This paper describes the main steps in the study of Senna spectabilis, as part of our bioprospection studies, with the isolation of spectaline, cassine and other piperidine alkaloids, as well as the preparation of semi-synthetic analogues, and the studies of their pharmacological and toxicological properties, which led to the establishment of two candidate drugs for the treatment of Alzheimer disease.

Objective: Early treatment in sepsis may improve outcome. The aim of this study was to evaluate how the delay in starting resuscitation influences the severity of sepsis and the treatment needed to achieve hemodynamic stability. Design: Prospective, randomized, controlled experimental study. Setting: Experimental laboratory in a university hospital. Subjects: Thirty-two anesthetized and mechanically ventilated pigs. Interventions: Pigs were randomly assigned (n = 8 per group) to a nonseptic control group or one of three groups in which fecal peritonitis (peritoneal instillation of 2 g/kg autologous feces) was induced, and a 48-hr period of protocolized resuscitation started 6 (Delta T-6 hrs), 12 (Delta T-12 hrs), or 24 (Delta T-24 hrs) hrs later. The aim of this study was to evaluate the impact of delays in resuscitation on disease severity, need for resuscitation, and the development of sepsis-associated organ and mitochondrial dysfunction. Measurements and Main Results: Any delay in starting resuscitation was associated with progressive signs of hypovolemia and increased plasma levels of interleukin-6 and tumor necrosis factor-alpha prior to resuscitation. Delaying resuscitation increased cumulative net fluid balances (2.1 +/- 0.5 mL/kg/hr, 2.8 +/- 0.7 mL/kg/hr, and 3.2 +/- 1.5 mL/kg/hr, respectively, for groups.T-6 hrs, Delta T-12 hrs, and.T-24 hrs; p < .01) and norepinephrine requirements during the 48-hr resuscitation protocol (0.02 +/- 0.04 mu g/kg/min, 0.06 +/- 0.09 mu g/kg/min, and 0.13 +/- 0.15 mu g/kg/min; p = .059), decreased maximal brain mitochondrial complex II respiration (p = .048), and tended to increase mortality (p = .08). Muscle tissue adenosine triphosphate decreased in all groups (p < .01), with lowest values at the end in groups Delta T-12 hrs and.T-24 hrs. Conclusions: Increasing the delay between sepsis initiation and resuscitation increases disease severity, need for resuscitation, and sepsis-associated brain mitochondrial dysfunction. Our results support the concept of a critical window of opportunity in sepsis resuscitation. (Crit Care Med 2012; 40:2841-2849)

This work describes the atropisomeric relationships of 3-methyl-5-(3-methyl-5-phenyl-1H-pyrazol-1-yl)-1-phenyl-1H-pyrazol-4-amine (2d), which belongs to series 4-aminobipyrazole derivatives designed as anti-inflammatory agents. The 1H nuclear magnetic resonance spectra obtained in the presence of a chiral lanthanide shift salt associated to chiral high-performance liquid chromatography analysis, X-ray diffraction, and molecular modeling tools confirmed that ortho bis-functionalized bipyrazole 2d exists as a mixture of aR,aS-atropisomers. These results provide useful information to understand the pharmacological profile of this derivative and of other 4-aminobipyrazole analogs. Chirality 24:463470, 2012. p38 mitogen-activated protein kinase (p38 MAPK) is an important signal transducing enzyme involved in many cellular regulations, including signaling pathways, pain and inflammation. Several p38 MAPK inhibitors have been developed as drug candidates to treatment of autoimmune disorders, such as rheumatoid arthritis. In this paper we reported the docking, synthesis and pharmacological activity of novel urea-derivatives (4a-e) designed as p38 MAPK inhibitors. These derivatives presented good theoretical affinity to the target p38 MAPK, standing out compound 4e (LASSBio-998), which showed a better score value compared to the prototype GK-00687. This compound was able to reduce in vitro TNF-alpha production and was orally active in a hypernociceptive murine model sensible to p38 MAPK inhibitors. Otherwise, compound 4e presented a dose-dependent analgesic effect in a model of antigen (mBSA)-induced arthritis and anti-inflammatory profile in carrageenan induced paw edema, indicating its potential as a new antiarthritis prototype. Hancornia speciosa Gomes is a fruit tree native from Brazil that belongs to Apocinaceae family, and is popularly known as Mangabeira. Its fruits are widely consumed raw or processed as fruit jam, juices and ice creams, which have made it a target of intense exploitation. The extractive activities and intense human activity on the environment of natural occurrence of H. speciosa has caused genetic erosion in the species and little is known about the ecology or genetic structure of natural populations. The objective of this research was the evaluation of the genetic diversity and genetic structure of H. speciosa var. speciosa. The genetic variability was assessed using 11 allozyme loci with a sample of 164 individuals distributed in six natural populations located in the States of Pernambuco and Alagoas, Northeastern Brazil. The results showed a high level of genetic diversity within the species (e= 0.36) seeing that the most of the genetic variability of H. speciosa var. speciosa is within its natural populations with low difference among populations ( or = 0.081). The inbreeding values within ( = -0.555) and among populations ( =-0.428) were low showing lacking of endogamy and a surplus of heterozygotes. The estimated gene flow ( m ) was high, ranging from 2.20 to 13.18, indicating to be enough to prevent the effects of genetic drift and genetic differentiation among populations. The multivariate analyses indicated that there is a relationship between genetic and geographical distances, which was confirmed by a spatial pattern analysis using Mantel test (r = 0.3598; p = 0.0920) with 1000 random permutations. The high genetic diversity index in these populations indicates potential for in situ genetic conservation.

Le ammine biogene sono il prodotto della decarbossilazione degli amminoacidi da parte di enzimi microbici. Tra essi vi è la tirosina decarbossilasi, caratterizzata dalla possibilità di utilizzare, in assenza di tirosina, la fenilalanina, ottenendo la 2-feniletilamina. In particolare, la tiramina è responsabile della comparsa di importanti sintomi tossicologici, raggruppati con il termine “cheese reaction”. In questa sperimentazione sono stati presi in considerazione 2 ceppi di Enterococcus mundtii (C46 e C53) coltivati in BHI in presenza o assenza di tirosina per caratterizzarne l’attività decarbossilasica. Sono state monitorate la crescita microbica, mediante densità ottica e la produzione di tiramina e 2-feniletilamina mediante tecnica HPLC. Dai risultati ottenuti è emerso che entrambi i ceppi producono tiramina sia in presenza che in assenza del precursore. La concentrazione massima rilevata per il ceppo C46 è stata di 797 mg/l e 767 mg/l per C53. È inoltre emerso che essi possono decarbossilare la fenilalanina, ma solo dopo 8 e 24 ore di incubazione per il ceppo C46 e C53. Per quanto concerne la crescita, entrambi i ceppi hanno raggiunto il massimo valore di densità ottica dopo 6-8 ore a 37°C, con una durata della fase lag ridotta, seguita da un rapido aumento della densità ottica. Non sono state riscontrate differenze significative in termini di massima densità ottica raggiunta (A) e durata della fase lag (λ) tra i due ceppi, mentre C53 ha presentato valori inferiori per quanto riguarda la velocità incremento della densità ottica in fase esponenziale (µmax). Dagli studi genici è emerso che l’organizzazione dell’operone dei ceppi considerati corrisponde con quella filogeneticamente riconosciuta per il genere Enterococcus, ma nonostante la similarità, l’operone manca del gene codificante per l’antiporto Na+/H+. È stata inoltre evidenziata nel genoma dei ceppi considerati un’altra regione che contiene geni codificanti per un ulteriore sistema decarbossilasico.
