BACKGROUND: Reference intervals for many laboratory parameters determined in 24-h urine collections are either not publicly available or based on small numbers, not sex specific or not from a representative sample. METHODS: Osmolality and concentrations or enzymatic activities of sodium, potassium, chloride, glucose, creatinine, citrate, cortisol, pancreatic α-amylase, total protein, albumin, transferrin, immunoglobulin G, α1-microglobulin, α2-macroglobulin, as well as porphyrins and their precursors (δ-aminolevulinic acid and porphobilinogen) were determined in 241 24-h urine samples of a population-based cohort of asymptomatic adults (121 men and 120 women). For 16 of these 24 parameters creatinine-normalized ratios were calculated based on 24-h urine creatinine. The reference intervals for these parameters were calculated according to the CLSI C28-A3 statistical guidelines. RESULTS: By contrast to most published reference intervals, which do not stratify for sex, reference intervals of 12 of 24 laboratory parameters in 24-h urine collections and of eight of 16 parameters as creatinine-normalized ratios differed significantly between men and women. For six parameters calculated as 24-h urine excretion and four parameters calculated as creatinine-normalized ratios no reference intervals had been published before. For some parameters we found significant and relevant deviations from previously reported reference intervals, most notably for 24-h urine cortisol in women. Ten 24-h urine parameters showed weak or moderate sex-specific correlations with age. CONCLUSIONS: By applying up-to-date analytical methods and clinical chemistry analyzers to 24-h urine collections from a large population-based cohort we provide as yet the most comprehensive set of sex-specific reference intervals calculated according to CLSI guidelines for parameters determined in 24-h urine collections.

The aim of this study was to evaluate the preventive effect of sodium bicarbonate on systemic acidosis due to ruminal acidosis, which was induced by ingestion of concentrate after prolonged fasting. Fourteen sheep were divided into three experimental groups: control group (Cg), with four sheep, submitted to fasting without development of ruminal acidosis; no-treated group (NTg), with five sheep with rumen acidosis without preventive treatment; and treated group (Tg), with five sheep with rumen acidosis and preventively treated with sodium bicarbonate. Assessments of ruminal pH and arterial hemogasometry were performed for 48 hours after ingestion of the concentrate. There was a reduction in the ruminal pH in all groups, whereas the Cg showed a reduction only after 24 hours. A reduction in the arterial pH, bicarbonate and base excess in all groups was also noted, indicating systemic metabolic acidosis, but the NTg presented the greatest alteration. It is concluded that sodium bicarbonate prevents systemic metabolic acidosis, reducing its severity in sheep subjected to ruminal acidosis.

Este trabalho foi realizado com o objetivo de avaliar o efeito do trifloxysulfuron-sodium no crescimento do algodoeiro cultivar Fabrika. As plantas foram submetidas ou não a 7,5 g ha¹ de trifloxysulfuron-sodium, aos 19 dias após a emergência, quando estavam com quatro folhas verdadeiras (estádio V4). Os índices de crescimento das plantas foram determinados aos 0, 7, 14, 24, 36, 52 e 73 dias após aplicação do herbicida (DAA). O delineamento experimental foi o inteiramente casualizado, com quatro repetições. A intoxicação causada pelo herbicida foi maior aos 20 dias após sua aplicação (DAA), com 35% de dano. O acúmulo de matéria seca do caule principal, do total de folhas, de folhas do ramo principal, da parte aérea e a área foliar aumentaram com o desenvolvimento da cultura. Entretanto, o ponto de máximo crescimento para essas características foi atingido mais precocemente nas plantas-controle, exceto para área foliar total. De modo geral, a taxa de crescimento absoluto, a taxa de crescimento relativo e a taxa assimilatória líquida foram menores nas avaliações iniciais, nas plantas submetidas ao herbicida. As plantas-controle apresentaram redução mais acentuada na razão de área foliar que as submetidas ao herbicida. O trifloxysulfuron-sodium também causou atraso no crescimento das maçãs, que apresentaram metade do acúmulo de matéria seca em relação às plantas-controle, aos 52 DAA; todavia, a produção de algodão em caroço foi semelhante entre os tratamentos. O trifloxysulfuron-sodium influenciou negativamente o crescimento do algodoeiro, principalmente nas primeiras semanas após sua aplicação. Entretanto, a rápida recuperação da planta tratada possibilitou produção de algodão em caroço semelhante àquela obtida com as plantas-controle.

Objetivou-se com este trabalho desenvolver tecnologia para manejo de plantas daninhas na cultura do algodoeiro, em sistema de plantio convencional, combinando os herbicidas S-metolachlor em pré-emergência com trifloxysulfuron-sodium em pós-emergência. Foram avaliados 14 tratamentos, em arranjo fatorial 3 x 4 (três doses de S-metolachlor: 384, 768 e 1.152 g ha-1 e quatro doses de trifloxysulfuron-sodium: 0,0; 2,625; 5,250; e 7,875 g ha-1), mais duas testemunhas (com e sem convivência com as plantas daninhas por todo o ciclo do algodoeiro), em delineamento de blocos casualizados, com quatro repetições. Na área, foi verificada a presença das seguintes espécies daninhas: Alternanthera tenella, representando mais de 80% do total, Bidens spp., Acanthospermum hispidum, Cenchrus echinatus, Digitaria horizontalis, Eleusine indica e Commelina benghalensis. S-metolachlor apresentou alta eficiência no controle de A. tenella, C. echinatus, D. horizontalis, E. indica e C. benghalensis. Trifloxysulfuron-sodium controlou as espécies dicotiledôneas eficientemente. Os tratamentos que proporcionaram melhor produtividade de algodão em caroço foram Smetolachlor (768 g ha-1) mais trifloxysulfuron-sodium (7,875 g ha-1 ) e S-metolachlor (1.152 g ha-1 ) mais trifloxysulfuron-sodium (5,250 e 7,875 g ha-1). O melhor controle de plantas daninhas na colheita do algodão foi obtido com 1.152 g ha-1 de S-metolachlor mais 7,875 g ha-1 de trifloxysulfuron-sodium.

Este trabalho teve por objetivo avaliar a suscetibilidade de cinco espécies de plantas daninhas do gênero Amaranthus a herbicidas aplicados em pós-emergência. As espécies avaliadas foram: A. deflexus (caruru-rasteiro), A. hybridus (caruru-roxo), A. retroflexus (caruru-gigante), A. spinosus (caruru-de-espinho) e A. viridis (caruru-de-mancha). O trabalho foi dividido em duas fases. Na primeira, as espécies de plantas daninhas foram submetidas à aplicação de 12 tratamentos herbicidas em pós-emergência. Na segunda, os herbicidas trifloxysulfuron-sodium e chlorimuron-ethyl foram avaliados com a metodologia de curvas de dose-resposta, repetida duas vezes. Os herbicidas foram aplicados sobre plantas com 5-6 folhas e as doses utilizadas na segunda fase foram: 16D, 4D, D, 1/4D, 1/16D, 1/64D e ausência do produto, em que D é a dose recomendada de cada herbicida. As doses utilizadas (D) foram de 3,75 e 7,5 g ha-1 para o herbicida trifloxysulfuron e 12,5 e 17,5 g ha-1 para chlorimuron, na primeira e na segunda condução, respectivamente. Na primeira fase, foram avaliados o controle percentual e a massa seca das parcelas aos 20 dias após a aplicação (DAA); na segunda, avaliou-se o controle percentual aos 20 DAA. As espécies de Amaranthus avaliadas neste trabalho apresentaram diferenças de suscetibilidade aos herbicidas aplicados em pósemergência, principalmente ao trifloxysulfuron e ao chlorimuron, em que A. deflexus foi a espécie menos suscetível, seguido por A. spinosus, A. viridis, A. hybridus e A. retroflexus.

A influência de concentrações de bispyribac-sodium, aplicadas na parte aérea ou nas raízes das plantas de arroz, foi avaliada. O experimento foi realizado em casa de vegetação, usando bispyribac-sodium nas doses de 0, 24, 48, 72, 96 e 120 ppb, aplicadas na parte aérea ou nas raízes de plantas de arroz BRS Pelota e BRS Bojuru. As unidades experimentais constaram de copos plásticos perfurados na lateral próximo ao fundo, com areia lavada, onde foram colocadas cinco sementes de arroz, e mantidas dentro de bandejas contendo água em nível imediatamente inferior ao nível de areia. Quando as plantas se encontravam no estádio de duas a três folhas definitivas, as soluções foram pulverizadas diretamente sobre as folhas. Para aplicação nas raízes, o herbicida foi colocado na água de irrigação. Quarenta dias após emergência, foram avaliados comprimento de plantas, matéria fresca e seca de parte aérea e raízes e volume do sistema radical. Os dados foram submetidos à análise de variância, com análise de regressão através de modelos polinomiais, quando significativos. O cultivar de arroz BRS Bojuru se mostrou mais sensível que o BRS Pelota ao incremento na dose de bispyribac-sodium, com efeito mais pronunciado quando aplicado diretamente às folhas. Em campo, devem-se evitar doses acima das recomendadas, principalmente quando do uso de cultivares japonica.

We determined whether ANP (atrial natriuretic peptide) concentrations, measured by radioimmunoassay, in the ANPergic cerebral regions involved in regulation of sodium intake and excretion and pituitary gland correlated with differences in sodium preference among 40 Wistar male rats (180-220 g). Sodium preference was measured as mean spontaneous ingestion of 1.5% NaCl solution during a test period of 12 days. The relevant tissues included the olfactory bulb (OB), the posterior and anterior lobes of the pituitary gland (PP and AP, respectively), the median eminence (ME), the medial basal hypothalamus (MBH), and the region anteroventral to the third ventricle (AV3V). We also measured ANP content in the right (RA) and left atrium (LA) and plasma. The concentrations of ANP in the OB and the AP were correlated with sodium ingestion during the preceding 24 h, since an increase of ANP in these structures was associated with a reduced ingestion and vice-versa (OB: r = -0.3649, P<0.05; AP: r = -0.3291, P<0.05). Moreover, the AP exhibited a correlation between ANP concentration and mean NaCl intake (r = -0.4165, P<0.05), but this was not the case for the OB (r = 0.2422). This suggests that differences in sodium preference among individual male rats can be related to variations of AP ANP level. Earlier studies indicated that the OB is involved in the control of NaCl ingestion. Our data suggest that the OB ANP level may play a role mainly in day-to-day variations of sodium ingestion in the individual rat

We have demonstrated that acute third ventricle injections of lead acetate (PbAc) exert a powerful antidipsogenic effect and induce a significant increase in renal sodium excretion. In the present study we confirm the antidipsogenic effect of lead and demonstrate that central administration of this metal, in minute amounts, significantly reduces salt intake both during dehydration and after central angiotensinergic stimulation. Adult male Wistar rats had the third ventricle cannulated seven days before the experiments. During this period they had free access to distilled water and hypertonic saline solution (1.5%). After a 24-h period of fluid deprivation, experimental animals received third ventricle injections of PbAc (0.3, N = 8 and 3.0 nmol/rat, N = 14) while controls received sodium acetate (NaAc; 3.0 nmol/rat, N = 10). Rats treated with PbAc at the highest dose showed a significant reduction (P<0.05) both in water and hypertonic saline intake when compared to controls. When the effect of lead administration on angiotensin II-induced water and salt intake was studied, normohydrated animals received third ventricle injections of angiotensin II (9.6 nmol/rat) after pretreatment with 3.0 nmol/rat of PbAc (experimental group, N = 10) or NaAc (controls, N = 8). The group pretreated with PbAc presented a significant reduction (P<0.05) in both water and salt intake compared to controls. Thus, this study confirms the antidipsogenic effect of central lead injections and demonstrates that the presence of lead in the brain exerts a significant inhibition of sodium appetite.

The present study determined the effect of an electrolytic lesion of the dorsal raphe nucleus (DRN) on water intake and sodium appetite. Male Wistar rats weighing 290-320 g with a lesion of the DRN (L-DRN), performed two days before experiments and confirmed by histology at the end of the experiments, presented increased sensitivity to the dehydration induced by fluid deprivation. The cumulative water intake of L-DRN rats reached 23.3 ± 1.9 ml (a 79% increase, N = 9) while sham-lesioned rats (SL-DRN) did not exceed 13.0 ± 1.0 ml (N = 11, P < 0.0001) after 5 h. The L-DRN rats treated with isoproterenol (300 µg kg-1 ml-1, sc) exhibited an increase in water intake that persisted throughout the experimental period (L-DRN, 15.7 ± 1.47 ml, N = 9 vs SL-DRN, 9.3 ± 1.8 ml, N = 11, P < 0.05). The L-DRN rats also showed an increased spontaneous sodium appetite during the entire period of assessment. The intake of 0.3 M NaCl after 12, 24, 36 and 72 h by the L-DRN rats was always higher than 20.2 ± 4.45 ml (N = 10), while the intake by SL-DRN was always lower than 2.45 ± 0.86 ml (N = 10, P < 0.00001). Sodium- and water-depleted L-DRN rats also exhibited an increased sodium appetite (13.9 ± 2.0 ml, N = 11) compared to SL-DRN (4.6 ± 0.64 ml, N = 11) after 120 min of observation (P < 0.02). The sodium preference of L-DRN rats in both conditions was always higher than that of SL-DRN rats. These results suggest that electrolytic lesion of the DRN overcomes a tonic inhibitory component of sodium appetite.

We determined if the dorsal raphe nucleus (DRN) exerts tonic control of basal and stimulated sodium and water intake. Male Wistar rats weighing 300-350 g were microinjected with phosphate buffer (PB-DRN, N = 11) or 1 µg/0.2 µl, in a single dose, ibotenic acid (IBO-DRN, N = 9 to 10) through a guide cannula into the DRN and were observed for 21 days in order to measure basal sodium appetite and water intake and in the following situations: furosemide-induced sodium depletion (20 mg/kg, sc, 24 h before the experiment) and a low dose of dietary captopril (1 mg/g chow). From the 6th day after ibotenic acid injection IBO-DRN rats showed an increase in sodium appetite (12.0 ± 2.3 to 22.3 ± 4.6 ml 0.3 M NaCl intake) whereas PB-DRN did not exceed 2 ml (P < 0.001). Water intake was comparable in both groups. In addition to a higher dipsogenic response, sodium-depleted IBO-DRN animals displayed an increase of 0.3 M NaCl intake compared to PB-DRN (37.4 ± 3.8 vs 21.6 ± 3.9 ml 300 min after fluid offer, P < 0.001). Captopril added to chow caused an increase of 0.3 M NaCl intake during the first 2 days (IBO-DRN, 33.8 ± 4.3 and 32.5 ± 3.4 ml on day 1 and day 2, respectively, vs 20.2 ± 2.8 ml on day 0, P < 0.001). These data support the view that DRN, probably via ascending serotonergic system, tonically modulates sodium appetite under basal and sodium depletion conditions and/or after an increase in peripheral or brain angiotensin II.

The present study evaluated the acute effect of the intraperitoneal (ip) administration of a whey protein hydrolysate (WPH) on systolic arterial blood pressure (SBP) and renal sodium handling by conscious spontaneously hypertensive rats (SHR). The ip administration of WPH in a volume of 1 ml dose-dependently lowered the SBP in SHR 2 h after administration at doses of 0.5 g/kg (0.15 M NaCl: 188.5 ± 9.3 mmHg vs WPH: 176.6 ± 4.9 mmHg, N = 8, P = 0.001) and 1.0 g/kg (0.15 M NaCl: 188.5 ± 9.3 mmHg vs WPH: 163.8 ± 5.9 mmHg, N = 8, P = 0.0018). Creatinine clearance decreased significantly (P = 0.0084) in the WPH-treated group (326 ± 67 µL min-1 100 g body weight-1) compared to 0.15 M NaCl-treated (890 ± 26 µL min-1 100 g body weight-1) and captopril-treated (903 ± 72 µL min-1 100 g body weight-1) rats. The ip administration of 1.0 g WPH/kg also decreased fractional sodium excretion to 0.021 ± 0.019% compared to 0.126 ± 0.041 and 0.66 ± 0.015% in 0.15 M NaCl and captopril-treated rats, respectively (P = 0.033). Similarly, the fractional potassium excretion in WPH-treated rats (0.25 ± 0.05%) was significantly lower (P = 0.0063) than in control (0.91 ± 0.15%) and captopril-treated rats (1.24 ± 0.30%), respectively. The present study shows a decreased SBP in SHR after the administration of WPH associated with a rise in tubule sodium reabsorption despite an angiotensin I-converting enzyme (ACE)-inhibiting in vitro activity (IC50 = 0.68 mg/mL). The present findings suggest a pathway involving ACE inhibition but measurements of plasma ACE activity and angiotensin II levels are needed to support this suggestion.

Since cellulose is a linear macromolecule it can be used as a material for regenerated cellulose fiber products e.g. in textile fibers or film manufacturing. Cellulose is not thermoformable, thus the manufacturing of these regenerated fibers is mainly possible through dissolution processes preceding the regeneration process. However, the dissolution of cellulose in common solvents is hindered due to inter- and intra-molecular hydrogen bonds in the cellulose chains, and relatively high crystallinity. Interestingly at subzero temperatures relatively dilute sodium hydroxide solutions can be used to dissolve cellulose to a certain extent. The objective of this work was to investigate the possible factors that govern the solubility of cellulose in aqueous NaOH and the solution stability. Cellulose-NaOH solutions have the tendency to form a gel over time and at elevated temperature, which creates challenges for further processing. The main target of this work was to achieve high solubility of cellulose in aqueous NaOH without excessively compromising the solution stability. In the literature survey an overview of the cellulose dissolution is given and possible factors contributing to the solubility and solution properties of cellulose in aqueous NaOH are reviewed. Furthermore, the concept of solution rheology is discussed. In the experimental part the focus was on the characterization of the used materials and properties of the prepared solutions mainly concentrating on cellulose solubility and solution stability.

Ablation of the area postrema/caudal nucleus of the tractus solitarius (NTS) complex increases sodium intake, but the effect of selective lesions of the caudal NTS is not known. We measured depletion-induced sodium intake in rats with electrolytic lesions of the commissural NTS that spared the area postrema. One day after the lesion, rats were depleted of sodium with furosemide (10 mg/kg body weight, sc) and then had access to water and a sodium-deficient diet for 24 h when 1.8% NaCl was offered. Water and saline intakes were measured for 2 h. Saline intake was higher in lesioned than in sham-lesioned rats (mean ± SEM: 20 ± 2 vs 11 ± 3 mL/2 h, P < 0.05, N = 6-7). Saline intake remained elevated in lesioned rats when the tests were repeated 6 and 14 days after the lesion, and water intake in these two tests was increased as well. Water intake seemed to be secondary to saline intake both in lesioned and in sham-lesioned rats. A second group of rats was offered 10% sucrose for 2 h/day before and 2, 7, and 15 days after lesion. Sucrose intake in lesioned rats was higher than in sham-lesioned rats only 7 days after lesioning. A possible explanation for the increased saline intake in rats with commissural NTS lesions could be a reduced gastrointestinal feedback inhibition. The commissural NTS is probably part of a pathway for inhibitory control of sodium intake that also involves the area postrema and the parabrachial nucleus.

High salt intake is related to an increase in blood pressure and development of hypertension. However, currently, there are no national representative data in Brazil using the gold standard method of 24-h urine collection to measure sodium consumption. This study aimed to determine salt intake based on 24-h urine collection in a sample of 272 adults of both genders and to correlate it with blood pressure levels. We used a rigorous protocol to assure an empty bladder prior to initiating urine collection. We excluded subjects with a urine volume <500 mL, collection period outside of an interval of 23-25 h, and subjects with creatinine excretion that was not within the range of 14.4-33.6 mg/kg (men) and 10.8-25.2 mg/kg (women). The mean salt intake was 10.4±4.1 g/day (d), and 94% of the participants (98% of men and 90% of women) ingested more than the recommended level of 5 g/d. We found a positive association between salt and body mass index (BMI) categories, as well as with salt and blood pressure, independent of age and BMI. The difference in systolic blood pressure reached 13 mmHg between subjects consuming less than 6 g/d of salt and those ingesting more than 18 g/d. Subjects with hypertension had a higher estimated salt intake than normotensive subjects (11.4±5.0 vs 9.8±3.6 g/d, P<0.01), regardless of whether they were under treatment. Our data indicate the need for interventions to reduce sodium intake, as well the need for ongoing, appropriate monitoring of salt consumption in the general population.
