Na+-Ca2+ exchanger and Ca2+ channel are two major sarcolemmal Ca2+-transporting proteins of cardiac myocytes. Although the Ca2+ channel is effectively regulated by protein kinase A-dependent phosphorylation, no enzymatic regulation of the exchanger protein has been identified as yet. Here we report that in frog ventricular myocytes, isoproterenol down-regulates the Na+-Ca2+ exchanger, independent of intracellular Ca2+ and membrane potential, by activation of the beta-receptor/adenylate-cyclase/cAMP-dependent cascade, resulting in suppression of transmembrane Ca2+ transport via the exchanger and providing for the well-documented contracture-suppressant effect of the hormone on frog heart. The beta-blocker propranolol blocks the isoproterenol effect, whereas forskolin, cAMP, and theophylline mimic it. In the frog heart where contractile Ca2+ is transported primarily by the Na+-Ca2+ exchanger, the beta-agonists' simultaneous enhancement of Ca2+ current, ICa, and suppression of Na+-Ca2+ exchanger current, INa-Ca would enable the myocyte to develop force rapidly at the onset of depolarization (enhancement of ICa) and to decrease Ca2+ influx (suppression of INa-Ca) later in the action potential. This unique adrenergically induced shift in the Ca2+ influx pathways may have evolved in response to paucity of the sarcoplasmic reticulum Ca2+-ATPase/phospholamban complex and absence of significant intracellular Ca2+ release pools in the frog heart.

The role of cAMP subcellular compartmentation in the progress of beta-adrenergic stimulation of cardiac L-type calcium current (ICa) was investigated by using a method based on the use of whole-cell patch-clamp recording and a double capillary for extracellular microperfusion. Frog ventricular cells were sealed at both ends to two patch-clamp pipettes and positioned approximately halfway between the mouths of two capillaries that were separated by a 5-micron thin wall. ICa could be inhibited in one half or the other by omitting Ca2+ from one solution or the other. Exposing half of the cell to a saturating concentration of isoprenaline (ISO, 1 microM) produced a nonmaximal increase in ICa (347 +/- 70%; n = 4) since a subsequent application of ISO to the other part induced an additional effect of nearly similar amplitude to reach a 673 +/- 130% increase. However, half-cell exposure to forskolin (FSK, 30 microM) induced a maximal stimulation of ICa (561 +/- 55%; n = 4). This effect was not the result of adenylyl cyclase activation due to FSK diffusion in the nonexposed part of the cell. To determine the distant effects of ISO and FSK on ICa, the drugs were applied in a zero-Ca solution. Adding Ca2+ to the drug-containing solutions allowed us to record the local effect of the drugs. Dose-response curves for the local and distant effects of ISO and FSK on ICa were used as an index of cAMP concentration changes near the sarcolemma. We found that ISO induced a 40-fold, but FSK induced only a 4-fold, higher cAMP concentration close to the Ca2+ channels, in the part of the cell exposed to the drugs, than it did in the rest of the cell. cAMP compartmentation was greatly reduced after inhibition of phosphodiesterase activity with 3-isobutyl-methylxanthine, suggesting the colocalization of enzymes involved in the cAMP cascade. We conclude that beta-adrenergic receptors are functionally coupled to nearby Ca2+ channels via local elevations of cAMP.

A eficiência e a racionalidade energética da iluminação pública têm relevante importância no sistema elétrico, porque contribui para diminuir a necessidade de investimentos na construção de novas fontes geradoras de energia elétrica e nos desperdícios energéticos. Apresenta-se como objetivo deste trabalho de pesquisa o desenvolvimento e aplicação do IDE (índice de desempenho energético), fundamentado no sistema de inferência nebulosa e indicadores de eficiência e racionalidade de uso da energia elétrica. A opção em utilizar a inferência nebulosa deve-se aos fatos de sua capacidade de reproduzir parte do raciocínio humano, e estabelecer relação entre a diversidade de indicadores envolvidos. Para a consecução do sistema de inferência nebulosa, foram definidas como variáveis de entrada: os indicadores de eficiência e racionalidade; o método de inferência foi baseado em regras produzidas por especialista em iluminação pública, e como saída um número real que caracteriza o IDE. Os indicadores de eficiência e racionalidade são divididos em duas classes: globais e específicos. Os indicadores globais são: FP (fator de potência), FC (fator de carga) e FD (fator de demanda). Os indicadores específicos são: FU (fator de utilização), ICA (consumo de energia por área iluminada), IE (intensidade energética) e IL (intensidade de iluminação natural). Para a aplicação deste trabalho, foi selecionada e caracterizada a iluminação pública da Cidade Universitária \"Armando de Salles Oliveira\" da Universidade de São Paulo. Sendo assim, o gestor do sistema de iluminação, a partir do índice desenvolvido neste trabalho, dispõe de condições para avaliar o uso da energia elétrica e, desta forma, elaborar e simular estratégias com o objetivo de economizá-la.

The Import Substitution Process in Latin Amer ica was an attempt to enhance GDP growth and productivity by rising trade barriers upon capital-intensive products. Our main goal is to analyze how an increase in import tariff on a particular type of good affects the production choices and trade pattern of an economy. We develop an extension of the dynamic Heckscher-Ohlin model – a combination of a static two goods, two-factor Heckscher-Ohlin model and a two-sector growth model – allowing for import tariff. We then calibrate the closed economy model to the US. The results show that the economy will produce less of both consumption and investment goods under autarky for low and high levels of capital stock per worker. We also find that total GDP may be lower under free trade in comparison to autarky.

The aim of this paper is to analyze what different types of populist communication styles emerged during the 2014 EP election campaign and under which conditions political parties selected specific populist communication styles. To do this, a comparative quantitative content analysis of press releases in the run-up of the latest EP elections has been conducted for parties in France, Germany, Austria and Greece. The paper presents a definition of populism based on the contemporary academic discourse, which focuses on the transnational nature of the European political field. It is shown that populist party communication is more pronounced on the fringes of the political spectrum and in countries struggling with severe macroeconomic difficulties. Contrary to intuitive expectations, the perceived populist rhetoric of exclusivity in the context of the European sovereign debt crisis, which is identified as a central feature of right-wing populism, barely takes place within populist party communication.

A blind nonlinear interference cancellation receiver for code-division multiple-access- (CDMA-) based communication systems operating over Rayleigh flat-fading channels is proposed. The receiver which assumes knowledge of the signature waveforms of all the users is implemented in an asynchronous CDMA environment. Unlike the conventional MMSE receiver, the proposed blind ICA multiuser detector is shown to be robust without training sequences and with only knowledge of the signature waveforms. It has achieved nearly the same performance of the conventional training-based MMSE receiver. Several comparisons and experiments are performed based on examining BER performance in AWGN and Rayleigh fading in order to verify the validity of the proposed blind ICA multiuser detector.

We consider blind signal detection in an asynchronous code-division multiple-access (CDMA) system employing short spreading sequences in the presence of unknown multipath fading. This approach is capable of countering the presence of multiple-access interference (MAI) in CDMA fading channels. The proposed blind multiuser detector is based on an independent component analysis (ICA) to mitigate both MAI and noise. This algorithm has been utilised in blind source separation (BSS) of unknown sources from their mixtures. It can also be used for estimating the basis vectors of BSS. The aim is to include an ICA algorithm within a wireless receiver in order to reduce the level of interference in wideband systems. This blind multiuser detector requires no training sequence compared with the conventional multiuser detection receiver. The proposed ICA blind multiuser detector is made robust with respect to knowledge of signature waveforms and the timing of the user of interest. Several experiments are performed in order to verify the validity of the proposed ICA algorithm.

Policy and social work practice currently lack a theoretical framework that adequately explains the emergence, diffusion, and continuance of the intercountry adoption (ICA) phenomenon. Using South Korea as a case study and the application of actor network theory to the ICA phenomenon, this paper introduces a theoretical approach that allows an examination of the complex interrelationships between the global and local influences of a country's engagement in ICA. This theoretical approach provides a different way of understanding the phenomenon, which, in turn, can better inform policies and practice that affect children and families across the globe.

It is well known that even slight changes in nonuniform illumination lead to a large image variability and are crucial for many visual tasks. This paper presents a new ICA related probabilistic model where the number of sources exceeds the number of sensors to perform an image segmentation and illumination removal, simultaneously. We model illumination and reflectance in log space by a generalized autoregressive process and Hidden Gaussian Markov random field, respectively. The model ability to deal with segmentation of illuminated images is compared with a Canny edge detector and homomorphic filtering. We apply the model to two problems: synthetic image segmentation and sea surface pollution detection from intensity images.
