The Caudal Cervical Spondylomyelopathy, also known as “Wobbler syndrome” is a neurological disorder that affects mainly breeds of large and giant size, especially Doberman pinsher and Great danes. Its aetiology is multifactorial and leads to a narrowing of the spinal canal by morphological and positional caudal cervical vertebrae (C5, C6 and C7), causing compression of the spinal cord and nerve roots. The clinical signs presented by the affected animals are progressive ataxia of hindlimbs and, later, the forelimbs, sometimes progressing to tetraparesis. Neck pain may be present. The diagnosis is made through the association of clinical signs and diagnostic imaging such as radiography, myelography, computed tomography and magnetic resonance imaging. According to the classification of the lesion obtained by imaging examinations, the conservative or surgical treatment is established and the prognosis is variable in accordance with the degree of affection of the spinal cord

Tumoral masses can cause several direct problems in the organism, such as invading organs and altering their functions, leading to other problems such as the Paraneoplastic syndrome. The paraneoplastic syndrome is an alteration in the structure and function of the body due to the non-invasive actions of the tumor, for example, liberating hormones, peptides, cytokines and leading to cross reactions between normal tissues. The syndrome can affect different locations in the body, being that some are indicative of specific tumors, however the interpretation of the clinical and pathological findings referring to this syndrome should be utilized in the diagnostic and treatment. In the hematological paraneoplastic syndrome, there are alterations that occur due to indirect actions of the tumor on the blood cellular elements and the coagulation system, and that are generally detected in routine clinical and laboratorial exams. This study objective is to aboard some hematological paraneoplastic syndromes in dogs such as: anemia, thrombocytopenia, neutrophilic leukocytosis, hypergammaglobulinemia and erythrocytosis, emphasizing the different etiologies that may assist in differential diagnosis and the principal neoplasm related to this syndrome. Besides this, the early discovery and treatment of the paraneoplatic syndrome is important as the tumor itself, as it improves the prognostic and quality of life of the patient

Aquaporins (AQPs) are membrane protein channels expressed in a wide variety of cells and tissues that are involved in the transport of water. In the male excurrent duct, the movement of fluid and solutes across the epithelium is essential for establishing the proper luminal environment in which sperm mature and are stored. The initial aim of the present propose is to localize AQP 2 e 9 in epithelium of the initial segment, caput, corpus and cauda epididymidis in Mongolian gerbil during postnatal development (10, 30, 60, 120 and 300 days), observing possible changes in the pattern of expression of the AQPs along the postnatal development. The animals will be killed by inhalation of carbon dioxide. After euthanasia, the epididymis will be removed and the different regions will be isolated. The biological material obtained will be processed in accordance with the routines of histological techniques and imunnohistochemistry to study the expression and localization of the AQPs 2 and 9. The results obtained will be analyzed and photographed in conventional light microscope

Mesenchymal stem cells (MSCs) are adult multipotent cells with fibroblastoid morphology and adherent to plastic. Furthermore, they can be obtained from different sources. Besides bone marrow, these cells are taken from umbilical cord blood, umbilical vein, saphenous vein, peripheral blood, arteries, liver and fetal pancreas, placenta, dental pulp and adipose tissue. MSCs derived from adipose tissue are important because of the abundant number of cells that can be obtained from this tissue, easy access and little discomfort to the patient. This study compared two techniques for obtaining MSCs from adipose tissue: mechanical dissociation (MD) and enzymatic digestion (ED). We also analyzed the inter-species cross-reactions using commercial monoclonal antibodies directed against surface antigens of stem cells from different species: mouse, horse, rabbit, monkey and human. We found that MD technique is favorable in relation to ED within 15 days of culture, and ED is more efficient in the first days of culture. The data also showed that MD causes less damage to cellular DNA. About inter-species cross-reactions, the monoclonal antibody A69 directed against stem cells from rabbits, which can be used in veterinary medicine, particularly in research involving horses

Osteosarcoma is the most common primary bone cancer in dogs. It affects most commonly dogs of big or giant breeds with 7 to 8 years and the etiology is unknown. Osteosarcoma is defined as a bone matrix-producing malignant mesenchymal tumor and has a predilection for the metaphyseal region of appendicular skeleton, however, it can affect axial skeleton and soft tissues. Distal radius is the most commonly affected site. The definitive diagnosis of osteosarcoma can be obtained with history, physical examination, radiographs and biopsy. Lung is the most common organ for metastatic disease. The mainly treatment for osteosarcoma is limb amputation and systemic chemotherapy for metastatic disease control. Limb-sparing surgery is a viable alternative to amputation for dogs with concomitant conditions that impede limb amputation. Palliative treatments for osteosarcoma have been studied such as local and systemic radiotherapy, immunotherapy and biphosphonates. This work will discuss how magnetic fields can be produced, either generated by magnets, natural, artificial, or even by an electric current going through a wire, as discovered by Oersted. Besides the theoretical content, experimental studies on magnetic induction and on the Laws of Faraday and Lenz will be performed. In the Magnetic Induction experiment, the electromotive force generated by varying the flow of the field B in a solenoid, depending on the variation of the current intensity and frequency associated with it will be measured; the experiment on the Laws of Faraday and Lenz the electromotive force produced by the relative movement of the magnet in relation to a coil. Thus, this study experimental verification of magnetic induction using solenoids and magnets; analysis of magnetic induction by Faraday's Law and Lenz's Law

O depósito de amilina é um achado histopatológico freqüente em pacientes portadores de diabetes melito tipo 2 (DM 2) e parece estar relacionado à disfunção da célula beta pancreática característica desta doença. Apenas as moléculas de amilina que sofrem agregação in vivo, tal como a amilina humana, são citotóxicas para as células beta, enquanto as variedades não agregantes, como a amilina de rato, não apresentam efeito deletério. Com o objetivo de compreender melhor os mecanismos de toxicidade associados à forma agregante da amilina, um projeto em andamento em nosso laboratório estudou, por micro-arranjos de cDNA, o perfil de genes modulados pela amilina humana em ilhotas pancreáticas murídeas, comparando duas situações específicas: ilhotas tratadas com fibrilas de amilina (“amilina madura”) e ilhotas tratadas com oligômeros de tamanho intermediário (“amilina fresca”), já que evidências recentes apontam para um efeito deletério apenas de oligômeros de amilina. As ilhotas foram isoladas a partir de ratos Wistar, mantidas em cultura por 24 horas e a seguir tratadas com 10

Os mecanismos pelos quais o hiper e hipotireoidismo afetam o osso durante a menopausa são pouco conhecidos. O objetivo deste estudo foi verificar a ação do hormônio triiodotironina (T3), em diferentes concentrações associado ao estrógeno (E2) em concentração infra-fisiológica (condição de menopausa), sobre a proliferação de células de rato ROS 17/2.8, através de uma curva de crescimento para obtenção do tempo de dobramento (TD). As células ROS 17/2.8 são consideradas modelo de células osteoblásticas e foram cultivadas em meio HAM F-12, e submetidas ao tratamento hormonal com as combinações das concentrações fisiológica, infrafisiológica e suprafisiológica de T3 e infra-fisiológica de E2. Utilizamos como grupo controle aquele tratado com as concentrações fisiológicas dos dois hormônios. Para cada concentração hormonal foram realizadas 5 repetições, e as células contadas manualmente nos dias 1, 3, 5 e 7, após o início da adição hormonal. A análise estatística utilizada foi a técnica da análise de variância não paramétrica, complementada com teste de comparações múltiplas. Baseando-se nos resultados obtidos, pode-se concluir que o crescimento das células ROS 17/2.8 é afetado pelos tratamentos hormonais, em especial quando a concentração infrafisiológica de E2 está associada à concentração suprafisiológica de T3, aumentando o tempo de dobramento celular. Este efeito poderia tornar o metabolismo do osso mais lento, prejudicando a neoformação óssea em pacientes menopausadas e hipertireoidéas

It is believed that epigenetic mechanisms such as DNA methylation are important for the tumorigenesis and maintenance of the altered state of tumor cells. DNA methylation occurs by the addition of a methyl group to carbon 5 of cytosine, catalyzed by the enzyme DNA methyl-transferase, which can change the expression of a gene, including the tumor suppressor genes. In human squamous cell carcinoma, several features have shown the etiological role of genes in tumor development. Among them, FOXE1 gene (forkhead box E1 - thyroid transcription factor) is presented with an important role in susceptibility to disease. Similarly the FOXE1 methylation pattern could alter the expression of this gene in dogs and predisposed to tumor on. Therefore, this study aims to investigate in dogs, the validity of the strategy employed in humans to analyze the FOXE1 methylation status. DNA extraction from fresh frozen tumoral samples was performed by Wizard Genomic® DNA Purification Kit. The methylation status was determined by MSP-PCR (methylation-specific polymerase chain reaction), using 2.0 ng of DNA treated with sodium bisulphate. One hundred micrograms of bisulphite-modified DNA was amplified using primers specific for either methylated or unmethylated DNA (primers sequences are available at http://pathology2.jhu.edu/pancreas/primer.pdf). The analysis of fragments was loaded on to 7% polyacrylamide gels and silver nitrate staining. In this stage of technical approach, 60% were FOXE1 hypermethylated. In conclusion, it was observed that the standard technique for assessing the methylation pattern of gene FOXE1 in humans can be used for the same evaluation in dogs. The correlation of these molecular data with clinical and histopathological parameters may have diagnostic and prognostic value and still be used as a tumor marker for therapeutic decision and surgical approach

A hipertensão arterial sistêmica é uma doença crônica degenerativa de etiologia multifatorial e poligênica, caracterizada pela presença de níveis tensionais elevados, sendo considerada um dos mais importantes fatores de risco para o desenvolvimento das doenças cardiovasculares. Estudos demonstram que cerca de um bilhão de indivíduos apresentam HAS em todo mundo, sendo esta patologia responsável por 7,1 milhões de morte a cada ano. Diversos fatores estão associados ao aumento da incidência da hipertensão arterial, como obesidade, dislipidemias, dieta rica em sal, alterações nos níveis hormonais e sedentarismo. Após o período da menopausa, observa-se significativa elevação nos valores de pressão arterial dessa população, sendo estes similares ou até maiores aos dos homens para esse mesmo período. O sistema renina-angiotensina (SRAA) desempenha importante papel no controle dos sistemas cardiovascular e renal e a angiotensina II é um potente vasoconstritor desse sistema, formado a partir da ação da Enzima Conversora de Angiotensina (ECA). Por outro lado, a recente descoberta da Enzima Conversora de Angiotensina 2 (ECA2), um homólogo da ECA, mudou todo o conceito do SRAA. A ECA 2 é responsável pela formação do peptídeo angiotensina- (1-7) a partir da clivagem do último aminoácido (fenilalanina) da angiotensina II. Recentes evidências têm demonstrado efeitos benéficos da angiotensina-(1-7) para as doenças cardiovasculares através da vasodilatação, efeito anti-arritmogênico, melhora na função contrátil pós-isquemia e inibição da proliferação celular do músculo liso vascular, sendo esses efeitos antagônicos àqueles observados pela angiotensina II. Hyphema is an intraocular hemorrhage that can result from many ocular and/or systemic disorders. It is an important clinical sign, since it may appear in the initial stages of some diseases or haematological disorders, acting as an initial marker for early diagnosis. The hematologic disturbances that predispose animals to present hyphema are: Thrombocytopenia, von Willebrand Disease, Scott Syndrome, Disseminated Intravascular Coagulation and Hyperviscosity Syndrome due to Multiple Myeloma. Each of the differential diagnoses due to hematological changes from the appearance of hyphema in dogs shows clinical relevance and particular ocular signs

Adequate testosterone levels are necessary for the development, growth and maintenance of the male reproductive system. Testosterone deficiency is common in men with diabetes in whom it may contribute to impaired performance, with consequent reduction of the activity of the androgen regulated organs, such as the prostate. However, little attention has been given to the plasma dihydrotestosterone (DHT) level, the most potent androgen, nor to the expression of the androgen receptor (AR), insulin-like growth factor type I (IGF-1) and receptor (IGF-1R) in target tissues. Here, we investigated the effect of type I diabetes mellitus on DHT plasma levels and on prostate AR, IGF-1 and IGF-1R expression during rat pubertal growth. Diabetes was induced in prepubertal male rats through administration of streptozotocin (STZ; 40 mg/kg). Diabetic, diabetic treated with insulin, and age-matched control animals were killed by overdoses of pentobarbital. The ventral prostatic lobe (VP) was dissected, weighed and processed for immunohistochemistry for AR, IGF-1 e IGF-1R; plasma T and DHT levels were also determined. Hyperglycemia at puberty reduced VP weight gain to about 50% and plasma T level to about 80% of the control levels. In contrast there were no changes in plasma DHT levels. Insulin replacement restored the VP weight gain, but not the plasma T levels, which remained 90% below the ones of controls. Immunohistochemistry showed that AR, IGF-1 and IGF-1R expression in the prostate epithelial cells did not change with hyperglycemia or insulin replacement. Thus, the AR expression in the prostate epithelial cells appears to be regulated by DHT, and to a minor extent it also controls glandular growth

Os distúrbios do sistema nervoso central mais comuns relatados no mundo são a ansiedade e a depressão, com grande incidência de casos nos quais aparecem juntos. Os tratamentos utilizados na clínica além de não serem efetivos na totalidade dos casos podem, por vezes, apresentar diversos efeitos não desejados. Assim, na busca novas drogas mais efetivas e com menor incidência de efeitos adversos, a Cananga odorata (Lam.) Hook. f. & Thomson aparece como uma alternativa. Esta espécie vegetal é utilizada na aromaterapia como na prevenção do estresse, ansiedade e depressão (BIRD, 2003). A busca de novas ferramentas terapêuticas depende do uso de animais de experimentação, e com as discussões na sociedade científica sobre o uso de animais, há uma maior preocupação em se desenvolver novas técnicas experimentais onde se possa aplicar os conceitos dos 3Rs objetivando a redução (Replacement) do número de animais, sua substituição (Reduction) e o refinamento (Refinement) das técnicas experimentais. Neste trabalho avaliou-se a atividade ansiolítica e antidepressiva do óleo essencial (OE) de Cananga odorata, em animais que foram sucessivamente submetidos a testes para avaliar a atividade motora, ansiolítica e antidepressiva. Camundongos machos foram tratados por via oral com OE nas doses de 0,1; 1, 10 e 100 mg/kg, e após 30 minutos foram submetidos ao teste do Campo Aberto, da Caixa Claro-Escuro e Suspensão pela Cauda, nesta ordem, com um intervalo de 30 minutos entre eles. Os resultados indicaram que a seqüência de testes é adequada para a avaliação destas atividades com economia de tempo, animais e drogas

The Canine Visceral Leishmaniasis is a chronic disease of endemic character, caused by Leishmania Chagasi in Americas. The inoculation of the promastigote form in the individual triggers a local and widespread immune reaction with formation of inflammatory infiltrates and deposition of immune complexes in tissues. Initial clinical symptoms of the disease are: weight reduction, hepatomegaly, splenomegaly and, according to the disease chronicity, signs such as alopecia, erythema, onychogryphosis, arthropathies, renal diseases, pyoderma, seborrheic dermatitis, muscle atrophy and Ocular diseases. Ocular diseases are often reported and are result of the direct parasitism or immune-mediated mechanisms caused by the disease. The Leishmania spp have greater affinity for the anterior segment, so that anterior uveitis is one of the most frequently diagnosed injuries. Blepharitis diffuse and Keratoconjunctivitis also appear as important ocular changes. In histological section, inflammatory infiltrates and macrophages with amastigote form are observed in all ocular tissues, with the exception of the retina and optical nerve. In the clinical analysis and disease diagnosis, should be considered the differential diagnosis, such as Ehrlichiosis and systemic hypertension, because these may cause some ocular manifestations similar to those observed in leishmaniasis

Há muito tempo pesquisadores estudam os malefícios à saúde causados pelo sedentarismo. Atualmente a inatividade física representa um grave problema de saúde pública. Segundo a Organização Mundial de Saúde – OMS (2009a), um estilo de vida sedentário é a principal causa de morte, doenças crônicos degenerativas e deficiências. Além disso, níveis insatisfatórios de atividade física associados a dietas hipercalóricas e com alto teor de gordura estão associados ao aumento de peso corporal, o que leva à desbalanços nos níveis lípedes circulantes, ou dislipidemia (DURSTINE, 2001). A disilipidemia está relacionada a alterações no metabolismo lipídico, incluindo níveis elevados de colesterol total, LDL colesterol, triglicerídeos e redução do HDL colesterol (DÂMASO, 2003). Em contrapartida, já é comprovado que a prática regular de atividade física aparece como uma ferramenta não medicamentosa na prevenção e tratamento de portadores de doenças crônicas. Estudos com indivíduos e grupos populacionais demonstram existir uma associação inversa entre os níveis de atividade física e a incidência de diversas doenças, como a hipertensão, a obesidade, os diabetes, a doença arterial coronariana e a depressão (NAHAS,2001). Sabe-se que a dislipidemia está associada a doenças de preocupação pública como obesidade, DM e doenças cardiovasculares. Por outro lado os benefícios da prática de atividade física já são consolidados. Assim, o objetivo deste estudo é verificar o efeito de um programa de atividade física moderado no perfil lipídico e na glicemia em mulheres acima de 50 anos através da análise do efeito de 7 meses de atividade física moderada nos seguintes parâmetros bioquímicos: colesterol, lipoproteína de baixa densidade (LDL), lipoproteína de alta densidade (HDL), triglicerídeos (TG) e glicemia.
