A mercury-free electrode chemically modified with carbon paste containing dimethylglyoxime was used for determination of nickel in fuel ethanol. The instrumental parameters and composition of the modified paste were optimized. The analytical curve for nickel determination from 5.0 x 10(-9) to 5.0 x10(-7) mol(-1) was obtained using 25 min of accumulation time. The detection limit and amperometric sensitivity obtained for this method were 2.7 x 10 mol(-1) and 5.2 x 10(8) mu A mol(-1) L, respectively. The values for nickel concentration in four commercial samples of fuel ethanol were obtained in the range of 1.1 x 10(-8) to 6.9 x 10(-8) mol(-1). A comparison to graphite furnace atomic absorption spectrometry (GFAAS) was performed for nickel determination in commercial samples of ethanol.

A exposição ao mercúrio tem sido estudada em áreas sob influência de garimpos de ouro, entretanto, em alguns aspectos epidemiológicos os estudos ainda são insuficientes. O objetivo principal deste estudo foi avaliar a intensidade e a prevalência da exposição ao mercúrio em comunidades ribeirinhas do Tapajós ao longo do período de 1994 a 2008. O estudo foi conduzido em duas comunidades situadas próximo a nascente do rio Tapajós através da análise de Hg total em amostras de cabelos, medidos pela espectrofotometria de absorção atômica, no Laboratório de Toxicologia Humana e Ambiental da Universidade Federal do Pará (UFPA). Os resultados indicaram que as comunidades possuem uma grande freqüência de mulheres em idade reprodutiva, a maioria dos habitantes tem um longo tempo de residência local e a dieta é caracterizada pela elevada frequência de refeições do pescado da região. O tempo de exposição ao mercúrio vem se estendendo há pelo menos 14 anos, com níveis que oferecem riscos à saúde, principalmente, para o grupo materno-infantil, e não há diferença de exposição entre as duas comunidades. A prevalência de exposição ao mercúrio (habitantes com níveis de Hg total (Hg total) > 10 μg/g) foi alta nas duas comunidades. Não houve correlação entre a produção de ouro e os níveis de Hg total em amostras de cabelo. Conclui-se que os níveis de exposição apresentados no período do estudo caracterizam uma exposição em longo prazo os quais são capazes de oferecem riscos à saúde, principalmente materno fetal; que a frequência na ingestão de peixes contaminados pode contribuir para o aparecimento de danos causados pelo mercúrio. Monitoramento toxicológico e clínico dos indivíduos expostos, bem como, aplicação de medidas educativas deverão ser fortalecidas com vista a prevenção da doença causada pelo mercúrio.

Neste trabalho é apresentada uma nova técnica para a realização do empilhamento sísmico, aplicada ao problema do imageamento de refletores fixos em um meio bidimensional, suavemente heterogêneo, isotrópico, a partir de dados de reflexão. Esta nova técnica chamada de imageamento homeomórfico tem como base a aproximação geométrica do raio e propriedades topológicas dos refletores. São utilizados, portanto, os conceitos de frente de onda, ângulo de incidência, raio de curvatura da frente de onda, cáustica e definição da trajetória do raio; de tal modo que a imagem obtida mantém relações de homeomorfismo com o objeto que se deseja imagear. O empilhamento sísmico é feito, nesta nova técnica de imageamento, aplicando-se uma correção local do tempo, ∆ t, ao tempo de trânsito, t, do raio que parte da fonte sísmica localizada em xo, reflete-se em um ponto de reflexão, Co, sendo registrado como uma reflexão primária em um geofone localizado em xg, em relação ao tempo de referência to no sismograma, correspondente ao tempo de trânsito de um raio central. A fórmula utilizada nesta correção temporal tem como parâmetros o raio de curvatura Ro, o ângulo de emergência βo da frente de onda, no instante em que a mesma atinge a superfície de observação, e a velocidade vo considerada constante nas proximidades da linha sísmica. Considerando-se uma aproximação geométrica seguido um círculo para a frente de onda, pode-se estabelecer diferentes métodos de imageamento homeomórfico dependendo da configuração de processamento. Sendo assim tem-se: 1) Método Elemento de Fonte (Receptor) Comum (EF(R)C). Utiliza-se uma configuração onde se tem um conjunto de sismogramas relacionado com uma única fonte (receptor), e considera-se uma frente de onda real (de reflexão); 2) Método Elemento de Reflexão Comum (ERC). Utiliza-se uma configuração onde um conjunto de sismogramas é relacionado com um único ponto de reflexão, e considera-se uma frente de onda hipoteticamente originada neste ponto; 3) Método Elemento de Evoluta Comum (EEC). Utiliza-se uma configuração onde cada sismograma está relacionado com um par de fonte e geofone coincidentemente posicionados na linha sísmica, e considera-se uma frente de onda hipoteticamente originada no centro de curvatura do refletor. Em cada um desses métodos tem-se como resultados uma seção sísmica empilhada, u(xo, to); e outras duas seções denominadas de radiusgrama, Ro (xo, to), e angulograma, βo(xo, to), onde estão os valores de raios de curvatura e ângulos de emergência da frente de onda considerada no instante em que a mesma atinge a superfície de observação, respectivamente. No caso do método denominado elemento refletor comum (ERC), a seção sísmica resultante do empilhamento corresponde a seção afastamento nulo. Pode-se mostrar que o sinal sísmico não sofre efeitos de alongamento como consequência da correção temporal, nem tão pouco apresenta problemas de dispersão de pontos de reflexão como consequência da inclinação do refletor, ao contrário do que acontece com as técnicas de empilhamento que tem por base a correção NMO. Além disto, por não necessitar de um macro modelo de velocidades a técnica de imageamento homeomórfico, de um modo geral, pode também ser aplicada a modelos heterogêneos, sem perder o rigor em sua formulação. Aqui também são apresentados exemplos de aplicação dos métodos elemento de fonte comum (EFC) (KEYDAR, 1993), e elemento refletor comum (ERC) (STEENTOFT, 1993), ambos os casos com dados sintéticos. No primeiro caso, (EFC), onde o empilhamento é feito tendo como referência um raio central arbitrário, pode-se observar um alto nível de exatidão no imageamento obtido, além do que é dada uma interpretação para as seções de radiusgrama e angulograma, de modo a se caracterizar aspectos geométricos do model geofísico em questão. No segundo caso, (ERC), o método é aplicado a série de dados Marmousi, gerados pelo método das diferenças finitas, e o resultado é comparado com aquele obtido por métodos convecionais (NMO/DMO) aplicados aos mesmos dados. Como consequência, observa-se que através do método ERC pode-se melhor detectar a continuidade de refletores, enquanto que através dos métodos convencionais caracterizam-se melhor a ocorrência de difrações. Por sua vez, as seções de radiusgrama e angulograma, no método (ERC), apresentam um baixo poder de resolução nas regiões do modelo onde se tem um alto grau de complexidade das estruturas. Finalmente, apresenta-se uma formulação unificada que abrange os diferentes métodos de imageamento homeomórfico citados anteriormente, e também situações mais gerais onde a frente de onda não se aproxima a um círculo, mas a uma curva quadrática qualquer.

A new selective sensor based on molecularly imprinted polymers (MIPs) was developed for the determination of hexazinone (HXZ) in environmental samples. MIPs were synthesized using a non-covalent approach, and selection of the monomers employed in the polymerization reaction was carried out by molecular modeling. Three functional monomers with high (2-vinylpyridine (MP17)) and intermediate (methacrylic acid (MP12) and acrylamide (MP5)) energies of binding to the template (HXZ) were selected for preparation of the MIPs, in order to conduct comparative studies and validate the theoretical data. For sensor construction, carbon pastes were modified with each MIP or NIP (non-imprinted polymer), and HXZ determination was performed using differential pulse adsorptive cathodic stripping voltammetry (DPAdCSV). All parameters affecting the sensor response were optimized. In HCl at pH 2.5, the sensor prepared with MP17 (5% w/w in the paste) showed a dynamic linear range between 1.9 × 10−11 and 1.1 × 10−10 mol L−1, and a detection limit of 2.6 × 10−12 mol L−1, under the following conditions: accumulation time of 200 s at a potential of −0.5V, scan rate of 50 mVs−1, pulse amplitude of 60 mV, and pulse width of 50 ms. The sensor was selective in the presence of other similar compounds, and was successfully applied to the analysis of HXZ in river water samples.

Aerial surveys were conducted in 1999 and 2000 to estimate the densities of ringed (Phoca hispida) and bearded (Erignathus barbatus) seals in the eastern Chukchi Sea. Survey lines were focused mainly on the coastal zone within 37 km of the shoreline, with additional lines flown 148–185 km offshore to assess how densities of seals changed as a function of distance from shore. Satellite-linked time-depth recorders were attached to ringed seals in both years to evaluate the time spent basking on the ice surface. Haulout patterns indicated that ringed seals transitioned to basking behavior in late May and early June, and that the largest proportion of seals (60–68%) was hauled out between 0830 and 1530 local solar time. Ringed seals were relatively common in nearshore fast ice and pack ice, with lower densities in offshore pack ice. The average density of ringed seals was 1.91 seals km-2 in 1999 (range 0.37– 16.32) and 1.62 seals km-2 in 2000 (range 0.42–19.4), with the highest densities of ringed seals found in coastal waters south of Kivalina and near Kotzebue Sound. The estimated abundance of ringed seals for the entire study area was similar in 1999 (252,488 seals, SE=47,204) and 2000 (208,857 seals, SE=25,502). Bearded seals were generally more common in offshore pack ice, with the exception of high bearded seal numbers observed near the shore south of Kivalina. Bearded seal densities were not adjusted for haulout behavior, and therefore, abundance was not estimated. Unadjusted average bearded seal density was 0.07 seals km-2 in 1999 (range 0.011–0.393) and 0.14 seals km-2 in 2000 (range 0.009– 0.652). Levels of primary productivity, benthic biomass, and fast ice distribution may influence the distributions of ringed and bearded seals in the Chukchi Sea. Information on movement and haulout behavior of ringed and bearded seals would be very useful for designing future surveys.

The abundance of harbor seals (Phoca vitulina richardii) has declined in recent decades at several Alaska locations. The causes of these declines are unknown, but there is concern about the status of the populations, especially in the Gulf of Alaska. To assess the status of harbor seals in the Gulf of Alaska, we conducted aerial surveys of seals on their haul-out sites in August-September 1996. Many factors influence the propensity of seals to haul out, including tides, weather, time of day, and time of year. Because these “covariates” cannot simultaneously be controlled through survey design, we used a regression model to adjust the counts to an estimate of the number of seals that would have been ashore during a hypothetical survey conducted under ideal conditions for hauling out. The regression, a generalized additive model, not only provided an adjustment for the covariates, but also confirmed the nature and shape of the covariate effects on haul-out behavior. The number of seals hauled out was greatest at the beginning of the surveys (mid-August). There was a broad daily peak from about 1100-1400 local solar time. The greatest numbers were hauled out at low tide on terrestrial sites. Tidal state made little difference in the numbers hauled out on glacial ice, where the area available to seals did not fluctuate with the tide. Adjusting the survey counts to the ideal state for each covariate produced an estimate of 30,035 seals, about 1.8 times the total of the unadjusted counts (16,355 seals). To the adjusted count, we applied a correction factor of 1.198 from a separate study of two haul-out sites elsewhere in Alaska, to produce a total abundance estimate of 35,981 (SE 1,833). This estimate accounts both for the effect of covariates on survey counts and for the proportion of seals that remained in the water even under ideal conditions for hauling out.

The stability of oil-in-water (O/W) emulsions used as metalworking fluids is a key factor for the economical and environmental balance of the entire metalworking process because used and broken fluids must be recycled or disposed. In this study, the ability of turbidimetric spectroscopy in the ultraviolet and visible light range to detect metalworking fluids destabilization was evaluated. Destabilization was achieved by adding calcium chloride, thus achieving accelerated aging, which leads to coalescence, creaming, and complete emulsion separation. The stability of the metalworking fluids at 5% volumetric concentration in deionized water was monitored using a spectroscopic turbidimeter composed of an optical probe for in-line measurements. Destabilization was also monitored by measuring the vertical profile of backscattered and transmitted light. The results of this offline measurement system were compared with those from the in-line spectroscopic sensor, indicating that the latter can provide local, real-time information on emulsion destabilization, thus enabling control actions.

Resultate dieser Arbeit zeigen, dass endotheliale und neuronale NO-Synthasen (eNOS und nNOS) ihr Substrat Arginin nicht ausschließlich aus extrazellulären, sondern auch aus intrazellulären Quellen beziehen. Das Substrat aus den intrazellulären Quellen scheint nicht über Membrantransporter in den Extrazellulärraum gelangen zu können. Dies deutet darauf hin, dass eine enge Assoziation der Arginin-bereitstellenden Enzyme mit eNOS bzw. nNOS vorliegen könnte. Dadurch würde das durch diese Enzyme generierte Arginin direkt an die NOS weitergereicht und nicht über Transporter gegen andere basische Aminosäuren (AS) im Extrazellulärraum ausgetauscht werden. Eine intrazelluläre Substrat-Quelle besteht aus dem so genannten „Recycling“, der Umwandlung des bei der NO-Synthese entstehenden Citrullins in Arginin. Eine Kopplung von Arginin-bereitstellenden „Recycling“-Enzymen mit NOS wird in Endothelzellen und teilweise auch in TGW-nu-I Neuroblastomzellen beobachtet, nicht jedoch in A673 Neuroepitheliomzellen. Die Kopplung scheint daher vom Zelltyp abhängig zu sein. Das zur Arginin-Regeneration benötigte Citrullin kann allen untersuchten Zellen durch den Austausch mit spezifischen neutralen AS, die ausschließlich zum Substratprofil des System N Transporters SN1 passen, entzogen werden. Die Anwesenheit von SN1-Substraten im Extrazellulärraum führt daher indirekt zu einer Depletion der Recycling-Quelle. SN1 mRNA ist in allen untersuchten Zellen nachweisbar. Aus Protein-Abbau stammendes Arginin stellt den zweiten Teil der intrazellulären Arginin-Quelle dar. Dieser ist in allen untersuchten eNOS- oder nNOS exprimierenden Zellen vorhanden. Das Arginin stammt dabei sowohl aus lysosomalem als auch proteasomalen Proteinabbau, wie der Einsatz spezifischer Inhibitoren zeigt. Extrazelluläres Histidin (aber keine andere Aminosäure) kann diese Arginin-Quelle depletieren. Wir vermuten deshalb, dass Histidin über den Peptid-Histidin-Transporter PHT1, der in allen untersuchten Zellen stark exprimiert ist, gegen die durch lysosomalen und proteasomalen Proteinabbau entstehenden Arginin-haltigen Di- und Tripeptide ausgetauscht wird. Der wichtigste endogene NOS-Inhibitor, asymmetrisches Dimethylarginin (ADMA), ein Marker für endotheliale Dysfunktion und Risikofaktor für kardiovaskuläre Erkrankungen, stammt ebenfalls aus Proteinabbau. Die Verfügbarkeit dieser intrazellulären Arginin-Quelle wird deshalb stark vom Methylierungsgrad des Arginins in den abgebauten Proteinen abhängen. Eine lokale ADMA-Anreicherung könnte eine Erklärung für das Arginin-Paradox sein, der unter pathophysiologischen Bedingungen beobachteten Verminderung der endothelialen NO-Synthese bei anscheinend ausreichenden intrazellulären Argininkonzentrationen. Da auch in neurodegenerativen Erkrankungen, wie Morbus Alzheimer, ADMA eine Rolle zu spielen scheint, könnte das Arginin-Paradox auch für die nNOS-vermittelte NO-Synthese von Bedeutung sein. Die Resultate demonstrieren, dass die Substratversorgung der beiden NOS-Isoformen nicht ausschließlich von kationischen Aminosäuretransportern abhängig ist, sondern auch von Transportern für neutrale Aminosäuren und Peptide, und außerdem von Arginin-bereitstellenden Enzymen. Der jeweilige Beitrag der verschiedenen Arginin-Quellen zur Substratversorgung der NOS ist daher abhängig vom Anteil der jeweiligen Aminosäuren und Peptide in der extrazellulären Flüssigkeit.

Repeat airborne laser altimeter measurements are used to derive surface elevation changes on parts of Whillans Ice Stream and Ice Stream C, West Antarctica. Elevation changes are converted to estimates of ice equivalent thickness change using local accumulation rates, surface snow densities and vertical bedrock motions. The surveyed portions of two major tributaries of Whillans Ice Stream are found to be thinning almost uniformly at an average rate of similar to 1 m a(-1). Ice Stream C has a complicated elevation-change pattern, but is generally thickening. These results are used to estimate the contribution of each surveyed region to the current rate of global sea-level rise.

This study presents an integrated mineralogical-geochemical data base on fine-grained sediments transported by all major rivers of southern Africa, including the Zambezi, Okavango, Limpopo, Olifants, Orange and Kunene. Clay mineralogy, bulk geochemistry, Sr and Nd isotopic signatures of river mud, considered as proxy of suspended load, are used to investigate the influence of source-rock lithology and weathering intensity on the composition of clay and silt produced in subequatorial to subtropical latitudes. Depletion in mobile alkali and alkaline-earth metals, minor in arid Namibia, is strong in the Okavango, Kwando and Upper Zambezi catchments, where recycling is also extensive. Element removal is most significant for Na, and to a lesser extent for Sr. Depletion in K, Ca and other elements, negligible in Namibia, is moderate elsewhere. The most widespread clay minerals are smectite, dominant in muds derived from Karoo or Etendeka flood basalts, or illite and chlorite, dominant in muds derived from metasedimentary rocks of the Damara Orogen or Zimbabwe Craton. Kaolinite represents 30-40% of clay minerals only in Okavango and Upper Zambezi sediments sourced in humid subequatorial Angola and Zambia. After subtracting the effects of recycling and of local accumulation of authigenic carbonates in soils, the regional distribution of clay minerals and chemical indices consistently reflect weathering intensity primarily controlled by climate. Bulk geochemistry identifies most clearly volcaniclastic sediments and mafic sources in general, but cannot discriminate the other sources of detritus in detail. Instead, Sr and Nd isotopic fingerprints are insensitive to weathering, and thus mirror faithfully the tectonic structure of the southern African continent. Isotopic tools thus represent a much firmer basis than bulk geochemistry or clay mineralogy in the provenance study of mudrocks.

Institutional Review Boards (IRBs) are the primary gatekeepers for the protection of ethical standards of federally regulated research on human subjects in this country. This paper focuses on what general, broad measures that may be instituted or enhanced to exemplify a "model IRB". This is done by examining the current regulatory standards of federally regulated IRBs, not private or commercial boards, and how many of those standards have been found either inadequate or not generally understood or followed. The analysis includes suggestions on how to bring about changes in order to make the IRB process more efficient, less subject to litigation, and create standardized educational protocols for members. The paper also considers how to include better oversight for multi-center research, increased centralization of IRBs, utilization of Data Safety Monitoring Boards when necessary, payment for research protocol review, voluntary accreditation, and the institution of evaluation/quality assurance programs. ^ This is a policy study utilizing secondary analysis of publicly available data. Therefore, the research for this paper focuses on scholarly medical/legal journals, web information from the Department of Health and Human Services, Federal Drug Administration, and the Office of the Inspector General, Accreditation Programs, law review articles, and current regulations applicable to the relevant portions of the paper. ^ Two issues are found to be consistently cited by the literature as major concerns. One is a need for basic, standardized educational requirements across all IRBs and its members, and secondly, much stricter and more informed management of continuing research. There is no federally regulated formal education system currently in place for IRB members, except for certain NIH-based trials. Also, IRBs are not keeping up with research once a study has begun, and although regulated to do so, it does not appear to be a great priority. This is the area most in danger of increased litigation. Other issues such as voluntary accreditation and outcomes evaluation are slowing gaining steam as the processes are becoming more available and more sought after, such as JCAHO accrediting of hospitals. ^ Adopting the principles discussed in this paper should promote better use of a local IRBs time, money, and expertise for protecting the vulnerable population in their care. Without further improvements to the system, there is concern that private and commercial IRBs will attempt to create a monopoly on much of the clinical research in the future as they are not as heavily regulated and can therefore offer companies quicker and more convenient reviews. IRBs need to consider the advantages of charging for their unique and important services as a cost of doing business. More importantly, there must be a minimum standard of education for all IRB members in the area of the ethical standards of human research and a greater emphasis placed on the follow-up of ongoing research as this is the most critical time for study participants and may soon lead to the largest area for litigation. Additionally, there should be a centralized IRB for multi-site trials or a study website with important information affecting the trial in real time. There needs to be development of standards and metrics to assess the performance of the IRBs for quality assurance and outcome evaluations. The boards should not be content to run the business of human subjects' research without determining how well that function is actually being carried out. It is important that federally regulated IRBs provide excellence in human research and promote those values most important to the public at large.^
