Background: The beneficial cardiovascular effects of vegetables may be underpinned by their high inorganic nitrate content. Objective: We sought to examine the effects of a 6-wk once-daily intake of dietary nitrate (nitrate-rich beetroot juice) compared with placebo intake (nitrate-depleted beetroot juice) on vascular and platelet function in untreated hypercholesterolemics. Design: A total of 69 subjects were recruited in this randomized, double-blind, placebo-controlled parallel study. The primary endpoint was the change in vascular function determined with the use of ultrasound flow-mediated dilatation (FMD). Results: Baseline characteristics were similar between the groups, with primary outcome data available for 67 patients. Dietary nitrate resulted in an absolute increase in the FMD response of 1.1% (an ∼24% improvement from baseline) with a worsening of 0.3% in the placebo group (P < 0.001). A small improvement in the aortic pulse wave velocity (i.e., a decrease of 0.22 m/s; 95% CI: −0.4, −0.3 m/s) was evident in the nitrate group, showing a trend (P = 0.06) to improvement in comparison with the placebo group. Dietary nitrate also caused a small but significant reduction (7.6%) in platelet-monocyte aggregates compared with an increase of 10.1% in the placebo group (P = 0.004), with statistically significant reductions in stimulated (ex vivo) P-selectin expression compared with the placebo group (P < 0.05) but no significant changes in unstimulated expression. No adverse effects of dietary nitrate were detected. The composition of the salivary microbiome was altered after the nitrate treatment but not after the placebo treatment (P < 0.01). The proportions of 78 bacterial taxa were different after the nitrate treatment; of those taxa present, 2 taxa were responsible for >1% of this change, with the proportions of Rothia mucilaginosa trending to increase and Neisseria flavescens (P < 0.01) increased after nitrate treatment relative to after placebo treatment. Conclusions: Sustained dietary nitrate ingestion improves vascular function in hypercholesterolemic patients. These changes are associated with alterations in the oral microbiome and, in particular, nitrate-reducing genera. Our findings provide additional support for the assessment of the potential of dietary nitrate as a preventative strategy against atherogenesis in larger cohorts. O estudo taxonômico do gênero Eleocharis R. Br. para o Rio Grande do Sul foi desenvolvido através dos métodos tradicionais em taxonomia. Os dados foram obtidos através da bibliografia, revisão de herbários regionais e coleta de exemplares a campo. O gênero está representado no Rio Grande do Sul por 27 espécies: Eleocharis acutangula (Roxb.) Schult., E. bonariensis Nees, E. contracta Maury, E. dunensis Kük., E. elegans (Kunth) Roem. & Schult., E. filiculmis Kunth, E. flavescens (Poir.) Urb., E. geniculata (L.) Roem. & Schult., E. interstincta (Vahl) Roem. & Schult., E. loefgreniana Boeck., E. maculosa (Vahl) Roem. & Schult., E. minima Kunth var. minima, E. montana (Kunth) Roem. & Schult., E. montevidensis Kunth, E. nudipes (Kunth) Palla, E. obtusetrigona (Lindl. & Nees) Steud., E. ochrostachys Steud., E. parodii Barros, E. aff. quinquangularis, E. quinquangularis Boeck., E. rabenii Boeck., E. radicans (Poir.) Kunth, E. sellowiana Kunth, E. squamigera Svenson, E. subarticulata (Nees) Boeck., E. viridans Kük. e Eleocharis sp. O trabalho apresenta descrições, ilustrações, dados sobre distribuição geográfica, habitat e períodos de floração e frutificação das espécies, além de uma chave dicotômica para diferenciá-las.

Realizaram-se estudos sobre sistemas de reprodução de Cassia spectabilis (L.) D C. (Leguminosae) observando-se a diversidade, freqüência e constância dos insetos visitantes em diferentes horários. Também testou-se a influência dos fatores ambientais em relação às visitas. Os resultados de polinização manual sugerem que C. spectabilis é autocompatível, porém, a xenogamia é o sistema de reprodução predominante. As inflorescências foram visitadas por uma grande quantidade de insetos, havendo predominância de abelhas. O horário de maior ocorrência dos insetos nas flores de C. spectabilis foi das 8 às 14 h e de menor ocorrência entre 7 e 8 h e das 17 às 18 h. Quanto ao comportamento dos insetos em relação à flor de C. spectabilis, observou-se que Xylocopa frontalis Olivier, X. suspecta Camargo & Moure, Bombus morio Swederus e Centris scopipes Friese possuem comportamento e morfologia adequados aos polinizadores legítimos; C. similis F., Oxaea flavescens Klug e Epicharis rustica flava Cockerell foram considerados polinizadores ocasionais. Pseudaugochloropsis graminea (F.), Tetragonisca angustula Latreille e A. mellifera L. foram considerados pilhadores. A polinização por vibração é o método usado pelas abelhas para coleta de pólen.

Paspalum dilatatum is a valuable forage grass in the subtropics. This species consists of several sexual (tetraploid) and apomict (penta- and hexaploid) biotypes. It has been proposed that the presence of a genome of unknown origin, the X genome, is responsible for apomixis in penta- and hexaploid biotypes. Here we evaluated the utility of random amplified polymorphic DNA (RAPD) markers for discriminating sexual and apomictic P dilatatum biotypes. DNA samples from nine accessions, including P. intermedium, P. juergensh, and P dilatatum (ssp. flavescens, and the common and Uruguayan biotypes) were analyzed with 86 RAPID primers. Three hundred sixty-two fragments were scored and genetic similarity estimates revealed that the penta- and hexaploid biotypes were highly similar (S,, greater than or equal to 0.913). Forty RAPDs were unique to the penta- and hexaploid biotypes. Overall RAPID markers were useful for assessing genetic variation among closely related P dilatatum genotypes as well as generating putative X genome markers.

Mycobacterium fortuitum is a rapidly-growing species of bacteria, ubiquitous in the environment and related to important human mycobacterioses. It has been isolated from blood, abscesses, the endocardium and surgical and traumatic wounds. This mycobacterium is hard to treat, being recognized in the literature as resistant even to the drugs used in the treatment of tuberculosis. The objective of this study was to screen extracts prepared from plants of the Brazilian cerrado (extended savanna-like belt) with known activity against M. fortuitum, employing the Microplate Alamar Blue Assay (MABA) as the analytical method. Out of 26 extracts tested against M. fortuitum, the nonpolar extract of Quassia amara (in methylene dichloride) gave the best result (MIC 62.5μg/ mL), followed by the nonpolar extracts of Syngonanthus macrolepsis, Davilla elliptica and Turnera ulmifolia, with equal MICs of 125μg/ml. The polar extracts (in ethanol and methanol) obtained from the same plants were considered inactive, since the MIC values determined were above 500μg/mL and not significantly different from those of extracts from other plants, without known activity.

Predation is a primary driver of tadpole assemblages, and the activity rate is a good predictor of the tadpoles' tolerance for predation risk. The conflicting demands between activity and exposure to predation can generate suboptimal behaviours. Because morphological components, such as body colouration, may affect the activity of tadpoles, we predict that environmental features that enhance or match the tadpole colouration should affect their survival or activity rate in the presence of a predator. We tested this prediction experimentally by assessing the mortality rate of tadpoles of Rhinella schneideri and Eupemphix nattereri and the active time on two artificial background types: one bright-coloured and one black-coloured. We found no difference in tadpole mortality due to the background type. However, R. schneideri tadpoles were more active than E. nattereri tadpoles, and the activity of R. schneideri was reduced less in the presence of the predator than that of E. nattereri. Although the background colouration did not affect the tadpole mortality rate, it was a stimulus that elicited behavioural responses in the tadpoles, leading them to adjust their activity rate to the type of background colour. © 2013 Dipartimento di Biologia, Università degli Studi di Firenze, Italia.

Background: Mycobacterium spp. is one of the most important species of zoonotic pathogens that can be transmitted from cattle to humans. The presence of these opportunistic, pathogenic bacteria in bovine milk has emerged as a public-health concern, especially among individuals who consume raw milk and related dairy products. To address this concern, the Brazilian control and eradication program focusing on bovine tuberculosis, was established in 2001. However, bovine tuberculosis continues to afflict approximately 1,3 percent of the cattle in Brazil. In the present study, 300 samples of milk from bovine herds, obtained from both individual and collective bulk tanks and informal points of sale, were cultured on Löwenstein-Jensen and Stonebrink media. Polymerase chain reaction (PCR)-based tests and restriction-enzyme pattern analysis were then performed on the colonies exhibiting phenotypes suggestive of Mycobacterium spp., which were characterized as acid-fast bacilli.Results: Of the 300 bovine milk samples that were processed, 24 were positively identified as Mycobacterium spp.Molecular identification detected 15 unique mycobacterial species: Mycobacterium bovis, M. gordonae, M. fortuitum, M. intracellulare, M. flavescens, M. duvalii, M. haemophilum, M. immunogenum, M. lentiflavum, M. mucogenicum, M. novocastrense, M. parafortuitum, M. smegmatis, M. terrae and M. vaccae. The isolation of bacteria from the various locations occurred in the following proportions: 9 percent of the individual bulk-tank samples, 7 percent of the collective bulk-tank samples and 8 percent of the informal-trade samples. No statistically significant difference was observed between the presence of Mycobacterium spp. in the three types of samples collected, the milk production profiles, the presence of veterinary assistance and the reported concerns about bovine tuberculosis prevention in the herds.Conclusion: The microbiological cultures associated with PCR-based identification tests are possible tools for the investigation of the presence of Mycobacterium spp. in milk samples. Using these methods, we found that the Brazilian population may be regularly exposed to mycobacteria by consuming raw bovine milk and related dairy products. These evidences reinforces the need to optimize quality programs of dairy products, to intensify the sanitary inspection of these products and the necessity of further studies on the presence of Mycobacterium spp. in milk and milk-based products. © 2013 Franco et al.; licensee BioMed Central Ltd.

Saccharomyces and non-Saccharomyces yeast species from a winery located in Brazil were identified by ribosomal gene-sequencing analysis. A total of 130 yeast strains were isolated from grape surfaces and musts during alcoholic fermentation from Isabel, Bordeaux, and Cabernet Sauvignon varieties. Samples were submitted to PCR-RFLP analysis and genomic sequencing. Thirteen species were identified: Candida quercitrusa, Candida stellata, Cryptococcus flavescens, Cryptococcus laurentii, Hanseniaspora uvarum, Issatchenkia occidentalis, Issatchenkia orientalis, Issatchenkia terricola, Pichia kluyveri, Pichia guilliermondii, Pichia sp., Saccharomyces cerevisiae, and Sporidiobolus pararoseus. A sequential substitution of species during the different stages of fermentation, with a dominance of non-Saccharomyces yeasts at the beginning, and a successive replacement of species by S. cerevisiae strains at the final steps were observed. This is the first report about the yeast distribution present throughout the alcoholic fermentation in a Brazilian winery, providing supportive information for future studies on their contribution to wine quality. © 2013 Springer Science+Business Media New York.
