The efficacy of liposome-encapsulated nisin and bacteriocin-like substance (BLS) P34 to control growth of Listeria monocytogenes in Minas frescal cheese was investigated. Nisin and BLS P34 were encapsulated in partially purified soybean phosphatidylcholine (PC-1) and PC-1-cholesterol (7:3) liposomes. PC-1 nanovesicles were previously characterized. PC-1-cholesterol encapsulated nisin and BLS P34 presented, respectively, 218 nm and 158 nm diameters, zeta potential of -64 mV and -53 mV, and entrapment efficiency of 88.9% and 100%. All treatments reduced the population of L monocytogenes compared to the control during 21 days of storage of Minas frescal cheese at 7 degrees C. However, nisin and BLS P34 encapsulated in PC-1-cholesterol liposomes were less efficient in controlling L monocytogenes growth in comparison with free and PC-1 liposome-encapsulated bacteriocins. The highest inhibitory effect was observed for nisin and BLS P34 encapsulated in PC-1 liposomes after 10 days of storage of the product The encapsulation of bacteriocins in liposomes of partially purified soybean phosphatidylcholine may be a promising technology for the control of food-borne pathogens in cheeses. Effective incorporation of a probiotic into foods requires the culture to remain viable all along processing and storage, without adverse alterations to sensory characteristics. The objective of this work was developing Minas-type fresh cheese with probiotic properties from buffalo milk. Four batches of Minas-type fresh cheese were prepared using buffalo milk: batch T1 in which neither culture nor lactic acid added; batch T3 in which only lactic acid added; batches T2 and T4 , both added of Lactobacillus acidophilus LAC 4, but T4 was also acidified. Resulting cheeses were evaluated for probiotic culture stability, texture profile, sensory acceptance, and changes in pH. The T4 probiotic cheese presented hardness, gumminess, and chewiness significantly lower than the other treatments. However, values for springiness and cohesiveness did not differ between all cheeses, and no sensory differences (p > 0.05) were found between treatments for texture, taste, and overall acceptance. The addition of probiotic to the acidified cheese (T4) yielded best aroma. The populations of L. acidophilus were greater than 10(6) CFU g-1 after 28 days of storage all products. Minas-type fresh cheese from buffalo milk is a suitable food for the delivery of L. acidophilus, since the culture remained viable during the shelf life of the products and did not negative affect analysed parameters.

OBJETIVO: Verificar a ocorrência de Staphylococcus aureus em uma amostra de queijo tipo Minas "frescal" comercializado na cidade de Poços de Caldas, MG, de modo a obter subsídios que permitam avaliar o risco potencial que este produto pode representar para a saúde da população consumidora. MÉTODOS: Foi investigada a presença e o número de cepas de Staphylococcus aureus em 80 amostras de queijo tipo Minas "frescal" produzido artesanalmente e comercializado na cidade de Poços de Caldas, MG, Brasil. RESULTADOS: Os resultados obtidos evidenciaram a presença de S. aureus em 40 (50,0%) amostras, cujas contagens revelaram valores médios em torno de 10(5)/g. CONCLUSÕES: Tais achados parecem ser extremamente preocupantes, pois além de se situarem acima do limite máximo de 10³/g estabelecido pelo Ministério da Saúde, estes valores mostraram-se muito próximos dos requeridos para a produção de enterotoxinas em quantidades suficientes para a ocorrência de surtos de intoxicação alimentar estafilocócica.

Foi avaliado o prazo de vida útil do queijo Minas frescal armazenado a 4 °C por 30 dias, através de análises microbiológicas e físico-químicas. Três lotes (A, B e C) de uma mesma marca foram analisados nos dias 1, 10, 20 e 30 após a fabricação. Foram feitas as contagens de microrganismos psicrotróficos, lipolíticos, proteolíticos, bactérias lácticas e mesófilas, que apresentaram um crescimento contínuo durante o período, atingindo contagens médias, respectivamente:, piscotróficos = 11,63; lipolíticos = 10,34; proteolíticos = 9,75 bactérias lácticas = 9,53; e mesófilos = 11,47 log UFC.g-1. A população de Escherichia coli desenvolveu-se em um dos lotes e Staphylococcus coagulase + e Salmonella não foram detectados. No mesmo período, observou-se a redução de 0,81 no valor do pH e o aumento do ácido láctico (0,026%). Não se observou redução na gordura (21,5%), embora a quantidade de ácidos graxos livres tenha aumentado em 0,22 mg KOH.g-1 de gordura. A proteína variou de 21,30 a 22,10% sem diferença significativa. O índice de extensão da proteólise aumentou 3% a cada 10 dias. Apenas em um dos lotes (C) no 30º dia o produto apresentou-se impróprio para o consumo com 1,7 x 10(4) NMP.g-1 de E. coli.

Foram isolados e identificados microrganismos patogênicos e deterioradores em três diferentes tipos de queijos, a saber, Prato, Minas Frescal e Provolone, adquiridos junto ao comércio de Piracicaba. As análises realizadas foram para Contagem Total, Coliformes Totais, Coliformes Fecais, Staphylococcus, Psicrotróficos, Termófilos, Proteolíticos, Lipolíticos, Enterococos, Salmonella, Fungos e Leveduras. As altas contagens verificadas para Contagem Total, Coliformes Totais, Coliformes Fecais, S. aureus, Enterococos, Proteolíticos, Lipolíticos e Psicrotróficos na maioria das amostras analisadas neste trabalho, e a presença de Salmonella em algumas delas, revelam que as mesmas devem ser consideradas impróprias ao consumo humano, uma vez que, a maioria das contagens de microrganismos patogênicos, está acima dos padrões estabelecidos pela Comissão Nacional de Normas e Padrões para Alimentos (1978).

Com o objetivo de avaliar os métodos de enriquecimento, semeadura direta e número mais provável (NMP), este último segundo o sistema do tubo Fung-Yu e segundo a técnica convencional, para a detecção de Listeria monocytogenes, foram analisadas 103 amostras de diferentes tipos de queijos (minas frescal, ricota, gorgonzola, roquefort, brie, camembert e cheddar) fabricados e comercializados na cidade do Rio de Janeiro, Brasil. O método de enriquecimento permitiu a detecção de L. monocytogenes em 11 (10.8%) amostras, enquanto o do NMP e da semeadura direta apenas em 3 (2.9%) e 2 (1.9%), respectivamente. O sistema Fung-Yu e a técnica convencional para proceder o método do NMP tiveram a mesma eficácia, em termos de sensibilidade e tempo, na detecção de L. monocytogenes e demais espécies do mesmo gênero. A eficiência dos meios seletivos indicadores testados variou em função da contaminação do queijo, sendo o ágar Mox mais sensível e mais seletivo que o Palcam para as amostras de queijos que apresentaram elevado nível de contaminação. Já para os demais tipos de queijos analisados ambos apresentaram a mesma eficiência. Das 103 amostras analisadas, 11 (10.8%) estavam contaminadas com L. monocytogenes, 13 (12.7%) com L. innocua, 6 (5.8%) com L grayi, e 1 (0.97%) com L. welshimeri, sendo L. monocytogenes evidenciada em amostras de queijos gorgonzola, roquefort e minas frescal.

In the present study, 24 samples of Minas Frescal cheese and 24 samples of Minas Padrao cheese produced in the North-east region of the state of Sao Paulo, Brazil, were analysed for aflatoxin M1 (AFM1) by high-performance liquid chromatography (HPLC) between March and August 2008. AFM1 was detected in 13 (27.1%) samples at concentrations ranging from 0.037 to 0.313 ng g-1. The mean concentrations of AFM1 in positive samples of Minas Frescal and Minas Padrao cheese were 0.142 +/- 0.118 and 0.118 +/- 0.054 ng g-1, respectively. It is concluded that the incidence of AFM1 in Minas cheese may contribute to an increase in the overall ingestion of aflatoxins in the diet, hence indicating the need for the adoption of a tolerance limit for AFM1 in cheese in Brazil.

The incidence of Listeria monocytogenes in three cheese manufacturing plants from the northeastern region of Sao Paulo, Brazil, was evaluated from October 2008 to September 2009. L. monocytogenes was found in samples from two plants, at percentages of 13.3% (n = 128) and 9.6% (n = 114). Samples of raw and pasteurized milk, water, and Minas Frescal cheese were negative for L. monocyto genes, although the pathogen was isolated from the surface of Prato cheese and in brine from one of the plants evaluated. L. monocytogenes was also isolated from different sites of the facilities, mainly in non food contact surfaces such as drains, floors, and platforms. Serotype 4b was the most predominant in the plants studied. The results of this study indicate the need for control strategies to prevent the dispersion of L. monocytogenes in the environment of cheese manufacturing plants.

This study reports the implementation of GMPs in a mozzarella cheese processing plant. The mozzarella cheese manufacturing unit is located in the Southwestern region of the state of Parana, Brazil, and processes 20,000 L of milk daily. The implementation of GMP took place with the creation of a multi-disciplinary team and it was carried out in four steps: diagnosis, report of the diagnosis and road map, corrective measures and follow-up of GMP implementation. The effectiveness of actions taken and GMP implementation was compared by the total percentages of non-conformities and conformities before and after implementation of GMR Microbiological indicators were also used to assess the implementation of GMP in the mozzarella cheese processing facility. Results showed that the average percentage of conformity after the implementation of GMP was significant increased to 66%, while before it was 32% (p < 0.05). The populations of aerobic microorganisms and total coliforms in equipment were significantly reduced (p < 0.05) after the implementation of GMP, as well as the populations of total coliforms in the hands of food handlers (p < 0.05). In conclusion, GMP implementation changed the overall organization of the cheese processing unity, as well as managers and food handlers' behavior and knowledge on the quality and safety of products manufactured. Intense physical activity results in a substantial volume of stress and hence a significant probability of immunosuppression in athletes, with milk proteins being, perhaps, the most recommended protein supplements. Consumption of a probiotic cheese can attenuate immune suppression induced by exhausting exercise in rats. A popular Brazilian fresh cheese (Minas Frescal cheese) containing Lactobacillus acidophilus LA14 and Bifidobacterium longum BL05 was fed for 2 wk to adult Wistar rats, which then were brought to exhaustion on the treadmill. Two hours after exhaustion, the rats were killed and material was collected for the determination of serum uric acid, total and high-density lipoprotein cholesterol fraction, total protein, triacylglycerols, aspartate aminotransferase, alanine aminotransferase, creatine kinase, and blood cell (monocyte, lymphocyte, neutrophil, and leukocyte) counts. Exercise was efficient in reducing lymphocyte counts, irrespective of the type of ingested cheese, but the decrease in the group fed the probiotic cheese was 22% compared with 48% in the animals fed regular cheese. Monocyte counts were unaltered in the rats fed probiotic cheese compared with a significant decrease in the rats fed the regular cheese. Most importantly, ingestion of the probiotic cheese resulted in a >100% increase in serum high-density lipoprotein cholesterol and a 50% decrease in triacylglycerols. We conclude that probiotic Minas Reseal cheese may be a viable alternative to enhance the immune system and could be used to prevent infections, particularly those related to the physical overexertion of athletes.

This study aimed to evaluate the effect of diet with selenium on the physico-chemical composition and somatic cell count of buffalo milk, and check the selenium waste in milk and minas kind fresh cheese. For the analysis of physical-chemical composition and SCC were used 2264 buffalo milk data from the Murrah, belonging to Tapuio Ltda, located in the agreste region of the state of Rio Grande do Norte, in the period 2010 to 2014. To check the amount of selenium residue in buffalo milk and fresh cheese mines were used 100 Murrah buffaloes divided into 5 lots. The batches were formed according to the production levels of animals, wherein the amount of concentrate fed varied within each lot, which received 0.08 ppm / Se / kg of concentrate. Was collected from 300 ml of tank milk from and each lot, as well as 300 gr minas cheese, from August to November 2014, with the collection of lots held only in the month of November. The samples were sent to the Pernambuco Tecnology Institute for selenium residue analysis. The effect of the seasons was contrasted in two ways, namely: Spring, Summer, Autumn and Winter season or drought and rainy season. The analyzes statistics were performed using the procedures available in the software SAS® (SAS, 2002). Selenium level provided to the animals in this study was not significant to the point of detection of their residue in milk and the cheeses. However, it influences the reduction of the CCS. The milk characteristics were not affected by selenium use only the seasons.
