The effectiveness of macrophages in the response to systemic candidiasis is crucial to an effective clearance of the pathogen. The secretion of proteins, mRNAs, non-coding RNAs and lipids through extracellular vesicles (EVs) is one of the mechanisms of communication between immune cells. EVs change their cargo to mediate different responses, and may play a role in the response against infections. Thus, we have undertaken the first quantitative proteomic analysis on the protein composition of THP1 macrophages-derived EVs during the interaction with Candida albicans. This study revealed changes in EVs sizes and in protein composition, and allowed the identification and quantification of 717 proteins. Of them, 133 proteins changed their abundance due to the interaction. The differentially abundant proteins were involved in functions relating to immune response, signaling, or cytoskeletal reorganization. THP1-derived EVs, both from control and from Candida-infected macrophages, had similar effector functions on other THP1-differenciated macrophages, activating ERK and p38 kinases, and increasing both the secretion of proinflammatory cytokines and the candidacidal activity; while in THP1 non-differenciated monocytes, only EVs from infected macrophages increased significantly the TNF-α secretion. Our findings provide new information on the role of macrophage-derived EVs in response to C. albicans infection and in macrophages communication.

The aim of the present work was to characterize the internal structure of nanogratings generated inside bulk fused silica by ultrafast laser processing and to study the influence of diluted hydrofluoric acid etching on their structure. The nanogratings were inscribed at a depth of 100 mu m within fused silica wafers by a direct writing method, using 1030 nm radiation wavelength and the following processing parameters: E = 5 mu J, tau = 560 fs, f = 10 kHz, and v = 100 mu m/s. The results achieved show that the laser-affected regions are elongated ellipsoids with a typical major diameter of about 30 mu m and a minor diameter of about 6 mu m. The nanogratings within these regions are composed of alternating nanoplanes of damaged and undamaged material, with an average periodicity of 351 +/- 21 nm. The damaged nanoplanes contain nanopores randomly dispersed in a material containing a large density of defects. These nanopores present a roughly bimodal size distribution with average dimensions for each class of pores 65 +/- 20 x 16 +/- 8 x 69 +/- 16 nm(3) and 367 +/- 239 x 16 +/- 8 x 360 +/- 194 nm(3), respectively. The number and size of the nanopores increases drastically when an hydrofluoric acid treatment is performed, leading to the coalescence of these voids into large planar discontinuities parallel to the nanoplanes. The preferential etching of the damaged material by the hydrofluoric acid solution, which is responsible for the pores growth and coalescence, confirms its high defect density. Population Structure and reproductive Maturity of females were investigated in the shrimp Artemesia longinaris Bate, 1888 from coastal waters of northern São Paulo State (Brazil. 23 degrees S) and Mar del Plata (Argentina, 38 degrees S) from June 2001 to May 2002. Monthly collections were taken by, commercial shrimp fishing boats equipped with bottom trawl nets. Population parameters from size frequency distributions and size (carapace length = CL) of female reproductive maturity were analyzed and compared front the two sampling areas. Latitudinal trends in reproductive parameters of A. longinaris were shown in overall body size and size of reproductive maturity, both of which were smaller in females from the tropical location than those front the cold-temperate sampling area. Largest females (> 30 turn CL) were collected in Argentina, while Brazilian specimens reached maximum size at 27 nun CL. The smallest Size of female sexual maturity was estimated at 13.6 nun CL in Brazilian samples compared to 22.1 mm CL calculated for those from Argentina. Populations from both regions exhibited a bimodal size distribution in the spring, with the peak at small body size probably corresponding to recent recruits and the peak at larger body, size to reproductive females or shrimps migrating in from deeper waters or other latitudinal regions. In late spring and summer. an intrusion of the cold South Atlantic Coastal Water mass was observed which lowered water temperature and stimulated plankton production. The primary food source for the larvae of a typically cold-temperate species such as A. longinaris. The trend of increasing body size and delay of sexual maturity with increasing latitude appears to be correlated with the decreasing water temperature and increasing plankton productivity at higher latitudes.

In dieser Arbeit wurden zwei Wege zur sauer katalysierten Synthese von Polyorganosiloxan-Kern-Schale-Nanopartikeln mit monomodaler Größenverteilung erarbeitet. Zum einen führt eine Erhöhung der Rührergeschwindigkeit auf 14000 rpm unter Verwendung eines Ultrathorax, während der ersten Stunde der Kondensation des Kernes zu Nanokugeln mit einer monomodalen Größenverteilung mit einem hydrodynamischen Radius von 33.1 nm +/- 22%. Zum anderen eröffnete die Reduktion des Flottenverhältnisses S, d.h. des Verhältnisses von Tensid- zu Monomermenge, von S=0.02 auf S=0.001 einen zweiten Weg zur Synthese von Nanokugeln mit monomodaler Größenverteilung. Der Radius dieser Kugeln beträgt 54.2 nm +/- 20%. Durch diese beiden Synthesewege sind Polyorganosiloxan-Nanokugeln in zwei verschiedenen Größen zugänglich. Durch 29Si-NMR-Messungen der Kerndispersion konnte gezeigt werden, dass die Ursache der bimodalen Größenverteilung der Polyorganosiloxan-Nanokugeln in der Synthese der Kerndispersion zu finden ist. Aus den hieraus ermittelten Ergebnissen geht hervor, dass sich während der sauer katalysierten Kondensation von Diethoxydimethylsilan neben den PDMS-Ketten auch zyklische Kondensationsprodukte bilden. Die während der Reaktion fortschreitende Bildung von Zyklen - insbesondere von Vierringen - bewirkt eine Phasenseparation der Dispersion. Dies führt zur beobachteten Bildung der bimodalen Größenverteilung der Polyorganosiloxan-Nanokugeln. Wird die Rührergeschwindigkeit auf 14000 rpm während der ersten Stunde der Kondensation des Kernes erhöht, wird eine verminderte Ringbildung gefunden. Der erhöhte Energieeintrag und die damit verbundene bessere Durchmischung der Dispersion während der ersten Stunde der Kondensation des Kerns führt bevorzugt zum Kettenwachstum, so dass die Tendenz zur Ringbildung verringert wird. Es tritt keine Phasenseparation auf, wodurch die beobachtete monomodale Größenverteilung der Nanokugeln erklärt wird. Wird das Flottenverhältnis reduziert und somit der pH-Wert der Lösung erhöht, werden bevorzugt offenkettige PDMS-Produkte gebildet. Die Bildung von Vierringen erfolgt nicht, Ringe höherer Ordnung werden nur in untergeordneter Menge gebildet. Es erfolgt keine Phasenseparation der Dispersion und eine monomodale Größenverteilung der Polyorganosiloxan-Nanokugeln wird erhalten. Die durch Erniedrigung des Flottenverhältnisses synthetisierten Polyorganosiloxan-Nanokugeln zeigten in AFM-Experimenten interessante Eigenschaften. So ist es möglich, die Nanokugeln auf einer Mica-Oberfläche mittels der AFM-Spitze zu manipulieren, ohne sie hierbei sie degradieren. Die sauer katalysierte Synthese ermöglicht die Einführung von basenlabilen Hydridgruppen in die Polyorganosiloxan-Nanokugeln. Ausgehend von in organischen Lösungsmitteln redispergierbaren Polyorganosiloxan-Nanokugeln, deren Oberfläche mit Hydridgruppen funktionalisiert wurde, konnten durch Hydrosilylierung mit allylterminiertem Polyethylenoxid wasserlösliche potentielle Nanokontainer synthetisiert werden.

Polymere Hohlstrukturen eignen sich um eine große Anzahl an Gastmolekülen zu verkapseln und bieten somit interessante Anwendungsmöglichkeiten, z.B. im Bereich kontrollierter Transportsysteme. Solche wohl definierten Strukturen lassen sich mittels des Sol-Gel-Prozesses durch Hydrolyse und Kondensation von Dialkoxydialkyl- und Trialkoxyalkylsilanen in wässriger Dispersion in Gegenwart von Tensiden synthetisieren. Die Methode ermöglicht den Aufbau verschiedener Kern-Schale-Systeme, inklusive Hohlkugelarchitekturen, mit Durchmessern von 10-100 nm. Abhängig von den eingestellten Parametern wird dabei eine bimodale Größenverteilung der Partikel beobachtet. Die bimodalen Proben wurden mittels der circularen asymmetrischen Fluss Feld-Fluss Fraktionierung (CAFFFE) fraktioniert. NMR-Untersuchungen deuten darauf hin, dass die Ursache der bimodalen Verteilung in der Synthese der Kerndispersion zu liegen scheint. MALDI-TOF-MS und GC-Messungen zeigen, dass der Kern der größeren Partikel ausschließlich aus zyklischen Kondensationsprodukten besteht, während im Kernmaterial der kleineren Partikel zusätzlich noch lineare Polydimethylsiloxan-Ketten vorhanden sind. Unter der Annahme, dass PDMS als Ultrahydrophob wirkt, lässt sich die Ostwaldreifung als Ursache der Bimodalität ausmachen. Eine Erhöhung des PDMS-Anteils, der zur Stabilisierung gegen den Reifungsprozess notwendig ist, führt zu einer monomodalen Verteilung der erhaltenen Partikel.

Friedreich’s ataxia, the most frequent inherited ataxia, is caused, in the vast majority of cases, by large GAA repeat expansions in the first intron of the frataxin gene. The normal sequence corresponds to a moderately polymorphic trinucleotide repeat with bimodal size distribution. Small normal alleles have approximately eight to nine repeats whereas a more heterogeneous mode of large normal alleles ranges from 16 to 34 GAA. The latter class accounts for ≈17% of normal alleles. To identify the origin of the expansion mutation, we analyzed linkage disequilibrium between expansion mutations or normal alleles and a haplotype of five polymorphic markers within or close to the frataxin gene; 51% of the expansions were associated with a single haplotype, and the other expansions were associated with haplotypes that could be related to the major one by mutation at a polymorphic marker or by ancient recombination. Of interest, the major haplotype associated with expansion is also the major haplotype associated with the larger alleles in the normal size range and was almost never found associated with the smaller normal alleles. The results indicate that most if not all large normal alleles derive from a single founder chromosome and that they represent a reservoir for larger expansion events, possibly through “premutation” intermediates. Indeed, we found two such alleles (42 and 60 GAA) that underwent cataclysmic expansion to pathological range in a single generation. This stepwise evolution to large trinucleotide expansions already was suggested for myotonic dystrophy and fragile X syndrome and may relate to a common mutational mechanism, despite sequence motif differences.

The work presented was conducted within the scope of a larger study investigating impacts of the Stuart Oil Shale project, a facility operating to the north of the industrial city of Gladstone, Australia. The aims of the investigations were threefold: (a) the identification of the plant signatures in terms of particle size distributions in the submicrometer range (13-830 nm) through stack measurements, (b) exploring the applicability of these signatures in tracing the source contributions at locations of interest, at a distance from the plant, and (c) assessing the contribution of the plant to the total particle number concentration at locations of interest. The stack measurements conducted for three different conditions of plant operation showed that the particle size distributions were bimodal with average modal count median diameters (CMDs) of 24 (SD 4) and 52 (SD 9) nm. The average of all the particle size distributions recorded within the plant sector at a site located 4.5 km from the plant, over the sampling period when the plant was operating, also showed a bimodal distribution. The modal CMDs in this case were 27 and 50 nm, similar to those at the stack. This bimodal size distribution is distinct from the size distribution of the most common ambient anthropogenic emission source, which is vehicle emissions, and can be considered as a signature of this source. The average contribution of the plant (for plant sector winds) was estimated to be (10.0 +/- 3.8) x 10(2) particles cm(-3) and constituted approximately a 50% increase over the local particle ambient concentration for plant sector winds. This increase in particle number concentration compared to the local background concentration, while high compared to the clean environment concentration, is not significant when compared to concentrations generally encountered in the urban environment of Brisbane.

Unique bimodal distributions of single crystal epitaxially grown In2O3 nanodots on silicon are shown to have excellent IR transparency greater than 87% at IR wavelengths up to 4 μm without sacrificing transparency in the visible region. These broadband antireflective nanodot dispersions are grown using a two-step metal deposition and oxidation by molecular beam epitaxy, and backscattered diffraction confirms a dominant (111) surface orientation. We detail the growth of a bimodal size distribution that facilitates good surface coverage (80%) while allowing a significant reduction in In2O3 refractive index. This unique dispersion offers excellent surface coverage and three-dimensional volumetric expansion compared to a thin film, and a step reduction in refractive index compared to bulk active materials or randomly porous composites, to more closely match the refractive index of an electrolyte, improving transparency. The (111) surface orientation of the nanodots, when fully ripened, allows minimum lattice mismatch strain between the In2O3 and the Si surface. This helps to circumvent potential interfacial weakening caused by volume contraction due to electrochemical reduction to lithium, or expansion during lithiation. Cycling under potentiodynamic conditions shows that the transparent anode of nanodots reversibly alloys lithium with good Coulombic efficiency, buffered by co-insertion into the silicon substrate. These properties could potentially lead to further development of similarly controlled dispersions of a range of other active materials to give transparent battery electrodes or materials capable of non-destructive in situ spectroscopic characterization during charging and discharging.

The absorption spectra of phytoplankton in the visible domain hold implicit information on the phytoplankton community structure. Here we use this information to retrieve quantitative information on phytoplankton size structure by developing a novel method to compute the exponent of an assumed power-law for their particle-size spectrum. This quantity, in combination with total chlorophyll-a concentration, can be used to estimate the fractional concentration of chlorophyll in any arbitrarily-defined size class of phytoplankton. We further define and derive expressions for two distinct measures of cell size of mixed populations, namely, the average spherical diameter of a bio-optically equivalent homogeneous population of cells of equal size, and the average equivalent spherical diameter of a population of cells that follow a power-law particle-size distribution. The method relies on measurements of two quantities of a phytoplankton sample: the concentration of chlorophyll-a, which is an operational index of phytoplankton biomass, and the total absorption coefficient of phytoplankton in the red peak of visible spectrum at 676 nm. A sensitivity analysis confirms that the relative errors in the estimates of the exponent of particle size spectra are reasonably low. The exponents of phytoplankton size spectra, estimated for a large set of in situ data from a variety of oceanic environments (~ 2400 samples), are within a reasonable range; and the estimated fractions of chlorophyll in pico-, nano- and micro-phytoplankton are generally consistent with those obtained by an independent, indirect method based on diagnostic pigments determined using high-performance liquid chromatography. The estimates of cell size for in situ samples dominated by different phytoplankton types (diatoms, prymnesiophytes, Prochlorococcus, other cyanobacteria and green algae) yield nominal sizes consistent with the taxonomic classification. To estimate the same quantities from satellite-derived ocean-colour data, we combine our method with algorithms for obtaining inherent optical properties from remote sensing. The spatial distribution of the size-spectrum exponent and the chlorophyll fractions of pico-, nano- and micro-phytoplankton estimated from satellite remote sensing are in agreement with the current understanding of the biogeography of phytoplankton functional types in the global oceans. This study contributes to our understanding of the distribution and time evolution of phytoplankton size structure in the global oceans.

We have studied the magnetic and power absorption properties of a series of magnetic nanoparticles (MNPs) of Fe(3)O(4) with average sizes < d > ranging from 3 to 26 rim. Heating experiments as a function of particle size revealed a strong increase in the specific power absorption (SPA) values for particles with < d > = 25-30 mn. On the other side saturation magnetization M(s) values of these MNPs remain essentially constant for particles with < d > above 10 rim, suggesting that the absorption mechanism is not determined by Ms. The largest SPA value obtained was 130 W/g, corresponding to a bimodal particle distribution with average size values of 17 and 26 nm.
