The influence of structural features of tropical river humic substances (HS) on their capability to reduce mercury(II) in aqueous solutions was studied. The HS investigated were conventionally isolated from Rio Negro water-Amazonas State/Brazil by means of the collector XAD 8. In addition, the isolated HS were on-line fractionated by tangential-flow multistage ultrafiltration (nominal molecular-weight cut-offs: 100, 50, 30, 10, 5 kDa) and characterized by potentiometry and UV/VIS spectroscopy. The reduction of Hg(II) ions to elemental Hg by size-fractions of Rio Negro HS was assessed by cold-vapor AAS (CVAAS). UV/VIS spectrometry revealed that the fractions of high molecular-size (F-1 > 100 kDa and F-2: 50-100 kDa) have a higher aromaticity compared to the fractions of small molecular-size (F-5: 5-10 kDa, F-6: < 5 kDa). In contrast, the potentiometric study showed different concentration of functional groups in the studied HS fractions. The reduction of Hg(II) by aquatic HS fractions at pH 5 proceeded in two steps (I, II) of slow first order kinetics (t(1/2) of I: 160 min, t(1/2) of II: 300 min) weakly influenced by the molecular-size, in contrast to the differing degree of Hg(II) reduction (F-5 > F-2 > > F-1 > F-3 > F-4 > > F-6). Accordingly, Hg(II) ions were preferably reduced by HS molecules having a relatively high ratio of phenolic/carboxylic groups and a small concentration of sulfur. From these results a complex 'competition' between reduction and complexation of mercury(II) by aquatic HS occurring in tropical rivers such as the Rio Negro can be suggested. (C) 2003 Elsevier B.V. The aim of the present work was to carry out experimental comparison between humic substances (HS) and representative α-amino acids (methionine, methionine sulfoxide and cysteine hydrochloride) in relation to the complexation of biologically active trace elements (Al, Cu, Pb, Mn, Zn, Cd and Ni). A mobile time-controlled tangential-flow UF technique was applied to differentiate between HS-metal and α-aminoacids-metal complexes. Metal determinations were conventionally carried out using a ICP-OES. The results showed that HS may be considered as a selective complexing agents with higher metal bonding capability in relation to Al, Cu and Pb, the fact that may be clinically important.

Purpose: The purpose of this paper is to study the interactions of sedimentary humic substances (SHS) from a sugarcane cultivation area with Cu(II) and Cr(III) and to evaluate the occurrence of these metals in the pore water and SHS. Materials and methods: For this study, the northwestern region of the State of São Paulo, Brazil, which is considered the region with the highest production of sugar cane in the state, was selected. Samples of sediment were collected from four sampling sites in the Preto, Turvo, and Grande rivers. The SHS and pore water were extracted from the sediment using the method suggested by the International Humic Substances Society and centrifugation, respectively. The complexing capacity (CC) of the SHS for Cu(II) and Cr(III) was determined by individually titrating these metals with an ultrafiltration system using tangential flow. The total concentrations of Cr and Cu were determined for the pore water, sediments, and humic substances with graphite furnace atomic absorption spectrometry and Zeeman background correction after an acid digestion, according to the methods described in US EPA Method 3050B. Results and discussion: The SHS from a site in the Turvo River, which is typically cultivated with sugarcane, possessed the highest concentration of Cu bound to SHS (25.0%), the largest CC (0.63 mmol Cu g-1 HS) and the highest concentration of this metal in the pore water (1.38 mg Cu Kg-1 sed.). For Cr, the SHS collected from a location on the Preto River dam had the largest CC (0.90 mmol Cr g-1 HS) and the lowest Cr content in the pore water (0.29 mg Cr Kg-1 sed.), indicating that there was an inverse relationship between the CC and the concentration of metal available in the pore water. Conclusions: Sedimentary humic substances might be one of the regulatory factors controlling the availability of Cu and Cr in the sediments found in a typical region that has been planted with sugarcane. Distinct behaviors were observed between the two elements investigated; higher CC and a larger fraction of Cu(II) were found in the pore water of samples originating from sugarcane crops. Micro-organismos fotossintetizantes, incluído aqui o gênero Arthrospira, vêm sendo amplamente produzidos em larga escala em vários países, detendo um mercado que gera mais de 1 bilhão de dólares ao ano. A produção industrial utiliza grande volume de água com alta concentração salina para produzir milhares de toneladas de biomassa microalgal. É crescente a utilização de tratamento de águas por processo de separação por membranas, demonstrando ser uma técnica que gera água de ótima qualidade, de instalação compacta e de fácil automação. No presente trabalho, foi avaliada esta tecnologia para o reaproveitamento do meio de cultura em novos cultivos de micro-organismos fotossintetizantes, visando contribuir para a sustentabilidade deste processo produtivo. O efluente do cultivo de Arthrospira platensis oriundo de processo descontínuo em minitanques foi submetido a tratamento por membranas de filtração tangencial, incluindo microfiltração (MF) (porosidades de 0,65 µm e de 0,22 µm) e ultrafiltração (UF) (peso molecular de corte de 5.000 Da), em pressões transmembrana (TMP) de 22,5 a 90 kPa. Os processos de MF levaram a reduções médias de 53,9±1,3 % e 93,1±1,1 % de matéria orgânica natural (NOM) e pigmentos nos meios residuais, respectivamente. Com o uso de processos de UF, cujos meios foram previamente tratados por MF (0,22 µm e 22,5 kPa), as reduções médias de NOM e pigmentos foram de 57,2±0,5 % e 94,0±0,8 %, respectivamente. Os processos de MF com TMP de 22,5 kPa levaram a concentrações celulares máximas (Xm) equivalentes às obtidas com meio novo. O uso de membrana de 0,65 µm e TMP de 22,5 kPa levou a uma perda média de 2,9 %, 22,7 % e 16,4% dos nutrientes carbonato, fosfato e nitrato, respectivamente, mas a correção desses valores aos mesmos do meio padrão levou à obtenção dos mais altos valores de Xm (3586,6±80 mg L-1), produtividade em células (505,0±11,6 mg L-1 d-1) e fator de conversão de nitrogênio em células (29,6±0,7 mg mg-1). O teor protéico da biomassa foi estatisticamente igual ao da biomassa obtida de cultivo com meio padrão novo. Os dados deste trabalho evidenciam que processos de filtração por membrana são promissores para o reuso de meio de micro-organismos fotossintetizantes.

Dewatering of microalgal culture is a major bottleneck towards the industrial-scale processing of microalgae for bio-diesel production. The dilute nature of harvested microalgal cultures poses a huge operation cost to dewater; thereby rendering microalgae-based fuels less economically attractive. This study explores the influence of microalgal growth phases and intercellular interactions during cultivation on dewatering efficiency of microalgae cultures. Experimental results show that microalgal cultures harvested during a low growth rate phase (LGRP) of 0.03 d-1 allowed a higher rate of settling than those harvested during a high growth rate phase (HGRP) of 0.11 d-1, even though the latter displayed a higher average differential biomass concentration of 0.2 g L-1 d-1. Zeta potential profile during the cultivation process showed a maximum electronegative value of -43.2 ± 0.7 mV during the HGRP which declined to stabilization at -34.5 ± 0.4 mV in the LGRP. The lower settling rate observed for HGRP microalgae is hence attributed to the high stability of the microalgal cells which electrostatically repel each other during this growth phase. Tangential flow filtration of 20 L HGRP culture concentrated 23 times by consuming 0.51 kWh/m3 of supernatant removed whilst 0.38 kWh/m3 was consumed to concentrate 20 L of LGRP by 48 times.

Filtration and cross-flow ultrafiltration techniques were used to separate culture media of Prorocentrum donghaiense at the exponential growth, stationary and decline stages into < 0.45 mu m filtrate, 100 kDa-0.45 mu m, 10-100 kDa and 1-10 kDa retentate and < 1 kDa ultrafiltrate fractions. The fluorescence. properties of different molecular weights of dissolved organic matter (DOM) were measured by excitation-emission matrix spectra. Protein-like and humic-like fluorophores were observed in the DOM produced by P. donghaiense. The central positions of protein-like fluorophores showed a red shift with prolonged growth duration, shifting from tyrosine-like properties at the exponential growth stage to tryptophan-like properties at the stationary and decline stages. The excitation wavelengths of protein-like fluorophores exhibited some change in the exponential growth and stationary stages with increased molecular size, but showed little change in the decline stage. However, the emission wavelengths in the decline stage exhibited a blue shift. Very distinct C type and A type peaks in humic-like fluorophores were observed. With a prolonged culture time, the intensities of both of the peaks became strong and the excitation wavelengths of peak A showed a red shift, while the A:C ratios fell. More than 94% of fluorescent DOM was in the lower than 1 kDa molecular weight fraction.

Les polymorphonucléaires neutrophiles (PMNs) représentent une arme primordiale dans la défense contre divers agents pathogènes; notamment les bactéries, les champignons, les cellules tumorales de même que les cellules infectées par des virus. Cependant, certaines pathologies reliées à l’inflammation chronique soulèvent l’implication des neutrophiles notamment dans l’arthrite rhumatoïde. La réponse inflammatoire persistante générée par l’activation et la survie des neutrophiles engendre une destruction des tissus environnants suite à la sécrétion non contrôlée de leurs produits cytotoxiques. Même si l’activation chronique des neutrophiles est néfaste dans plusieurs pathologies, elle pourrait s’avérer un bon outil en cas de neutropénie, comme c’est souvent le cas les patients ayant reçu des traitements de chimiothérapie. Ce projet fait suite aux travaux doctoraux de Lagraoui (1999). Il vise à identifier le(s) facteur(s) du liquide synovial qui augmente la survie des neutrophiles ainsi que le mécanisme d’action impliqué dans ce processus. Similairement au facteur semi-pur isolés par Lagraoui (1999), le milieu conditionné concentré (MCC) augmente la survie des PMNs de 75% (39% ± 9.5 vs 68% ± 2.5, p<0.01). Suivant le séquençage du MCC parallèlement au facteur semi-pur actif, deux protéines ont été identifiées à la fois dans le MCC et dans le facteur semi-pur soient : l’albumine et la fétuine. Notre projet vise donc à comparer les effets de l’albumine et de la fétuine à ceux du GM-CSF dans l’optique d’une thérapie alternative au GM-CSF en tant qu’adjuvant de chimiothérapie. La présence d’albumine, de fétuine ou de GM-CSF chez les PMNs incubés 24 heures avec la Mutamycin® induit une diminution du nombre de cellules en apoptose par rapport à la Mutamycin® (Ctrl : 43% ± 10; A : 74% ± 3; F : (82% ± 6 et GM : 74% ± 7; p<0.01). L’effet de l’albumine dépend de la voie de la kinase PI3 mais également celle la kinase ERK, alors que celle de la fétuine dépend de la kinase PI3. Similairement l’EPO, l’albumine et la fétuine supporte la différentiation des HSCs en précurseurs érythrocytaires de type BFU-E. Dans un modèle murin de chiomioprotection, l’albumine augmente la concentration cellulaire rapport au groupe contrôle des leukocytes de la rate (66 ±8 x106c/ml vs 81 ±16 x106c/ml) et du sang (3.6 ±0.4 x106c/ml vs 5.7 ±2.3 x106c/ml). Donc, in vitro, l’albumine et la fétuine sont comparables au GM-CSF au niveau fonctionalité et mécansimes d’action. Cependant, vu leur manque de spécificité, l’application thérapeutique en tant qu’adjuvant de chiomiothérapie de l’albumine et la fétuine est peu prometteuse. Par contre, les maladies dégénératives et les évènements ischémiques pourraient s’avérer de bonnes cibles thérapeutiques, principalement pour l’albumine.

This paper reports the results of the investigations of 2006-2007 on the distribution and migration forms of artificial radionuclides and chemical elements in the Ob-Irtysh water system. Three regions were studied. One of them is a local segment of the Ob River upstream from the confluence with the Irtysh River; its investigation allowed us to estimate the general radioecological state of the aquatic environment affected by the activity of the Tomsk 7 plant. The second region is a local segment of the Irtysh River upstream from its confluence with the Ob River, where the influence of emissions from the NPO Mayak could be estimated. The third region is the water area of the Ob River after its confluence with the Irtysh River. It characterizes the real level of radioactive and chemical contamination of the middle reaches of the Ob River. In order to explain horizontal variations in the distribution of radionuclides in the upper layer of bottom sediments collected at various sites, the results of sorption-kinetic experiments with radioactive tracers in the precipitate-solution system were used. The investigation of the migration forms of trace elements and radionuclides occurring in river water was based on the method of tangential-flow membrane filtration. Chemical element contents were determined in 400-ml water samples. A set of Millipore polysulfone membranes with pore sizes of 8, 1.2, 0.45, 0.1, and 0.025 µm was employed. Taking into account the ultralow specific concentrations of radionuclides in the water, they were analyzed in 300-500 litre samples using Millipore polysulfone membranes with pore sizes of 0.45 µm and 15 kDa. This allowed us to estimate the percentages of cesium-137 and plutonium-239, 240 in the suspended particulate fraction, colloids, and dissolved species.

Nanoparticles offer an ideal platform for the delivery of small molecule drugs, subunit vaccines and genetic constructs. Besides the necessity of a homogenous size distribution, defined loading efficiencies and reasonable production and development costs, one of the major bottlenecks in translating nanoparticles into clinical application is the need for rapid, robust and reproducible development techniques. Within this thesis, microfluidic methods were investigated for the manufacturing, drug or protein loading and purification of pharmaceutically relevant nanoparticles. Initially, methods to prepare small liposomes were evaluated and compared to a microfluidics-directed nanoprecipitation method. To support the implementation of statistical process control, design of experiment models aided the process robustness and validation for the methods investigated and gave an initial overview of the size ranges obtainable in each method whilst evaluating advantages and disadvantages of each method. The lab-on-a-chip system resulted in a high-throughput vesicle manufacturing, enabling a rapid process and a high degree of process control. To further investigate this method, cationic low transition temperature lipids, cationic bola-amphiphiles with delocalized charge centers, neutral lipids and polymers were used in the microfluidics-directed nanoprecipitation method to formulate vesicles. Whereas the total flow rate (TFR) and the ratio of solvent to aqueous stream (flow rate ratio, FRR) was shown to be influential for controlling the vesicle size in high transition temperature lipids, the factor FRR was found the most influential factor controlling the size of vesicles consisting of low transition temperature lipids and polymer-based nanoparticles. The biological activity of the resulting constructs was confirmed by an invitro transfection of pDNA constructs using cationic nanoprecipitated vesicles. Design of experiments and multivariate data analysis revealed the mathematical relationship and significance of the factors TFR and FRR in the microfluidics process to the liposome size, polydispersity and transfection efficiency. Multivariate tools were used to cluster and predict specific in-vivo immune responses dependent on key liposome adjuvant characteristics upon delivery a tuberculosis antigen in a vaccine candidate. The addition of a low solubility model drug (propofol) in the nanoprecipitation method resulted in a significantly higher solubilisation of the drug within the liposomal bilayer, compared to the control method. The microfluidics method underwent scale-up work by increasing the channel diameter and parallelisation of the mixers in a planar way, resulting in an overall 40-fold increase in throughput. Furthermore, microfluidic tools were developed based on a microfluidics-directed tangential flow filtration, which allowed for a continuous manufacturing, purification and concentration of liposomal drug products.

The axisymmetric steady laminar compressible boundary layer swirling flow of a gas with variable properties in a nozzle has been investigated. The partial differential equations governing the non-similar flow have been transformed into new co-ordinates having finite ranges by means of a transformation which maps an infinite range into a finite one. The resulting equations have been solved numerically using an implicit finite-difference scheme. The computations have been carried out for compressible swirling flow through a convergent conical nozzle. The results indicate that the swirl exerts a strong influence on the longitudinal skin friction, but its effect on the tangential skin friction and heat transfer is comparatively small. The effect of the variation of the density-viscosity product across the boundary layer is appreciable only at low-wall temperature. The results are in good agreement with those of the local-similarity method for small values of the longitudinal distance.

The paper deals with the flow and heat-transfer problem of a steady axisymmetric laminar incompressible boundary layer swirling flow of a fluid through a conical hydrocyclone. The implicit finitedifference scheme is used to solve the partial differential equations governing the flow. The effect of swirl is found to be more pronounced on the longitudinal skin friction than on the tangential skin friction and heat transfer. The skin friction and heat transfer increase with swirl or with longitudinal distance. Swirl also gives rise to velocity overshoot in the longitudinal velocity profiles and the magnitude of the velocity overshoot increases as the swirl parameter increases. The results are found to be in good agreement with those of the local nonsimilarity and momentum integral methods but they differ appreciably from those of the local similarity method except for the longitudinal skin friction which is fairly in good agreement with that of the local similarity method.Die Arbeit beschäftigt sich mit der Strömung und dem Wärmeübergang in einem konischen Zyklon unter der Voraussetzung stationärer, achsensymmetrischer, laminarer, inkompressibler Grenzschichtströmung. Ein implizites Differenzenverfahren wird benutzt, um die partiellen Differentialgleichungen zu lösen. Der Einfluß des Dralls ist besonders ausgeprägt auf die longitudinale Komponente der Oberflächenreibung, weniger dagegen bei der tangentialen Komponente und beim Wärmeübergang. Die Oberflächenreibung und der Wärmeübergang nehmen zu mit dem Drall, sowie mit dem longitudinalen Abstand. Der Drall erzeugt ein Überschießen der Geschwindigkeit in der longitudinalen Abstand. Der Drall erzeugt ein Überschießen der Geschwindigkeit in der Längsrichtung. Die Größe des Überschusses nimmt mit wachsendem Drallparameter zu. Die Resultate stimmen gut mit den Ergebnissen der Theorie der lokalen Nichtähnlichkeit und der Impulsintegralmethode überein. Dagegen weichen sie mit Ausnahme der longitudinalen Komponente der Oberflächenreibung beträchtlich von den Resultaten der Theorie der lokalen Ähnlichkeit ab.

The modification of the axisymmetric viscous flow due to relative rotation of the disk or fluid by a translation of the boundary is studied. The fluid is taken to be compressible, and the relative rotation and translation velocity of the disk or fluid are time-dependent. The nonlinear partial differential equations governing the motion are solved numerically using an implicit finite difference scheme and Newton's linearisation technique. Numerical solutions are obtained at various non-dimensional times and disk temperatures. The non-symmetric part of the flow (secondary flow) describing the translation effect generates a velocity field at each plane parallel to the disk. The cartesian components of velocity due to secondary flow exhibit oscillations when the motion is due to rotation of the fluid on a translating disk. Increase in translation velocity produces an increment in the radial skin friction but reduces the tangential skin friction.

The stability of the Hagen-Poiseuille flow of a Newtonian fluid in a tube of radius R surrounded by an incompressible viscoelastic medium of radius R < r < HR is analysed in the high Reynolds number regime. The dimensionless numbers that affect the fluid flow are the Reynolds number Re = (rho VR/eta), the ratio of the viscosities of the wall and fluid eta(r) = (eta(s)/eta), the ratio of radii H and the dimensionless velocity Gamma = (rho V-2/G)(1/2). Here rho is the density of the fluid, G is the coefficient of elasticity of the wall and V is the maximum fluid velocity at the centre of the tube. In the high Reynolds number regime, an asymptotic expansion in the small parameter epsilon = (1/Re) is employed. In the leading approximation, the viscous effects are neglected and there is a balance between the inertial stresses in the fluid and the elastic stresses in the medium. There are multiple solutions for the leading-order growth rate s((0)), all of which are imaginary, indicating that the fluctuations are neutrally stable, since there is no viscous dissipation of energy or transfer of energy from the mean flow to the fluctuations due to the Reynolds stress. There is an O(epsilon(1/2)) correction to the growth rate, s((1)), due to the presence of a wall layer of thickness epsilon(1/2)R where the viscous stresses are O(epsilon(1/2)) smaller than the inertial stresses. An energy balance analysis indicates that the transfer of energy from the mean flow to the fluctuations due to the Reynolds stress in the wall layer is exactly cancelled by an opposite transfer of equal magnitude due to the deformation work done at the interface, and there is no net transfer from the mean flow to the fluctuations. Consequently, the fluctuations are stabilized by the viscous dissipation in the wall layer, and the real part of s(1) is negative. However, there are certain values of Gamma and wavenumber k where s((1)) = 0. At these points, the wall layer amplitude becomes zero because the tangential velocity boundary condition is identically satisfied by the inviscid flow solution. The real part of the O(epsilon) correction to the growth rate s((2)) turns out to be negative at these points, indicating a small stabilizing effect due to the dissipation in the bulk of the fluid and the wall material. It is found that the minimum value of s((2)) increases proportional to (H-1)(-2) for (H-1) much less than 1 (thickness of wall much less than the tube radius), and decreases proportional to H-4 for H much greater than 1. The damping rate for the inviscid modes is smaller than that for the viscous wall and centre modes in a rigid tube, which have been determined previously using a singular perturbation analysis.
