PURPOSE: 3'-deoxy-3'-[(18)F]fluorothymidine ([(18)F]FLT), a cell proliferation positron emission tomography (PET) tracer, has been shown in numerous tumors to be more specific than 2-deoxy-2-[(18)F]fluoro-D-glucose ([(18)F]FDG) but less sensitive. We studied the capacity of a nontoxic concentration of 5-fluoro-2'-deoxyuridine (FdUrd), a thymidine synthesis inhibitor, to increase uptake of [(18)F]FLT in tumor xenografts. METHODS: The duration of the FdUrd effect in vivo on tumor cell cycling and thymidine analogue uptake was studied by varying FdUrd pretreatment timing and holding constant the timing of subsequent flow cytometry and 5-[(125)I]iodo-2'-deoxyuridine biodistribution measurements. In [(18)F]FLT studies, FdUrd pretreatment was generally performed 1 h before radiotracer injection. [(18)F]FLT biodistributions were measured 1 to 3 h after radiotracer injection of mice grafted with five different human tumors and pretreated or not with FdUrd and compared with [(18)F]FDG tumor uptake. Using microPET, the dynamic distribution of [(18)F]FLT was followed for 1.5 h in FdUrd pretreated mice. High-field T2-weighted magnetic resonance imaging (MRI) and histology were used comparatively in assessing tumor viability and proliferation. RESULTS: FdUrd induced an immediate increase in tumor uptake of 5-[(125)I]iodo-2'-deoxyuridine, that vanished after 6 h, as also confirmed by flow cytometry. Biodistribution measurements showed that FdUrd pretreatment increased [(18)F]FLT uptake in all tumors by factors of 3.2 to 7.8 compared with controls, while [(18)F]FDG tumor uptake was about fourfold and sixfold lower in breast cancers and lymphoma. Dynamic PET in FdUrd pretreated mice showed that [(18)F]FLT uptake in all tumors increased steadily up to 1.5 h. MRI showed a well-vascularized homogenous lymphoma with high [(18)F]FLT uptake, while in breast cancer, a central necrosis shown by MRI was inactive in PET, consistent with the histomorphological analysis. CONCLUSION: We showed a reliable and significant uptake increase of [(18)F]FLT in different tumor xenografts after low-dose FdUrd pretreatment. These results show promise for a clinical application of FdUrd aimed at increasing the sensitivity of [(18)F]FLT PET.

Sebastià Juan Arbó (1902-1984) era un escriptor en llengua catalana fins a la Guerra Civil espanyola. Arran del desenllaç de la guerra, Arbó va esdevenir un escriptor tant en llengua catalana com en llengua castellana. Aquest pas d’una llengua a una altra va tenir un pas intermedi: l’autotraducció. A partir de la seva primera autotraducció al castellà (Tierras del Ebro, 1940), Sebastià Juan Arbó es va introduir en les lletres castellanes. Aquesta tasca autotraductora, que encetà el 1940, no l’abandonà en la resta de la seva obra catalana i mai no l’exercí en la seva obra escrita originalment en llengua castellana.Amb aquest treball de recerca es vol establir, en primer lloc, un mètode d’anàlisi per a l’autotraducció de Terres de l’Ebre, de Sebastià Juan Arbó, que pugui ser exportable a l’anàlisi d’altres autotraduccions, i, en segon lloc, analitzar un fragment de les primeres edicions en català (1932) i castellà (1940) de Terres de l’Ebre, que permeti reconèixer-hi les tendències autotraductores de Sebastià Juan Arbó.

The development of RGD-based antagonist of αvβ3 integrin receptor has enhanced the interest in PET probes to image this receptor for the early detection of cancer, to monitor the disease progression and the response to therapy. In this work, a novel prosthetic group (N-(4-fluorophenyl)pent-4-ynamide or FPPA) for the (18)F-labeling of an αvβ3 selective RGD-peptide was successfully prepared. [(18)F]FPPA was obtained in three steps with a radiochemical yield of 44% (decay corrected). Conjugation to c(RGDfK(N3)) by the Cu(II) catalyzed Huisgen azido alkyne cycloaddition provided the [(18)F]FPPA-c(RGDfK) with a radiochemical yield of 29% (decay corrected), in an overall synthesis time of 140min.

Tämän toiminnallisen opinnäytetyön tarkoitus oli toteuttaa suun terveyden edistämisnäytelmä. HUS:n Kipinä ja Kunttari kuntoutumisosastoille, joiden asukkaat ovat 14-18-vuotiaita mielenterveyskuntoutujia. Opinnäytetyön tavoitteena oli lisätä nuorten suun terveydenlukutaitoa. Näytelmän tavoitteena oli innostaa nuoria suun terveydenhoitoon. Terveydenedistämismenetelmänä käytettiin draamaa. Lukuisat tutkimukset viittaavat siihen, että nuorten elintavat ovat muuttuneet epäterveellisempään suuntaan. Alueelliset terveyserot heijastuvat myös suunterveyteen. Helsingissä on moniongelmaisia perheitä, nuoria ja lapsia suhteellisesti enemmän kuin muualla Suomessa. Opinnäytetyössä sovellettiin Precede-Proceed -terveyden edistämismallia, joka näkyy työmme suunnittelu-, toteutus- ja arviointivaiheessa. Suun terveydenedistämisnäytelmä ”Liian hapokasta” suunniteltiin ja toteutettiin opinnäytetyöryhmässä yhteistyössä työelämäyhteistyökumppanin kanssa. Näytelmä oli luonteeltaan osallistava. Tämä tarkoittaa sitä, että yleisö sai itse vaikuttaa näytelmän kulkuun valitsemalla kahdesta eri vaihtoehdosta sopivimman. Näytelmän keskeiset aihealueet valittiin näyttöön ja kokemukseen perustuen ja ne olivat: suun hoito, tupakointi, iensairaudet, karies ja ravitsemus. Tälle työlle sovellus- ja jatkomahdollisuudet voisivat olla tutkia millaisen terveyden lukutaidon nuoret saavuttivat. Työmme voi toimia myös esimerkkinä tulevien terveyden edistämisnäytelmien toteutuksille ja käsikirjoituksille.

DNA that survives in museum specimens, bones and other tissues recovered by archaeologists is invariably fragmented and chemically modified. The extent to which such modifications accumulate over time is largely unknown but could potentially be used to differentiate between endogenous old DNA and present-day DNA contaminating specimens and experiments. Here we examine mitochondrial DNA sequences from tissue remains that vary in age between 18 and 60,000 years with respect to three molecular features: fragment length, base composition at strand breaks, and apparent C to T substitutions. We find that fragment length does not decrease consistently over time and that strand breaks occur preferentially before purine residues by what may be at least two different molecular mechanisms that are not yet understood. In contrast, the frequency of apparent C to T substitutions towards the 5'-ends of molecules tends to increase over time. These nucleotide misincorporations are thus a useful tool to distinguish recent from ancient DNA sources in specimens that have not been subjected to unusual or harsh treatments.
