Die optische Eigenschaften sowie der Oberflächenverstärkungseffekt von rauen Metalloberflächen sowie Nanopartikeln wurden intensiv für den infraroten Bereich des Spektrums in der Literatur diskutiert. Für die Präparation solcher Oberflächen gibt es prinzipiell zwei verschiedene Strategien, zum einen können die Nanopartikel zuerst ex-situ synthetisiert werden, der zweite Ansatz beruht darauf, dass die Nanopartikel in-situ hergestellt und aufgewachsen werden. Hierbei wurden beide Ansätze ausgetestet, dabei stellte sich heraus, dass man nur mittels der in-situ Synthese der Goldnanopartikel in der Lage ist nanostrukturierte Oberflächen zu erhalten, welche elektronisch leitfähig sind, nicht zu rau sind, um eine Membranbildung zu ermöglichen und gleichzeitig einen optimalen Oberflächenverstärkungseffekt zeigen. Obwohl keine ideale Form der Nanopartikel mittels der in-situ Synthese erhalten werden können, verhalten sich diese dennoch entsprechend der Theorie des Oberflächenverstärkungseffekts. Optimierungen der Form und Grösse der Nanopartikel führten in dieser Arbeit zu einer Optimierung des Verstärkungseffekts. Solche optimierten Oberflächen konnten einfach reproduziert werden und zeichnen sich durch eine hohe Stabilität aus. Der so erhaltene Oberflächenverstärkungseffekt beträgt absolut 128 verglichen mit dem belegten ATR-Kristall ohne Nanopartikel oder etwa 6 mal, verglichen mit der Oberfläche, die bis jetzt auch in unserer Gruppe verwendet wurde. Daher können nun Spektren erhalten werden, welche ein deutlich besseres Signal zu Rauschverhältnis (SNR) aufweisen, was die Auswertung und Bearbeitung der erhaltenen Spektren deutlich vereinfacht und verkürzt.rnNach der Optimierung der verwendeten Metalloberfläche und der verwendeten Messparameter am Beispiel von Cytochrom C wurde nun an der Oberflächenbelegung der deutlich größeren Cytochrom c Oxidase gearbeitet. Hierfür wurde der DTNTA-Linker ex-situ synthetisiert. Anschließend wurden gemischte Monolagen (self assembeld monolayers) aus DTNTA und DTP hergestellt. Die NTA-Funktionalität ist für die Anbindung der CcO mit der his-tag Technologie verantwortlich. Die Kriterien für eine optimale Linkerkonzentration waren die elektrischen Parameter der Schicht vor und nach Rekonstitution in eine Lipidmembran, sowie Elektronentransferraten bestimmt durch elektrochemische Messungen. Erst mit diesem optimierten System, welches zuverlässig und reproduzierbar funktioniert, konnten weitere Messungen an der CcO begonnen werden. Aus elektrochemischen Messungen war bekannt, dass die CcO durch direkten Elektronentransfer unter Sauerstoffsättigung in einen aktivierten Zustand überführt werden kann. Dieser aktivierte Zustand zeichnet sich durch eine Verschiebung der Redoxpotentiale um etwa 400mV gegenüber dem aus Gleichgewichts-Titrationen bekannten Redoxpotential aus. Durch SEIRAS konnte festgestellt werden, dass die Reduktion bzw. Oxidation aller Redoxzentren tatsächlich bei den in der Cyclovoltammetrie gemessenen Potentialen erfolgt. Außerdem ergaben die SEIRA-Spektren, dass durch direkten Elektronentransfer gravierende Konformationsänderungen innerhalb des Proteins stattfinden. rnBisher war man davon ausgegangen, aufgrund des Elektronentransfers mittels Mediatoren, dass nur minimale Konformationsänderungen beteiligt sind. Vor allem konnte erstmaligrnder aktivierte und nicht aktivierte Zustand der Cytochrom c Oxidase spektroskopisch nachweisen werden.rn

Kernkollaps-Supernovae werden von einem massiven Ausbruch niederenergetischer Neutrinos begleitet. Sie zählen zu den energiereichsten Erscheinungen im Universum und stellen die derzeit einzig bekannte Quelle extrasolarer Neutrinos dar.rnDie Detektion einer solchen Neutrinosignatur würde zu einem tieferen Verständnis des bislang unzureichend bekannten stellaren Explosionsmechanismus führen. rnDarüber hinaus würden neue Einblicke in den Bereich der Teilchenphysik und der Supernova-Modellierung ermöglicht. Das sich zur Zeit am geographischen Südpol im Aufbau befindliche Neutrinoteleskop IceCube wird 2011 fertig gestellt sein.rnIceCube besteht im endgültigen Ausbau aus 5160 Photovervielfachern, die sich in gitterförmiger Anordnung in Tiefen zwischen 1450m und 2450m unter der Eisoberfläche befinden. Durch den Nachweis von Tscherenkow-Photonenrnim antarktischen Gletscher ist es in der Lage, galaktische Supernovae über einen kollektiven Anstieg der Rauschraten in seinen Photonenvervielfachern nachzuweisen.rnIn dieser Arbeit werden verschiedene Studien zur Implementierung einer künstlichen Totzeit vorgestellt, welche korreliertes Rauschen unterdrücken und somit das Signal-Untergund-Verhältnis maximieren würden.rnEin weiterer Teil dieser Dissertation bestand in der Integration der Supernova-Datenakquise eine neue Experiment-Steuerungssoftware.rnFür den Analyseteil der Arbeit wurde ein Monte-Carlo für IceCube entwickelt und Neutinooszillations-Mechanismen und eine Reihe von Signalmodellen integriert. Ein Likelihoodhypothesen-Test wurde verwendet, um die Unterscheidbarkeit verschiedener Supernova- beziehungsweise Neutrinooszillations-Szenarien zu untersuchen. Desweiteren wurde analysiert inwieweit sich Schock-Anregungen und QCD-Phasenübergnag im Verlauf des Explosionsprozesses detektieren lassen.

With the increasing importance that nanotechnologies have in everyday life, it is not difficult to realize that also a single molecule, if properly designed, can be a device able to perform useful functions: such a chemical species is called chemosensor, that is a molecule of abiotic origin that signals the presence of matter or energy. Signal transduction is the mechanism by which an interaction of a sensor with an analyte yields a measurable form of energy. When dealing with the design of a chemosensor, we need to take into account a “communication requirement” between its three component: the receptor unit, responsible for the selective analyte binding, the spacer, which controls the geometry of the system and modulates the electronic interaction between the receptor and the signalling unit, whose physico-chemical properties change upon complexation. A luminescent chemosensor communicates a variation of the physico-chemical properties of the receptor unit with a luminescence output signal. This thesis work consists in the characterization of new molecular and nanoparticle-based system which can be used as sensitive materials for the construction of new optical transduction devices able to provide information about the concentration of analytes in solution. In particular two direction were taken. The first is to continue in the development of new chemosensors, that is the first step for the construction of reliable and efficient devices, and in particular the work will be focused on chemosensors for metal ions for biomedical and environmental applications. The second is to study more efficient and complex organized systems, such as derivatized silica nanoparticles. These system can potentially have higher sensitivity than molecular systems, and present many advantages, like the possibility to be ratiometric, higher Stokes shifts and lower signal-to-noise ratio.

In case of violation of CPT- and Lorentz Symmetry, the minimal Standard Model Extension (SME) of Kostelecky and coworkers predicts sidereal modulations of atomic transition frequencies as the Earth rotates relative to a Lorentz-violating background field. One method to search for these modulations is the so-called clock-comparison experiment, where the frequencies of co-located clocks are compared as they rotate with respect to the fixed stars. In this work an experiment is presented where polarized 3He and 129Xe gas samples in a glass cell serve as clocks, whose nuclear spin precession frequencies are detected with the help of highly sensitive SQUID sensors inside a magnetically shielded room. The unique feature of this experiment is the fact that the spins are precessing freely, with transverse relaxation times of up to 4.4 h for 129Xe and 14.1 h for 3He. To be sensitive to Lorentz-violating effects, the influence of external magnetic fields is canceled via the weighted difference of the 3He and 129Xe frequencies or phases. The Lorentz-violating SME parameters for the neutron are determined out of a fit on the phase difference data of 7 spin precession measurements of 12 to 16 hours length. The result of the fit gives an upper limit for the equatorial component of the neutron parameter b_n of 3.7×10^(−32) GeV at the 95% confidence level. This value is not limited by the signal-to-noise ratio, but by the strong correlations between the fit parameters. To reduce the correlations and therewith improve the sensitivity of future experiments, it will be necessary to change the time structure of the weighted phase difference, which can be realized by increasing the 129Xe relaxation time.
Bau und Entwicklung eines Applikators zur Verabreichung hyperpolarisierter Gase in der MRT der Lunge

Im Rahmen meiner Dissertation habe ich gemäß dem Medizinproduktegesetz ein MR-kompatibles Verabreichungsgerät entwickelt, um Patienten hyperpolarisierte Gas Boli (He-3, Xe-129) als Kontrasgas zur MRT der Lunge zu applizieren. Das Gerät wurde dazu optimiert, die Gase oder Gasmischungen (z. B. HP Gas + N2) in definierten Mengen und zu definierten Zeitpunkten während der Inspiration mit hoher Reproduzierbarkeit und Zuverlässigkeit zu verabreichen, ohne dabei die MR-Qualität zu beeinträchtigen. Aus Sicherheitsgründen und zur späteren Datenanalyse werden die Atemflusskurven der Patienten kontinuierlich angezeigt und aufgezeichnet. Ein Kolbenkompressor ist integriert, um die gesamte gespeicherte He-3-Menge nutzen zu können. Weiterhin ist es möglich, die Polarisation vor Ort zu bestimmen und das abgeatmete He-3 zur späteren Rückgewinnung aufzufangen. Diese Auffangeffizienz konnte durch Untersuchungen mit He-4 (als He-3-Ersatz) gesteigert werden.rnDie ersten MR-Aufnahmen von 10 gesunden Probanden wurden mit diesem Aufbau imrnRahmen einer klinischen Studie durchgeführt. Die Analyse der bestimmten MR-Parameter Signal-Rausch-Verhältnis (SNR), Sauerstoffpartialdruck (pO2), Scheinbarer Diffusionskoeffizient (ADC=apparent diffusion coefficient) sowie die Signalanstiegszeit (rise-time) zeigt eine deutlich bessere Reproduzierbarkeit bei der Verabreichung des He-3 mit dem Applikators anstelle eines Tedlarbags (= kleine heliumdichte Plastiktüte).

Die Invarianz physikalischer Gesetze unter Lorentztransformationen ist eines der fundamentalen Postulate der modernen Physik und alle Theorien der grundlegenden Wechselwirkungen sind in kovarianter Form formuliert. Obwohl die Spezielle Relativitätstheorie (SRT) in einer Vielzahl von Experimenten mit hoher Genauigkeit überprüft und bestätigt wurde, sind aufgrund der weitreichenden Bedeutung dieses Postulats weitere verbesserte Tests von grundsätzlichem Interesse. Darüber hinaus weisen moderne Ansätze zur Vereinheitlichung der Gravitation mit den anderen Wechselwirkungen auf eine mögliche Verletzung der Lorentzinvarianz hin. In diesem Zusammenhang spielen Ives-Stilwell Experimente zum Test der Zeitdilatation in der SRT eine bedeutende Rolle. Dabei wird die hochauflösende Laserspektroskopie eingesetzt, um die Gültigkeit der relativistischen Dopplerformel – und damit des Zeitdilatationsfaktors γ – an relativistischen Teilchenstrahlen zu untersuchen. Im Rahmen dieser Arbeit wurde ein Ives-Stilwell Experiment an 7Li+-Ionen, die bei einer Geschwindigkeit von 34 % der Lichtgeschwindigkeit im Experimentierspeicherring (ESR) des GSI Helmholtzzentrums für Schwerionenforschung gespeichert waren, durchgeführt. Unter Verwendung des 1s2s3S1→ 1s2p3P2-Übergangs wurde sowohl Λ-Spektroskopie als auch Sättigungsspektroskopie betrieben. Durch die computergestützte Analyse des Fluoreszenznachweises und unter Verwendung optimierter Kantenfilter für den Nachweis konnte das Signal zu Rauschverhältnis entscheidend verbessert und unter Einsatz eines zusätzlichen Pumplasers erstmals ein Sättigungssignal beobachtet werden. Die Frequenzstabilität der beiden verwendeten Lasersysteme wurde mit Hilfe eines Frequenzkamms spezifiziert, um eine möglichst hohe Genauigkeit zu erreichen. Die aus den Strahlzeiten gewonnen Daten wurden im Rahmen der Robertson-Mansouri-Sexl-Testtheorie (RMS) und der Standard Model Extension (SME) interpretiert und entsprechende Obergrenzen für die relevanten Testparameter der jeweiligen Theorie bestimmt. Die Obergrenze für den Testparameter α der RMS-Theorie konnte gegenüber den früheren Messungen bei 6,4 % der Lichtgeschwindigkeit am Testspeicherring (TSR) des Max-Planck-Instituts für Kernphysik in Heidelberg um einen Faktor 4 verbessert werden.

Um die in der Atmosphäre ablaufenden Prozesse besser verstehen zu können, ist es wichtig dort vorhandene Partikel gut charakterisieren zu können. Dazu gehört unter anderem die Bestimmung der chemischen Zusammensetzung der Partikel. Zur Analyse insbesondere organischer Partikel wurde dazu in einer früheren Promotion das Aerosol-Ionenfallen-Massenspektrometer (AIMS) entwickelt.Im Rahmen dieser Arbeit wurden Entwicklungsarbeiten durchgeführt, um die Charakteristiken des Prototypen zu verbessern sowie es für den Feldeinsatz tauglich zu machen. Die durchgeführten Veränderungen betreffen mechanische und elektrische Komponenten sowie das LabView Steuerungsprogramm. So wurde z.B. die Ionenquelle derart modifiziert, dass die Ionen nicht mehr permanent erzeugt werden, sondern nur innerhalb des Zeitraums wenn sie auch in der Ionenfalle gespeichert werden können. Durch diese Modifikation konnte das Signal-zu-Rausch Verhältnis deutlich verbessert werden. Nach Beendigung der Umbauten wurden in ausführlichen Laborstudien die einzelnen Instrumentenparameter detailliert charakterisiert. Neben den Spannungen die zur Fokussierung oder zur Speicherung der Ionen in der Ionenfalle dienen, wurden die unterschiedlichen Arten der resonanten Anregung, mittels der die Ionen in der Ionenfalle gezielt zu Schwingungen angeregt werden können, sehr genau untersucht. Durch eine gezielte Kombination der unterschiedlichen Arten der resonanten Anregung ist es möglich MSn-Studien durchzuführen. Nach erfolgreicher Charakterisierung konnte in weiteren Laborstudien die MSn-Fähigkeit des AIMS demonstriert werden. Für Tryptophan (C11H12N2O2) wurde anhand von MS4-Studien ausgehend von m/z 130 ein möglicher Fragmentierungsweg identifiziert. Für die einzelnen Stufen der MS4-Studien wurden die Nachweisgrenzen abgeschätzt. Im Rahmen der PARADE (PArticles and RAdicals: Diel observations of the impact of urban and biogenic Emissions) Messkampagne im August/September 2011 auf dem kleinen Feldberg in der Nähe von Frankfurt am Main wurde die Feldtauglichkeit des AIMS demonstriert. Die Nachweisgrenzen liegen für eine Mittelungszeit von 60 Minuten für Organik bei 1,4 µg m-3, für Nitrat bei 0,5 µg m-3 und für Sulfat bei 0,7 µg m-3, was ausreichend ist um atmosphärisches Aerosol messen zu können. Dies ist ein signifikanter Fortschritt im Vergleich zum Prototypen, der aufgrund schlechter Reproduzierbarkeit und Robustheit noch nicht feldtauglich war. Im Vergleich zum HR-ToF-AMS, einem Standard-Aerosolmassenspektrometer, zeigte sich, dass beide Instrumente vergleichbare Trends für die Spezies Nitrat, Sulfat und Organik messen.

This thesis investigates metallic nanostructures exhibiting surface plasmon resonance for the amplification of fluorescence signal in sandwich immunoassays. In this approach, an analyte is captured by an antibody immobilized on a plasmonic structure and detected by a subsequently bound fluorophore labeled detection antibody. The highly confined field of surface plasmons originates from collective charge oscillations which are associated with high electromagnetic field enhancements at the metal surface and allow for greatly increased fluorescence signal from the attached fluorophores. This feature allows for improving the signal-to-noise ratio in fluorescence measurements and thus advancing the sensitivity of the sensor platform. In particular, the thesis presents two plasmonic nanostructures that amplify fluorescence signal in devices that rely on epifluorescence geometry, in which the fluorophore absorbs and emits light from the same direction perpendicular to the substrate surface.rnThe first is a crossed relief gold grating that supports propagating surface plasmon polaritons (SPPs) and second, gold nanoparticles embedded in refractive index symmetric environment exhibiting collective localized surface plasmons (cLSPs). Finite-difference time-domain simulations are performed in order to design structures for the optimum amplification of established Cy5 and Alexa Fluor 647 fluorophore labels with the absorption and emission wavelengths in the red region of spectrum. The design takes into account combined effect of surface plasmon-enhanced excitation rate, directional surface plasmon-driven emission and modified quantum yield for characteristic distances in immunoassays. Homebuilt optical instruments are developed for the experimental observation of the surface plasmon mode spectrum, measurements of the angular distribution of surface plasmon-coupled fluorescence light and a setup mimicking commercial fluorescence reading systems in epifluorescence geometry.rnCrossed relief grating structures are prepared by interference lithography and multiple copies are made by UV nanoimprint lithography. The fabricated crossed diffraction gratings were utilized for sandwich immunoassay-based detection of the clinically relevant inflammation marker interleukin 6 (IL-6). The enhancement factor of the crossed grating reached EF=100 when compared to a flat gold substrate. This result is comparable to the highest reported enhancements to date, for fluorophores with relatively high intrinsic quantum yield. The measured enhancement factor excellently agrees with the predictions of the simulations and the mechanisms of the enhancement are explained in detail. Main contributions were the high electric field intensity enhancement (30-fold increase) and the directional fluorescence emission at (4-fold increase) compared to a flat gold substrate.rnCollective localized surface plasmons (cLSPs) hold potential for even stronger fluorescence enhancement of EF=1000, due to higher electric field intensity confinement. cLSPs are established by diffractive coupling of the localized surface plasmon resonance (LSPR) of metallic nanoparticles and result in a narrow resonance. Due to the narrow resonance, it is hard to overlap the cLSPs mode with the absorption and emission bands of the used fluorophore, simultaneously. Therefore, a novel two resonance structure that supports SPP and cLSP modes was proposed. It consists of a 2D array of cylindrical gold nanoparticles above a low refractive index polymer and a silver film. A structure that supports the proposed SPP and cLSP modes was prepared by employing laser interference lithography and the measured mode spectrum was compared to simulation results.rn

The study was arranged to manifest its objectives through preceding it with an intro-duction. Particular attention was paid in the second part to detect the physical settings of the study area, together with an attempt to show the climatic characteristics in Libya. In the third part, observed temporal and spatial climate change in Libya was investigated through the trends of temperature, precipitation, relative humidity and cloud amount over the peri-ods (1946-2000), (1946-1975), and (1976-2000), comparing the results with the global scales. The forth part detected the natural and human causes of climate change concentrat-ing on the greenhouse effect. The potential impacts of climate change on Libya were ex-amined in the fifth chapter. As a case study, desertification of Jifara Plain was studied in the sixth part. In the seventh chapter, projections and mitigations of climate change and desertification were discussed. Ultimately, the main results and recommendations of the study were summarized. In order to carry through the objectives outlined above, the following methods and approaches were used: a simple linear regression analysis was computed to detect the trends of climatic parameters over time; a trend test based on a trend-to-noise-ratio was applied for detecting linear or non-linear trends; the non-parametric Mann-Kendall test for trend was used to reveal the behavior of the trends and their significance; PCA was applied to construct the all-Libya climatic parameters trends; aridity index after Walter-Lieth was shown for computing humid respectively arid months in Libya; correlation coefficient, (after Pearson) for detecting the teleconnection between sun spot numbers, NAOI, SOI, GHGs, and global warming, climate changes in Libya; aridity index, after De Martonne, to elaborate the trends of aridity in Jifara Plain; Geographical Information System and Re-mote Sensing techniques were applied to clarify the illustrations and to monitor desertifi-cation of Jifara Plain using the available satellite images MSS, TM, ETM+ and Shuttle Radar Topography Mission (SRTM). The results are explained by 88 tables, 96 figures and 10 photos. Temporal and spatial temperature changes in Libya indicated remarkably different an-nual and seasonal trends over the long observation period 1946-2000 and the short obser-vation periods 1946-1975 and 1976-2000. Trends of mean annual temperature were posi-tive at all study stations except at one from 1946-2000, negative trends prevailed at most stations from 1946-1975, while strongly positive trends were computed at all study stations from 1976-2000 corresponding with the global warming trend. Positive trends of mean minimum temperatures were observed at all reference stations from 1946-2000 and 1976-2000, while negative trends prevailed at most stations over the period 1946-1975. For mean maximum temperature, positive trends were shown from 1946-2000 and from 1976-2000 at most stations, while most trends were negative from 1946-1975. Minimum tem-peratures increased at nearly more than twice the rate of maximum temperatures at most stations. In respect of seasonal temperature, warming mostly occurred in summer and au-tumn in contrast to the global observations identifying warming mostly in winter and spring in both study periods. Precipitation across Libya is characterized by scanty and sporadically totals, as well as high intensities and very high spatial and temporal variabilities. From 1946-2000, large inter-annual and intra-annual variabilities were observed. Positive trends of annual precipi-tation totals have been observed from 1946-2000, negative trends from 1976-2000 at most stations. Variabilities of seasonal precipitation over Libya are more strikingly experienced from 1976-2000 than from 1951-1975 indicating a growing magnitude of climate change in more recent times. Negative trends of mean annual relative humidity were computed at eight stations, while positive trends prevailed at seven stations from 1946-2000. For the short observation period 1976-2000, positive trends were computed at most stations. Annual cloud amount totals decreased at most study stations in Libya over both long and short periods. Re-markably large spatial variations of climate changes were observed from north to south over Libya. Causes of climate change were discussed showing high correlation between tempera-ture increasing over Libya and CO2 emissions; weakly positive correlation between pre-cipitation and North Atlantic Oscillation index; negative correlation between temperature and sunspot numbers; negative correlation between precipitation over Libya and Southern Oscillation Index. The years 1992 and 1993 were shown as the coldest in the 1990s result-ing from the eruption of Mount Pinatubo, 1991. Libya is affected by climate change in many ways, in particular, crop production and food security, water resources, human health, population settlement and biodiversity. But the effects of climate change depend on its magnitude and the rate with which it occurs. Jifara Plain, located in northwestern Libya, has been seriously exposed to desertifica-tion as a result of climate change, landforms, overgrazing, over-cultivation and population growth. Soils have been degraded, vegetation cover disappeared and the groundwater wells were getting dry in many parts. The effect of desertification on Jifara Plain appears through reducing soil fertility and crop productivity, leading to long-term declines in agri-cultural yields, livestock yields, plant standing biomass, and plant biodiversity. Desertifi-cation has also significant implications on livestock industry and the national economy. Desertification accelerates migration from rural and nomadic areas to urban areas as the land cannot support the original inhabitants. In the absence of major shifts in policy, economic growth, energy prices, and con-sumer trends, climate change in Libya and desertification of Jifara Plain are expected to continue in the future. Libya cooperated with United Nations and other international organizations. It has signed and ratified a number of international and regional agreements which effectively established a policy framework for actions to mitigate climate change and combat deserti-fication. Libya has implemented several laws and legislative acts, with a number of ancil-lary and supplementary rules to regulate. Despite the current efforts and ongoing projects being undertaken in Libya in the field of climate change and desertification, urgent actions and projects are needed to mitigate climate change and combat desertification in the near future.

Lo scopo di questo lavoro è la caratterizzazione fisica del flat panel PaxScan4030CB Varian, rivelatore di raggi X impiegato in un ampio spettro di applicazioni cliniche, dalla radiografia generale alla radiologia interventistica. Nell’ambito clinico, al fine di una diagnosi accurata, è necessario avere una buona qualità dell’immagine radiologica mantenendo il più basso livello di dose rilasciata al paziente. Elemento fondamentale per ottenere questo risultato è la scelta del rivelatore di radiazione X, che deve garantire prestazioni fisiche (contrasto, risoluzione spaziale e rumore) adeguati alla specifica procedura. Le metriche oggettive che misurano queste caratteristiche sono SNR (Signal-to-Noise Ratio), MTF (Modulation Transfer Function) ed NPS (Noise Power Spectrum), che insieme contribuiscono alla misura della DQE (Detective Quantum Efficiency), il parametro più completo e adatto a stabilire le performance di un sistema di imaging. L’oggettività di queste misure consente anche di mettere a confronto tra loro diversi sistemi di rivelazione. La misura di questi parametri deve essere effettuata seguendo precisi protocolli di fisica medica, che sono stati applicati al rivelatore PaxScan4030CB presente nel laboratorio del Centro di Coordinamento di Fisica Medica, Policlinico S.Orsola. I risultati ottenuti, conformi a quelli dichiarati dal costruttore, sono stati confrontati con successo con alcuni lavori presenti in letteratura e costituiscono la base necessaria per la verifica di procedure di ottimizzazione dell’immagine radiologica attraverso interventi sul processo di emissione dei raggi X e sul trattamento informatico dell’immagine (Digital Subtraction Angiography).

The problem of localizing a scatterer, which represents a tumor, in a homogeneous circular domain, which represents a breast, is addressed. A breast imaging method based on microwaves is considered. The microwave imaging involves to several techniques for detecting, localizing and characterizing tumors in breast tissues. In all such methods an electromagnetic inverse scattering problem exists. For the scattering detection method, an algorithm based on a linear procedure solution, inspired by MUltiple SIgnal Classification algorithm (MUSIC) and Time Reversal method (TR), is implemented. The algorithm returns a reconstructed image of the investigation domain in which it is detected the scatterer position. This image is called pseudospectrum. A preliminary performance analysis of the algorithm vying the working frequency is performed: the resolution and the signal-to-noise ratio of the pseudospectra are improved if a multi-frequency approach is considered. The Geometrical Mean-MUSIC algorithm (GM- MUSIC) is proposed as multi-frequency method. The performance of the GMMUSIC is tested in different real life computer simulations. The performed analysis shows that the algorithm detects the scatterer until the electrical parameters of the breast are known. This is an evident limit, since, in a real life situation, the anatomy of the breast is unknown. An improvement in GM-MUSIC is proposed: the Eye-GMMUSIC algorithm. Eye-GMMUSIC algorithm needs no a priori information on the electrical parameters of the breast. It is an optimizing algorithm based on the pattern search algorithm: it searches the breast parameters which minimize the Signal-to-Clutter Mean Ratio (SCMR) in the signal. Finally, the GM-MUSIC and the Eye-GMMUSIC algorithms are tested on a microwave breast cancer detection system consisting of an dipole antenna, a Vector Network Analyzer and a novel breast phantom built at University of Bologna. The reconstruction of the experimental data confirm the GM-MUSIC ability to localize a scatterer in a homogeneous medium.

The level of improvement in the audiological results of Baha(®) users mainly depends on the patient's preoperative hearing thresholds and the type of Baha sound processor used. This investigation shows correlations between the preoperative hearing threshold and postoperative aided thresholds and audiological results in speech understanding in quiet of 84 Baha users with unilateral conductive hearing loss, bilateral conductive hearing loss and bilateral mixed hearing loss. Secondly, speech understanding in noise of 26 Baha users with different Baha sound processors (Compact, Divino, and BP100) is investigated. Linear regression between aided sound field thresholds and bone conduction (BC) thresholds of the better ear shows highest correlation coefficients and the steepest slope. Differences between better BC thresholds and aided sound field thresholds are smallest for mid-frequencies (1 and 2 kHz) and become larger at 0.5 and 4 kHz. For Baha users, the gain in speech recognition in quiet can be expected to lie in the order of magnitude of the gain in their hearing threshold. Compared to its predecessor sound processors Baha(®) Compact and Baha(®) Divino, Baha(®) BP100 improves speech understanding in noise significantly by +0.9 to +4.6 dB signal-to-noise ratio, depending on the setting and the use of directional microphone. For Baha users with unilateral and bilateral conductive hearing loss and bilateral mixed hearing loss, audiological results in aided sound field thresholds can be estimated with the better BC hearing threshold. The benefit in speech understanding in quiet can be expected to be similar to the gain in their sound field hearing threshold. The most recent technology of Baha sound processor improves speech understanding in noise by an order of magnitude that is well perceived by users and which can be very useful in everyday life.

Although magnetic resonance spectroscopy can be used as a unique tool to study molecular diffusion, it is rarely used to measure the diffusion properties of intramyocellular and extramyocellular lipids. Lipids have very low apparent diffusion coefficients (ADCs), which make these measurements difficult and necessitate strong diffusion gradients and long diffusion times. Consequence is that these measurements have inherently low signal-to-noise ratio and are prone to artifacts. The addition of physiological triggering and individual storage and processing of the spectra is seen to be a possible approach to maximize signal intensity and achieve high reproducibility of the experiments. Thus, the optimized measurement protocol was used to investigate the diffusion properties of lipids in human skeletal muscle in vivo. At a diffusion time of about 110 ms, intramyocellular lipids show a significantly lower ADC (2.0 × 10(-6) mm(2)/s, 95% confidence interval 1.10 × 10(-6) to 2.94 × 10(-6) mm(2)/s) than extramyocellular lipids (1.58 × 10(-5) mm(2)/s, 95% confidence interval 1.41 × 10(-5) to 1.75 × 10(-5) mm(2)/s). Because the chemical properties of both lipid pools can be assumed to be similar, the difference can only be attributed to restricted or severely hindered diffusion in the intramyocellular droplets.

With the increasing advances in hip joint preservation surgery, accurate diagnosis and assessment of femoral head and acetabular cartilage status is becoming increasingly important. Magnetic resonance imaging (MRI) of the hip does present technical difficulties. The fairly thin cartilage lining necessitates high image resolution and high contrast-to-noise ratio (CNR). With MR arthrography (MRA) using intraarticular injected gadolinium, labral tears and cartilage clefts may be better identified through the contrast medium filling into the clefts. However, the ability of MRA to detect varying grades of cartilage damage is fairly limited and early histological and biochemical changes in the beginning of osteoarthritis (OA) cannot be accurately delineated. Traditional MRI thus lacks the ability to analyze the biological status of cartilage degeneration. The technique of delayed gadolinium-enhanced MRI of cartilage (dGEMRIC) is sensitive to the charge density of cartilage contributed by glycosaminoglycans (GAGs), which are lost early in the process of OA. Therefore, the dGEMRIC technique has a potential to detect early cartilage damage that is obviously critical for decision-making regarding time and extent of intervention for joint-preservation. In the last decade, cartilage imaging with dGEMRIC has been established as an accurate and reliable tool for assessment of cartilage status in the knee and hip joint.This review outlines the current status of dGEMRIC for assessment of hip joint cartilage. Practical modifications of the standard technique including three-dimensional (3D) dGEMRIC and dGEMRIC after intra-articular gadolinium instead of iv-dGEMRIC will also be addressed.

A 20-channel phased-array coil for MRI of mice has been designed, constructed, and validated with bench measurements and high-resolution accelerated imaging. The technical challenges of designing a small, high density array have been overcome using individual small-diameter coil elements arranged on a cylinder in a hexagonal overlapping design with adjacent low impedance preamplifiers to further decouple the array elements. Signal-to-noise ratio (SNR) and noise amplification in accelerated imaging were simulated and quantitatively evaluated in phantoms and in vivo mouse images. Comparison between the 20-channel mouse array and a length-matched quadrature driven small animal birdcage coil showed an SNR increase at the periphery and in the center of the phantom of 3- and 1.3-fold, respectively. Comparison with a shorter but SNR-optimized birdcage coil (aspect ratio 1:1 and only half mouse coverage) showed an SNR gain of twofold at the edge of the phantom and similar SNR in the center. G-factor measurements indicate that the coil is well suited to acquire highly accelerated images.
