O Brasil é dos países líderes na importação de peixes salgados e secos. Em 2008, importou da Noruega cerca de 30 mil toneladas destes produtos, a um custo de aproximadamente 190 milhões de dólares. O bacalhau é um produto salgado e seco bastante apreciado no país, mas espécies afins, de menor valor comercial, são também utilizadas na fabricação de peixes salgados secos e muitas vezes comercializadas como o bacalhau do Atlântico (Gadus morhua). A variação da razão de isótopos estáveis de carbono (13C/12C) e de nitrogênio (15N/14N) é medida com alta precisão por espectrômetro de massa de razão isotópica (IRMS) e nos últimos 20 anos tem aumentado significativamente a aplicação desta tecnologia na avaliação de fraudes e adulterações dos alimentos. O objetivo deste trabalho foi utilizar isótopos estáveis de 15N e 13C na identificação de bacalhau e espécies afins. Utilizando a análise isotópica, em músculos e ossos, foi possível diferenciar o bacalhau (do Atlântico e do Pacífico) de Ling, Zarbo e Saithe (p < 0,01), que são três produtos salgados e secos de menor valor comercial. Com a pele não foi possível tal distinção entre bacalhau do Pacífico e Ling (p > 0,05). A combinação de análises de valores de 15N de diferentes tecidos – músculo, pele e ossos – possibilitou a separação das espécies. A comparação gráfica de valores de 15N de músculo vs. pele, músculo vs. ossos e pele vs. ossos permite estabelecer três regiões de confiança: uma com bacalhau do Atlântico e bacalhau do Pacífico; outra com Ling e Zarbo; e finalmente, uma terceira região com o Saithe isolada de todos. Os resultados obtidos demonstram que a metodologia é viável para a distinção entre bacalhau e outros peixes salgados secos. No entanto, conclusões mais seguras serão obtidas com uma base de dados, para que, isotopicamente, se faça um histórico da origem desses peixes.

Huntington's disease (HD) is an autosomal dominant neurodegenerative disorder caused by an expansion of CAG repeats in the huntingtin (Htt) gene. Despite intensive efforts devoted to investigating the mechanisms of its pathogenesis, effective treatments for this devastating disease remain unavailable. The lack of suitable models recapitulating the entire spectrum of the degenerative process has severely hindered the identification and validation of therapeutic strategies. The discovery that the degeneration in HD is caused by a mutation in a single gene has offered new opportunities to develop experimental models of HD, ranging from in vitro models to transgenic primates. However, recent advances in viral-vector technology provide promising alternatives based on the direct transfer of genes to selected sub-regions of the brain. Rodent studies have shown that overexpression of mutant human Htt in the striatum using adeno-associated virus or lentivirus vectors induces progressive neurodegeneration, which resembles that seen in HD. This article highlights progress made in modeling HD using viral vector gene transfer. We describe data obtained with of this highly flexible approach for the targeted overexpression of a disease-causing gene. The ability to deliver mutant Htt to specific tissues has opened pathological processes to experimental analysis and allowed targeted therapeutic development in rodent and primate pre-clinical models.

BACKGROUND: Spirometry reference values are important for the interpretation of spirometry results. Reference values should be updated regularly, derived from a population as similar to the population for which they are to be used and span across all ages. Such spirometry reference equations are currently lacking for central European populations. OBJECTIVE: To develop spirometry reference equations for central European populations between 8 and 90 years of age. MATERIALS: We used data collected between January 1993 and December 2010 from a central European population. The data was modelled using "Generalized Additive Models for Location, Scale and Shape" (GAMLSS). RESULTS: The spirometry reference equations were derived from 118'891 individuals consisting of 60'624 (51%) females and 58'267 (49%) males. Altogether, there were 18'211 (15.3%) children under the age of 18 years. CONCLUSION: We developed spirometry reference equations for a central European population between 8 and 90 years of age that can be implemented in a wide range of clinical settings.

Introduction: Vitamin D plays a major role in bone metabolism and neuromuscular function. Supplementation with vitamin D is effective to reduce the risk of fall and of fracture. However adherence to oral daily vitamin D supplementation is low. Screening and correcting vitamin D insufficiency in a general rheumatologic population could improve both morbidity and quality of life in these patients with chronic painful disorders and at high risk of osteoporosis. After determining the prevalence of vitamin D deficiency in this population, we evaluated if supplementation with a single high dose of oral 25-OH vitamin D3 was sufficient to correct this abnormality. Methods: During one month (November 2009), levels of 25-OH vitamin D were systematically determined in our rheumatology outpatient clinic and classified into three groups: vitamin D deficiency (<10 μg/l), vitamin D insufficiency (10 to 30 μg/l) or normal vitamin D (>30 μg/l). Patients with insufficiency or deficiency received respectively a single high dose of 300000 IU or 600000 IU oral vitamin D3. In addition, all patients with osteoporosis were prescribed daily supplement of calcium (1 g) and vitamin D (800 IU). 25-OH vitamin D levels were reevaluated after 3 months. Results: Vitamin D levels were initially determined in 292 patients (mean age 53, 211 women, 87% Caucasian). 77% had inflammatory rheumatologic disease (IRD), 20% osteoporosis (OP) and 12% degenerative disease (DD). Vitamin D deficiency was present in 20 (6.8%), while 225 (77.1%) had insufficiency. Of the 245 patients with levels <30 μg/l, a new determination of vitamin D level was available in 173 (71%) at 3 months. Conclusion: Vitamin D insufficiency is highly prevalent in our rheumatologic population (84%), and is not adequately corrected by a single high dose of oral vitamin D3 in more than half of the patients with IRD and DD. In patients with OP, despite association of a single high dose with daily oral vitamin D supplementation, 40% of patients are still deficient when reevaluated at 3 months.

The 26S proteasome constitutes an essential degradation apparatus involved in the consistent recycling of misfolded and damaged proteins inside cells. The aberrant activation of the proteasome has been widely observed in various types of cancers and implicated in the development and progression of carcinogenesis. In the era of targeted therapies, the clinical use of proteasome inhibitors necessitates a better understanding of the molecular mechanisms of cell death responsible for their cytotoxic action, which are reviewed here in the context of sensitization of malignant gliomas, a tumor type particularly refractory to conventional treatments.
