Radical livre é qualquer substância, átomo ou molécula capaz de existir independente, e que possua elétrons desemparelhados em seu último orbital energético. Uma vez formados, começam uma série de reações, podendo levar a danos em biomoléculas (lipídeos, proteínas e também o DNA). Os radicais livres podem ser gerados, entre outras formas, pelo exercício físico aeróbio, que eleva o consumo de oxigênio (VO2) entre 10-15 vezes mais que em situação de repouso, essa elevação induz uma maoir atividade mitocondrial, onde aproximadamente 5% do oxigênio utilizado na mitocôndria, como aceptor de eletrons, é liberado na forma de superóxido. Porém, especula-se que as espécies reativas de oxigênio, são geradas no exercício anaeróbio por um aumento na atividade da xantina oxidase, pela liberação de prótons, provocada pela acidose láctica (que em estudos in vitro, mostrou ser um potente fator pró-oxidante), por uma atividade aumentada da óxido nítrico sintase, pela autooxidação de catecolaminas, pela síndrome de esquemia/reperfusão, entre outras fontes. O organismo, para se proteger desses danos oxidativos, possui dois tipos de proteção antioxidante, a enzimática: como a catalase, a superóxido dismutase e a glutationa peroxidase, e o sistema antioxidante não-enzimatico, onde, podemos citar dentro de uma vasta lista: ácido úrico, vitaminas E, A e C, bilirrubina, albumina e compostos fenólicos, entre outros. O treinamento físico induz adaptações antioxidante ao organismo dos indivíduos, onde os sujeitos são expostos cronicamente a condição de estresse oxidativo, que é onde a formação de espécies reativas de oxigênio é maior que a capacidade protetora, e isto faz com que ocorra um aumento na atividade ou conteúdo dos antioxidantes, ou então que a produção desses oxidantes seja menor. Com isso, o objetivo desse estudo foi comparar o estresse oxidativo induzido pelo exercício, através de aspectos bioquímicos e fisiológicos comparando atletas profissionais de voleibol de quadra, jogadores de vôlei de praia e indivíduos não treinados. Todos os sujeitos foram voluntários, do sexo masculino, não fumantes, sem fazer uso de drogas/suplementos/medicamentos, não ingeriram bebidas alcoólicas, assim como também não praticaram atividade física exaustiva 48 h antes dos testes. Os sujeitos executaram um teste máximo de carga progressiva para determinar o consumo máximo de oxigênio, servindo para determinação da carga do teste aeróbio submáximo de 1 hora, que foi igual para todos os voluntários (10% abaixo do segundo limiar ventilatório), além do teste anaeróbio Wingate com 30 segundos de duração. Os indivíduos receberam a prescrição de uma dieta padrão, que se constituiu 100% da RDA para cada indivíduo.

Considerando à lacuna sobre as informações da biologia reprodutiva dos crustáceos de água doce no Brasil, principalmente dos lagostins, a presente pesquisa teve como objetivo caracterizar aspectos da dinâmica da reprodução, como a descrição e desenvolvimento das gônadas, e analisar as variações bioquímicas do metabolismo intermediário de Parastacus varicosus Faxon, 1898 para relacioná-las ao período reprodutivo. Os animais foram amostrados de junho de 2004 a maio de 2005 em um arroio pertencente à Bacia do Rio Gravataí, localidade Cova do Touro, Gravataí, Rio Grande do Sul. As fêmeas foram separadas em três estágios gonadais de acordo com a maturação. Todos animais apresentaram os dois pares de aberturas e dutos genitais. Foram identificados histologicamente 40 machos, 32 fêmeas e 6 espécimes transicionais. Os índices gonadossomático (IG) e hepatossomático (IH) foram determinados. Ocorreu um aumento do IG e uma diminuição no IH, no período de passagem da primavera para o verão, indicando uma maior atividade reprodutiva no verão e uma possível transferência das reservas do hepatopâncreas para a gônada nesse período. Nas análises bioquímicas da hemolinfa, hepatopâncreas, músculo e gônadas os níveis de glicose, glicogênio, proteínas totais, colesterol e lipídeos totais foram quantificados. As variações sazonais observadas mostraram estar relacionada com o período reprodutivo, mostrando uma transferência de parte das reservas energéticas do hepatopâncreas e do músculo para o desenvolvimento das gônadas durante período reprodutivo. Nessa pesquisa portanto foi observada uma correlação entre os resultados obtidos das investigações das gônadas e das análises bioquímicas indicando assim uma conexão entre essas pesquisas e contribuindo para um melhor entendimento da biologia reprodutiva de P. varicosus.

Introdução: A incidência da doença arterial coronária é uma das principais causas de morbidade e motalidade em diversos países e o estudo dos fatores de risco têm grande importância na prevenção e no tratamento dessa enfermidade. Entre outros fatores, a obesidade e a obesidade abdominal têm sido associadas com a maior incidência de DAC. A ingestão diária de nutrientes também pode estar relacionada com essa doença, porém, uma vez que a alimentação é complexa e contém diversos nutrientes, ainda não foi possível elucidar o impacto da alimentação no risco de desenvolver a doença arterial coronária. Objetivo: Avaliar a relação entre o consumo alimentar diário, a presença de obesidade abdominal e achados angiográficos de obstrução arterial em pacientes portadores de cardiopatia isquêmica, submetidos a cateterismo cardíaco. Métodos: Foi realizado um estudo transversal, com 284 pacientes submetidos a cateterismo cardíaco, da unidade de hemodinâmica de um hospital universitário. Foi avaliada a RCQ, o IMC, a ingestão alimentar diária através de um inquérito nutricional, a análise bioquímica do sangue e a avaliação do laudo do cateterismo cardíaco. Resultados: Dos pacientes avaliados, 172 indivíduos (60,6%) apresentavam alterações em uma ou mais artérias coronárias. A ingestão média diária de calorias foi de 2450,56 Kcal/dia. O consumo de proteínas foi em média 1,66 g/Kg/dia, de carboidratos foi de 3,83 g/Kg/dia e de lipídeos foi de 1,21 g/Kg/dia. A idade, o sexo masculino, os níveis séricos de triglicerídeos, o consumo de álcool e a glicemia em jejum foram estatisticamente significativos na análise multivariada. Conclusão: Nos pacientes avaliados, o consumo diário de calorias encontra-se adequado, porém a ingestão de proteínas, carboidratos e lipídeos estão inadequados. Em relação aos fatores de risco para DAC, as mulheres apresentaram maior associação para desenvolver a síndrome metabólica do que os homens.

Nanoemulsões catiônicas têm sido recentemente consideradas como potenciais sistemas de liberação de oligonucleotídeos. O objetivo do presente trabalho foi avaliar a influência da adição de quantidades crescentes dos lipídeos catiônicos estearilamina (EA), oleilamina (OA) ou DOTAP (DT) sobre propriedades físico-químicas de nanoemulsões como sistemas de liberação de um oligonucleotídeo modelo (pdT16). As formulações foram preparadas pelo procedimento de emulsificação espontânea. As nanoemulsões constituídas de triglicerídeos de cadeia média, lecitina de gema de ovo, glicerol, quantidades crescentes dos lipídeos catiônicos (até 20 mM) e água foram caracterizadas em termos de diâmetro médio, pH, potencial zeta e viscosidade. Baseado nos resultados obtidos, as formulações contendo EA, OA e DT na concentração de 2 mM foram selecionadas para a continuidade do trabalho, uma vez que apresentam diâmetro de gotícula e potencial zeta de cerca de 250 nm e 40-50 mV, respectivamente. A taxa de associação do pdT16 às nanoemulsões foi determinada, indiretamente, pelo doseamento deste, na fase aquosa externa após separação em membranas de ultrafiltração/centrifugação, por meio de espectrofotometria no UV. A maior taxa de associação do pdT16 foi detectada para a formulação contendo o lipídeo catiônico DT (até ~70 mg/g de fase interna). Evidências adicionais da associação do pdT16 com as nanoemulsões foram detectadas pelo aumento do diâmetro de gotícula, inversão do potencial zeta e morfologia das gotículas. Em uma última etapa, um estudo preliminar da liberação do pdT16 foi realizado especialmente para a formulação contendo o lipídeo catiônico DT. O fator de diluição e relação de cargas [+/-] foram identificados como os principais parâmetros que influenciam a liberação do pdT16 a partir das nanoemulsões. Em conclusão, o conjunto dos resultados obtidos demonstra a influência da concentração e da natureza do lipídeo catiônico empregado sobre as propriedades físico-químicas de nanoemulsões catiônicas como sistemas de liberação de oligonucleotídeos.

The mushrooms have been object of intense research in view of its potential raising of application in different sectors of the pharmacology and alimentary industry. Among diverse bioactive composites of polyssacharides nature that exist in the fungus the glucans are much searched. These are polymers of glucose and classified as the type of glicosidic linking [α, β]. Peroxisome proliferator-activated receptors (PPARs), ranscription factors belonging to the family of nuclear receptors that bind themselves o specific agonists, have shown their importance in controlling the inflammatory process. The aim of this study was to perform a chemical characterization of extract rom the mushroom Caripia montagnei, assess its antiinflammatory and antibacterial effect and determine if this effect occurs via PPAR. This mushroom is composed of carbohydrates (63.3±4.1%), lipids (21.4l±0.9%) and proteins (2.2± 0.3%). The aqueous solution resulting from the fractionation contained carbohydrates (98.7±3.3%) and protein (1.3±0.25%). Analyses of infrared spectrophotometry and of nuclear magnetic esonance demonstrated that the extract of mushroom C. montagnei is rich in β-glucans. In hioglycolate-induced peritonitis, the C. montagnei glucans (50 mg/kg) educed the inflammatory process in 65.5±5.2% and agonists, pharmacological igands, for PPAR: Wy-14643 (49.3±6.1%), PFOA (48.9±3.8%) and clofibrate in 45.2±3.2%. Sodium diclofenac showed a reduction of 81.65±0.6%. In the plantar edema, the glucans from C. montagnei (50 mg/kg) and L-NAME reduced the edema to a similar degree 91.4±0.3% and 92.8±0,5 %, respectively. In all the groups tested, nitric oxide (NO), an inflammation mediator, showed a significant reduction in the nitrate/nitrite levels when compared to the positive control (P<0.001). The C. montagnei glucans did not show cytotoxicity in the concentrations tested (2.5, 5.0, 10.0, 20.0 and 40.0 µg/100 µL). Antibacterial activity demonstrated that, unlike total extract, there was no inhibition of bacterial growth. The C. montagnei glucans show great potential for antiinflammatory applications. This effect suggests that it is mediated by PPAR activation and by COX and iNOS inhibition

The plants are often exposed to variations in environmental conditions that may trigger metabolic disturbances leading to a consequent loss in productivity of crops. These stressful conditions usually induce an accumulation of reactive oxygen species (ROS) in the cell, a condition known how oxidative stress. Among these species, hydrogen peroxide (H2O2) is an important molecule involved in numerous signaling mechanisms. The present study aimed to understand the relationship between the different enzymatic mechanisms of elimination of H2O2 by catalase (CAT) and ascorbate peroxidase (APX) in leaf tissues of seedlings of the species Vigna unguiculata L. Walp, under conditions of oxidative stress induced by application of CAT inhibitor, 3-amino-1,2,4-triazole (3-AT), and H2O2 itself on the roots. Three experiments were conducted. The first experiment was performed applying the compound 3-AT (5 mM) during the time (hours). In the second experiment, seedlings were exposed to different concentrations of H2O2 (2.5, 5.0, 7.5, 10 mM) for 48 h. The third strategy included the pre-treatment with H2O2 (2.5 mM) for 24 h, followed by subsequent treatment with the inhibitor 3-AT and recovery control condition. Treatment with 3-AT causes a strong inhibition of CAT activity in leaf tissues accompanied by an increase of activity of APX. However a decrease in oxidative damage to lipids is not observed as indicated by TBARS. It was observed that activity of APX is directly linked to the content of peroxide. Inductions in the activities of CAT and APX were observed mainly in the seedlings treated with 2.5 mM H2O2. This can be associated with a decrease in oxidative damage to lipids. In contrast, one same tendency was not observed in treatments with higher concentrations of this ROS. These results suggest that the concentration of 2.5 mM H2O2 can induce responses antioxidants later in seedling cowpea. This concentration when applied as pre-treatment for 24 h promoted an induction systems removers CAT and APX, both in activity and in terms of gene expression. However this increment was not observed in the recovered plants and the plants subsequently subjected to 3-AT. Additionally, the pretreatment was not sufficient to attenuate the inhibition of CAT activity and oxidative damage to lipids caused by the subsequent application of this inhibitor. The results showed that the application of 3-AT and H2O2 in the root systems of seedlings of cowpea promote changes in the parameters analyzed in leaf tissues that indicate a direct response to the presence of these factors or systemic signaling mecanisms. H2O2 appears to activate the responses of two antioxidant systems in this study thar does not promote greater protection in case of additional treatment with 3-AT. This demonstrates the importance of the CAT system. In this work, complete results indicate that there is a difference between the signaling and the effects caused by exposure to H2O2 and by treatment with 3-AT

Gene therapy is one of the major challenges of the post-genomic research and it is based on the transfer of genetic material into a cell, tissue or organ in order to cure or improve the patient s clinical status. In general, gene therapy consists in the insertion of functional genes aiming substitute, complement or inhibit defective genes. The achievement of a foreigner DNA expression into a population of cells requires its transfer to the target. Therefore, a key issue is to create systems, vectors, able to transfer and protect the DNA until it reaches the target. The disadvantages related to the use of viral vectors have encouraged efforts to develop emulsions as non-viral vectors. In fact, they are easy to produce, present suitable stability and enable transfection. The aim of this work was to evaluate two different non-viral vectors, cationic liposomes and nanoemulsions, and the possibility of their use in gene therapy. For the two systems, cationic lipids and helper lipids were used. Nanoemulsions were prepared using sonication method and were composed of Captex® 355; Tween® 80; Spam® 80; cationic lipid, Stearylamine (SA) or 1,2-dioleoyl-3-trimethylammoniumpropane (DOTAP) and water (Milli-Q®). These systems were characterized by average droplet size, Polidispersion Index (PI) and Zeta Potential. The stability of the systems; as well as the DNA compaction capacity; their cytotoxicity and the cytotoxicity of the isolated components; and their transfection capacity; were also evaluated. Liposomes were made by hydration film method and were composed of DOTAP; 1,2-dioleoyl-sn-glycero-3-phosphoethanolamine (DOPE), containing or not Rhodaminephosphatidylethanolamine (PE- Rhodamine) and the conjugate Hyaluronic Acid DOPE (HA-DOPE). These systems were also characterized as nanoemulsions. Stability of the systems and the influence of time, size of plasmid and presence or absence of endotoxin in the formation of lipoplexes were also analyzed. Besides, the ophthalmic biodistribution of PE-Rhodamine containing liposomes was studied after intravitreal injection. The obtained results show that these systems are promising non-viral vector for further utilization in gene therapy and that this field seems to be very important in the clinical practice in this century. The genus Saccharum belongs to Poaceae family. Sugarcane has become important monocultures in Brazil due to their products: ethanol and sugar. The production may change between different regions from Brazil. This difference is related to soil, climatic conditions and temperature that promotes oxidative stress that may induce an early flowering. The aim of this work was to identify the effects of oxidative stress. In order to analyse this, sugarcane plants were submitted to oxidative stress using hydrogen peroxide. After this treatment, the oxidative stress were analyzed Then, the plant responses were analyzed under different approaches, using morphophysiological, biochemical and molecular tools. Thus, sugarcane plants were grown under controlled conditions and until two months they were subjected first to a hydroponics condition for 24 hours in order to acclimation. After this period, these plants were submitted to oxidative stresse using 0 mM, 10 mM, 20 mM and 30 mM hydrogen peroxide during 8 hours. The histomorphometric analysis allowed us to verify that both root and leaf tissues had a structural changes as it was observed by the increased in cell volume, lignin accumulation in cell walls. Besides, this observation suggested that there was a change in redox balance. Also, it was analyzed the activity of the SOD, CAT and APX enzymes. It was observed an increase in the SOD activity in roots and it was also observed a lipid peroxidation in leaves and roots. Then, in order to identify proteins that were differently expressed in this conditions it was used the proteomic tool either by bidimensional gel or by direct sequencing using the Q-TOF EZI. The results obtained with this approach identified more than 3.000 proteins with the score ranging from 100-5000 ions. Some of the proteins identified were: light Harvesting; oxygenevolving; Thioredoxin; Ftsh-like protein Pftf precusor; Luminal-binding protein; 2 cys peroxiredoxin e Lipoxygenase. All these proteins are involved in oxidative stress response, photsynthetic pathways, and some were classified hypothetical proteins and/or unknown (30% of total). Thus, our data allows us to propose that this treatment induced an oxidative stress and the plant in response changed its physiological process, it made changes in tissue, changed the redox response in order to survival to this new condition

The dyslipidemia and excess weight in adolescents, when combined, suggest a progression of risk factors for cardiovascular disease (CVD). Besides these, the dietary habits and lifestyle have also been considered unsuitable impacting the development of chronic diseases. The study objectives were: (1) estimate the prevalence of lipid profile and correlate with body mass index (BMI), waist circumference (WC) and waist / height ratio (WHR) in adolescents, considering the maturation sexual, (2) know the sources of variance in the diet and the number of days needed to estimate the usual diet of adolescents and (3) describe the dietary patterns and lifestyle of adolescents, family history of CVD and age correlates them with the patterns of risk for CVD, adjusted for sexual maturation. A cross-sectional study was performed with 432 adolescents, aged 10-19 years from public schools of the Natal city, Brazil. The dyslipidemias were evaluated considering the lipid profile, the index of I Castelli (TC / HDL) and II (LDL / HDL) and non-HDL cholesterol. Anthropometric indicators were BMI, WC and WHR. The intake of energy, nutrients including fiber, fatty acids and cholesterol was estimated from two 24-hour recalls (24HR). The variables of lipid profile, anthropometric and clinical data were used in the models of Pearson correlation and linear regression, considering the sexual maturation. The variance ratio of the diet was calculated from the component-person variance, determined by analysis of variance (ANOVA). The definition of the number of days to estimate the usual intake of each nutrient was obtained by taking the hypothetical correlation (r) ≥ 0.9, between nutrient intake and the true observed. We used the principal component analysis as a method of extracting factors that 129 accounted for the dependent variables and known cardiovascular risk obtained from the lipid profile, the index for Castelli I and II, non-HDL cholesterol, BMI, and WC the WHR. Dietary patterns and lifestyle were obtained from the independent variables, based on nutrients consumed and physical activity weekly. In the study of principal component analysis (PCA) was investigated associations between the patterns of cardiovascular risk factors in dietary patterns and lifestyle, age and positive family history of CVD, through bivariate and multiple logistic regression adjusted for sexual maturation. The low HDL-C dyslipidemia was most prevalent (50.5%) for adolescents. Significant correlations were observed between hypercholesterolemia and positive family history of CVD (r = 0.19, p <0.01) and hypertriglyceridemia with BMI (r = 0.30, p <0.01), with the CC (r = 0.32, p <0.01) and WHR (r = 0.33, p <0.01). The linear model constructed with sexual maturation, age and BMI explained about 1 to 10.4% of the variation in the lipid profile. The sources of variance between individuals were greater for all nutrients in both sexes. The reasons for variances were  1 for all nutrients were higher in females. The results suggest that to assess the diet of adolescents with greater precision, 2 days would be enough to R24h consumption of energy, carbohydrates, fiber, saturated and monounsaturated fatty acids. In contrast, 3 days would be recommended for protein, lipid, polyunsaturated fatty acids and cholesterol. Two cardiovascular risk factors as have been extracted in the ACP, referring to the dependent variables: the standard lipid profile (HDL-C and non-HDL cholesterol) and "standard anthropometric index (BMI, WC, WHR) with a power explaining 75% of the variance of the original data. The factors are representative of two independent variables led to dietary patterns, "pattern 130 western diet" and "pattern protein diet", and one on the lifestyle, "pattern energy balance". Together, these patterns provide an explanation power of 67%. Made adjustment for sexual maturation in males remained significant variables: the associations between puberty and be pattern anthropometric indicator (OR = 3.32, CI 1.34 to 8.17%), and between family history of CVD and the pattern lipid profile (OR = 2.62, CI 1.20 to 5.72%). In females adolescents, associations were identified between age after the first stage of puberty with anthropometric pattern (OR = 3.59, CI 1.58 to 8.17%) and lipid profile (OR = 0.33, CI 0.15 to 0.75%). Conclusions: The low HDL-C was the most prevalent dyslipidemia independent of sex and nutritional status of adolescents. Hypercholesterolemia was influenced by family history of CVD and sexual maturation, in turn, hypertriglyceridemia was closely associated with anthropometric indicators. The variance between the diets was greater for all nutrients. This fact reflected in a variance ratio less than 1 and consequently in a lower number of days requerid to estimate the usual diet of adolescents considering gender. The two dietary patterns were extracted and the pattern considered unhealthy lifestyle as healthy. The associations were found between the patterns of CVD risk with age and family history of CVD in the studied adolescents

Este trabalho teve como objetivo avaliar o efeito da levedura desidratada de álcool (Saccharomyces cerevisae) como substituto do suplemento vitamínico, em dietas de tilápia-do-nilo (Oreochromis niloticus). Trezentos alevinos machos, sexualmente revertidos, com peso médio de 1,5 g, foram distribuídos igualmente em 20 aquários de fibra de vidro, equipados com filtro biológico. Cinco dietas isoprotéicas com 32% de proteína bruta na primeira fase (40 dias) e com 28% de proteína bruta na segunda fase (74 dias) foram fornecidas ad libitum, duas vezes ao dia. A dieta-controle não conti-nha levedura, e as demais apresentavam 10% de levedura. O delineamento foi inteiramente casualizado, com cinco tratamentos e quatro repetições. Não houve influência significativa dos tratamentos sobre o ganho de peso e taxa de crescimento específico. Foi observada, nos peixes alimentados com dietas sem suplemento vitamínico, alta mortalidade. Os peixes que receberam levedura apresentaram menor conteúdo corporal de proteína, e maior, de lipídeos. A levedura desidratada de álcool pode ser usada como fonte de vitaminas hidrossolúveis em dietas para alevinos de tilápia-do-nilo.
