Background and purpose: The inflammation-resolving lipid mediator resolvin E1 (RvE1) effectively stops inflammation-induced bone loss in vivo in experimental periodontitis. It was of interest to determine whether RvE1 has direct actions on osteoclast (OC) development and bone resorption. Experimental approach: Primary OC cultures derived from mouse bone marrow were treated with RvE1 and analysed for OC differentiation, cell survival and bone substrate resorption. Receptor binding was measured using radiolabelled RvE1. Nuclear factor (NF)-kappa B activation and Akt phosphorylation were determined with western blotting. Lipid mediator production was assessed with liquid chromatography tandem mass spectrometry. Key results: OC growth and resorption pit formation were markedly decreased in the presence of RvE1. OC differentiation was inhibited by RvE1 as demonstrated by decreased number of multinuclear OC, a delay in the time course of OC development and attenuation of receptor activator of NF-kappa B ligand-induced nuclear translocation of the p50 subunit of NF-kappa B. OC survival and apoptosis were not altered by RvE1. Messenger RNA for both receptors of RvE1, ChemR23 and BLT(1) is expressed in OC cultures. Leukotriene B(4) (LTB(4)) competed with [(3)H] RvE1 binding on OC cell membrane preparations, and the LTB(4) antagonist U75302 prevented RvE1 inhibition of OC growth, indicating that BLT(1) mediates RvE1 actions on OC. Primary OC synthesized the RvE1 precursor 18R-hydroxy-eicosapentaenoic acid and LTB(4). Co-incubation of OC with peripheral blood neutrophils resulted in transcellular RvE1 biosynthesis. Conclusions and implications: These results indicate that RvE1 inhibits OC growth and bone resorption by interfering with OC differentiation. The bone-sparing actions of RvE1 are in addition to inflammation resolution, a direct action in bone remodelling.

The effects of single or repeated amphetamine (AMPH) treatment and those of AMPH withdrawals on immune-mediated lung inflammatory response were studied in rats. Two experiments were done. In the first, rats egg-albumin (OVA) sensitized were singularly or repeatedly (21 days, once daily) treated with AMPH (1.0 mg/kg) or with a similar number and volume of 0.9% NaCl. The OVA aerosol challenge was performed 12 h after the single or last repeated AMPH treatment and also 72 and 120 h after AMPH withdrawal. In the second experiment, the effects of reserpine (1.0 mg/kg/day for 5 consecutive days) on single AMPH actions on lung allergic response of rats were analyzed. Single and repeated AMPH treatment induced opposite actions on Bronchoalveolar lavage fluid (BAL) cellularity of allergic rats: single treatment decreased and repeated treatment increased the total number of cells as well as those of macrophages, neutrophils and eosinophils. Our data also showed that single but not repeated AMPH treatment decreased the number of neutrophils, monocytes and lymphocytes in the peripheral blood, and increased the total number of bone marrow cells in rats sensitized and challenged with OVA. Furthermore, it was shown that reserpine treatment precluded the effects of single AMPH treatment on cellular migration to the lung of OVA-sensitized and challenged rats. It was concluded that AMPH effects on lung inflammatory response and cell recruitment to the lung in allergic rats rely at least partially on corticosterone serum levels. The possible involvement of vesicular monoamine transporter type 2 (VMAT2) with these observed effects was discussed. (c) 2008 Elsevier B.V. All rights reserved.

Mineralization of the articular cartilage is a pathological condition associated with age and certain joint diseases in humans and other mammals. In this work, we describe a physiological process of articular cartilage mineralization in bullfrogs. Articular cartilage of the proximal and distal ends of the femur and of the proximal end of the tibia-fibula was studied in animals of different ages. Mineralization of the articular cartilage was detected in animals at 1 month post-transformation. This mineralization, which appeared before the hypertrophic cartilage showed any calcium deposition, began at a restricted site in the lateral expansion of the cartilage and then progressed to other areas of the epiphyseal cartilage. Mineralized structures were identified by von Kossa's staining and by in vivo incorporation of calcein green. Element analysis showed that calcium crystals consisted of poorly crystalline hydroxyapatite. Mineralized matrix was initially spherical structures that generally coalesced after a certain size to occupy larger areas of the cartilage. Alkaline phosphatase activity was detected at the plasma membrane of nearby chondrocytes and in extracellular matrix. Apoptosis was detected by the TUNEL (TDT-mediated dUTP-biotin nick end-labeling) reaction in some articular chondrocytes from mineralized areas. The area occupied by calcium crystals increased significantly in older animals, especially in areas under compression. Ultrastructural analyses showed clusters of needle-like crystals in the extracellular matrix around the chondrocytes and large blocks of mineralized matrix. In 4-year-old animals, some lamellar bone (containing bone marrow) occurred in the same area as articular cartilage mineralization. These results show that the articular cartilage of R. catesbeiana undergoes precocious and progressive mineralization that is apparently stimulated by compressive forces. We suggest that this mineralization is involved in the closure of bone extremities, since mineralization appears to precede the formation of a rudimentary secondary center of ossification in older animals.

The effect of Ketoconazole (KTZ) on the hamster experimental intratesticular paracoccidioidomycosis was studied employing different treatment schedules. KTZ long course treatment beginning at an early stage of the infection was effective in preventing fungal proliferation, dissemination to lymph nodes, spleen and kidneys, and in maintaining low levels of humoral and cellular specific immune responses. KTZ short course treatment starting at an advanced stage of disease resulted in a more severe histopathological picture without significant changes in the immunological profile. The drug prolonged the life span of hamsters infected with Paracoccidioides brasiliensis, but did not prevent mortality. Toxic necrosis of the bone marrow occurred in normal animals receiving 120 mg/kg/day of KTZ but with lower doses no morphologic alterations were observed in heart, lungs, kidneys, adrenals, spleen, liver, intestine or bone marrow. © 1984 Dr W. Immunohistochemistry was applied to identify the nature of the nucleated cells that accumulate in the vasa rectae of the corticomedullary junction in acute tubular necrosis. In all 6 cases studied, there were intravascular cells that reacted with monoclonal antibodies to erythroblast, macrophages, myeloid cells, T and B lymphocytes and rare megakaryocytes. The findings are consistent with the occurrence of intravascular haematopoiesis in the renal medulia in acute tubular necrosis.

The localization of peroxidase activity in different cell regions is used as a criterion for classifying the stage of maturity of mammalian mononuclear phagocytes, with a positive peroxidase reaction indicating the presence of monoblasts, promonocytes, monocytes, and macrophages. Peroxidase activity was observed ultrastructurally in the circulating blood of pacu fish (Piaractus mesopotamicus), identifying monoblasts, promonocytes, monocytes, and macrophages. These observations suggest that differentiation of mononuclear phagocytes occurs in the blood circulation of fish, whereas in mammals, monoblasts and promonocytes are detected in bone marrow, with only monocytes detected in circulating blood and differentiation into macrophages occurring in other body compartments.

Background: There is some evidence showing that cyclosporin A (CsA) and nifedipine (NIF) affect bone metabolism. The purpose of this work was to study the effects of CsA and NIF, given alone or concurrently, on alveolar bone of rats of different ages. Methods: Rats 15, 30, 60, and 90 days old were treated daily with 10 mg/kg body weight of CsA subcutaneously injected and/or 50 mg/kg body weight of NIF/day given orally for 60 days. Alveolar bone of the first lower molars was morphologically and stereologically evaluated in serial 5 μm bucco-lingual paraffin sections, stained with hematoxylin and eosin. Serum calcium and alkaline phosphatase levels were measured in all animals at the end of the experimental period. Results: Rats treated with CsA or NIF alone or CsA and NIF concurrently showed decreased alveolar bone density. CsA was more effective than NIF. A significant decrease in serum calcium was found only in animals treated with CsA or CsA/NIF. The results were similar regardless of age. Conclusions: These results indicate that the decrease in the alveolar bone volume in rats caused by CsA and NIF alone or concurrently is not age dependent. Furthermore, NIF (50 mg/kg) did not further increase the loss of alveolar bone volume induced by CsA (10 mg/kg).

The noxious effects of low or effective dose exposure to single or mixed pesticides on macrophage activity and the lymphohematopoietic organs were investigated. Male Wistar rats were orally exposed to dichlorvos, dicofol, endosulfan, dieldrin and permethrin, either as single or combined mixtures during a 28-day study containing eight groups: one group received a semipurified diet (non-treated); two groups received a semipurified diet containing low dose mixture (dieldrin 0.025 mg/kg, endosulfan, 0.6 mg/kg, dicofol 0.22 mg/kg, dichlorvos 0.23 mg/kg, permethrin 5 mg/kg) or an effective dose mixture (dichlorvos 2.3 mg/kg, dicofol 2.5 mg/kg, endosulfan 2.9 mg/kg, dieldrin 0.05 mg/kg and permethrin 25.0 mg/kg), respectively; the other five groups received a semipurified diet containing each single pesticide in effective doses. At sacrifice, the thymus, spleen, mesenteric lymph nodes, Payer's patches and bone marrow were removed for histological analysis. Peritoneal macrophages were obtained to determine the phagocytosis and spreading indexes and tumoral necrosis factor alpha (TNF-α), nitric oxide (NO) and H2O2 production. Exposure to pesticide mixtures did not alter the percentage of macrophage phagocytosis and spreading, TNF-α production or the NO and H2O2 release when compared to the non-treated group. Neither was there any apparent evidence that a pesticide mixture at low or effective doses altered the histological structure of the lymphohematopoietic organs. The findings indicate that short-term treatment with pesticide mixtures did not induce an apparent immunotoxic effect in male Wistar rats. © 2013 Copyright Taylor and Francis Group, LLC.

This study was undertaken to understand how Lentinula edodes modulates in vivo mutagenesis induced by alkylating agents in bone marrow and peripheral blood as described in our previous article. Male Swiss mice were pretreated for 15 consecutive days with aqueous extracts prepared from L. edodes, after which, the number of circulating blood cells, normal erythroid bone marrow cell cycling, and phagocytosis of micronucleated reticulocyte (MNRET) and activation of spleen macrophages were assessed. The results indicate that the antimutagenicity seen in bone marrow and peripheral blood is exerted by distinct compounds with different actions. The antimutagenic effect in bone marrow is exerted by compounds subject to degradation at deep-freeze storage temperature of -20 C. On the other hand, compounds responsible for antimutagenicity in peripheral blood are not subject to degradation at -20 C. The results also indicate that the antimutagenic action in peripheral blood leading to the reduction of circulating MNRET occurs in the spleen primarily through a phagocytic activity due to higher macrophage numbers and probably not due to the enhanced activation state of individual cells. © Mary Ann Liebert, Inc.

Diversos estudos sugerem que a tetraciciclina semi-sintética minociciclina e o transplante de células mononucleares da medula óssea (CMMOs) induzem neuroproteção em modelos experimentais de acidente vascular encefálico (AVENC). No entanto, poucos investigaram, comparativamente, os efeitos destas duas abordagens terapêuticas após AVENC induzido por microinjeções de endotelina – 1 (ET -1). Nesta dissertação, objetivou-se comparar os efeitos do bloqueio microglial com minociclina com os obtidos pelo transplante intraestriatal de CMMOs na fase aguda após acidente vascular encefálico experimental, sobre a área de lesão, neuroproteção, apoptose de recuperação funcional. Ratos machos adultos, da raça Wistar, pesando entre 250 e 350g, foram distribuídos em quatro grupos experimentais: controle (chamado de Salina) - isquêmico tratado com salina (N=4), isquêmico tratado com minociclina (N=4), isquêmico tratado com CMMOs (N=3) e doador de CMMOs (N=2). Testes comportamentais foram realizados em 1, 3 e 7 dias pós-isquemia para avaliar a recuperação funcional entre os grupos. Animais tratados com minociclina receberam 2 doses diárias de 50mg/kg nos 2 primeiros dias, e 5 aplicações únicas de 25mg/kg (i.p) nos dias subsequentes até o sexto dia após a indução isquêmica. 1x106 de CMMOs foram obtidas de ratos da mesma linhagem e transplantadas diretamente no estriato, 24h após a lesão isquêmica. Todos os animais foram perfundidos 7 dias após a indução isquêmica. Secções coronais foram coradas por violeta de cresila para análise histopatológica geral, e por imunohistoquímica para a identificação de corpos neuronais (neuN), microglia/macrófagos ativados (ED1) e células apoptóticas (Caspase-3). A análise histopatológica geral mostrou grande palor, perda tecidual e intensa ativação microglial/ macrofágica no estriato de animais tratados com solução salina estéril. O tratamento com CMMO foi mais eficaz do que a minociclina (P<0,05, ANOVA-Tukey) na redução do número de microglia/macrófagos ativados (salina 276,3 ± 9,3); CMMOs 133,8 ± 6,8) e minociclina 244,6 ± 7,1). CMMOs e minociclina reduziram a área de lesão, em 67,75% e 69,1%, respectivamente. Os dois tratamentos promoveram o mesmo nível de preservação neuronal (p< 0,05) em relação ao controle, 61,3 ± 1,5); 86,8 ± 3,4) e 81 ± 3,4). As CMMOs reduziram de forma mais eficaz (p<0,01) o número de células apoptóticas em relação à minociclina e grupo controle (26,5 ± 1,6); 13,1 ± 0,7) e 19,7 ± 1,1). Ambas as abordagens terapêuticas promoveram recuperação funcional dos animais isquêmicos. Os resultados sugerem que o tratamento com CMMOs é mais eficaz na modulação da resposta microglial e na diminuição da apoptose do que o tratamento com minociclina, apesar de ambos serem igualmente eficazes para indução da neuroproteção. Estudos futuros devem investigar se o tratamento com minociclina associado ao transplante de CMMOs produzem efeitos sinérgicos, o que poderia amplificar os níveis de neuroproteção observados.

A lobomicose é uma infecção subcutânea crônica, granulomatosa, causada pela implantação traumática do fungo Lacazia loboi nos tecidos cutâneo e subcutâneo. Ocorre predominantemente na região Amazônica e atinge qualquer grupo populacional. Histologicamente, observa-se reação inflamatória crônica caracterizada por intensa histiocitose e fibroplasia, abundante número de macrófagos, células gigantes multinucleadas do tipo corpo estranho e presença de considerável número de células leveduriformes. Os macrófagos são células fagocíticas que participam do reconhecimento e da resposta a patógenos através da fagocitose, da apresentação de antígenos aos linfócitos T e da produção de citocinas. As células de Langerhans (LC) são um grupo de Células dendríticas (CD) derivadas da medula óssea situadas principalmente em uma camada suprabasal da epiderme. Estudos envolvendo a interação fungo-hospedeiro na doença de Jorge Lobo são escassos. Assim, Este estudo é um passo importante para o melhor entendimento da biologia e patogenia do L. loboi, e para o estudo da imunopatologia da interação patógeno versus hospedeiro desta doença emergente e pouco conhecida. O objetivo do presente trabalho foi analisar a interação in vitro entre macrófagos peritoneais não ativados e/ou LC, isolados de camundongos BALB/c, com L. loboi recém-isolado de pacientes com doença de Jorge Lobo, bem como determinar os índices de infecção, fagocitose e fusão, e medir a produção das citocinas TNF-α, IL-4, IL-6, IL-10 e IL-12. Os resultados demonstraram que L. loboi é fagocitado por macrófagos, mas não por LC. O índice de infecção na interação entre macrófagos e L. loboi foi semelhante à interação entre macrófagos, LC e L. loboi em todos os tempos analisados. A média do número de fungos por macrófago também foi praticamente igual entre as interações e ao longo do tempo, variando de 1,2 a 1,6 fungos/macrófagos. Não houve a formação de células gigantes em macrófagos cultivados ou LC cultivadas isoladamente e em nenhum dos co-cultivos. Não houve diferença significante na produção de IL-4, IL-2 e IL-10 nas interações estudadas. Os níveis de TNF-α diminuem ao longo do tempo na interação entre macrófagos e L. loboi, enquanto a adição de LC induz aumento da produção de TNF-α, principalmente após 48 horas. LC modulam negativamente a produção de IL-6 por macrófagos e L. loboi também inibem essa produção por macrófagos isoladamente ou em co-cultivo com LC. L. loboi estimulam significativamente a produção de IL-12 por macrófagos co-cultivados com LC, mas não em LC ou macrófagos isoladamente.
