Aim of the study: Mycorrhizal fungi in Mediterranean forests play a key role in the complex process of recovery after wildfires. A broader understanding of an important pyrophytic species as Pinus pinaster and its fungal symbionts is thus necessary for forest restoration purposes. This study aims to assess the effects of ectomycorrhizal symbiosis on maritime pine seedlings and how fire severity affects fungal colonization ability. Area of study: Central Spain, in a Mediterranean region typically affected by wildfires dominated by Pinus pinaster, a species adapted to fire disturbance. Material and Methods: We studied P. pinaster root apexes from seedlings grown in soils collected one year after fire in undisturbed sites, sites moderately affected by fire and sites highly affected by fire. Natural ectomycorrhization was observed at the whole root system level as well as at two root vertical sections (0-10 cm and 10-20 cm). We also measured several morphometric traits ( tap root length, shoot length, dry biomass of shoots and root/shoot ratio), which were used to test the influence of fire severity and soil chemistry upon them. Main results: Ectomycorrhizal colonization in undisturbed soils for total and separated root vertical sections was higher than in soils that had been affected by fire to some degree. Inversely, seedling vegetative size increased according to fire severity. Research highlights: Fire severity affected soil properties and mycorrhizal colonization one year after occurrence, thus affecting plant development. These findings can contribute to a better knowledge of the factors mediating successful establishment of P. pinaster in Mediterranean forests after wildfires.

Proanthocyanidins from P. contorta leaves and from a commercial quebracho extract were isolated and characterized. Flavonoids, catechins and gallic acid were also identified in the extracts of P. contorta. Compounds were evaluated for their antioxidant properties and for their antiviral activity against an acyclovir-resistant herpes simplex virus type 1 strain. The low molecular weight phenolic derivatives and the proanthocyanidins from P. contorta showed the highest antioxidant activity. Purified proanthocyanidins from both P. contorta and quebracho showed the same maximum non toxic concentrations (25 µg/mL), with 82.2% and 100% of virus inhibition, respectively.

The essential oils of leaves, aerial and underground stems of Senecio crassiflorus var. crassiflorus were extracted by hydrodistillation and their analyses were performed by GC/MS. Qualitative and quantitative differences have been found between the analyzed parts. The main compounds of the essentials oils of the leaves were found to be oxygenated sesquiterpenes, while the oils from the aerial and underground stems showed sesquiterpene hydrocarbons as major constituents. Furthermore, the essential oils were tested against Staphylocccus aureus, Escherichia coli, Pseudomonas aeruginosa and Bacillus cereus. The essential oil of the leaves was shown to be the most active.

From leaves of Senna spectabilis var. excelsa were isolated caffeine, the triterpenes lupeol, α-amyrin, β-amyrin, cycloeucalenol, friedelin and ursolic, oleanolic and betulinic acids, besides the steroids sitosterol and stigmasterol and their respective glucosides. The structures of these compounds were established by spectroscopic analysis including two-dimensional NMR methods and comparison with published spectral data. This paper deals with the first report of these compounds in S. spectabilis var. excelsa.

Distribution and stocks of soil organic matter (SOM) compartments after Pinus monoculture introduction in a native pasture area of a Cambisol, Santa Catarina, Brazil, were investigated. Pinus introduction increased soil acidity, content of exchangeable Al+3 and diminished soil nutrients. Nevertheless, soil C stock increased in all humic fractions of the 0-5 cm layer after Pinus afforestation. In the subsurface, the vegetation change only promoted SOM redistribution from the NaOH-extractable humic substances to a less hydrophobic humin fraction. Under Pinus, soil organo-mineral interactions were relevant up to a 15 cm depth, while in pasture environment, this mechanism occurred mainly in the surface layer.

Este trabalho teve por objetivo verificar a viabilidade de utilização da técnica de PCR, usando primers considerados específicos, para identificação de Xanthomonas axonopodis pv. phaseoli (Xap) e x. axonopodis pv. phaseoli var. fuscans (Xapf), visando criar bases para sua utilização em diagnose rotineira e em programas de certificação de sementes. Foram incluídos no estudo 42 isolados da bactéria provenientes de locais distintos, além de outros gêneros, espécies e patovares, para ser verificada a especificidade dos primers. Os resultados obtidos permitem concluir que os primers utilizados são adequados para identificação de Xap e Xapf, embora dois isolados patogênicos não tenham sido amplificados. Foi observada também a amplificação de uma banda fraca para X. axonopodis pv. vitians, com o mesmo número de pares de bases, o que sugere existir homologia entre estes patovares, na região amplificada.

Foi estudado o processo de infecção de Colletotrichum guaranicola em clones de guaranazeiro (Paullinia cupana var. sorbilis) resistente e suscetível, por meio de microscopia de luz. Avaliaram-se quantitativamente os eventos de pré-penetração em folhas novas e velhas dos clones. Observou-se diferença quantitativa quanto à formação de apressório em folhas novas e velhas dos clones, sendo maior em folhas novas do clone suscetível. Estruturas de pré-penetração e infecção foram semelhantes nos dois clones. O processo de infecção teve inicio após a germinação do fungo e formação de apressório na superfície do hospedeiro, seguido de penetração direta da cutícula e da parede celular das células epidérmicas. Dentro da célula, a hifa de infecção deu origem a uma vesícula, que em seguida formou a hifa primária que se ramificou, originando a hifa secundária, que colonizou intra e intercelularmente a epiderme e o parênquima, causando desorganização dos tecidos e necrose. Observou-se uma diferença temporal na colonização dos tecidos dos clones. No clone suscetível, com 48 h após a inoculação, as células da epiderme e do parênquima estavam colonizadas por hifas intra e intercelulares. No quinto dia após a inoculação, observou-se o surgimento dos sintomas. No clone resistente, a colonização só foi observada no quarto dia após a inoculação. Somente no sétimo dia após a inoculação, foram observados os primeiros sintomas típicos da doença.

O mal-do-pé em arroz (Oryza sativa) foi constatado em lavouras de terras altas nos municípios de Unaí (MG), Palmeiras (GO), Itaberaí (GO), Humaitá (AM) e em lavouras irrigadas nos Estados de Goiás, Tocantins e Rio Grande de Sul. O agente causal foi identificado como Gaeumannomyces graminis var. graminis baseado em características morfológicas, culturais e testes de patogenicidade utilizando diferentes isolados brasileiros. O método de inoculação e avaliação da doença em de casa de vegetação foi descrito. Este é o primeiro registro desta enfermidade na cultura do arroz no Brasil.

Fungal diseases in cotton (Gossypium hirsutum), such as anthracnose caused by Colletotrichum gossypii and ramulose caused by C. gossypii var. cephalosporioides, are responsible for large yield losses. These pathogens are seed borne and morphologically similar although they induce different symptoms, which can lead to misdiagnosis using the blotter testing method. The present study was carried out to assess the viability of using Amplified Fragment Length Polymorphism (AFLP) markers to differentiate these pathogens. Five isolates, for each pathogen, were classified according to pathogenicity on cotton plants, and mycelial growth morphology. Conidial suspensions were sprayed on 30-day-old cotton plants and the symptoms assessed ten and 40 days after inoculation. For growth morphology 200 cottonseeds were inoculated with seven-day-old pure cultures, and the mycelial traits observed under a stereoscopic microscope seven days after inoculation. The DNA for AFLP analysis was obtained from seven-day-old fungal mycelia grown in liquid medium, using the Dneasy Qiagen protocol. Using the AFLP technique 318 polymorphic bands were selected to estimate similarities using Dice's Coefficient. The results clearly distinguished between ramulose and anthracnose isolates, which agreed with morphological and pathogenicity testing.
