CD4(+) Foxp3(+) regulatory T cells inhibit the production of interferon-?, which is the major mediator of protection against Mycobacterium tuberculosis infection. In this study, we evaluated whether the protection conferred by three different vaccines against tuberculosis was associated with the number of spleen and lung regulatory T cells. We observed that after homologous immunization with the 65 000 molecular weight heat-shock protein (hsp 65) DNA vaccine, there was a significantly higher number of spleen CD4(+) Foxp3(+) cells compared with non-immunized mice. Heterologous immunization using bacillus Calmette Guerin (BCG) to prime and DNA-hsp 65 to boost (BCG/DNA-hsp 65) or BCG to prime and culture filtrate proteins (CFP)-CpG to boost (BCG/CFP-CpG) induced a significantly higher ratio of spleen CD4(+)/CD4(+) Foxp3(+) cells compared with non-immunized mice. In addition, the protection conferred by either the BCG/DNA-hsp 65 or the BCG/CFP-CpG vaccines was significant compared with the DNA-hsp 65 vaccine. Despite the higher ratio of spleen CD4(+)/CD4(+) Foxp3(+) cells found in BCG/DNA-hsp 65-immunized or BCG/CFP-CpG-immunized mice, the lungs of both groups of mice were better preserved than those of DNA-hsp 65-immunized mice. These results confirm the protective efficacy of BCG/DNA-hsp 65 and BCG/CFP-CpG heterologous prime-boost vaccines and the DNA-hsp 65 homologous vaccine. Additionally, the prime-boost regimens assayed here represent a promising strategy for the development of new vaccines to protect against tuberculosis because they probably induce a proper ratio of CD4(+) and regulatory (CD4(+) Foxp3(+)) cells during the immunization regimen. In this study, this ratio was associated with a reduced number of regulatory cells and no injury to the lungs.

Trypanosoma cruzi is an organism highly resistant to ionizing radiation. Following a dose of 500 Gy of gamma radiation, the fragmented genomic DNA is gradually reconstructed and the pattern of chromosomal bands is restored in less than 48 hours. Cell growth arrests after irradiation but, while DNA is completely fragmented, RNA maintains its integrity. In this work we compared the transcriptional profiles of irradiated and non-irradiated epimastigotes at different time points after irradiation using microarray. In total, 273 genes were differentially expressed; from these, 160 were up-regulated and 113 down-regulated. We found that genes with predicted functions are the most prevalent in the down-regulated gene category. Translation and protein metabolic processes, as well as generation of precursor of metabolites and energy pathways were affected. In contrast, the up-regulated category was mainly composed of obsolete sequences (which included some genes of the kinetoplast DNA), genes coding for hypothetical proteins, and Retrotransposon Hot Spot genes. Finally, the tyrosyl-DNA phosphodiesterase 1, a gene involved in double-strand DNA break repair process, was up-regulated. Our study demonstrated the peculiar response to ionizing radiation, raising questions about how this organism changes its gene expression to manage such a harmful stress.

Visceral leishmaniasis (VL) is a zoonotic disease characterized by infection of mononuclear phagocytes by Leishmania chagasi. The primary vector is Lutzomyia longipalpis and the dog is the main domestic reservoir. The control and current treatment of dogs using synthetic drugs have not shown effectiveness in reducing the incidence of disease in man. In attempt to find new compounds with leishmanicidal action, plant secondary metabolites have been studied in search of treatments of VL. This study aimed to evaluate the leishmanicidal activity of Musa paradisiaca (banana tree) and Spondias mombin (cajazeira) chemical constituents on promastigotes and amastigotes of L. chagasi. Phytochemical analysis by column chromatography was performed on ethanol extracts of two plants and fractions were isolated. Thin layer chromatography was used to compare the fractions and for isolation the substances to be used in vitro tests. The in vitro tests on promastigotes of L chagasi used the MTT colorimetric method and the method of ELISA in situ was used against amastigotes besides the cytotoxicity in RAW 264.7 cells. Of the eight fractions tested, Sm1 and Sm2 from S. mombin had no action against promastigotes, but had good activity against amastigotes. The fractions Mp1 e Mp4 of M. paradisiaca were very cytotoxic to RAW 264.7 cells. The best result was obtained with the fraction Sm3 from S. mombin with IC50 of 11.26 mu g/ml against promastigotes and amastigotes of 0.27 mu g/ml. The fraction Sm3 characterized as tannic acid showed the best results against both forms of Leishmania being a good candidate for evaluation in in vivo tests. (C) 2012 Published by Elsevier B.V.

A resiliência física de solos é proveniente de processos regenerativos que incluem ciclos de umedecimento e secamento, congelamento e descongelamento, assim como as atividades biológicas. Este estudo testou a hipótese de que as propriedades físicas do solo, como a permeabilidade do solo ao ar, densidade do solo, porosidade de aeração e porosidade total, são indicadores físicos eficientes para quantificar a resiliência de solos de diferentes texturas submetidos ao estresse mecânico (compactação) e após subsequentes ciclos de umedecimento e secamento. O objetivo foi avaliar o comportamento e a resiliência do solo por meio de propriedades físicas de dois Latossolos Vermelhos. Foram retiradas 25 amostras indeformadas (0,00-0,05 m) de dois solos: solo I, com textura argilosa, e solo II, com textura franco-argilo-arenosa, realizando as determinações das propriedades físicas nos tratamentos: antes da compactação (A), depois da compactação (C0) e após ciclos de umedecimento e secamento (C1, C2, C3 e C4). As propriedades densidade do solo e porosidade total não apresentaram recuperação da condição inicial após a compactação nos solos I e II; as propriedades conteúdo volumétrico de água e porosidade de aeração mostraram recuperação parcial apenas no solo I; e a permeabilidade do solo ao ar foi a propriedade que apresentou a melhor recuperação e a maior resiliência. Em relação ao distinto comportamento dos dois solos, observou-se que o solo I foi mais resiliente que o solo II nas propriedades que apresentaram recuperação.

The increased incidence of visceral leishmaniasis (VL) in Brazil is due to a lack of effective disease control measures. In addition to that, no effective treatment exists for canine VL in response to synthetic drugs. Thus, the objective of this study was to evaluate the effect of the essential oils of Coriandrum sativum and Lippia sidoides, and oleoresin from Copaifera reticulata, on Leishmania chagasi promastigotes and amastigotes. We also examined the toxicity of these treatments on the murine monocyte cell line RAW 264.7. To determine the IC50 a MTT test (3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) was performed on promastigotes, and an in situ ELISA assay was conducted on amastigotes. Here, we demonstrate that oleoresin from C. reticulata was effective against both promastigotes (IC50 of 7.88 µg.mL-1) and amastigotes (IC50 of 0.52 µg.mL-1), and neither of the two treatments differed significantly (p > 0.05) from pentamidine (IC50 of 2.149 µg.mL-1) and amphotericin B (IC50 of 9.754 µg.mL-1). Of the three plant oils tested, only oleoresin showed no toxicity toward monocyte, with 78.45% viability after treatment. Inhibition of promastigote and amastigote growth and the lack of cytotoxicity by C. reticulata demonstrate that oleoresin may be a viable option for analyzing the in vivo therapeutic effects of leishmanicidal plants

Los foraminíferos bentónicos representan un excelente proxy geológico, ya que sus caparazones son incorporados en el sedimento y pasan a comportarse como granos sedimentares durante los eventos de transporte y erosión, permitiendo así, el registro de condiciones y eventos que ocurren en los océanos. De este modo, son usados para describir patrones de hidrodinámica y evaluar índices de deposición, erosión y retrabajamiento del sedimento en los ambientes marinos. Concretamente, las regiones costeras representan zonas de alta complejidad ambiental, debido a que en ellas se da la mezcla de agua tanto de origen continental como marino, y a su vez, al efecto de la descarga fluvial que aporta material particulado del continente para el océano. El Río de la Plata (RdLP) representa el cuarto mayor estuario del mundo y el segundo del continente americano, lo cual convierte al sistema RdLP-Océano Atlántico en un complejo gradiente estuarino de interés en estudios sedimentológicos con posteriores aplicaciones paleo-climáticas y paleo-ecológicas del Cuaternario Superior. En el presente estudio, se registra a partir de las asociaciones muertas, para la región del estuario entrada de agua marina hasta la zona del frente termohalino. Dichas asociaciones se caracterizaron por especies típicas de plataforma y aguas frías. La región marina del RdlP registró los mayores porcentajes de abrasión y fragmentación de caparazones, indicando una zona de alta energía. Por otro lado, en la región más oceánica las asociaciones muertas reflejaron un transporte en dirección norte, influenciado por la pluma del RdlP. Cabe resaltar la presencia en esta región de una isla barrera, vestigio de una paleo-costa producto de una regresión Holocénica. Este local se caracterizó por un aumento de la abrasión y fragmentación mostrando la gran hidrodinámica presente. En todos los casos existió correlación entre las señales tafonómicas y los datos geo-químicos (sedimentología, pH, oxígeno disuelto, carbono orgánico y nitrógeno total).
