertiinae (Gentianaceae), the internal transcribed spacer (ITS) region of nrDNA for 17 species and one outgroup was sequenced. Our data suggest that corolla type, gland shape, and corolla appendage are poorly correlated with the ITS phylogeny. The genus Swertia s.l. and the rotate group previously recognized based on the corolla types and gland shapes are polyphyletic. Four genera with simple protruding glands and three taxa with corolla appendages are not clustered as the monophyletic groups. Four separate clades, corresponding to the four sections, were identified in Swertia s.1. Lomatogoniopsis with the simple protruding gland type of the tubular group closely related to Lomatogonium of the rotate group. The deeply lobing corolla and concave foveae may be ancestral in the Swertiinae, while the tubular corolla and the protruding glands may have undergone convergent evolution.

Uma das principais limitações para a abacaxicultura na região Extremo Sul da Bahia é a ocorrência de floração natural nos meses de junho e julho, que resulta na contratação da colheita, em dezembro, época de grande oferta e, consequentemente, de preços baixos. Combinações de época de plantio e de tamanho de mudas são usadas para tentar evitar a concentração da safra e conseguir deslocar a produção para épocas de preços mais altos. Resultados de pesquisa indicaram que o plantio de março a maio, com adubação baseada na análise de solo e conduzido sob condiçõe de sequeiro, possibilitou a indução artificial do florescimento das plantas a partir dos dez meses de idade, o que permite programar as colheitas no período de junho a outubro do ano subsequente ao plantio. O plantio no mês de janeiro possibilitou a colheita de grande parte da produção no período da entressafra, pois resultou na incidência de florações naturais a partir de julho do mesmo ano e colheita dos frutos durante o primeiro semestre do ano seguinte. A indução do florescimento do abacaxizeiro plantado em janeiro deve ser realizada nos meses de setembro ou outubro em plantas sem diferenciação floral natural. Esse procedimento permite a colheita dos frutos no período de preços mais favoráveis (até março) e a floração das plantas em período pouco favorável à incidêcia da fusariose nos frutos (novembro/dezembro), em decorrência da menor umidade relativa do ar e temperatura mais alta.

This study selected six geographically-similar villages with traditional and alternative cultivation methods (two groups of three, one traditional and two alternatives) in two counties of Henan Province, China—a representative area of the Huang-huai-hai Plain representing traditional rural China. Soil heavy metal concentrations, floral and faunal biodiversity, and socio-economic data were recorded. Heavy metal concentrations of surface soils from three sites in each village were analysed using Inductively Coupled Plasma Mass Spectrometry (ICP-MS, chromium, nickel, copper, cadmium, and lead) and Atomic Absorption Spectrophotometer (AAS, zinc). The floral biodiversity of four land-use types was recorded following the Braun-Blanquet coverage-abundance method using 0.5×0.5m quadrats. The faunal biodiversity of two representative farmland plots was recorded using 0.3×0.3m quadrats at four 0.1m layers. The socio-economic data were recorded through face-to-face interviews of one hundred randomly selected households at each village. Results demonstrate different cultivation methods lead to different impact on above variables. Traditional cultivation led to lower heavy metal concentrations; both alternative managements were associated with massive agrochemical input causing heavy metal pollution in farmlands. Floral distribution was significantly affected by village factors. Diverse cultivation supported high floral biodiversity through multi-scale heterogeneous landscapes containing niches and habitats. Faunal distribution was also significantly affected by village factor nested within soil depth. Different faunal groups responded differently, with Acari being taxonomically diverse and Collembola high in densities. Increase in manual labour and crop number in villages using alternative cultivation may positively affect biodiversity. The results point to the conservation potential of diverse cultivation methods in traditional rural China and other regions under social and political reforms, where traditional agriculture is changing to unified, large-scale mechanized agriculture. This study serves as a baseline for conservation in small-holding agricultural areas of China, and points to the necessity of further studies at larger and longer scales.

Light is a critical environmental signal that regulates every phase of the plant life cycle, from germination to floral initiation. Of the many light receptors in the model plant Arabidopsis thaliana, the red- and far-red light-sensing phytochromes (phys) are arguably the best studied, but the earliest events in the phy signaling pathway remain poorly understood. One of the earliest phy signaling events is the translocation of photoactivated phys from the cytoplasm to the nucleus, where they localize to subnuclear foci termed photobodies; in continuous light, photobody localization correlates closely with the light-dependent inhibition of embryonic stem growth. Despite a growing body of evidence supporting the biological significance of photobodies in light signaling, photobodies have also been shown to be dispensable for seedling growth inhibition in continuous light, so their physiological importance remains controversial; additionally, the molecular components that are required for phy localization to photobodies are largely unknown. The overall goal of my dissertation research was to gain insight into the early steps of phy signaling by further defining the role of photobodies in this process and identifying additional intragenic and extragenic requirements for phy localization to photobodies.

Even though the domain structure of phys has been extensively studied, not all of the intramolecular requirements for phy localization to photobodies are known. Previous studies have shown that the entire C-terminus of phys is both necessary and sufficient for their localization to photobodies. However, the importance of the individual subdomains of the C-terminus is still unclear. For example a truncation lacking part of the most C-terminal domain, the histidine kinase-related domain (HKRD), can still localize to small photobodies in the light and behaves like a weak allele. However, a point mutation within the HKRD renders the entire molecule completely inactive. To resolve this discrepancy, I explored the hypothesis that this point mutation might impair the dimerization of the HKRD; dimerization has been shown to occur via the C-terminus of phy and is required for more efficient signaling. I show that this point mutation impairs nuclear localization of phy as well as its subnuclear localization to photobodies. Additionally, yeast-two-hybrid analysis shows that the wild-type HKRD can homodimerize but that the HKRD containing the point mutation fails to dimerize with both itself and with wild-type HKRD. These results demonstrate that dimerization of the HKRD is required for both nuclear and photobody localization of phy.

Studies of seedlings grown in diurnal conditions show that photoactivated phy can persist into darkness to repress seedling growth; a seedling's growth rate is therefore fastest at the end of the night. To test the idea that photobodies could be involved in regulating seedling growth in the dark, I compared the growth of two transgenic Arabidopsis lines, one in which phy can localize to photobodies (PBG), and one in which it cannot (NGB). Despite these differences in photobody morphology, both lines are capable of transducing light signals and inhibiting seedling growth in continuous light. After the transition from red light to darkness, the PBG line was able to repress seedling growth, as well as the accumulation of the growth-promoting, light-labile transcription factor PHYTOCHROME INTERACTING FACTOR 3 (PIF3), for eighteen hours, and this correlated perfectly with the presence of photobodies. Reducing the amount of active phy by either reducing the light intensity or adding a phy-inactivating far-red pulse prior to darkness led to faster accumulation of PIF3 and earlier seedling growth. In contrast, the NGB line accumulated PIF3 even in the light, and seedling growth was only repressed for six hours; this behavior was similar in NGB regardless of the light treatment. These results suggest that photobodies are required for the degradation of PIF3 and for the prolonged stabilization of active phy in darkness. They also support the hypothesis that photobody localization of phys could serve as an instructive cue during the light-to-dark transition, thereby fine-tuning light-dependent responses in darkness.

In addition to determining an intragenic requirement for photobody localization and further exploring the significance of photobodies in phy signaling, I wanted to identify extragenic regulators of photobody localization. A recent study identified one such factor, HEMERA (HMR); hmr mutants do not form large photobodies, and they are tall and albino in the light. To identify other components in the HMR-mediated branch of the phy signaling pathway, I performed a forward genetic screen for suppressors of a weak hmr allele. Surprisingly, the first three mutants isolated from the screen were alleles of the same novel gene, SON OF HEMERA (SOH). The soh mutations rescue all of the phenotypes associated with the weak hmr allele, and they do so in an allele-specific manner, suggesting a direct interaction between SOH and HMR. Null soh alleles, which were isolated in an independent, tall, albino screen, are defective in photobody localization, demonstrating that SOH is an extragenic regulator of phy localization to photobodies that works in the same genetic pathway as HMR.

In this work, I show that dimerization of the HKRD is required for both the nuclear and photobody localization of phy. I also demonstrate a tight correlation between photobody localization and PIF3 degradation, further establishing the significance of photobodies in phy signaling. Finally, I identify a novel gene, SON OF HEMERA, whose product is necessary for phy localization to photobodies in the light, thereby isolating a new extragenic determinant of photobody localization. These results are among the first to focus exclusively on one of the earliest cellular responses to light - photobody localization of phys - and they promise to open up new avenues into the study of a poorly understood facet of the phy signaling pathway.

La temperatura y las precipitaciones son factores claves en la fenología de especies vegetales y su productividad. Los pastizales de la provincia de San Luis son especialmente interesantes porque coexisten gramíneas con vías metabólicas C3 (invernales) y C4 (estivales). Para evaluar cómo se relacionan la producción y la fenología reproductiva de las especies frente a cambios de temperatura y disponibilidad de agua, evalué cuatro gramíneas representativas de la región (dos C3 y dos C4) en dos estudios: 1) observacional a campo, y 2) manipulativo en invernáculo con dos niveles de temperatura y dos niveles de agua. En el estudio observacional comparé diez años de relevamientos fenológicos históricos realizados entre 1976 y 1986, con relevamientos fenológicos propios entre 2008 y 2010, todos llevados a cabo sobre el mismo sitio de estudio. En esta comparación encontré que una de las cuatro especies (Poa ligularis), atrasó significativamente su ciclo reproductivo (38 días la floración y 16 días la diseminación de semillas). La temperatura tuvo menor efecto sobre la producción, pero reflejó un claro control sobre la fenología en las cuatro especies estudiadas. En general, altas temperaturas estivales atrasaron el fin y aumentaron el largo del ciclo reproductivo de todas las especies. En cambio, primaveras más cálidas adelantaron el inicio reproductivo de las C4, pero retrasaron el de las C3. Por otra parte, la mayor disponibilidad de agua incrementó la producción, adelantó el comienzo floral y extendió entre 15 y 30 días el ciclo reproductivo de las cuatro especies. Inviernos lluviosos adelantaron el inicio floral de las especies C3, mientras que años con elevadas precipitaciones estivales retrasaron y extendieron el ciclo reproductivo de las C4. Estos resultados aportan valiosa información sobre las respuestas de la vegetación al clima, y pueden servir de insumo en el diseño de estrategias de manejo sustentable de estos pastizales.

La pérdida de la biodiversidad que genera la disminución de interacciones bióticas y procesos ecosistémicos es uno de los principales problemas ecológicos en la actualidad. Esta situación se ha relacionado a la simplificación de los sistemas naturales y a la intensificación de los sistemas productivos agrícolas, prácticas que incluso pueden ir en detrimento del rendimiento de los campos de cultivo. Sin embargo, las huertas podrían ser reservorios de biodiversidad y recursos alimenticios que provean hábitats de alta calidad para muchos organismos. En este trabajo, se relaciona el tipo de manejo agronómico (convencional y orgánico) en huertas aguacateras con la riqueza y la composición de las comunidades de visitantes florales, acarreadores de polen y plantas herbáceas con el fin de estudiar la diversidad asociada a dichos sistemas y la incidencia del manejo agronómico sobre estas comunidades. El trabajo se compone de 3 capítulos. El primer capítulo plantea la relación entre el manejo agronómico y la diversidad de visitantes florales y plantas herbáceas a partir de la generación de Unidades Básicas de Clasificación (UBC). En el segundo capítulo, se aborda la ecología reproductiva del aguacatero, en donde se estimó la oferta floral, la diversidad de la comunidad de visitantes florales y acarreadores de polen, su eficiencia de estos en el acarreo de polen y su contribución a la producción de frutos bajo el manejo agronómico orgánico y convencional. En el tercer capítulo, se construyeron y analizaron seis redes de interacción de polen acarreado-acarreador en dos momentos del cultivo: i) durante la floración del aguacatero y ii) en ausencia de floración del aguacatero. El estudio se llevó a cabo durante dos años consecutivos en el pico de floración 2010/2011 y 2011/2012 en 6 huertas orgánicas y 4 convencionales (...) Los resultados de este trabajo mostraron como el manejo agronómico puede influir en la estructura de la comunidad de visitantes, acarreadores de polen y herbáceas. Considerando que el cultivo del aguacatero presenta la floración en una época específica del año, es importante que existan de forma continua otros recursos florales para los acarreadores como son las herbáceas. Los recursos provenientes de estas plantas pueden mantener durante el año la funcionalidad de la comunidad de visitantes y acarredores, proporcionando el servicio ecosistémico de la polinización de plantas silvestres y de las cultivadas. A su vez, la tesis contribuye al fortalecimiento del conocimiento de patrones que proponen que la disponibilidad de recursos florales en las huertas derivados de manejos agronómicos más integrales y menos intensivos que favorecen el incremento de la riqueza de visitantes de flores y acarreadores de polen nativos

Maximizar la productividad en las plantaciones densas de olivo requiere de un manejo apropiado de la iluminación del canopeo. Pero el limitado conocimiento acerca de la respuesta del rendimiento a la radiación fotosintéticamente activa (PAR) dificulta su logro. Además, los periodos potencialmente críticos, donde el rendimiento sería sensible al sombreado, y el efecto de la poda sobre la respuesta a la PAR, han sido escasamente estudiados. Esta tesis se centró sobre las relaciones entre los determinantes y componentes del rendimiento, y la PAR, utilizando dos aproximaciones. En primer lugar, se caracterizó el ambiente lumínico en distintas posiciones de un seto en dos estaciones de crecimiento y se estableció su relación con los determinantes y componentes del rendimiento. Las yemas axilares e inflorescencias respondieron bilinealmente a la PAR aunque la poda alteró esta respuesta. El peso seco del fruto y la concentración de aceite respondieron linealmente a la irradiancia. En segundo lugar, se manipuló directamente el nivel de PAR (3 a 70 de PAR incidente) con redes mediasombra en árboles semi-aislados, focalizando las mediciones en una posición externa de la copa. Combinando ambos enfoques, se contrastaron las funciones de respuestas derivadas de distintas posiciones en el seto (primera aproximación) con aquellas obtenidas en una posición externa de la copa (segunda aproximación). Utilizando la segunda aproximación se demostró que la inducción floral, el cuaje y llenado del fruto son periodos críticos para el rendimiento bajo PAR limitante. También fue posible establecer que los frutos evidenciaron una mejor capacidad de recuperación después de retirar los tratamientos, cuando el periodo de post-sombreo precedió y/o aconteció durante la división celular del fruto. La información aquí generada podría mejorar el modelado de la estructura óptima del canopeo, que asegura una óptima intercepción de la PAR y un alto rendimiento.

El comportamiento bienal facultativo de Oenothera biennis representa una limitante para su explotación comercial extensiva. Los individuos de esta especie pueden florecer en la primera estación de crecimiento (ciclo anual) o retrasar su floración hasta la siguiente (ciclo bianual). Este comportamiento estaría modulado por la capacidad endógena de las plantas para responder a las señales ambientales que desencadenan la floración, la cual dependería del logro de un umbral de biomasa. El objetivo de esta tesis fue contribuir al entendimiento de los factores que controlan la floración de Oenothera biennis haciendo énfasis en la relación entre el tamaño de las plantas y su respuesta a las señales inductoras de la reproducción. Se realizaron experimentos en macetas a campo donde se manipularon factores candidatos a inducir la floración (vernalización y fotoperíodo). Se modificó la disponibilidad de nutrientes mediante fertilización mineral, para lograr variabilidad en los indicadores de la condición endógena de las plantas (área y tasa de crecimiento de las rosetas). La vernalización, el fotoperíodo (días largos) y la fertilización redujeron la incidencia de plantas de comportamiento bienal (hasta 25 por ciento respecto al control) y adelantaron la floración entre 3 y 31 días (según tratamiento y condiciones experimentales) en las plantas de comportamiento anual La respuesta a la vernalización fue facultativa y no dependió de una biomasa umbral, ya que se vernalizaron plántulas recién germinadas. La respuesta al fotoperíodo tampoco requirió de un tamaño umbral, pero estuvo condicionada por la tasa de crecimiento de las plantas, la cual fue modulada por la fertilización y el área de las rosetas al inicio de las condiciones fotoinductivas. Se identificó a la incidencia de temperaturas elevadas (T media menor a 25ºC; Tmáx menor a 30ºC), alrededor de la iniciación floral, como un posible factor causal de la bianualidad que inhibiría la reproducción, de una proporción de los individuos, en fechas de siembra tardías. Se concluye que los factores que aceleran el desarrollo (vernalización, fotoperíodos extendidos artificialmente y nutrición mineral) contribuyen indirectamente a reducir la bianualidad, evitando que las condiciones inductivas de la floración coincidan con el período estival de alta temperatura.
