The antenna presented in this article will be developed for satellite communications onboard systems based on the recommendations ITU-R S.580-6 and ITU-R S.465-5. The antenna consists of printed elements grouped in an array, this terminal works in a frequency band from 7.25 up to 8.4 GHz (14.7% of bandwidth), where both bands, reception (7.25 - 7.75 GHz) and transmission (7.9 - 8.4 GHz), are included simultaneously. The antenna reaches a gain about 31 dBi, and it has a radiation pattern with a beamwidth smaller than 10° and a dual circular polarization. The antenna has the capability to steer in elevation from 90° to 40° electronically and 360° in azimuth with a motorized junction.

A new and effective method for reduction of truncation errors in partial spherical near-field (SNF) measurements is proposed. The method is useful when measuring electrically large antennas, where the measurement time with the classical SNF technique is prohibitively long and an acquisition over the whole spherical surface is not practical. Therefore, to reduce the data acquisition time, partial sphere measurement is usually made, taking samples over a portion of the spherical surface in the direction of the main beam. But in this case, the radiation pattern is not known outside the measured angular sector as well as a truncation error is present in the calculated far-field pattern within this sector. The method is based on the Gerchberg-Papoulis algorithm used to extrapolate functions and it is able to extend the valid region of the calculated far-field pattern up to the whole forward hemisphere. To verify the effectiveness of the method, several examples are presented using both simulated and measured truncated near-field data.

This paper investigates the propagation of airblast from quarry blasting. Peak overpressure is calculated as a function of blasting parameters (explosive mass per delay and velocity at which the detonation sequence proceeds along the bench) and polar coordinates of the point of interest (distance to the blast and azimuth with respect to the free face of the blast). The model is in the form of the product of a classical scaled distance attenuation law times a directional correction factor. The latter considers the influence of the bench face, and attenuates overpressure at the top level and amplifies it at the bottom. Such factor also accounts for the effect of the delay by amplifying the pressure in the direction of the initiation sequence if the velocity of initiation exceeds half the speed of sound and up to an initiation velocity in the range of the speed of sound. The model has been fitted to an empirical data set composed by 134 airblast records monitored in 47 blasts at two quarries. The measurements were made at distances to the blast less than 450 m. The model is statistically significant and has a determination coefficient of 0.869

In this work, a dual circular polarized steering antenna for satellite communications in X band is presented. The antenna consists of printed elements grouped in an array. This terminal works in a frequency band from 7.25 GHz up to 8.4 GHz (15% of bandwidth), where both bands, reception (RX) and transmission (TX) are included simultaneously and Left Handed Circular Polarization (LHCP) and Right Handed Circular Polarization (RHCP) are interchangeable. The antenna is compact, narrow bandwidth and reaches a gain of 16 dBi. It has the capability to steer in elevation to 45±, 75±, 105± and 135± electronically with a Butler matrix and 360± in azimuth with a motorized junction.

Digital image correlation (DIC) is applied to analyzing the deformation mechanisms under transverse compression in a fiber-reinforced composite. To this end, compression tests in a direction perpendicular to the fibers were carried out inside a scanning electron microscope and secondary electron images obtained at different magnifications during the test. Optimum DIC parameters to resolve the displacement and strain field were computed from numerical simulations of a model composite and they were applied to micrographs obtained at different magnifications (250_, 2000_, and 6000_). It is shown that DIC of low-magnification micrographs was able to capture the long range fluctuations in strain due to the presence of matrix-rich and fiber-rich zones, responsible for the onset of damage. At higher magnification, the strain fields obtained with DIC qualitatively reproduce the non-homogeneous deformation pattern due to the presence of stiff fibers dispersed in a compliant matrix and provide accurate results of the average composite strain. However, comparison with finite element simulations revealed that DIC was not able to accurately capture the average strain in each phase.

Transverse galloping is a type of aeroelastic instability characterised by large amplitude, low frequency oscillation of a structure in the direction normal to the mean wind direction. It normally appears in bodies with small stiffness and structural damping, provided the incident flow velocity is high enough. In the simplest approach transverse galloping can be considered as a one-degree-of-freedom oscillator subjected to aerodynamic forces, which in turn can be described by using a quasi-steady description. In this frame it has been demonstrated that hysteresis phenomena in transverse galloping is related to the existence of inflection points in the curve giving the dependence with the angle of attack of the aerodynamic coefficient normal to the incident flow. Aiming at experimentally checking such a relationship between these inflection points and hysteresis, wind tunnel experiments have been conducted. Experiments have been restricted to isosceles triangular cross-section bodies, whose galloping behaviour is well documented. Experimental results show that, according to theoretical predictions, hysteresis takes place at the angles of attack where there are inflection points in the lift coefficient curve, provided that the body is prone to gallop at these angles of attack.

A number of short-to-mid height RC buildings with wide beams have been constructed in moderate-seismicity areas of Spain. The seismic behavior in the direction of the wide beams appears to be deficient because of low lateral strength, low ductility of the wide beams, big strut compressive forces inside the column-beam connections, and unreliable contribution of the spandrel zones of the wide beams. In the orthogonal direction, the behavior is worse since only the joists and the façade beams contribute to the lateral resistance. The objective is to assess the seismic capability of these structures; further research will involve proposing retrofit strategies. The research approach consists of selecting a number of representative buildings and evaluating their vulnerability by code-type, push-over and dynamic analyses. The cooperation of the masonry infill walls is accounted for. The main conclusion is that the seismic behavior of these buildings is inadequate in most of the situations.

Nowadays one of the challenges of materials science is to find new technologies that will be able to make the most of renewable energies. An example of new proposals in this field are the intermediate-band (IB) materials, which promise higher efficiencies in photovoltaic applications (through the intermediate band solar cells), or in heterogeneous photocatalysis (using nanoparticles of them, for the light-induced degradation of pollutants or for the efficient photoevolution of hydrogen from water). An IB material consists in a semiconductor in which gap a new level is introduced [1], the intermediate band (IB), which should be partially filled by electrons and completely separated of the valence band (VB) and of the conduction band (CB). This scheme (figure 1) allows an electron from the VB to be promoted to the IB, and from the latter to the CB, upon absorption of photons with energy below the band gap Eg, so that energy can be absorbed in a wider range of the solar spectrum and a higher current can be obtained without sacrificing the photovoltage (or the chemical driving force) corresponding to the full bandgap Eg, thus increasing the overall efficiency. This concept, applied to photocatalysis, would allow using photons of a wider visible range while keeping the same redox capacity. It is important to note that this concept differs from the classic photocatalyst doping principle, which essentially tries just to decrease the bandgap. This new type of materials would keep the full bandgap potential but would use also lower energy photons. In our group several IB materials have been proposed, mainly for the photovoltaic application, based on extensively doping known semiconductors with transition metals [2], examining with DFT calculations their electronic structures. Here we refer to In2S3 and SnS2, which contain octahedral cations; when doped with Ti or V an IB is formed according to quantum calculations (see e.g. figure 2). We have used a solvotermal synthesis method to prepare in nanocrystalline form the In2S3 thiospinel and the layered compound SnS2 (which when undoped have bandgaps of 2.0 and 2.2 eV respectively) where the cation is substituted by vanadium at a ?10% level. This substitution has been studied, characterizing the materials by different physical and chemical techniques (TXRF, XRD, HR-TEM/EDS) (see e.g. figure 3) and verifying with UV spectrometry that this substitution introduces in the spectrum the sub-bandgap features predicted by the calculations (figure 4). For both sulphide type nanoparticles (doped and undoped) the photocatalytic activity was studied by following at room temperature the oxidation of formic acid in aqueous suspension, a simple reaction which is easily monitored by UV-Vis spectroscopy. The spectral response of the process is measured using a collection of band pass filters that allow only some wavelengths into the reaction system. Thanks to this method the spectral range in which the materials are active in the photodecomposition (which coincides with the band gap for the undoped samples) can be checked, proving that for the vanadium substituted samples this range is increased, making possible to cover all the visible light range. Furthermore it is checked that these new materials are more photocorrosion resistant than the toxic CdS witch is a well know compound frequently used in tests of visible light photocatalysis. These materials are thus promising not only for degradation of pollutants (or for photovoltaic cells) but also for efficient photoevolution of hydrogen from water; work in this direction is now being pursued.

Leonhardt demonstrated (2009) that the 2D Maxwell Fish Eye lens (MFE) can focus perfectly 2D Helmholtz waves of arbitrary frequency, i.e., it can transport perfectly an outward (monopole) 2D Helmholtz wave field, generated by a point source, towards a "perfect point drain" located at the corresponding image point. Moreover, a prototype with λ/5 superresolution (SR) property for one microwave frequency has been manufactured and tested (Ma et al, 2010). Although this prototype has been loaded with an impedance different from the "perfect point drain", it has shown super-resolution property. However, neither software simulations nor experimental measurements for a broad band of frequencies have yet been reported. Here we present steady state simulations for two cases, using perfect drain as suggested by Leonhardt and without perfect drain as in the prototype. All the simulations have been done using a device equivalent to the MFE, called the Spherical Geodesic Waveguide (SGW). The results show the super-resolution up to λ/3000, for the system loaded with the perfect drain, and up to λ/500 for a not perfect load. In both cases super-resolution only happens for discrete number of frequencies. Out of these frequencies, the SGW does not show super-resolution in the analysis carried out.

A significant number of short-to-mid height RC buildings with wide beams have been constructed in areas of moderate seismicity of Spain, mainly for housing and administrative use. The buildings have a framed structure with one-way slabs; the wide beams constitute the distinctive characteristic, their depth being equal to that of the rest of the slab, thus providing a flat lower surface, convenient for construction and the layout of facilities. Seismic behavior in the direction of the wide beams appears to be deficient because of: (i) low lateral strength, mainly because of the small effective depth of the beams, (ii) inherent low ductility of the wide beams, generated by high amount of reinforcement, (iii) the big strut compressive forces developed inside the column-beam connections due to the low height of the beams, and (iv) the fact that the wide beams are wider than the columns, meaning that the contribution of the outer zones to the resistance of the beam-column joints is unreliable because there is no torsion reinforcement. In the orthogonal direction, the behavior is worse since the only members of the slabs that contribute to the lateral resistance are the joists and the façade beams. Moreover, these buildings were designed with codes that did not include ductility requirements and required only a low lateral resistance; indeed, in many cases, seismic action was not considered at all. Consequently, the seismic capacity of these structures is not reliable. The objective of this research is to assess numerically this capability, whereas further research will aim to propose retrofit strategies. The research approach consists of: (i) selecting a number of 3-story and 6-story buildings that represent the vast majority of the existing ones and (ii) evaluating their vulnerability through three types of analyses, namely: code-type, push-over and nonlinear dynamic analysis. Given the low lateral resistance of the main frames, the cooperation of the masonry infill walls is accounted for; for each representative building, three wall densities are considered. The results of the analyses show that the buildings in question exhibit inadequate seismic behavior in most of the examined situations. In general, the relative performance is less deficient for Target Drift CP (Collapse Prevention) than for IO (Immediate Occupancy). Since these buildings are selected to be representative of the vast majority of buildings with wide beams that were constructed in Spain without accounting for any seismic consideration, our conclusions can be extrapolated to a broader scenario.

Foliage Penetration (FOPEN) radar systems were introduced in 1960, and have been constantly improved by several organizations since that time. The use of Synthetic Aperture Radar (SAR) approaches for this application has important advantages, due to the need for high resolution in two dimensions. The design of this type of systems, however, includes some complications that are not present in standard SAR systems. FOPEN SAR systems need to operate with a low central frequency (VHF or UHF bands) in order to be able to penetrate the foliage. High bandwidth is also required to obtain high resolution. Due to the low central frequency, large integration angles are required during SAR image formation, and therefore the Range Migration Algorithm (RMA) is used. This project thesis identifies the three main complications that arise due to these requirements. First, a high fractional bandwidth makes narrowband propagation models no longer valid. Second, the VHF and UHF bands are used by many communications systems. The transmitted signal spectrum needs to be notched to avoid interfering them. Third, those communications systems cause Radio Frequency Interference (RFI) on the received signal. The thesis carries out a thorough analysis of the three problems, their degrading effects and possible solutions to compensate them. The UWB model is applied to the SAR signal, and the degradation induced by it is derived. The result is tested through simulation of both a single pulse stretch processor and the complete RMA image formation. Both methods show that the degradation is negligible, and therefore the UWB propagation effect does not need compensation. A technique is derived to design a notched transmitted signal. Then, its effect on the SAR image formation is evaluated analytically. It is shown that the stretch processor introduces a processing gain that reduces the degrading effects of the notches. The remaining degrading effect after processing gain is assessed through simulation, and an experimental graph of degradation as a function of percentage of nulled frequencies is obtained. The RFI is characterized and its effect on the SAR processor is derived. Once again, a processing gain is found to be introduced by the receiver. As the RFI power can be much higher than that of the desired signal, an algorithm is proposed to remove the RFI from the received signal before RMA processing. This algorithm is a modification of the Chirp Least Squares Algorithm (CLSA) explained in [4], which adapts it to deramped signals. The algorithm is derived analytically and then its performance is evaluated through simulation, showing that it is effective in removing the RFI and reducing the degradation caused by both RFI and notching. Finally, conclusions are drawn as to the importance of each one of the problems in SAR system design.

El primer procesamiento estricto realizado con el software científico Bernese y contemplando las más estrictas normas de cálculo recomendadas internacionalmente, permitió obtener un campo puntual de alta exactitud, basado en la integración y estandarización de los datos de una red GPS ubicada en Costa Rica. Este procesamiento contempló un total de 119 semanas de datos diarios, es decir unos 2,3 años, desde enero del año 2009 hasta abril del año 2011, para un total de 30 estaciones GPS, de las cuales 22 están ubicadas en el territorio nacional de Costa Rica y 8 internaciones pertenecientes a la red del Sistema Geocéntrico para las Américas (SIRGAS). Las denominadas soluciones semilibres generaron, semana a semana, una red GPS con una alta exactitud interna definida por medio de los vectores entre las estaciones y las coordenadas finales de la constelación satelital. La evaluación semanal dada por la repetibilidad de las soluciones brindó en promedio errores de 1,7 mm, 1,4 mm y 5,1 mm en las componentes [n e u], confirmando una alta consistencia en estas soluciones. Aunque las soluciones semilibres poseen una alta exactitud interna, las mismas no son utilizables para fines de análisis cinemático, pues carecen de un marco de referencia. En Latinoamérica, la densificación del Marco Internacional Terrestre de Referencia (ITRF), está representado por la red de estaciones de operación continua GNSS de SIRGAS, denominada como SIRGAS-CON. Por medio de las denominadas coordenadas semanales finales de las 8 estaciones consideradas como vínculo, se refirió cada una de las 119 soluciones al marco SIRGAS. La introducción del marco de referencia SIRGAS a las soluciones semilibres produce deformaciones en estas soluciones. Las deformaciones de las soluciones semilibres son producto de las cinemática de cada una de las placas en las que se ubican las estaciones de vínculo. Luego de efectuado el amarre semanal a las coordenadas SIRGAS, se hizo una estimación de los vectores de velocidad de cada una de las estaciones, incluyendo las de amarre, cuyos valores de velocidad se conocen con una alta exactitud. Para la determinación de las velocidades de las estaciones costarricenses, se programó una rutina en ambiente MatLab, basada en una ajuste por mínimos cuadrados. Los valores obtenidos en el marco de este proyecto en comparación con los valores oficiales, brindaron diferencias promedio del orden de los 0,06 cm/a, -0,08 cm/a y -0,10 cm/a respectivamente para las coordenadas [X Y Z]. De esta manera se logró determinar las coordenadas geocéntricas [X Y Z]T y sus variaciones temporales [vX vY vZ]T para el conjunto de 22 estaciones GPS de Costa Rica, dentro del datum IGS05, época de referencia 2010,5. Aunque se logró una alta exactitud en los vectores de coordenadas geocéntricas de las 22 estaciones, para algunas de las estaciones el cálculo de las velocidades no fue representativo debido al relativo corto tiempo (menos de un año) de archivos de datos. Bajo esta premisa, se excluyeron las ocho estaciones ubicadas al sur de país. Esto implicó hacer una estimación del campo local de velocidades con solamente veinte estaciones nacionales más tres estaciones en Panamá y una en Nicaragua. El algoritmo usado fue el denominado Colocación por Mínimos Cuadrados, el cual permite la estimación o interpolación de datos a partir de datos efectivamente conocidos, el cual fue programado mediante una rutina en ambiente MatLab. El campo resultante se estimó con una resolución de 30' X 30' y es altamente constante, con una velocidad resultante promedio de 2,58 cm/a en una dirección de 40,8° en dirección noreste. Este campo fue validado con base en los datos del modelo VEMOS2009, recomendado por SIRGAS. Las diferencias de velocidad promedio para las estaciones usadas como insumo para el cálculo del campo fueron del orden los +0,63 cm/a y +0,22 cm/a para los valores de velocidad en latitud y longitud, lo que supone una buena determinación de los valores de velocidad y de la estimación de la función de covarianza empírica, necesaria para la aplicación del método de colocación. Además, la grilla usada como base para la interpolación brindó diferencias del orden de -0,62 cm/a y -0,12 cm/a para latitud y longitud. Adicionalmente los resultados de este trabajo fueron usados como insumo para hacer una aproximación en la definición del límite del llamado Bloque de Panamá dentro del territorio nacional de Costa Rica. El cálculo de las componentes del Polo de Euler por medio de una rutina programa en ambiente MatLab y aplicado a diferentes combinaciones de puntos no brindó mayores aportes a la definición física de este límite. La estrategia lo que confirmó fue simplemente la diferencia en la dirección de todos los vectores velocidad y no permitió reveló revelar con mayor detalle una ubicación de esta zona dentro del territorio nacional de Costa Rica. ABSTRACT The first strict processing performed with the Bernese scientific software and contemplating the highest standards internationally recommended calculation, yielded a precise field of high accuracy, based on the integration and standardization of data from a GPS network located in Costa Rica. This processing watched a total of 119 weeks of daily data, is about 2.3 years from January 2009 to April 2011, for a total of 30 GPS stations, of which 22 are located in the country of Costa Rica and 8 hospitalizations within the network of Geocentric System for the Americas (SIRGAS). The semi-free solutions generated, every week a GPS network with high internal accuracy defined by vectors between stations and the final coordinates of the satellite constellation. The weekly evaluation given by repeatability of the solutions provided in average errors of 1.7 mm 1.4 mm and 5.1 mm in the components [n e u], confirming a high consistency in these solutions. Although semi-free solutions have a high internal accuracy, they are not used for purposes of kinematic analysis, because they lack a reference frame. In Latin America, the densification of the International Terrestrial Reference Frame (ITRF), is represented by a network of continuously operating GNSS stations SIRGAS, known as SIRGAS-CON. Through weekly final coordinates of the 8 stations considered as a link, described each of the solutions to the frame 119 SIRGAS. The introduction of the frame SIRGAS to semi-free solutions generates deformations. The deformations of the semi-free solutions are products of the kinematics of each of the plates in which link stations are located. After SIRGAS weekly link to SIRGAS frame, an estimate of the velocity vectors of each of the stations was done. The velocity vectors for each SIRGAS stations are known with high accuracy. For this calculation routine in MatLab environment, based on a least squares fit was scheduled. The values obtained compared to the official values, gave average differences of the order of 0.06 cm/yr, -0.08 cm/yr and -0.10 cm/yr respectively for the coordinates [XYZ]. Thus was possible to determine the geocentric coordinates [XYZ]T and its temporal variations [vX vY vZ]T for the set of 22 GPS stations of Costa Rica, within IGS05 datum, reference epoch 2010.5. The high accuracy vector for geocentric coordinates was obtained, however for some stations the velocity vectors was not representative because of the relatively short time (less than one year) of data files. Under this premise, the eight stations located in the south of the country were excluded. This involved an estimate of the local velocity field with only twenty national stations plus three stations in Panama and Nicaragua. The algorithm used was Least Squares Collocation, which allows the estimation and interpolation of data from known data effectively. The algorithm was programmed with MatLab. The resulting field was estimated with a resolution of 30' X 30' and is highly consistent with a resulting average speed of 2.58 cm/y in a direction of 40.8° to the northeast. This field was validated based on the model data VEMOS2009 recommended by SIRGAS. The differences in average velocity for the stations used as input for the calculation of the field were of the order of +0.63 cm/yr, +0.22 cm/yr for the velocity values in latitude and longitude, which is a good determination velocity values and estimating the empirical covariance function necessary for implementing the method of application. Furthermore, the grid used as the basis for interpolation provided differences of about -0.62 cm/yr, -0.12 cm/yr to latitude and longitude. Additionally, the results of this investigation were used as input to an approach in defining the boundary of Panama called block within the country of Costa Rica. The calculation of the components of the Euler pole through a routine program in MatLab and applied to different combinations of points gave no further contributions to the physical definition of this limit. The strategy was simply confirming the difference in the direction of all the velocity vectors and not allowed to reveal more detail revealed a location of this area within the country of Costa Rica.

El tomate (Solanum lycopersicum L.) es considerado uno de los cultivos hortícolas de mayor importancia económica en el territorio Español. Sin embargo, su producción está seriamente afectada por condiciones ambientales adversas como, salinidad, sequía y temperaturas extremas. Para resolver los problemas que se presentan en condiciones de estrés, se han empleado una serie de técnicas culturales que disminuyen sus efectos negativos, siendo de gran interés el desarrollo de variedades tolerantes. En este sentido la obtención y análisis de plantas transgénicas, ha supuesto un avance tecnológico, que ha facilitado el estudio y la evaluación de genes seleccionados en relación con la tolerancia al estrés. Estudios recientes han mostrado que el uso de genes reguladores como factores de transcripción (FTs) es una gran herramienta para obtener nuevas variedades de tomate con mayor tolerancia a estreses abióticos. Las proteínas DOF (DNA binding with One Finger) son una familia de FTs específica de plantas (Yangisawa, 2002), que están involucrados en procesos fisiológicos exclusivos de plantas como: asimilación del nitrógeno y fijación del carbono fotosintético, germinación de semilla, metabolismo secundario y respuesta al fotoperiodo pero su preciso rol en la tolerancia a estrés abiótico se desconoce en gran parte. El trabajo descrito en esta tesis tiene como objetivo estudiar genes reguladores tipo DOF para incrementar la tolerancia a estrés abiotico tanto en especies modelo como en tomate. En el primer capítulo de esta tesis se muestra la caracterización funcional del gen CDF3 de Arabidopsis, así como su papel en la respuesta a estrés abiótico y otros procesos del desarrollo. La expresión del gen AtCDF3 es altamente inducido por sequía, temperaturas extremas, salinidad y tratamientos con ácido abscísico (ABA). La línea de inserción T-DNA cdf3-1 es más sensible al estrés por sequía y bajas temperaturas, mientras que líneas transgénicas de Arabidopsis 35S::AtCDF3 aumentan la tolerancia al estrés por sequía, osmótico y bajas temperaturas en comparación con plantas wild-type (WT). Además, estas plantas presentan un incremento en la tasa fotosintética y apertura estomática. El gen AtCDF3 se localiza en el núcleo y que muestran una unión específica al ADN con diferente afinidad a secuencias diana y presentan diversas capacidades de activación transcripcional en ensayos de protoplastos de Arabidopsis. El dominio C-terminal de AtCDF3 es esencial para esta localización y su capacidad activación, la delección de este dominio reduce la tolerancia a sequía en plantas transgénicas 35S::AtCDF3. Análisis por microarray revelan que el AtCDF3 regula un set de genes involucrados en el metabolismo del carbono y nitrógeno. Nuestros resultados demuestran que el gen AtCDF3 juega un doble papel en la regulación de la respuesta a estrés por sequía y bajas temperaturas y en el control del tiempo de floración. En el segundo capítulo de este trabajo se lleva a cabo la identificación de 34 genes Dof en tomate que se pueden clasificar en base a homología de secuencia en cuatro grupos A-D, similares a los descritos en Arabidopsis. Dentro del grupo D se han identificado cinco genes DOF que presentan características similares a los Cycling Dof Factors (CDFs) de Arabidopsis. Estos genes son considerados ortólogos de Arabidopsis CDF1-5, y han sido nombrados como Solanum lycopersicum CDFs o SlCDFs. Los SlCDF1-5 son proteínas nucleares que muestran una unión específica al ADN con diferente afinidad a secuencias diana y presentan diversas capacidades de activación transcripcional in vivo. Análisis de expresión de los genes SlCDF1-5 muestran diferentes patrones de expresión durante el día y son inducidos de forma diferente en respuesta a estrés osmótico, salino, y de altas y bajas temperaturas. Plantas de Arabidopsis que sobre-expresan SlCDF1 y SlCDF3 muestran un incremento de la tolerancia a la sequía y salinidad. Además, de la expresión de varios genes de respuesta estrés como AtCOR15, AtRD29A y AtERD10, son expresados de forma diferente en estas líneas. La sobre-expresión de SlCDF3 en Arabidopsis promueve un retardo en el tiempo de floración a través de la modulación de la expresión de genes que controlan la floración como CONSTANS (CO) y FLOWERING LOCUS T (FT). En general, nuestros datos demuestran que los SlCDFs están asociados a funciones aun no descritas, relacionadas con la tolerancia a estrés abiótico y el control del tiempo de floración a través de la regulación de genes específicos y a un aumento de metabolitos particulares. ABSTRACT Tomato (Solanum lycopersicum L.) is one of the horticultural crops of major economic importance in the Spanish territory. However, its production is being affected by adverse environmental conditions such as salinity, drought and extreme temperatures. To resolve the problems triggered by stress conditions, a number of agricultural techniques that reduce the negative effects of stress are being frequently applied. However, the development of stress tolerant varieties is of a great interest. In this direction, the technological progress in obtaining and analysis of transgenic plants facilitated the study and evaluation of selected genes in relation to stress tolerance. Recent studies have shown that a use of regulatory genes such as transcription factors (TFs) is a great tool to obtain new tomato varieties with greater tolerance to abiotic stresses. The DOF (DNA binding with One Finger) proteins form a family of plant-specific TFs (Yangisawa, 2002) that are involved in the regulation of particular plant processes such as nitrogen assimilation, photosynthetic carbon fixation, seed germination, secondary metabolism and flowering time bur their precise roles in abiotic stress tolerance are largely unknown. The work described in this thesis aims at the study of the DOF type regulatory genes to increase tolerance to abiotic stress in both model species and the tomato. In the first chapter of this thesis, we present molecular characterization of the Arabidopsis CDF3 gene as well as its role in the response to abiotic stress and in other developmental processes. AtCDF3 is highly induced by drought, extreme temperatures, salt and abscisic acid (ABA) treatments. The cdf3-1 T-DNA insertion mutant was more sensitive to drought and low temperature stresses, whereas the AtCDF3 overexpression enhanced the tolerance of transgenic plants to drought, cold and osmotic stress comparing to the wild-type (WT) plants. In addition, these plants exhibit increased photosynthesis rates and stomatal aperture. AtCDF3 is localized in the nuclear region, displays specific binding to the canonical DNA target sequences and has a transcriptional activation activity in Arabidopsis protoplast assays. In addition, the C-terminal domain of AtCDF3 is essential for its localization and activation capabilities and the deletion of this domain significantly reduces the tolerance to drought in transgenic 35S::AtCDF3 overexpressing plants. Microarray analysis revealed that AtCDF3 regulated a set of genes involved in nitrogen and carbon metabolism. Our results demonstrate that AtCDF3 plays dual roles in regulating plant responses to drought and low temperature stress and in control of flowering time in vegetative tissues. In the second chapter this work, we carried out to identification of 34 tomato DOF genes that were classified by sequence similarity into four groups A-D, similar to the situation in Arabidopsis. In the D group we have identified five DOF genes that show similar characteristics to the Cycling Dof Factors (CDFs) of Arabidopsis. These genes were considered orthologous to the Arabidopsis CDF1 - 5 and were named Solanum lycopersicum CDFs or SlCDFs. SlCDF1-5 are nuclear proteins that display specific binding to canonical DNA target sequences and have transcriptional activation capacities in vivo. Expression analysis of SlCDF1-5 genes showed distinct diurnal expression patterns and were differentially induced in response to osmotic, salt and low and high temperature stresses. Arabidopsis plants overexpressing SlCDF1 and SlCDF3 showed increased drought and salt tolerance. In addition, various stress-responsive genes, such as AtCOR15, AtRD29A and AtERD10, were expressed differently in these lines. The overexpression of SlCDF3 in Arabidopsis also results in the late flowering phenotype through the modulation of the expression of flowering control genes such CONSTANS (CO) and FLOWERING LOCUS T (FT). Overall, our data connet SlCDFs to undescribed functions related to abiotic stress tolerance and flowering time through the regulation of specific target genes and an increase in particular metabolites.
