Low temperature (77K) linear dichroism spectroscopy was used to characterize pigment orientation changes accompanying the light state transition in the cyanobacterium, Synechococcus sp. pee 6301, and cold-hardening in winter rye (Secale cereale L. cv. Puma). Samples were oriented for spectroscopy using the gel squeezing method (Abdourakhmanov et aI., 1979) and brought to 77K in liquid nitrogen. The linear dichroism (LD) spectra of Synechococcus 6301 phycobilisome/thylakoid membrane fragments cross-linked in light state 1 and light state 2 with glutaraldehyde showed differences in both chlorophyll a and phycobilin orientation. A decrease in the relative amplitude of the 681nm chlorophyll a positive LD peak was observed in membrane fragments in state 2. Reorientation of the phycobilisome (PBS) during the transition to state 2 resulted in an increase in core allophycocyanin absorption parallel to the membrane, and a decrease in rod phycocyanin parallel absorption. This result supports the "spillover" and "PBS detachment" models of the light state transition in PBS-containing organisms, but not the "mobile PBS" model. A model was proposed for PBS reorientation upon transition to state 2, consisting of a tilt in the antenna complex with respect to the membrane plane. Linear dichroism spectra of PBS/thylakoid fragments from the red alga, Porphyridium cruentum, grown in green light (containing relatively more PSI) and red light (containing relatively more PSll) were compared to identify chlorophyll a absorption bands associated with each photosystem. Spectra from red light - grown samples had a larger positive LD signal on the short wavelength side of the 686nm chlorophyll a peak than those from green light - grown fragments. These results support the identification of the difference in linear dichroism seen at 681nm in Synechococcus spectra as a reorientation of PSll chromophores. Linear dichroism spectra were taken of thylakoid membranes isolated from winter rye grown at 20°C (non-hardened) and 5°C (cold-hardened). Differences were seen in the orientation of chlorophyll b relative to chlorophyll a. An increase in parallel absorption was identified at the long-wavelength chlorophyll a absorption peak, along with a decrease in parallel absorption from chlorophyll b chromophores. The same changes in relative pigment orientation were seen in the LD of isolated hardened and non-hardened light-harvesting antenna complexes (LHCII). It was concluded that orientational differences in LHCII pigments were responsible for thylakoid LD differences. Changes in pigment orientation, along with differences observed in long-wavelength absorption and in the overall magnitude of LD in hardened and non-hardened complexes, could be explained by the higher LHCII monomer:oligomer ratio in hardened rye (Huner et ai., 1987) if differences in this ratio affect differential light scattering properties, or fluctuation of chromophore orientation in the isolated LHCII sample.

Les effets de la marijuana, un médicament utilisé par l’homme depuis des millénaires, sur le système visuel sont peu connus. Une meilleure connaissance de la distribution du système endocannabinoïde (eCB) de la rétine pourrait expliquer comment cette drogue affecte la vision. Cette étude vise à caractériser la distribution du récepteur cannabinoïde CB1 (CB1R) et de l’enzyme de dégradation FAAH (“fatty acid amide hydrolase”) des ligands du CB1R dans la rétine du singe Vert (Chlorocebus sabaeus). De plus, elle vise à déterminer quelles sous-populations cellulaires de la rétine expriment ces composantes. La plupart des études à ce jour ont été conduites surtout sur les rongeurs et peu de travaux ont été réalisés chez le singe. Notre étude vient donc combler cette carence. Par le biais de méthodes immunohistochimiques, nous avons investigué la localisation du CB1R et de l’enzyme FAAH à différentes excentricités rétiniennes, de la fovéa centralis vers la périphérie. Nos résultats, en accord avec notre hypothèse de travail, démontrent que CB1R et FAAH sont exprimés à travers toute la rétine mais avec, cependant, des différences notoires. Au niveau de la couche des photorécepteurs, CB1R est exprimé préférentiellement dans les cônes et ce patron d’expression suit la distribution des photorécepteurs centre-périphérie. De plus, CB1R se retrouve surtout dans les pédicules des cônes de la couche plexiforme externe. CB1R et FAAH sont abondants dans les cellules bipolaires tant au centre qu’en périphérie. Le soma et l’axone des cellules ganglionnaires expriment aussi CB1R et FAAH. Ces données suggèrent que le système eCB est présent à travers toute la rétine du primate et pourrait expliquer les perturbations visuelles entrainées par la marijuana, telles la photosensibilité et la vision des couleurs.

Résumé La recherche présentée ici porte sur la manière dont les protestants conservateurs francophones du Québec évaluent la compatibilité entre leurs croyances religieuses et les lois qui limitent le recours au châtiment corporel à l’égard des enfants. Plus précisément, elle s’intéresse à la façon dont ils résolvent les conflits éventuels entre leurs croyances puisées dans la Bible et ces lois. En ce sens, la Bible prescrit dans plusieurs de ses versets, notamment dans le proverbe 22 :15, d’utiliser le châtiment corporel comme moyen pour chasser une inclination au mal qui serait innée chez les enfants et d’effectuer ce châtiment à l’aide d’une verge. De ce fait, de nombreux protestants conservateurs emploient des objets (cuillers en bois, bâtons, baguettes) pour administrer ce châtiment à leurs enfants. Or, ces pratiques entrent en contradiction avec l’article 43 du Code criminel du Canada qui limite et encadre le recours au châtiment corporel et avec la Loi sur la protection de la jeunesse du Québec qui protège les enfants contre des traitements pouvant s’apparenter à de la maltraitance et qui risquent de compromettre leur développement. La méthodologie utilisée est une méthodologie qualitative mixte basée d’abord sur une série d’observations non participantes in situ à des services religieux et des ateliers d’enseignement doctrinal dans quatre congrégations protestantes conservatrices (deux Églises évangéliques, une Église pentecôtiste et une Église baptiste) suivie d’une série d’entretiens auprès de trente-neuf protestants conservateurs québécois francophones appartenant à ces congrégations. Ce matériel a été complété par une analyse documentaire des écrits produits par ces groupes et des écrits d’autres organisations conservatrices consultés par ces groupes. L’analyse des données a permis de dégager chez les protestants conservateurs à l’étude trois différentes attitudes face à l’incompatibilité entre leurs préceptes religieux et les lois séculières : une attitude de conciliation qui se traduit par un effort d’accommodement de ces préceptes à la loi ; une attitude d’omission face à la loi séculière où l’individu opte pour une désobéissance passive de la loi ; et une attitude contestataire face aux autorités où la désobéissance aux lois est envisagée comme une forme de militance. Nous examinons les éléments qui influencent ces différents positionnements face aux lois. En plus de répondre aux objectifs visés, la présente recherche constitue une étude approfondie du discours des protestants conservateurs québécois francophones sur le châtiment corporel des enfants et de leurs pratiques.

Selenoproteins are proteins containing selenium in the form of the 21st amino acid, selenocysteine. Selenocysteine (Sec) is directly synthesized onto its cognate tRNA (tRNA[Ser]Sec or tRNASec) and inserted into selenoproteins co-translationally with the help of various cis- and trans-acting factors. Among those factors, SecP43 has been reported to possibly play an essential role in the methylation at the 2’-hydroxylribosyl moiety in the wobble position (Um34) of Sec-tRNA[Ser]Sec and consequently reduce the expression of glutathione peroxidase 1. SecP43 also called tRNASec-associated protein has also been reported to interact in with SepSecS and tRNASec in vivo and the targeted removal of one of these proteins affected the binding of the other to the Sec-tRNASec. The initial aim of the project was to solve the structure of SecP43 by means of x-ray crystallography. Secondly, we were interested in characterizing the interaction of the latter with some of the components of the selenocysteine insertion machinery. These factors are SepSecS and tRNASec. We were able to optimize the expression and the purification of soluble form of the human homologue of SecP43 and of SepSecS by using an adapted auto-induction protocol. This was a major challenge considering that full length SecP43 has not been expressed and purify to date. We did not succeed in crystallizing SecP43. Our failure to crystallize SecP43 is probably due to the fact that it is a partially folded protein as we were able to demonstrate by SAXS (Small Angle X-ray Scattering). The SecP43 envelope calculated by SAXS displayed a rod-shape like structure. In order to enhance the stability of SecP43 required for crystallization, binding affinity studies were conducted to characterize the interaction between SecP43, tRNASec and SepSecS. We did not detect an interaction between SecP43 and tRNASec by using EMSA (Electrophoretic Mobility Shift Assay) and gel filtration. We also could not detect an interaction between SecP43 and SepSecS using a cross-linking assay. In contrast, the tRNASec/SepSecS interaction was demonstrated by EMSA and the addition of SecP43 seemed to reduce the binding affinity. Therefore, SecP43 might induce a conformational change in SepSecS in the presence of tRNASec.

Study Design Retrospective study of surgical outcome. Objectives To evaluate quantitatively the changes in trunk surface deformities after scoliosis spinal surgery in Lenke 1A adolescent idiopathic scoliosis (AIS) patients and to compare it with changes in spinal measurements. Summary of Background Data Most studies documenting scoliosis surgical outcome used either radiographs to evaluate changes in the spinal curve or questionnaires to assess patients health-related quality of life. Because improving trunk appearance is a major reason for patients and their parents to seek treatment, this study focuses on postoperative changes in trunk surface deformities. Recently, a novel approach to quantify trunk deformities in a reliable, automatic, and noninvasive way has been proposed. Methods Forty-nine adolescents with Lenke 1A idiopathic scoliosis treated surgically were included. The back surface rotation and trunk lateral shift were computed on trunk surface acquisitions before and at least 6 months after surgery. We analyzed the effect of age, height, weight, curve severity, and flexibility before surgery, length of follow-up, and the surgical technique. For 25 patients with available three-dimensional (3D) spinal reconstructions, we compared changes in trunk deformities with changes in two-dimensional (2D) and 3D spinal measurements. Results The mean correction rates for the back surface rotation and the trunk lateral shift are 18% and 50%, respectively. Only the surgical technique had a significant effect on the correction rate of the back surface rotation. Direct vertebral derotation and reduction by spine translation provide a better correction of the rib hump (22% and 31% respectively) than the classic rod rotation technique (8%). The reductions of the lumbar Cobb angle and the apical vertebrae transverse rotation explain, respectively, up to 17% and 16% the reduction of the back surface rotation. Conclusions Current surgical techniques perform well in realigning the trunk; however, the correction of the deformity in the transverse plane proves to be more challenging. More analysis on the positive effect of vertebral derotation on the rib hump correction is needed. Level of evidence III.

This 'study' deals with a preliminary study of automatic beam steering properly in conducting polyaniline . Polyaniline in its undoped and doped .state was prepared from aniline by the chemical oxidative polymerization method. Dielectric properties of the samples were studied at S-band microwave frequencies using cavity perturbation technique. It is found that undoped po/vanihne is having greater dielectric loss and conductivity contpared with the doped samples. The beam steering property is studied using a perspex rod antenna and HP 85/OC vector network analyzer. The shift in the radiated beam is studied for different do voltages. The results show that polyaniline is a good nutterial far beam steering applications.

A new approach, the multipole theory (MT) method, is presented for the computation of cutoff wavenumbers of waveguides partially filled with dielectric. The MT formulation of the eigenvalue problem of an inhomogeneous waveguide is derived. Representative computational examples, including dielectric-rod-loaded rectangular and double-ridged waveguides, are given to validate the theory, and to demonstrate the degree of its efficiency

Preparation of an appropriate optical-fiber preform is vital for the fabrication of graded-index polymer optical fibers (GIPOF), which are considered to be a good choice for providing inexpensive high bandwidth data links, for local area networks and telecommunication applications. Recent development of the interfacial gel polymerization technique has caused a dramatic reduction in the total attenuation in GIPOF, and this is one of the potential methods to prepare fiber preforms for the fabrication of dye-doped polymer-fiber amplifiers. In this paper, the preparation of a dye-doped graded-index poly(methyl methacrylate) (PMMA) rod by the interfacial gel polymerization method using a PMMA tube is reported. An organic compound of high-refractive index, viz., diphenyl phthalate (DPP), was used to obtain a graded-index distribution, and Rhodamine B (Rh B), was used to dope the PMMA rod. The refractive index profile of the rod was measured using an interferometric technique and the index exponent was estimated. The single pass gain of the rod was measured at a pump wavelength of 532 nm. The extent of doping of the Rh B in the preform was studied by axially exciting a thin slice of the rod with white light and measuring the spatial variation of the fluorescence intensity across the sample.

The thesis presents the results of the studies carried out on certain diseases encountered in the larvae and postlarvae of penaeid prawns raised in the hatcheries at Cochin, Madras and Mandapam Camp during September 1985- April 1988. In the preliminary survey carried out to understand the common diseases occurring in the penaeid larvae and postlarvae, seven cases of diseases and abnormalities were encountered. These included ciliate infestation, Nit_zschia closteriurn infestation, parasitic protozoaninfection, parasitic dinoflagellate infection, appendage necrosis, heteromorphic eye and abnormal eggs and deformed nauplii .The clinical signs, seasonal occurrence and incidence of each of the above cases were provided along with the information on environmental factors such as salinity, dissolved oxygen, temperature and pH of the rearing medium. The Thésis is presented in nine chapters. Chapter 1 surveys the literature on the diseases of penaeid larvae, postlarvae and adult prawns from India and abroad. This is followed by a chapter on the material and methods employed during the present investigation. In the third chapter, seven cases of diseases and abnormalities encountered in the larvae and postlarvae of Penaeus indicus and p. semisulcatus during the survey carried out in the hatcheries located at different centres of Central‘ Marine Fisheries Research Institute are presented and discussed .A bacterium responsible for appendage necrosis was isolated and its taxonomy was studied. It was Gram-negative, fermentative and motile rod. It was sensitive to vibriostatic compound, 0/129. This bacterium was found to be a new isolate of vibrio on the basis of its morphological, biological, physiological and biochemical characters and comparison of these characters with those described for other related vibrios. This new isolate of vibrio was deposited in vibro Referrence Laboratory, Centres for Disease Control, Georgia, U.S.A. and coded as vibrio sp. 2448-88.

Investigations on the design and development of certain new hollow dielectric hom antennas of rectangular cross section have been carried out. The main shortcoming of the existing ordinary hollow dielectric hom antenna (HDH) is the abrupt discontinuity at the feed-end. A new launching technique using a dielectric rod is introduced to overcome this limitation. Also a strip loading technique is employed for further modification of the antenna. Radiation parameters of new I-IDH antennas of Eplane sectoral, H-plane sectoral and pyramidal types were studied and are found to be very attractive. Theoretical approach based on Marcatili’s principle and two aperture theory along with diffraction theory and image theory is used to support the experimental findings. The HDH is considered as solid horn of effective dielectric constant and the aperture field is evaluated. The antenna is excited by the open waveguide in the dominant TE1o mode and so the existence of any hybrid mode is mled-out. The theoretical results are observed to be in good agreement with the experimental results.

The application vistas of superconductors have widened very much since the discovery of high TC superconductors (HTS) as many of the applications can be realised at 77 K rather than going down to 4.2 K, the liquid He temperature. One such application is the HTS current lead which is used to connect a superconducting system with a room temperature power source. Minimising heat leak to the cryogenic environment is the main advantage of introducing current leads into superconducting systems. The properties of HTSS likes zero resistance (avoiding joule heating) and very low thermal conductivity (minimized conductive heat transfer) make them ideal candidates to be used as current leads. There are two forms of HTS current leads. (i) bulk form (tube or rod) prepared either from YBCO or BSCCO and (ii) tape form prepared from Bi-2223 multifilamentary tapes. The tape form of current leads has many advantages with respect to the mechanical and thermal stability related criteria. Crucial information on various aspects of HTS current lead development are not available in the literature as those are kept proprietary by various companies around the world. The present work has been undertaken to tailor the properties of multifilamentary tapes for the current lead application and to optimise the processing parameters of the same for enhanced critical current density and field tolerance. Also it is the aim of the present investigation is to prepare prototype current leads engineered for operation in conduction cooled mode and test them for operational stability

Mathematical models are often used to describe physical realities. However, the physical realities are imprecise while the mathematical concepts are required to be precise and perfect. The 1st chapter give a brief summary of the arithmetic of fuzzy real numbers and the fuzzy normed algebra M(I). Also we explain a few preliminary definitions and results required in the later chapters. Fuzzy real numbers are introduced by Hutton,B [HU] and Rodabaugh, S.E[ROD]. Our definition slightly differs from this with an additional minor restriction. The definition of Clementina Felbin [CL1] is entirely different. The notations of [HU]and [M;Y] are retained inspite of the slight difference in the concept.the 3rd chapter In this chapter using the completion M'(I) of M(I) we give a fuzzy extension of real Hahn-Banch theorem. Some consequences of this extension are obtained. The idea of real fuzzy linear functional on fuzzy normed linear space is introduced. Some of its properties are studied. In the complex case we get only a slightly weaker analogue for the Hahn-Banch theorem, than the one [B;N] in the crisp case
