Ao calcular o diâmetro médio ponderado (DMP) ou o diâmetro médio geométrico (DMG) de uma amostra de solo, ponderam-se o diâmetro médio aritmético (no caso do DMP) ou o logarítmico (no caso do DMG) de cada classe pela massa de agregados nela retida. A média aritmética de cada classe, multiplicada pela massa de agregados retidos na respectiva classe, apenas é correta, quando ocorrer uma distribuição simétrica de massa de agregados dentro de cada classe, o que é muito pouco provável. O objetivo deste trabalho, desenvolvido no final de 1996 no Departamento de Solos da UFRGS, Porto Alegre, é propor um novo método para o cálculo do diâmetro médio dos agregados, presumindo ser a distribuição dentro de cada classe semelhante à distribuição dos agregados de toda a amostra. Essa proposta pressupõe ser o diâmetro médio da classe mais bem estimado por aquele diâmetro que divide os agregados contidos nela em duas partes de massa igual. Para isso foi apresentado um procedimento de cálculo, e o diâmetro médio assim encontrado foi denominado "diâmetro médio ajustado" (DMA). Para testar o método, utilizaram-se dados existentes de vários solos, condições de manejo do solo e rotações de culturas. Considerando os resultados, justifica-se a utilização do parâmetro DMA no lugar do DMP ou DMG, por representar mais corretamente o diâmetro médio dos agregados de uma amostra de solo.

Na coleta da solução de solos alagados, é importante evitar o contato do oxigênio com a solução para prevenir reações redox que podem modificar sua composição química. Neste trabalho, realizado em casa de vegetação, foram feitos estudos sobre o estado de oxirredução e sobre a composição química de um Planossolo, em diferentes profundidades e tempos de alagamento, visando propor um novo método para a coleta da solução de solos alagados. O estudo foi feito em vasos plásticos com 7 kg de material de solo proveniente da camada superficial (0-20 cm), num delineamento inteiramente casualizado, com três repetições. O dispositivo de coleta da solução do solo consistiu de um tubo de polietileno com 70 cm de comprimento e 0,6 cm de diâmetro interno, com duas fileiras de orifícios de 1 mm de diâmetro, recoberto com tela de nylon de 400 mesh, construído na forma de espiral e acomodado nas profundidades de 2, 5 e 10 cm. A este dispositivo foi acoplado um sifão de vidro para a coleta da solução. Após a instalação do sistema coletor, o solo foi mantido alagado por 109 dias, tendo sido feitas coletas semanais da solução nas três profundidades. Foram analisados os valores de Eh e pH e os teores de Mn2+, Fe2+, Ca2+, Mg2+ e K+ na solução. As condições de oxirredução e os teores de cátions na solução nas diferentes profundidades, durante o período em que o solo se manteve alagado, permitiram concluir que o método utilizado pode ser indicado para coleta da solução de solos alagados, embora haja necessidade de estudos mais aprofundados sobre o assunto.

ABSTRACT Pneumocystis jirovecii is a fungus that causes severe pneumonia in immunocompromised patients. However, its study is hindered by the lack of an in vitro culture method. We report here the genome of P. jirovecii that was obtained from a single bronchoalveolar lavage fluid specimen from a patient. The major challenge was the in silico sorting of the reads from a mixture representing the different organisms of the lung microbiome. This genome lacks virulence factors and most amino acid biosynthesis enzymes and presents reduced GC content and size. Together with epidemiological observations, these features suggest that P. jirovecii is an obligate parasite specialized in the colonization of human lungs, which causes disease only in immune-deficient individuals. This genome sequence will boost research on this deadly pathogen. IMPORTANCE Pneumocystis pneumonia is a major cause of mortality in patients with impaired immune systems. The availability of the P. jirovecii genome sequence allows new analyses to be performed which open avenues to solve critical issues for this deadly human disease. The most important ones are (i) identification of nutritional supplements for development of culture in vitro, which is still lacking 100 years after discovery of the pathogen; (ii) identification of new targets for development of new drugs, given the paucity of present treatments and emerging resistance; and (iii) identification of targets for development of vaccines.

High-fructose diet stimulates hepatic de novo lipogenesis (DNL) and causes hypertriglyceridemia and insulin resistance in rodents. Fructose-induced insulin resistance may be secondary to alterations of lipid metabolism. In contrast, fish oil supplementation decreases triglycerides and may improve insulin resistance. Therefore, we studied the effect of high-fructose diet and fish oil on DNL and VLDL triglycerides and their impact on insulin resistance. Seven normal men were studied on four occasions: after fish oil (7.2 g/day) for 28 days; a 6-day high-fructose diet (corresponding to an extra 25% of total calories); fish oil plus high-fructose diet; and control conditions. Following each condition, fasting fractional DNL and endogenous glucose production (EGP) were evaluated using [1-13C]sodium acetate and 6,6-2H2 glucose and a two-step hyperinsulinemic-euglycemic clamp was performed to assess insulin sensitivity. High-fructose diet significantly increased fasting glycemia (7 +/- 2%), triglycerides (79 +/- 22%), fractional DNL (sixfold), and EGP (14 +/- 3%, all P < 0.05). It also impaired insulin-induced suppression of adipose tissue lipolysis and EGP (P < 0.05) but had no effect on whole- body insulin-mediated glucose disposal. Fish oil significantly decreased triglycerides (37%, P < 0.05) after high-fructose diet compared with high-fructose diet without fish oil and tended to reduce DNL but had no other significant effect. In conclusion, high-fructose diet induced dyslipidemia and hepatic and adipose tissue insulin resistance. Fish oil reversed dyslipidemia but not insulin resistance.

OBJECTIVES: This study was designed to compare the long-term clinical outcome of coronary artery bypass grafting (CABG) with intracoronary stenting of patients with isolated proximal left anterior descending coronary artery. BACKGROUND: Although numerous trials have compared coronary angioplasty with bypass surgery, none assessed the clinical evaluation in the long term. METHODS: We evaluated the 10-year clinical outcome in the SIMA (Stent versus Internal Mammary Artery grafting) trial. Patients were randomly assigned to stent implantation versus CABG. RESULTS: Of 123 randomized patients, 59 underwent CABG and 62 received a stent (2 patients were excluded). Follow-up after 10 years was obtained for 98% of the randomized patients. Twenty-six patients (42%) in the percutaneous coronary intervention group and 10 patients (17%) in the CABG group reached an end point (p < 0.001). This difference was due to a higher need for additional revascularization. The incidences of death and myocardial infarction were identical at 10%. Progression of the disease requiring additional revascularization was rare (5%) and was similar for the 2 groups. Stent thrombosis occurred in 2 patients (3%). Angina functional class showed no significant differences between the 2 groups. CONCLUSIONS: Both stent implantation and CABG are safe and highly effective in relieving symptoms in patients with isolated, proximal left anterior descending coronary artery stenosis. Stenting with bare-metal stents is associated with a higher need for repeat interventions. The long-term prognosis for these patients is excellent with either mode of revascularization.

Similar to human chronic lymphocytic leukemia (CLL), the de novo New Zealand Black (NZB) mouse model has a genetically determined age-associated increase in malignant B-1 clones and decreased expression of microRNAs miR-15a and miR-16 in B-1 cells. In the present study, lentiviral vectors were employed in vivo to restore miR-15a/16, and both the short-term single injection and long-term multiple injection effects of this delivery were observed in NZB. Control lentivirus without the mir-15a/16 sequence was used for comparison. We found that in vivo lentiviral delivery of mir-15a/16 increased miR-15a/16 expression in cells that were transduced (detected by GFP expression) and in sera when compared with control lentivirus treatment. More importantly, mice treated with the miR-expressing lentivirus had decreased disease. The lentivirus had little systemic toxicity while preferentially targeting B-1 cells. Short-term effects on B-1 cells were direct effects, and only malignant B-1 cells transduced with miR-15a/16 lentivirus had decreased viability. In contrast, long-term studies suggested both direct and indirect effects resulting from miR-15a/16 lentivirus treatment. A decrease in B-1 cells was found in both the transduced and non-transduced populations. Our data support the potential use of systemic lentiviral delivery of miR-15a/16 to ameliorate disease manifestations of CLL.

Obesity results from chronic energy surplus and excess lipid storage in white adipose tissue (WAT). In contrast, brown adipose tissue (BAT) efficiently burns lipids through adaptive thermogenesis. Studying mouse models, we show that cyclooxygenase (COX)-2, a rate-limiting enzyme in prostaglandin (PG) synthesis, is a downstream effector of beta-adrenergic signaling in WAT and is required for the induction of BAT in WAT depots. PG shifted the differentiation of defined mesenchymal progenitors toward a brown adipocyte phenotype. Overexpression of COX-2 in WAT induced de novo BAT recruitment in WAT, increased systemic energy expenditure, and protected mice against high-fat diet-induced obesity. Thus, COX-2 appears integral to de novo BAT recruitment, which suggests that the PG pathway regulates systemic energy homeostasis.

The induction of fungal metabolites by fungal co-cultures grown on solid media was explored using multi-well co-cultures in 2 cm diameter Petri dishes. Fungi were grown in 12-well plates to easily and rapidly obtain the large number of replicates necessary for employing metabolomic approaches. Fungal culture using such a format accelerated the production of metabolites by several weeks compared with using the large-format 9 cm Petri dishes. This strategy was applied to a co-culture of a Fusarium and an Aspergillus strain. The metabolite composition of the cultures was assessed using ultra-high pressure liquid chromatography coupled to electrospray ionisation and time-of-flight mass spectrometry, followed by automated data mining. The de novo production of metabolites was dramatically increased by nutriment reduction. A time-series study of the induction of the fungal metabolites of interest over nine days revealed that they exhibited various induction patterns. The concentrations of most of the de novo induced metabolites increased over time. However, interesting patterns were observed, such as with the presence of some compounds only at certain time points. This result indicates the complexity and dynamic nature of fungal metabolism. The large-scale production of the compounds of interest was verified by co-culture in 15 cm Petri dishes; most of the induced metabolites of interest (16/18) were found to be produced as effectively as on a small scale, although not in the same time frames. Large-scale production is a practical solution for the future production, identification and biological evaluation of these metabolites.

O semivariograma possibilita uma avaliação visual da dependência espacial, mas sem resultar diretamente em um valor numérico único que expresse a mensuração de tal dependência. Contudo, a partir dos parâmetros estimados do modelo teórico ajustado ao semivariograma experimental, é possível construir uma medida dessa dependência espacial. Atualmente, há dois índices na literatura, com uso cada vez mais frequente; porém, há inadequações nesses índices existentes. O objetivo deste trabalho foi propor um novo índice para medir a dependência espacial de dados geoestatísticos, que supere as incipiências dos atuais. Esse novo índice utiliza a relação existente entre o semivariograma e o correlograma, contemplando dessa forma todos os aspectos da dependência espacial. Realizaram-se uma comparação, por simulação, entre o índice proposto e os índices já existentes e também verificação da aplicabilidade do índice proposto utilizando pesquisas reais publicadas, em que ocorreram ajustes dos modelos teóricos esférico, exponencial e gaussiano. Verificou-se que o índice proposto foi melhor que os índices existentes. Além disso, observou-se que os índices existentes podem levar a equívocos nas interpretações do grau de dependência espacial, evidenciando que devem ser evitados. Em decorrência, recomenda-se a utilização do novo índice proposto para medir o grau da dependência espacial.
