The peritubular zone of the rat testis has an extensive extracellular matrix (ECM). Fibronectin (FN) is distributed primarily in the basal lamina of the seminiferous tubule boundary tissue and is synthesized by peritubular myoid cells. Several extracellular changes are mediated by growth factors and these changes occur at the time of hormone mediated testicular development, particularly in the peritubular zone. The effects of serum or dibutyryl cyclic AMP (cAMP) on FN production by the mesenchymal peritubular myoid cells were evaluated. Rats of various ages (10, 15, 20, 40 and 80 days) were employed for immunofluorescent localization of rat testicular FN in frozen sections. In all age groups tested, FN was primarily present in a broad layer around each seminiferous tubule, and blood vessel, and in variable distribution throughout the interstitial stroma. By day 20 there was no clear distinction in FN staining between the peritubular zone and the interstitial tissue. This indicates an involvement of FN in the ECM developments which occur in the peritubular zone of the testis at this time. The peritubular myoid cells were isolated from 20-22 day old rat testis and cultured on glass coverslips. These cells were grown to confluence with 10% fetal calf serum (FCS) in medium until day 4 and then subcultured to have secondary monocultures maintained with or without serum. By means of immunofluorescence and cytochemistry using avidin-biotin peroxidase complex it was observed that peritubular myoid cells were positive for FN and most of the FN was localized in the perinuclear region. Subcultured peritubular myoid cells maintained for 4 days in medium containing FCS developed an extensive interconnecting FN matrix. In the presence of 0.5 mM cAMP in culture, FN became localized along the filamentous process of peritubular myoid cells and more prominently in the areas of triangulated multi-cell aggregates as well as on the surface of the contracted small spherical cells. The addition of cAMP in the presence of FCS, also caused a noticeable change in the staining pattern; FN was detected along the filamentous process developing into a complex network of cells encased in an extensive matrix. It would appear that the translocation of FN in the cytoplasmic extensions of peritubular myoid cells may be a direct consequence of morphological changes associated with metabolic regulation of cAMP. This may also be related to the puberty associated development of in vivo changes in the ECM produced by peritubular myoid cells.

A leptina tem um papel importante na regulação do sistema reprodutivo além de seu papel principal na regulação do peso corporal e ingestão alimentar. O objetivo deste estudo foi avaliar o efeito da administração de leptina durante o período neonatal na função testicular da prole adulta. Vinte e quatro filhotes de 12 mães foram divididos em 2 grupos: Grupo leptina: injetados com 50 L de leptina (80ng/gPC, subcutânea) nos primeiros 10 dias de vida e Grupo Controle: injetados com o mesmo volume de solução salina. Todos os animais foram sacrificados aos 90 dias de vida. Parâmetros analisados: consumo alimentar, massa corporal, crescimento linear, data de início da puberdade, perfil lipídico, níveis séricos de estradiol e testosterona, expressão gênica (PCR em tempo real) e expressão proteica (Western blot) de ObRa, OBRb, aromatase, AR, ER, morfometria testicular, número, morfologia e viabilidade de espermatozoides. Os dados foram expressos como média erro padrão. A significância estatística foi determinada pelo teste t de Student. A Injeção de leptina levou a uma redução (p≤0,008) no consumo alimentar a partir do dia 26 ao 40 e do dia 70 em diante, enquanto que a massa corporal (p≤0,03) e o crescimento linear (p≤0,05) foram reduzidos do dia 26 até o dia 45. O peso da hipófise (p≤0,0006), hipotálamo (p≤0,01), próstata (p≤0,003), testículo (p≤0,008) e epidídimo (p≤0,004) e bexiga (p≤0,009) foram significativamente reduzidos pela injeção de leptina. A Injeção de leptina adiantou o início da puberdade (C=45,0 0,3; L=41,6 0,3; dias, P≤0,0001). Em relação ao perfil lipídico, a administração de leptina gerou um aumento nos níveis séricos de TG (C= 116,5 15,9; L=172,6 19,7; ng/dL, P≤0,05) e uma redução nos níveis de HDL (C=33 1,5; L = 24 3,5; ng/dL, P≤0,02). Os níveis séricos de testosterona também foram reduzidos pela leptina (C=5,2 1,0; L=1,1 0,3; ng/mL, P≤0,003). Todos os genes avaliados por PCR em tempo real mostraram um aumento na sua expressão: Obra (C=0,32 0,04; L=0,69 0,16; P≤0,04), OBRb (C=0,37 0,06; L=0,71 0,16; P≤0,03), AR (C=0,28 0,02; L=0,71 0,16; P≤0,02), Aromatase (C=0,31 0,04; L=0,53 0,09; P≤0,04), ER-α (C=0,79 0,03; L=0,93 0,03; P≤0,01), ER-β (C=0,29 0,03; L=0,73 0,16; P≤ 0,02). Por outro lado, a expressão proteica de OBR (C=4,4 0,29; L=6,6 0,84; P≤0,05), ER-α (C=0,4 0,02; L=0,6 0,05; P≤0,03) e aromatase (C=0,4 0,03; L=0,5 0,02; P≤0,04) aumentaram, enquanto que a expressão proteica de AR (C=0,16 0,01; L=0,09 0,01; P≤0,009) foi reduzida pela administração de leptina. A análise morfométrica mostrou que a leptina levou a um aumento da área total do túbulo seminífero (C=64,6 3,1; L=56,1 2,1;μm, P≤0,01), aumento da área luminal (C=40,6 2,3; L= 48,7 0,9; μm P≤ 0,004) e na altura do epitélio (C=21,5 1,2; L=24,6 1,1; μm, P≤0,03), enquanto que o comprimento do túbulo seminífero foi reduzido no grupo tratado (C=2200 350; L= 1100 110;cm, P≤0,006). A administração de leptina levou a um aumento no número total de espermatozoides (C=20x107 2x107; L=30x107 5x107;Cls/mL, P≤0,009) e no número de anormalidades (C=40,6 1,9; L=45,8 1,2, P≤0,049). Podemos concluir que a leptina tem um papel importante na morfologia e função testicular. A leptina parece ter efeito direto neste tecido uma vez que a expressão gênica e proteica de OBR, AR, ER e aromatase foram alterados pela administração da leptina.

Estudos demonstram que o sobrepeso e a obesidade podem afetar a fertilidade masculina. Além disso, a função tireóidea também esta associada à alteração da função testicular, entretanto, o papel fisiológico dos hormônios tireoideanos na regulação do sistema reprodutor masculino ainda não esta claro. Aos 21 dias de idade, ratos machos receberam uma dieta manipulada contendo diferentes concentrações de óleo de soja até a idade de 30 e 60 dias, os grupos controle (C30, n=6 e C60, n=6), receberam dieta contendo 7% e os grupos hiperlipídicos (HL30, n=6 e HL60, n=6), receberam dieta contendo 19% deste óleo. Ao final de cada período, os animais foram avaliados por DXA (Absorciometria de Raios-x em Duas Energias) e sacrificados por exsanguinação. Para avaliar alterações na estrutura dos tecidos testicular e tireóideo, foram realizados a morfologia e a estereologia. No plasma, para determinar os perfis bioquímico e hormonal, foram avaliados, triglicerídeos, colesterol total, HDL-colesterol, VLDL, glicose e albumina, por métodos colorimétricos e leptina, insulina, T4, T3, TSH e testosterona por radioimunoensaio (RIE). Para evidenciar a expressão de receptor androgênico (AR) em testículos, foi realizado imunomarcação, com anti-AR. Durante todo o período experimental, foram analisados a massa corporal, a ingestão alimentar e o comprimento corporal, os quais permaneceram inalterados. No grupo HL, a massa magra foi menor aos 30 dias, já a gordura corporal total foi maior no mesmo grupo, aos 60 dias nenhuma diferença foi notada entre os grupos. No grupo HL30 não houve diferença quanto à massa dos tecidos, já no grupo HL60, o peso do epidídimo, fígado e gordura visceral mantiveram-se aumentados. No grupo HL30 não houve diferença em relação ao perfil bioquímico, já no grupo HL60, os níveis de glicose, mantiveram-se altos. Quanto às dosagens hormonais no grupo HL30, TSH e leptina estiveram aumentados e T3 reduzido, e no grupo HL60, T3 e leptina estiveram aumentados. Os dados morfométricos e estereológicos de testículo no grupo HL30 mostram aumento no número de túbulos seminíferos e da densidade de comprimento (Lv), já no grupo HL60, há redução no número de túbulos seminíferos e no diâmetro do mesmo. Quanto à expressão de receptor androgênico nas células testiculares, não parece haver diferença entre os grupos independente da idade de consumo da dieta. A dieta hiperlipídica promoveu alterações metabólicas aos 30 dias e modificações na morfologia do tecido tireoidiano e testicular em ambas as idades, o que indica reflexos na função reprodutora.

The reproductive biology of male franciscanas (Pontoporia blainvillei), based on 121 individuals collected in Rio Grande do Sul State, southern Brazil, was studied. Estimates on age, length, and weight at attainment of sexual maturity are presented. Data on the reproductive seasonality and on the relationship between some testicular characteristics and age, size, and maturity status are provided. Sexual maturity was assessed by histological examination of the testes. Seasonality was determined by changes in relative and total testis weight, and in seminiferous tubule diameters. Testis weight, testicular index of maturity, and seminiferous tubule diameters were reliable indicators of sexual maturity, whereas testis length, age, length, and weight of the dolphin were not. Sexual maturity was estimated to be attained at 3.6 years (CI 95% =2.7–4.5) with the DeMaster method and 3.0 years with the logistic equation. Length and weight at attainment of sexual maturity were 128.2 cm (CI 95%=125.3–131.1 cm) and 26.4 kg (CI 95% =24.7–28.1 kg), respectively. It could not be verified that there was any seasonal change in the testis weight and in the seminiferous tubule diameters in mature males. It is suggested that at least some mature males may remain reproductively active throughout the year. The extremely low relative testis weight indicates that sperm competition does not occur in the species. On the other hand, the absence of secondary sexual characteristics, the reversed sexual size dimorphism, and the small number of scars from intrassexual combats in males reinforce the hypothesis that male combats for female reproductive access may be rare for franciscana. It is hypothesized that P. blainvillei form temporary pairs (one male copulating with only one female) during the reproductive period.

Abstract BACKGROUND: Each year 40,000 men have a vasectomy in the UK whilst another 2400 request a reversal to begin a second family. Sperm can now be obtained by testicular biopsy and subsequently used in assisted conception with intracytoplasmic sperm injection (ICSI). The study aims were to compare sperm yields of men post-vasectomy or with obstructive azoospermia (OA) of unknown aetiology with fertile men and to assess any alteration in the clinical pregnancy rates after ICSI. METHODS: Testicular tissue was obtained by Trucut needle from men who had undergone a vasectomy >5yrs previously, had OA from other causes and from fertile men during vasectomy. Seminiferous tubules were milked to measure sperm yields. Numbers of Sertoli cells, spermatids and thickness of the seminiferous tubule walls were assessed using quantitative computerized analysis. RESULTS and CONCLUSIONS: Sperm yields/g testis were significantly decreased in men post-vasectomy and in men with OA, relative to fertile men. Significant reductions were also observed in early (40%) and mature (29%) spermatid numbers and an increase of 31% was seen in the seminiferous tubule wall (basal membrane and collagen thickness) of vasectomised men compared to fertile men. Clinical pregnancy rates in couples who had had a vasectomy were also significantly reduced.

The study proposed to describe sexual development in pelagic stage loggerhead sea turtles Caretta caretta and compare this to hatchlings and adults. It is meant as an ontogenic approach, in order to understand reproductive development and population composition and their dynamics in the pelagic environment. The study focused on the pelagic loggerheads that are found in the waters offshore Madeira Island (Portugal) in the North-eastern Atlantic and use it as a developmental habitat. The innovating character of this work relied on the lack of any description regarding the gonad ontogenesis and reproductive development for the pelagic stage in any of the 7 existing sea turtle species, all of them in danger of extinction. Three methods were used to diagnose the sex of each juvenile individual and asses the level of reproductive development: (1) laparoscopy, (2) gonad biopsy and (3) the assessment of two sex steroids circulating levels, namely testosterone and estradiol. In order to cover all life stages and compare data obtained for the juvenile stage, hatchlings and nesting female adults were sampled at the nearest nesting rookery at Boa Vista Island in the Cape Verde Archipelago. Gonads from dead hatchlings were collected for gonad histology and blood was collected from nesting females for sex steroids assessment. Laparoscopies revealed to be a valid sexing method for the juvenile stage, since gonads are morphologically differentiated at these size classes. Moreover, laparoscopy was validated using gonad histology. Gonad histology of juveniles showed that gonads are already completely differentiated into ovaries or testes at the size classes examined, but development seems to be quiescent. Males present already developed seminiferous tubules with spermatogonia lining the interior of the seminiferous tubule. Female gonads present oocytes at different development stages, but only oocytes up to stage III were observed. The maximum oocyte diameter in each individual correlated with body size, suggesting that reproductive development is an on-going process in juvenile females. The circulating levels of both testosterone and estradiol in juveniles of both sexes were very low and consistently lower than the ones observed in the nesting females from Boa Vista Island. No bimodal distribution was found for any of the sex steroids analysed and thus circulating hormone levels were not a reliable tool for sexing juvenile individuals with a non-invasive technique. The ratio testosterone:estradiol did not show a bimodal distribution either. The levels of testosterone correlated with sea surface temperature. The fact that temperatures observed during this study were below 24ºC might have hindered a differential testosterone pattern between juvenile males and females. Sex ratios for this population were generated according to laparoscopy results and compared among years and size classes. An overall sex ratio of 2 females for each male was found, but they varied among size classes but not among years. Possible causes for the sex ratios observed are discussed. This study is a contribution to our knowledge on the pelagic stage of loggerhead turtles, namely on the population structure regarding sex ratio, which is a vital tool for implementing conservation strategies.

The seminiferous tubules of Prochilodus scrofa present a coiled morphological arrangement with intertubular anastomoses and unrestricted spermatogonial distribution. The structural pattern of the seminiferous tubules is cystic, with cysts formed by cytoplasmic prolongations of Sertoli cells. Inside the cysts are observed different types of germ cells. The seminiferous tubules open individually on the ventral surface of the main testicular duct present in each testis. Each main testicular duct prolongs as a spermatic duct, fusing with the spermatic duct of the opposite side to form the common spermatic duct which opens into the urogenital papilla. The mature sperm cysts break and extravasate their content into the lumen of the seminiferous tubules from which the seminal fluid and the spermatozoa penetrate the main testicular duct, the spermatic duct and the common spermatic duct for semen ejaculation.

In Nysius californicus (family Lygaeidae, subfamily Orsillinae), a pest commonly known as the seed bug, the chromosome complement is 2n = 16 (12A + 2m + XY), testes are formed by seven seminiferous tubules covered by an orange-colored membrane, and spermatogenesis is cystic. At prophase, sex chromosomes are heteropycnotic and autosomes usually show a chiasma. At metaphase, sex chromosomes along with microchromosomes may be seen located at the center of a ring formed by the remaining autosomes. A characteristic specific of N. californicus was the presence of nucleolar material observed from the cystic cell to the completely differentiated spermatozoon. Variations in size, shape and location of the nucleolar material occur during this process, denoting a variable degree of activity in the different stages. ©FUNPEC-RP.

Although they are of economic importance, there have been few cytogenetic studies of the Gerridae (Heteroptera) in Brazil. We examined spermatogenesis (meiosis and spermiogenesis) and nucleolar behavior in three species of the family Gerridae. Brachymetra albinerva and Halobatopsis platensis were found to have a chromosome complement of 2n = 25 (24A + X0) and Cylindrostethus palmaris 2n = 29 (28A + X0) chromosomes. Fifteen individuals of these species were collected from the reservoir of São José do Rio Preto, SP, using screens and were transported in pots containing water to the laboratory, where cytogenetic preparations were made. The polyploidy nuclei are formed by several heteropyknotic regions; cells in meiotic prophase have a heteropyknotic region that is probably the sex chromosome, and the chromosomes from chiasmata. The spermatids are rounded and have a heteropyknotic region at the periphery of the nucleus; the sperm head is small, with a long tail. Silver impregnation of meiotic cells showed one or more disorganized bodies around the perichromosomal sheath. The round spermatids had two bodies next to each other, but these were elongated; one of the bodies remained in the head and the other migrated to the initial part of the tail at the end of spermagenesis, when the staining was no longer evident. The meiotic cells appear during spermatogenesis and have very similar silver-impregnation patterns in different species of Heteroptera.

Bullfrog stem spermatogonia, also named primordial germ cells (PGCs), show strong testosterone immunolabeling in winter, but no or weak testosterone immunoexpression in summer. Thus, the role of testosterone in these cells needs to be clarified. In this study, we proposed to evaluate whether PGCs express aromatase and estrogen receptors, and verify a possible role of estrogen in PGCs seasonal proliferation. Testes of male adult bullfrogs, collected in winter (WG) and summer (SG), were fixed and embedded in historesin, for quantitative analysis, or paraffin for immunohistochemistry (IHC). The number of haematoxylin/eosin stained PGCs/lobular area was obtained. Proliferating cell nuclear antigen (PCNA), aromatase, estrogen receptor β (ERβ) and PCNA/ERβ double immunolabeling were detected by IHC. The number of PCNA-positive PGCs and the histological score (HSCORE) of aromatase and ERβ immunolabeled PGCs were obtained. Although the number of PGCs increased significantly in WG, a high number of PCNA-positive PGCs was observed in summer. Moreover, aromatase and ERβ HSCORE was higher in SG than WG. The results indicate that PGCs express a seasonal proliferative activity; the low mitotic activity in winter is related to the maximal limit of germ cells which can be supported in the large lobules. In SG, the increased ERβ and aromatase HSCORE suggests that testosterone is converted into estrogen from winter to summer. Moreover, the parallelism between the high PGCs mitotic activity and ERβ immunoexpression suggest a participation of estrogen in the control of the PGCs seasonal proliferative activity which guarantee the formation of new germ cysts from summer to next autumn. © 2012 Elsevier Inc.

The β-catenin/Lef/Tcf-mediated Wnt pathway is central to the developmental of all animals, stem cell renewal, and cancer progression. Prior studies in frogs and mice have indicated that the ligand Wnt-4 is essential for the mesenchyme to epithelial transition that generates tubules in the context of kidney organogenesis. More recently, Wnt-9b in mice, was likewise found to be required. Yet despite the importance of Wnt signals in renal development, the corresponding Frizzled receptor(s) and downstream signaling mechanim(s) are unclear. My work addresses these knowledge gaps using in vitro (Madin-Darby Canine Kidney cells) and in vivo (Xenopus laevis and zebrafish pronephros) tubulogenic kidney model systems. Employing established reporter constructs of Wnt/β-catenin pathway activity, I have determined that MDCK cells are highly responsive to Wnt-4, -1, and -3A, but not to Wnt-5A and control conditions. I have confirmed that Wnt-4's canonical signaling activity in MDCK cells is mediated by downstream effectors of the Wnt/β-catenin pathway using β-Engrailed and dnTCF-4, constructs that suppress this pathway. I have further found that MDCK cells express the Frizzled-6 receptor, and that Wnt-4 forms a biochemical complex with Frizzled-6, yet does not appear to transduce Wnt-4's canonical signal. Additionally, I demonstrate that standard Hepatocyte Growth Factor (HGF)-mediated (non-physiologic) induction of MDCK tubulogenesis in collagen matrices is not altered by activation or suppression of β-catenin signaling activity; however, β-catenin signaling maintains cell survival in this in vitro system. Using a Wnt/β-catenin signaling reporter in Xenopus laevis, I detect β-catenin signaling activity in the early pronephric epithelial kidney tubules. By inhibiting the Wnt/β-catenin signaling pathway in both zebrafish and Xenopus , a significant loss of kidney tubulogenesis is observed with little or no effect on adjoining axis or somite development. This inhibition also leads to the appearance of severe edema that phenocopies embryos depleted for Wnt-4. Tubulogenic loss does not appear to be caused by increased cell death in the Xenopus pronephric field, but rather by lessened expression of tubule epithelium genes associated with cellular differentiation. Together, my results show that Wnt/β-catenin signaling is required for renal tubule development and that Wnt-4 is a strong candidate for activating this pathway. ^

Kidney cortex is a main target for circulating vitamin B12 (cobalamin) in complex with transcobalamin (TC). Ligand blotting of rabbit kidney cortex with rabbit 125I-TC-B12 and human TC-57Co-B12 revealed an exclusive binding to megalin, a 600-kDa endocytic receptor present in renal proximal tubule epithelium and other absorptive epithelia. The binding was Ca2+ dependent and inhibited by receptor-associated protein (RAP). Surface plasmon resonance analysis demonstrated a high-affinity interaction between purified rabbit megalin and rabbit TC-B12 but no measurable affinity of the vitamin complex for the homologous alpha 2-macroglobulin receptor (alpha 2MR)/low density lipoprotein receptor related protein (LRP). 125I-TC-B12 was efficiently endocytosed in a RAP-inhibitable manner in megalin-expressing rat yolk sac carcinoma cells and in vivo microperfused rat proximal tubules. The radioactivity in the tubules localized to the endocytic compartments and a similar apical distribution in the proximal tubules was demonstrated after intravenous injection of 125I-TC-B12. The TC-B12 binding sites in the proximal tubule epithelium colocalized with megalin as shown by ligand binding to cryosections of rat kidney cortex, and the binding was inhibited by anti-megalin polyclonal antibody, EDTA, and RAP. These data show a novel nutritional dimension of megalin as a receptor involved in the cellular uptake of vitamin B12. The expression of megalin in absorptive epithelia in the kidney and other tissues including yolk sac and placenta suggests a role of the receptor in vitamin B12 homeostasis and fetal vitamin B12 supply.
